There are multiple stressors derived from urbanizations that result in frequent disturbances on streams and rivers reducing water quality and threatens aquatic biota. P-glycoprotein (P-gp)-mediated multixenobiotic resistance (MXR) is a defence mechanism analogous to multidrug resistance (MDR), which has been demonstrated in several aquatic organisms. This system protects cells against the entry and the accumulation of xenobiotics and has been proposed as a biomarker for pollution assessment. We conducted a study in a post-urban reach of Esquel stream (Chubut Province) downstream a wastewater treatment plant, in order to assess the presence and activity of MXR in five freshwater macroinvertebrate species (Helobdella michaelseni, Helobdella simplex, Patagoniobdella variabilis, Hyalella curvispina and Chironomus riparius). We measured the accumulation of the model P-gp substrate rhodamine B (RB) in organisms previously exposed to pollution. Our results described the activity of the MXR system in the three species of leeches suggesting their suitability as the in vivo biomonitoring. We also identified a dependence of the transporter activity with the development stage in H. simplex, highlighting the importance of using organisms of similar size classes since it may affect observed results. Finally, we concluded that benthic freshwater macroinvertebrates possess different species-specific levels of MXR activity possibly influencing their natural distribution as well as their survival. 相似文献

Purpose
The purpose of this study was to investigate relationships between chemical and thermal stabilities of Cu–humic complexes. The study of the chemical stability was based on pedological methods used for the determination of the bond strength of metal ions in soils by chemical leaching agents. The samples with various contents of the Cu(II) ions and their bond strength were put to the thermal analysis in order to correlate their thermo-oxidative behavior with their stability determined by leaching.Materials and methods
The humic acid was extracted from the South-Moravian lignite by standard alkaline extraction. The humic sample was used in two different forms: as the solid powder and as the hydrogel prepared by the acidic precipitation of humate. Six various concentrations of copper(II) solutions were used for the complexation of the humic powder and the hydrogel, in order to study the influence of their initial concentration on both the determined stabilities of the prepared complexes. Their chemical stability was assessed in terms of the Cu(II) ions release from the humic acid structure into two different extraction agents (MgCl2 and HCl solutions). Their thermo-oxidative behavior was investigated employing the thermogravimetry.Results and discussion
The complexation capacity of the humic hydrogel was higher in comparison with the humic powder. The amounts extractable from the Cu–humic complexes by the used leaching agents are higher for the humic powder, which shows on the lower chemical stability. The thermal degradation of the prepared complexes proceeds in several steps and this character remains also after the removal of the mobile and the ion-exchangeable fractions by the MgCl2. The elimination of these fractions as well as the extraction of the strongly bound Cu(II) ions shift the thermal degradation to higher temperatures. The incombustible residue increases with the Cu(II) content in the complexes except for the samples extracted by the HCl.Conclusions
The form of humic sample used for the preparation of the Cu–humic complexes influences both the chemical stability and the thermal one. The main reason is probably a better accessibility of the functional groups in the humic gel, which enables forming stronger binding copper(II) ions. The results showed that the thermal and chemical stabilities are closely related, which corresponds with the shift of the thermal degradation to higher temperatures after removing the less stable fractions from the humic complexes. 相似文献613.

Differences achieved in 18 selected morphological, phenological and grain parameters attributable to the wheat breeding process were evaluated on the basis of 262 original Czech and Slovak winter bread wheat cultivars (Triticum aestivum L.) developed during 1911–2008 and cultivated in the field and climatic conditions of Crop Research Institute Prague-Ruzyně for 3 years (2010–2012). The wheat breeding process unambiguously has improved grain production parameters, lodging and mildew resistance compared to wheat landraces and obsolete cultivars. Only wheat landrace ‘Visnovska Hustoklasa’ showed a higher resistance to lodging (8 points), no old cultivars registered till 1930 were resistant to mildew. Nevertheless, these economically positive properties of modern wheat cultivars are negatively reflected in lower crude protein content, protein content’s lower stability year to year, and decreased stability of Zeleny sedimentation. The breeding of wheat cultivars during 1961–1990 points to a key period responsible for these parameter changes. At the same time, within this somewhat older wheat group (from 1961–1990), several perspective wheat cultivars with acceptable spike productivity (e.g., ‘Regina’) and high crude protein stability (e.g., ‘Diana II’) were also identified. 相似文献

Results are reported of the influence of soilless culture upon the biological value of vegetables grown on gravel in glasshouses: 1. Genetic differences of cultivars used in the experiments will also be obvious by means of contents of nutrients by this method of cultivation. 2. Comparing greenhouse crops in soil as usual with those of soilless culture differences as regard the biological value are found sometimes, as follows:
- In cucumbers and radishes a slight decrease of dry matter and sugar contents, but no differences in ascorbic acid.
- In kohlrabi a slight decrease of all nutrients investigated.
- In cauliflower — no differences at all.
- In capsicum fruits — higher contents in soiless grown culture, especially of ascorbic acid.
- In tomatoes have been detected some features, which are obviously due to cultivation in greenhouses: A decrease of ascorbic acid contents when harvested in summertime and still more distinct when harvested in autumn. Furthermore a decrease of ‘relative’ sugar contents in dry matter in fruits of yield in autumn compared with that of summer yield.

