In tropical soils, the high turnover rate and mineralization of organic matter (OM) associated with intensive agricultural use, generally leads to faster soil degradation than that observed in temperate climatic zones. The application of sewage sludge to the soils is one proposed method of maintaining soil organic matter, and is also an alternative method of disposing of this waste product. As well as containing large quantities of OM, sludge is also a significant source of supplementary nitrogen, phosphorus and other essential nutrients for plant growth. However, it is necessary to understand the qualitative and quantitative changes that take place in the OM in soil treated with sewage sludge. The approach of the present study was intended to identify possible structural changes caused by sewage sludge applications on soil humic acids (HAs). The HAs extracted from a Typic Achrortox under sewage sludge applications were characterized by electron paramagnetic resonance (EPR) spectroscopy. The soil samples were collected from a field experiment designed to evaluate the effects of different doses of sewage sludge on corn growth and development in Brazil. The sewage sludge originated from urban waste treated at the sewage sludge treatment station in the city of Franca, state of Sao Paulo, Brazil. The following soil treatments were studied: control (non-cultivated soil under natural vegetation (NC)), control soil amended with NPK (conventional corn fertilization) and four treatments N1, N2, N4 and N8 with applications of 3.5, 7, 14 and 28 Mg ha−1 of sewage sludge (dry matter), respectively. HAs were extracted from the surface layer using the methodology of the International Humic Substance Society (IHSS). Fe3+ and VO2+ ions complexed with HAs, and also semiquinone-type free radical (SFR) at concentrations of approximately 2.0 × 1018 spins g−1 HA were identified in EPR spectra. The levels of SFR were lower for treatments where the applied sewage sludge doses were equivalent to four and eight times the normal doses of N mineral fertilization, reaching values of 1.7 × 1018 and 1.24 × 1018 spins g−1 HA, respectively. The observed decrease in SFR content as sewage sludge dose was increased, was probably associated with the incorporation of less aromatic components into HAs originating from the sewage sludge. 相似文献

A variety of popular instant, ground, and brewed coffees were analyzed using a modified liquid chromatography-tandem mass spectrometry (LC-MS/MS) method specifically developed for the determination of acrylamide in foods. Coffee test portions were spiked with 13C3-labeled acrylamide as an internal standard prior to their extraction and cleanup. Ground coffees (1 g) and instant coffees (0.5 g) were extracted by shaking with 9 mL of water for 20 min. Brewed coffee test portions (9 mL) were taken through the cleanup procedure without further dilution with extraction solvent. Coffee test portions were cleaned up by passing 1.5 mL first through an Oasis HLB (hydrophilic/lipophilic copolymer sorbent) solid phase extraction (SPE) cartridge and then a Bond Elut-Accucat (cation and anion exchange sorbent) SPE cartridge. The cleaned up extracts were analyzed by positive ion electrospray LC-MS/MS. The MS/MS data was used to detect, confirm, and quantitate acrylamide. The limit of quantitation of the method was 10 ng/g for ground and instant coffees and 1.0 ng/mL for brewed coffee. The levels of acrylamide ranged from 45 to 374 ng/g in unbrewed coffee grounds, from 172 to 539 ng/g in instant coffee crystals, and from 6 to 16 ng/mL in brewed coffee. 相似文献

Composting of pruning waste, leaves and grass clippings was monitored by different parameters. A windrow composting pile, having the dimensions 2.5 m (height) x 30 m (length) was establish. The maturation of pruning waste compost was accompanied by a decline in NH4 +-N concentration, water soluble C (WSC) and an increase in NO3 –-N content. Both organic matter (OM) content and total N (TN) losses during composting followed a first-order kinetic equation. These results were in agreement with the microbiological activity measured either by the CO2 respiration or dehydrogenase (DH-ase) activity during the process. Statistically significant correlations were found between DH-ase activity, easily biodegradable organic C forms, NH4 +-N and NO3 –-N concentrations and organic matter content and N losses. For this reason, DH-ase activity and the CO2 evolution could be used as good indicators of pruning waste compost maturity. In contrast, humification parameters data from the organic matter fractionation did not agree with the initially expected values and did not contribute to the assessment of compost maturity. Neither the cation exchange capacity nor the germination index showed a clear tendency during the composting time, suggesting that these parameters are not suitable for evaluating the dynamics of the process. 相似文献

