Enzymatic extracts from olive pulp (Olea europea L.) were used to characterize lipoxygenase (LOX) activity in order to determine its role in the biogenesis of the volatile compounds that influence the aroma of extra virgin olive oil. The LOX activity was tested spectrophotometrically at an optimal pH of 6.0 in three olive cultivars, Ascolana Tenera, Kalamata, and FS17. The trend of the LOX activity was determined as a function of pH and temperature; the kinetic constants of the enzyme were also determined. The highest LOX activity was observed in the FS17 fruit, which had the highest concentrations of C(5) and C(6) compounds (aldehydes, alcohols, and ketones), followed by Kalamata and Ascolana T., respectively. Given the direct relationship between enzymatic activity and the quantity of aromas measured in the fruit, it is hypothesized that olive LOX is involved in the formation of C(5) and C(6) volatile compounds. To study the mechanism of the movement of the aromas from the fruit to the oil, which was obtained by simple mechanical extraction, the headspace of the oil for each cultivar was analyzed as well as the aromatic composition in order to compare it with the aromas of the fruit.  相似文献

Since in the aged heart an increased basal production of reactive oxygen species (ROS) has been demonstrated, and the resistance to ROS attack could be ameliorated by antioxidant supplementation, we verified the protective effect of selenium, as sodium selenite (SS) or seleno methionine (SM), in cultured rat cardiomyocytes aged in vitro. In normoxia, glutathione peroxidase (GPx) activity and total antioxidant activity were higher in old than in young cardiomyocytes, suggesting the existence of a compensatory increase of antioxidant defenses. When aged cells were submitted to hypoxia/reoxygenation, GPx activity was not modified; while total antioxidant activity decreased, conjugated diene level increased. Selenium supplementation, particularly as SM, was able to increase GPx, and consequently total antioxidant activity, and to decrease conjugated diene production. The observed ability of selenium supplementation to protect aged cardiomyocytes from hypoxia/reoxygenation damage underlines the importance of an optimal selenium dietary intake, particularly in the elderly.  相似文献

Random-amplified polymorphic DNA (RAPD) markers were used to evaluate genetic variability among populations of an Italian strawberry ecotype, and to determinate genetic relationships between genotypes and their putative ancestor. A total of 65 selections and one cultivar ‘Madame Moutot’ (MM), were analysed to evaluate genetic variability present in Etna mountain area and to confirm as MM was one of the cultivars that originated the ecotype. A total of 222 RAPD markers was obtained using 16 decamer primers and 6 longer primers, 90.8% of the markers obtained by selected primers resulted polymorphic at least within analysed genotypes. RAPDs were used to calculate genetic similarity coefficients and to generate dendrograms representing genetic relationships among genotypes analysed. Cluster analysis displays as RAPD polymorphisms were able to characterize the genotype variability among closely related groups. The data show as MM could be considered the ancestral genotypes introduced in that area. The results obtained confirm that RAPD markers could be used as reliable markers to perform phylogenetic studies in Fragaria×ananassa Duch. ex Rozier. Monitoring the environmental impact of anthropogenic disturbance on soil ecosystem is of great importance for optimizing strategies for soil use, conservation and remediation. The aim of this study was to assess whether and to what extent a long-term, human-induced disturbance could have affected main chemical and biological properties in an agricultural soil. The study site was a hazel (Corylus avellana L.) orchard located in the area surrounding the volcanic apparatus of Somma-Vesuvius (Southern Italy). For the last two decades, the site has been repeatedly subjected to floodings by wastewaters containing not only alluvial sediments but also potentially hazardous compounds from illegally disposed wastes. Soil disturbance was assessed by a multitechnique approach, which combined chemical, biochemical and physiological (Biolog®) methods together with community fingerprinting by denaturing gradient gel electrophoresis (DGGE) and amplified ribosomal DNA restriction analysis (ARDRA). A hazel site never subjected to flooding provided the control soil. Soil sampling was repeated three times over a 1-year period. The effect of flooding by wastewaters, sampling time and their interaction were statistically evaluated. Under wastewater flooding, soil pH and most organic matter-related pools, i.e. total organic C, total N, and active soil C-resources such as basal (SBR) and substrate-induced respiration (SIR) and microbial biomass C (MBC) were all increased; whereas sampling time mostly affected two active-N pools, namely K2SO4-extractable N (Extr-N) and potentially mineralizable N that varied unconcurrently in tested soils. Also the electrical conductivity varied across samplings. Parameters related to microbial maintenance energy (ATP and qCO2) were higher in the flooded soil, even though they were not statistically affected by wastewater flooding or by sampling time. The Biolog® method evidenced that under recurrent flooding, soil microbial populations became functionally more uniform when compared to the control soil. Molecular fingerprinting of PCR-amplified 16S rDNA targets revealed that, along with seasonal shifts, a marked change in the genetic structure of total bacterial community occurred in the flooded soil. Furthermore, compositional shifts in the actinomycete community were less marked and mostly influenced by seasonal effects. Yet, a decreased genetic diversity in the ammonia-oxidizing bacteria community was evidenced in the flooded soil by ARDRA. Thus both the genetic and the functional structure of native soil bacterial populations were changed under repeated flooding by wastewaters. Repeated sampling over a 1-year period allowed us to reveal soil disturbance effects beyond seasonal variations.  相似文献

