Myrothecium verrucaria and Fusarium compactum were isolated from diseased Orobanche ramosa plants collected in southern Italy to find potential biocontrol agents of this parasitic weed. Both fungi grown in liquid culture produced metabolites that inhibited the germination of O. ramosa seeds at 1-10 muM. Eight metabolites were isolated from M. verrucaria culture extracts. The main metabolite was identified as verrucarin E, a disubstituted pyrrole not belonging to the trichothecene group. Seven compounds were identified by spectroscopic methods as macrocyclic trichothecenes, namely, verrucarins A, B, M, and L acetate, roridin A, isotrichoverrin B, and trichoverrol B. The main metabolite produced by F. compactum was neosoloaniol monoacetate, a trichothecene. All the trichothecenes proved to be potent inhibitors of O. ramosa seed germination and possess strong zootoxic activity when assayed on Artemia salina brine shrimps. The atmospheric concentrations of greenhouse gases have increased to unprecedented levels during last decades, raising concerns about their effect on climate change. Agriculture and land use change play an important role in atmospheric CO2 emission and fixation, especially by affecting the soil carbon (C) storage. In this context, agroforestry systems (AFSs) could play an important role contributing to climate change mitigation. Given the importance of olive cultivation in the Mediterranean region, it is important to investigate soil C stock in olive groves, and to assess which agronomic practices could improve the soil C stock in these systems. For this reason, a study was conducted in different olive groves, including conventional and organic management, and a typical silvopastoral AFS. Furthermore, an abandoned olive grove and nearby forest were examined as a comparison. Soil samples were collected in each farm and analysed for C content and physico-chemical characteristics. This study indicates that, irrespective of the management, olive groves in the Umbria region of Italy are characterised by a high level of soil C stock if compared to those growing in other areas and to forest ecosystems, indicating that the practices adopted in the area are not negatively affecting soil C storage. A slightly lower soil C stock was measured in the silvopastoral AFS in comparison to the other farms, while high soil C stock was associated with the use of pomace, suggesting that this practice can be used to further increase soil C stock in olive orchards.
Seedlings of Phillyrea latifolia L., a Mediterranean shrub, were exposed for 90 days to 110 nl l(-1) ozone (O(3)). Comparison of the cDNA-amplified fragment length polymorphism (cDNA-AFLP) patterns for treated and control plants allowed the identification and cloning of 88 differential sequences induced by O(3). The differential expression of 67 cloned sequences was further confirmed by RT-PCR. The functions of 36 cloned sequences, corresponding to seven of the twelve gene functional classes of Arabidopsis, were presumed on the basis of their homology with characterized gene sequences. Ozone induction of genes homologous to 24 of the clones has been reported in other plant species, whereas the induction of the 12 remaining sequences has not been observed before. Ozone activation of these newly identified genes could be a result of the chronic exposure to low O(3) concentration, because in most previous studies, acute treatments, involving high O(3) dosages, were applied. Possible roles of the cloned sequences in the response of P. latifolia to O(3) and other causes of oxidative stress are discussed. 相似文献
Xylem vulnerability to cavitation is a promising criterion for identifying trees with high drought tolerance, but traditional techniques for measuring cavitation resistance are unsuitable for screening large numbers of genotypes. We tested the potential of the new Cavitron technique for high throughput screening of cavitation resistance in five poplar (Populus spp.) and four willow (Salix spp.) clones. The Cavitron technique enabled the screening of three to four clones per day with sufficient accuracy to reveal significant differences between clones. Because intraspecific screening may be better carried out through the identification of correlated and more easily measured traits, we attempted to identify accessible parameters that correlate to cavitation resistance. Variability in vulnerability to cavitation across clones was poorly correlated with anatomical traits such as vessel diameter, vessel wall strength, wood density and fiber wall thickness; however, a striking correlation was established between cavitation resistance and aboveground biomass production, indicating a possible trade-off between xylem safety and growth potential. 相似文献
Scots pine is a highly diverse species, extended across Europe from Scandinavia to Spain, Italy, Greece and Turkey. It is
also a valuable species, used in many commercial monoculture plantations in Great Britain and particularly in Scotland. Because
of the diversity of growing environments and its commercial importance, it is necessary to identify the combination of significant
factors affecting the observed variability of growth. Temperature, mainly during the growing season, is quite commonly considered
as the most important factor in knowledge-based or empirical models. However, in highly oceanic climates like that of Scotland,
the impact of temperature may have a less significant impact on growth. Here we argue that other factors, such as incoming
winter solar radiation, frost, drought and management also have a significant effect on the growth of Scots pine. In addition,
we argue that the already developed Ecological Site Classification knowledge-based model, used as a forest management tool
in Great Britain, should be updated to incorporate our findings. Furthermore, we discuss the need to include management impact
and possibly more physiological based components in its growth modelling routines, as these would allow the introduction of