Clothe drape depends, on the one hand, on the mechanical and constructional properties of the textile materials and, on the
other hand, on the form — construction of the cloth, and their connected positions at the seams. This article analyses the
influence of seam types and directions on fabric drape. The drape coefficient together with the number and distribution of
folds were determined using a Cusick Drape meter with a video camera and Drape Analyser. The results of our investigation
showed that the number of folds on the samples with seams was greater, or in some cases, equal to samples without seams. The
distribution and form of the folds also changed. Drape coefficient was, in most cases, greater on samples with seams than
samples without seams. Furthermore it was discovered that by measurement repetition the angle values are similar even when

Microbial communities were studied in redoximorphic microsites of highly heterogeneous Gleysol at a mm scale using 16S and 18S amplicon sequencing to demonstrate if the composition of soil microbes reflects the differences in ferric and ferrous micro-sites. In both explored gley horizons with redoximorphic features (Bg2 and Cg), ferric mottles were significantly enriched with total P and Fe and depleted of O, Si, Al, K and Ca compared with the adjacent ferrous groundmass (SEM–EDS). Ferric mottles were determined as Fe oxide coatings and hypocoatings. In Bg2, both prokaryotic and micro-eukaryotic communities differed significantly between mottles and groundmass in composition of operational taxonomic units (OTUs) and in proportions of phyla, reflecting heterogeneities in the soil properties there. Mottles in Bg2 were characterized by increased proportion of Proteobacteria, decreased proportion of Acidobacteriota among prokaryotes and by dominance of a single proteobacterial OTU from Anaplasmataceae compared to all other samples. The composition of micro-eukaryotes showed an opposite trend, as micro-eukaryotes of Bg2 groundmass were unique among the other horizons, while micro-eukaryotes of Bg2 mottles had similar composition to neighbouring horizons. Microbial communities of adjacent samples were not more similar to each other than communities of randomly selected ones in Bg2 horizon. That suggests that at mm scale, the sample distance does not represent the driving factor of microbial community composition and that the adjacent samples differ rather due to physicochemical factors. The spatial organization of microbial communities revealed in Bg2 has not reappeared in similarly organized Cg horizon, probably due to other overriding factors. The differences revealed between Bg2 and Cg horizons, including granulometric composition, content of crystalline Fe, exchangeable Al, and organic carbon, as well as exposition to groundwater, were discussed as possible reasons of the distinct organization in Cg. The similarity of pro−/eukaryotic communities of adjacent and non-adjacent couples suggests no distance decay pattern at a mm scale. The agreement between patchiness in soil properties and microbial communities was revealed for the first time and confirms the importance of microscale patterns in soil. 相似文献

Trichinella spiralis and Trichinella britovi are species that are frequently found in domestic pigs and various sylvatic animals in Croatia. During routine trichinoscopy, non-encapsulated larvae were detected in the muscle tissue of a domestic pig. Artificial digestion revealed a larvae burden of 602 muscle larvae per gram of tissue. Tissue section analysis confirmed the presence of non-encapsulated larvae. Multiplex PCR identified the larvae as T. pseudospiralis. This observation is consistent with the reports of a local veterinary inspector who described the presence of non-encapsulated Trichinella in four individual cases over the last 2 years. This is the first report of T. pseudospiralis in Croatia and one of very few cases of T. pseudospiralis infection described in domestic pigs. The detection of non-encapsulated larvae stresses the need for implementation of artificial digestion instead of trichinoscopy for the detection and identification of Trichinella infections. 相似文献

In mammals, proteases are present in sperm acrosome and play key role in fertilization. Sturgeon sperm has an acrosome, but its physiology, biochemistry, and potential role in fertilization are unknown. In the present study, we have observed high protease activity in acidic extract of intact sperm compared to that of seminal plasma in sterlet (Acipenser ruthenus). The protease activity was decreased and increased in acidic extract of motility-activated sperm and in the activation medium, respectively. Molecular analysis revealed total protease and serine (acrosin) protease activities in sperm acidic extract which was accumulated in a protein band with relative molecular mass of 35 kDa. Immunoelectron microscopy using an affinity-purified polyclonal antibody for boar acrosin localized the protease at the acrosome region. Moreover, initiation of sperm motility was inhibited after activation in the presence of inhibitors for both trypsin-like and chymotrypsin-like proteases, while the effects of protease inhibitors on sperm velocity were uncertain. Our results indicate similarities in physiology and biochemistry of acrosome between sturgeon and mammals and suggest potential role of protease in the initiation of sperm motility in sturgeon. 相似文献

The effects of farming on planktonic microbial communities were investigated at the coastal sea bass/sea bream farm in the oligotrophic middle Adriatic Sea. Analyses of nutrients, chlorophyll a, cyanobacteria (Prochlorococcus), heterotrophic bacteria and nanoflagellates (both pigmented and heterotrophic) were carried out on samples taken during six seasonal cruises at different water depths. Farming activity increased the natural concentrations of some nutrients (ammonium, nitrite, phosphate, dissolved inorganic nitrogen) and changed their seasonal pattern. The values still remained typical for oligotrophic environ‐ments demonstrating no risk for eutrophication. Enhanced nutrient supply provoked an immediate increase in abundances of both autotrophic and heterotrophic microbial groups (except pigmented nanoflagellates) and chlorophyll a. The effect of farming was more visible for the heterotrophic microbial component. Results from this study reveal a potential common pattern of microbial response to farming in the oligotrophic environments. It also suggests the importance of heterotrophic microbial web in transferring the matter and energy released from the fish farms in nutrient poor environments. 相似文献