Isomers of 1,2,3,4,5,6-hexachlorocyclohexane (HCH) were some of the most widely used pesticides. Despite reduction in their production and use, HCH isomers present a serious environmental hazard. In this study, two bacterial isolates (LIN-1 and LIN-3) that can grow on gamma-HCH as a sole source of carbon and energy were isolated from an enrichment culture. In liquid cultures of LIN-1 and LIN-3, 25.0 and 45.5% removal of gamma-HCH, respectively, were achieved in 2 weeks. LIN-3 was identified as Pandoraea sp. by 16S rRNA gene sequence analysis (99% identity). Pandoraea sp. substantially degraded both gamma- and alpha-HCH isomers at concentrations of 10-200 mg L(-1) in liquid cultures. After 8 weeks of incubation in liquid culture, 89.9 and 93.3% of the gamma- and alpha-HCH isomers declined, respectively, at an initial concentration of 150 mg L(-1). In soil slurry cultures of Pandoraea sp., simulating a soil slurry phase bioremediation treatment, substantial decreases in the levels of the HCH isomers were observed at concentrations of 50-200 mg L(-1). After 9 weeks, 59.6 and 53.3% biodegradations of gamma- and alpha-HCH isomers, respectively, were achieved at 150 mg L(-1). Using two 23-mer oligonucloetide primers targeting the 330 bp region of the 16S rRNA gene of Pandoraea sp., an approximately 330 bp PCR product was successfully amplified from DNA templates prepared from bacterial colonies and soil slurry culture. This system provides a direct and rapid PCR-based molecular tool for tracking Pandoraea sp. strain LIN-3 in water and soils. These results have implied implications for the treatment of soils and water contaminated with HCH isomers.  相似文献

The productive potential of two different ecotypes of Rosmarinus officinalis (Cevoli and Lunigiana) cultivated in the littoral area near Pisa (northern Tuscany, Italy) and the differences in the yield and composition of the essential oils of leaves, flowers, and stems obtained from different positions of the plants were used to characterize the two ecotypes. The Cevoli ecotype plant produced the highest yield of dry matter (221 g plant-1) in comparison to the Lunigiana ecotype (72 g plant-1). There were significant differences in dry matter production of different organs of both ecotypes. The essential oil contents of Cevoli and Lunigiana ecotypes were similar. In contrast, the oil contents of the different plant parts showed marked differences. The apical part of the plant and the leaves gave the highest essential oil yields. The major difference between the oils of the two ecotypes consisted in the 1,8-cineole contents (6.6 and 37.9% in Cevoli and Lunigiana, respectively). The Cevoli ecotype was determined to be the most suitable for essential oil extraction because it was characterized by a preponderance of flowers and leaves in the apical portion. The Cevoli ecotype could be classifited as an alpha-pinene chemotype, whereas Lunigiana is a 1,8-cineole chemotype.  相似文献

The concentrations of trans-resveratrol, (+)-catechin, (-)-epicatechin, and quercetin were evaluated by means of high-performance liquid chromatography-diode array detection in red wines obtained from Aglianico, Piedirosso, and Nerello Mascalese grapes. The trans-resveratrol and epicatechin concentrations did not differ significantly between experimental wines. The concentration of quercetin in Nerello Mascalese wines was more than twice that observed in Aglianico and Piedirosso wines. Nerello Mascalese wines also significantly differed from other wines in the (+)-catechin content, which was significantly higher than those found in the other two wines. During maceration, the maximum extraction of trans-resveratrol was reached after 12 days for Aglianico and Piedirosso, after which a decline was observed. On the contrary, in the case of Nerello Mascalese, the concentration of trans-resveratrol increased steadily throughout the whole maceration process. After 2 days of maceration, the maximum concentration of quercetin was observed in Aglianico must, whereas the maximum quercetin extraction was reached after 12 days for Piedirosso and 17 days for Nerello Mascalese. The maximum levels of (+)-catechin and (-)-epicatechin were generally observed after 12 days of maceration for all wines, although a decline of (-)-epicatechin occurred after maximum extraction in Aglianico and Piedirosso wines. Following marc pressing, a significant increase in the concentration of trans-resveratrol for Aglianico, (+)-catechin and (-)-epicatechin for Piedirosso, and (-)-epicatechin for Nerello Mascalese was observed.  相似文献

This study evaluates the toxic, genotoxic/mutagenic, and antimutagenic effects of propolis extract from Amaicha del Valle, Tucumán, Argentina. The cytotoxicity assays carried out with the lethality test of Artemia salina revealed that the LD50 was around 100 microg/mL. Propolis extracts showed no toxicity to Salmonella typhimurium TA98 and TA100 strains and Allium cepa at concentrations that have antibiotic and antioxidant activities. Otherwise, for the testing doses, neither genotoxicity nor mutagenicity was found in any sample. The propolis extracts were able to inhibit the mutagenesis of isoquinoline (IQ) and 4-nitro o-phenylenediamine (NPD) with ID50 values of 40 and 20 microg/plate, respectively. From this result, the studied propolis may be inferred to contain some chemical compounds capable of inhibiting the mutagenicity of direct-acting and indirect-acting mutagens. A compound isolated from Amaicha del Valle propolis, 2',4'-dihydroxychalcone, showed cytotoxic activity (LC50 values of 0.5 microg/mL) but was not genotoxic or mutagenic. Furthermore, this compound was able to inhibit the mutagenicity of IQ (ID50 values of 1 microg/plate) but was unable to inhibit the mutagenicity of NPD. Our results suggest a potential anticarcinogenic activity of Amaicha del Valle propolis and the chalcone isolated from it.  相似文献