The stand density index,one of the most important metrics for managing site occupancy,is generally calculated from empirical data by means of a coefficient derived from the“self-thinning rule”or stand density model.I undertook an exploratory analysis of model fitting based on simulated data.I discuss the use of the logarithmic transformation(i.e.,linearisation)of the relationship between the total number of trees per hectare(N)and the quadratic mean diameter of the stand(QMD).I compare the classic method used by Reineke(J Agric Res 46:627–638,1933),i.e.,linear OLS model fitting after logarithmic transformation of data,with the“pure”powerlaw model,which represents the native mathematical structure of this relationship.I evaluated the results according to the correlation between N and QMD.Linear OLS and nonlinear fitting agreed in the estimation of coefficients only for highly correlated(between-1 and-0.85)or poorly correlated([-0.39)datasets.At average correlation values(i.e.,between-0.75 and-0.4),it is probable that for practical applications,the differences were relevant,especially concerning the key coefficient for Reineke’s stand density index calculation.This introduced a non-negligible bias in SDI calculation.The linearised log–log model always estimated a lower slope term than did the exponent of the nonlinear function except at the extremes of the correlation range.While the logarithmic transformation is mathematically correct and always equivalent to a nonlinear model in case of prediction of the dependent variable,the difference detected in my studies between the two methods(i.e.,coefficient estimation)was directly related to the correlation between N and QMD in each simulated/disturbed dataset.In general,given the power law as the“natural”structure of the N versus QMD relationship,the nonlinear model is preferred,with a logarithmic transformation used only in the case of violation of parametric assumptions(e.g.data distributed non-normally). 相似文献
A study was carried out to compare the efficacy of two injectable formulations of ivermectin, Ivomec,(1) Merial (IVM reference) and Ivogell,(2) Intervet (IVM generic) in the treatment of psoroptic mange (Psoroptes ovis) in Charollais feedlot cattle. A total of 22 animals were ranked in order of the severity of mange and allocated to 11 replicates of 2 animals each. Within each replicate, one animal was randomly allocated to IVM reference product treatment (Group 1) and one to IVM generic (Group 2). Animals were treated on Day 0 and on Day 8 at the recommended dosage of 200 microg ivermectin/kg bodyweight. The pharmacokinetics profiles (pK) of both IVM formulations were evaluated in plasma samples taken from 6 cattle randomly chosen per group on Day 0, before treatment, and then at 6, 12, 24 hours and daily from Day 2 to Day 7 after the treatment on Day 0. Additionally, the severity of mange lesions was assessed and mites were counted in skin scrapings on Days 0, 8, 15 and 25. Animals were weighed on Day 0 and 25 and body weight and average daily gains (ADG) were evaluated. No statistical differences were found between the cattle of the two groups in any pK parameters, although the mean IVM plasma concentrations in cattle treated with the IVM reference product were consistently higher than those found in cattle treated with the generic compound. By Day 25, all animals in Group 1 had recovered clinically and parasitologically from psoroptic mange while cattle from Group 2 still had mange lesions and, in two animals, living mites were found in the skin scrapings; these differences were significant (P<0.001). The mean body weight of the two groups was significantly different on Day 25 (P<0.01) when animals in Group 1 weighed 20 kg more than those in Group 2. In conclusion, despite similarities in their pharmacokinetic profiles and formulations, the clinical efficacy of the two injectable formulations of IVM differed significantly in their therapeutic efficacy against psoroptic mange in feedlot cattle up to 25 days after treatment: this difference in response was reflected in an incomplete clinical and parasitological response in Group 2 and a slower growth rate. 相似文献
This paper reports a very simple and fast method to collect eluates with high amounts of hydroxytyrosol, biotransforming Olea europaea L. leaf extract by a thermophilic beta-glycosidase immobilized on chitosan. Some phenolic compounds in the leaf tissue and in the eluates obtained by biotransformation are identified. To propose the eluates as natural substances from a vegetal source, their antioxidant properties have been compared with those of the leaf extract from which they are originated. The eluates possess a higher concentration of simple phenols, characterized by a stronger antioxidant capacity, than those available in extra virgin olive oils and in many tablets of olive leaf extracts, commercially found as dietetic products and food integrators. 相似文献
Over the past 70 years, heat has been widely used as a tracer for estimating the flow of water in woody and herbaceous plants. However, most commercially available techniques for monitoring whole plant water use are invasive and the measurements are potentially flawed because of wounding of the xylem tissue. The study of photosynthate transport in the phloem remains in its infancy, and little information about phloem transport rates is available owing to the fragility of the vascular tissue. The aim of our study was to develop a compact, stand-alone non-invasive system allowing for direct detection of phloem and xylem sap movement. The proposed method uses a heat pulse as a tracer for sap flow. Heat is applied to the surface of the stem with a near-infrared laser source, and heat propagation is monitored externally by means of an infrared camera. Heat pulse velocities are determined from the thermometric data and related to the more useful quantity, mass flow rate. Simulation experiments on the xylem tissue of severed silver birch (Betula pendula Roth.) branch segments were performed to assess the feasibility of the proposed approach, highlight the characteristics of the technique and outline calibration strategies. Good agreement between imposed and measured flow rates was achieved leading to experimentation with live silver birch and oak (Quercus robur L.) saplings. It was demonstrated that water flow through xylem vessels can be monitored non-invasively on an intact stem with satisfactory accuracy despite simultaneous sugar transport in the phloem. Two 5-month-old golden retriever dogs were referred for the evaluation of a mass arising from the left ventrolateral conjunctiva of the lower eyelid. The masses were externally located, deemed to be cystic in nature, and were surgically removed, and in one case, 0.15 mL of clear fluid was collected. Cytological examination of the aspirated fluid revealed proteinaceous, basophilic amorphous material, and few epithelial cells with foamy cytoplasm and small nuclei. No micro-organisms were detected. Histological examination of the cysts showed a wall composed of single to double cell layers. The cells were cuboidal to flattened, nonciliated, lined with mild submucosal connective tissues and slight inflammation composed of lymphocytes, plasma cells, and rare macrophages. In some areas, lobules of acinar glandular tissue were observed. To the authors' knowledge, this is the first case report of conjunctival dacryops in dogs. The complete surgical removal of the cysts was curative in both cases. 相似文献