The study was conducted in three regional states of Ethiopia: Amhara, Oromia, and Addis Ababa from August 2008 to April 2009
with the objectives of identifying the genetic diversity of serotypes and topotypes in Ethiopia, and determining the attack
rate and associations of potential risk factors with foot-and-mouth disease (FMD) seropositivity. A total of 496 cattle were
clinically and serologically examined for presence of specific lesions and nonstructural protein for FMD, respectively. Of
which, 140 (28.2%) manifested clinical signs and lesions suggestive of FMD, and 219 (44.2%) were seropositive. From a total
of 7,781 animals observed and recorded on a designed format in six districts, 1,409 (19.6%) were infected, and 15 (0.12%)
died during outbreaks of FMD. Epidemiological investigations revealed that the morbidity rate of the disease was 21.1% in
Akaki-kality sub-city, but the mortality rate was <2% in all districts. Furthermore, the mortality and case fatality rates
were relatively higher, 1.6% and 8.9% in calves than the other age groups, respectively. From a total of 33 bovine epithelial
tissue-cultured samples, 19 (57.6%) showed CPE for FMD virus, in which 16 samples had serotype O and EA-3 topotype, while
three samples had found serotype A, Africa topotype, and G-VII strain. Various strains of FMD viruses were isolated in Ethiopia
in this study, and therefore, further detailed studies on the evaluation of available vaccines and the development of a vaccine

Congenital hyperplastic goiter and cretinism were documented in a 16 month-old male North American black bear (Ursus americanus). The cub was captured at approximately 8 months of age and maintained for an additional 8 months in captivity. Clinical signs included growth retardation, clumsiness, and facial dysmorphism. Hypothyroidism was documented by determining serum triiodothyronine (T3) and thyroxine (T4) levels. Lysosomal storage disease was ruled out by measuring various lysosomal enzyme activities. Instrumented shoes were used to measure the vertical forces exerted by horses moving at a variety of gaits. Two types of shoes were used; one contained a single transducer positioned over the center of the frog and the second contained transducers located at the toe and both sides of the heel. Horses were shod with these instrumented shoes and walked and trotted over a force plate. Forces were simultaneously recorded from the transducers in the shoes and from the force plate. Comparisons were made between the amount and duration of the forces exerted on the transducers and the vertical and horizontal forces recorded from the force plate. Forces recorded from the single transducer shoes showed strong correlations with the forces recorded from the force plate for horses moving at the walk; however, at the trot only moderate correlations occurred between these forces. At both the walk and trot, forces recorded from each side of the heel and the total forces occurring on all three transducers from the front hooves of horses shod with three transducer shoes showed strong correlations to the vertical forces recorded from the force plate.Vertical forces were also recorded from the instrumented shoes as horses walked, trotted and galloped on a track straightaway. Forces recorded from normal horses shod with single transducer shoes on all four feet were greater on the forelimbs than the rear limbs at the walk and trot. At the gallop, forces were highest on the lead front followed by the nonlead front, lead rear and nonlead rearlimb, respectively. Forces recorded from a three transducer shoe on the right front hoof of a horse walking, trotting and galloping in a right lead were highest on the medial side of the heel and occurred during the middle of the support period. Peak forces on the toe occurred at or near the time of heel lift.The results of these studies indicate that these instrumented shoes have advantages over the methods previously used to measure locomotor forces. These instrumented shoes can be used to simultaneously record the temporal components and the amount and distribution of vertical forces exerted during consecutive strides of horses moving at a variety of gaits. 相似文献

2, 4, or 8°C was studied in relation to several catalytic activities. Bintje tubers accumulated sugars during storage at 2
or 4°C, whereas KW77-2916 showed reduced cold-sweetening at 2°C. The increase in glucose 6-phosphate and sucrose occurred
concurrently and preceded the increase in reducing sugar concentration. Phosphorylase activity showed a strong interaction
with temperature, storage duration and sugar accumulation in both genotypes. Invertase activity increased in Bintje concomitantly
with the increase in reducing sugars, but this effect was less obvious in KW77-2916. The activities of other glycolytic and
Krebs cycle enzymes showed no obvious correlation with sugar accumulation. It is suggested that the increase in phosphorylase

An HPLC method with on-line cleanup coupled to the separation column is described for determination of (-)-norephedrine, (+)-norpseudoephedrine, (-)-ephedrine, (+)-pseudoephedrine, (-)-N-methylephedrine, (+)-N-methylpseudoephedrine, and (+/-)-synephrine in finished dietary supplement products. Test portions were extracted in acidified aqueous acetone. A filtered aliquot was cleaned up on a strong cation exchange (SCX) precolumn that later was automatically coupled to the SCX analytical column. Measurement was by full-scan UV spectra for confirmation of identity by spectral matching and real-time integration of three wavelength signals for multiple quantitation. (+/-)-Synephrine was also quantitated by native fluorescence. Recovery averaged 95-100%. Determination of the major ingredients (-)-ephedrine, (+)-pseudoephedrine, and (+/-)-synephrine compared favorably to findings by an independent LC-MS analysis for a set of 25 samples. The results of a survey were reported for total ephedrine alkaloid and synephrine content and were compared to content declaration, for approximately 48 finished products. 相似文献
