Electron tomography of vitrified cells is a noninvasive three-dimensional imaging technique that opens up new vistas for exploring the supramolecular organization of the cytoplasm. We applied this technique to Dictyostelium cells, focusing on the actin cytoskeleton. In actin networks reconstructed without prior removal of membranes or extraction of soluble proteins, the cross-linking of individual microfilaments, their branching angles, and membrane attachment sites can be analyzed. At a resolution of 5 to 6 nanometers, single macromolecules with distinct shapes, such as the 26S proteasome, can be identified in an unperturbed cellular environment. 相似文献

The early-1990s’ war in former Yugoslavia led to severe damage to the urban and peri-urban forests of Sarajevo, capital of Bosnia and Herzegovina. During the Siege of Sarajevo (April 1992–March 1996), after the energy supplies to the city had been cut off, over three-quarters of all urban trees and nearly all peri-urban trees within the siege line were cut down for firewood by the desperate residents. After the war, the city trees were quickly and effectively replanted, and Sarajevo provides a useful case study of urban forest recovery. This study presents the observations and measurements, of trees that survived the war and the trees planted after the war, made in Sarajevo in May 2008. We summarize the lessons learned from the Sarajevo experience, regarding both damage and replanting, which include the close relationship between the urban tree damage and the specifics of military operations, the importance of the initial planting stock, and the advantages of collaboration between academic researchers and urban forest managers. 相似文献

Genetic diversity within and among 12 populations of the dioecious tropical tree species Hagenia abyssinica (Bruce) J.F. Gmel. in Ethiopia was examined with eight inter simple sequence repeat (ISSR) primers. A total of 104 clearly
scorable bands were generated, among which 84 (81%) were polymorphic. Jaccard similarity coefficient was calculated for pairwise
comparisons among all 120 individuals and ranged from 0.30 to 0.88 while average within-population similarity ranged from
0.53 to 0.66. Within-population variability was estimated as percentage polymorphic loci (ranging from 52% to 87%), Shannon’s
information index (0.30–0.50) and Nei’s genetic diversity (0.21–0.35). The highest variability values were obtained for one
recently planted population and for one wild population growing in an undisturbed primary forest area. Significant overall
differentiation among populations was detected by both Shannon’s information index (0.26) and G
ST (0.25). Relatedness among samples was estimated with a principal coordinate analysis, and relatedness among populations was
estimated with a cluster analysis (UPGMA). A Mantel test indicated a significant association between genetic and geographic
distances, and an autocorrelation analysis showed significant evidence of gene flow over distances up to 30 km. This study
is the first of its kind for H. abyssinica, which has decreased recently in Ethiopia and now must be regarded as an endangered species. Both within-population and between-population
diversity estimates are typical of outcrossing, longlived and late successional species, suggesting that recent anthropogenic
disturbances have not yet had much impact on population genetic parameters. DNA marker data can, however, be used to identify

The expansion of spruce-dominated forestry in Southern Sweden during the twentieth century has led to a considerable amount of oak (Quercus robur L.) woodlands being converted into stands dominated by planted spruce. The thinning of spruces around oak trees is currently done in Sweden to improve local diversity of insects, oak growing conditions and eventually decrease their mortality. To evaluate the effect of these treatments, we dendrochronologically studied growth of old (100–200 years old) oaks subjected to thinning of different intensity at nine locations in southern Sweden, and compared them to oaks located in nearby pastures. The overall pattern suggests that commonly adopted thinning intensities do not significantly affect oak growth. Oak growth was positively related to oak age and negatively to the amount of dead oak crown. Analyses of correlations between oak growth and summer drought conditions, as reflected by location-specific chronologies of the Monthly Drought Code (MDC), indicated that older trees exhibited generally negative correlations, whereas the correlation remained generally positive for the younger trees, both inside and outside forest stands. Ninety‐seven specimens of spotfin hatchetfish, Thoracocharax stellatus, an ornamental freshwater species from the Amazon basin, were captured in the basin of the Guamá River in the municipality of Belém, in northern Brazil, and analysed for coccidiosis infection. Overall, 26 of the specimens were infected by apicomplexan parasites of the genus Goussia, with unsporulated forms being found in the gastric epithelium and sporulated forms in the intestinal lumen. The spheroid oocysts (mean diameter: 13.2 ± 1.7 μm) have four elliptical sporocysts. A partial sequence of the SSU rDNA of the new species was obtained, which contained 1,121 base pairs, with 43.8% guanine + cytosine (G + C), and the bases distributed as follows: A = 28.1%, C = 18.3%, G = 25.5% and T = 28.1%. The combined analysis of the morphometric and phylogenetic evidence confirmed that the specimens represented the genus Goussia and were allocated to a new species, Goussia guamaensis n. sp., which is described here. 相似文献

- Bivalves are important components of freshwater ecosystems; however, they are also one of the most threatened animal groups, especially members of the order Unionida. The main threats to freshwater bivalves are habitat modification and invasive species. Protected areas are a common way to minimize impacts and preserve native species, but they are rarely designated with a focus on freshwater invertebrates.
- The main goal of this article was to describe the distribution of freshwater bivalves in the state of Rio de Janeiro, and the relationship between these species and bioclimatic areas, land use and protected areas.
- Five native and two invasive bivalve species have been reported in Rio de Janeiro. They occur mainly in bioclimatic areas related to the Paraíba do Sul River and Campista Lowlands. The few records of bivalves inside protected areas are in areas of sustainable use that offer a lower level of protection, and frequently in sympatry with invasive bivalves.
- Thus, the established protected areas in Rio de Janeiro are not adequately effective for freshwater bivalve conservation, and some species remain under threat even within them. Freshwater bivalves thus remain largely unprotected in the state.
- The development of new protected areas and management plans should consider other faunal groups that are usually ignored, such as freshwater bivalves, to achieve more inclusive and effective protection.
