Mycobacterium tuberculosis (Mtb) and Mycobacterium bovis (M. bovis) are the etiological agents of human and bovine tuberculosis (TB, bTB) respectively, and share genetic identity over 99% at the whole genome level. Progress has been made towards explaining how mycobacteria and their infected hosts remain in balance without producing clinical symptoms of disease, a phenomenon referred to as latency or persistence, which can be mimicked by certain in vitro conditions. Latency/persistence has mainly been studied using Mtb, where the two-component signalling system, dosRS, has been assigned an instrumental role, and even constitutes the current basis for development of new diagnostic methods and treatment addressing this particular stage of TB. M. bovis conserves homolog genes that in Mtb play a role in human latent TB infection and that, by analogy, would allow it to enter a persistent state in infected cattle; nevertheless, little attention has been paid to this stage in bovine hosts. We suggest that many of the advances acquired through the study of Mtb can and should be taken into consideration by research groups and veterinary professionals dealing with bTB. The study of the infection in bovines, paying particular attention to defining the molecular and cellular markers of a M. bovis persistent infection in cattle, presents great opportunities for the development and trial of new diagnostic tests and vaccines, tools that will surely help in promoting eradication of bTB in high-burden settings.  相似文献

Abstract: A 15‐month‐old, female mongrel dog was presented with a 6‐week history of inappetence, weight loss, and tetraparesis. Physical examination revealed weakness, poor body condition, mild fever, pale mucous membranes, and diffuse muscle atrophy. The right hind limb was painful and edematous, with large ecchymoses. The femur was irregular on palpation and moderate popliteal lymphadenopathy was evident. Results of a CBC showed severe anemia with mild regeneration, an inflammatory leukogram with 90% of neutrophils parasitized by Hepatozoon sp. gamonts, and moderate thrombocytopenia. A bone marrow aspirate had myeloid hyperplasia and contained a few extracellular Hepatozoon meronts and a few intracellular gamonts within neutrophils. Serum chemistry abnormalities included hypoalbuminemia, hyperglobulinemia, hypoglycemia, hypercalcemia, hyperphosphatemia, and elevated alkaline phosphatase activity. Radiologic findings of the right femur included periosteal bone proliferation and lesions compatible with osteomyelitis. A fine needle aspirate specimen from the bone lesion had neutrophilic inflammation; 36% of the neutrophils contained Hepatozoon gamonts. Results of cerebrospinal fluid analysis included a protein concentration of 37 mg/dL and marked mononuclear pleocytosis (243 cell/μL) with a predominance of lymphocytes. An ELISA was positive for Hepatozoon canis and PCR results with DNA sequencing confirmed infection with this organism. A diagnosis of hepatozoonosis with skeletal involvement and meningoencephalomyelitis was made. The dog recovered almost completely neurologically and had no gamonts in the blood after 60 days of therapy with imidocarb dipropionate and prednisone. This is an unusual case of canine hepatozoonosis involving neurologic signs and a periosteal reaction more typical of H. americanum infection and rarely reported in dogs infected with H. canis.  相似文献

During its first flyby of Mercury, the MESSENGER spacecraft measured the planet's near-equatorial magnetic field. The field strength is consistent to within an estimated uncertainty of 10% with that observed near the equator by Mariner 10. Centered dipole solutions yield a southward planetary moment of 230 to 290 nanotesla RM3 (where RM is Mercury's mean radius) tilted between 5 degrees and 12 degrees from the rotation axis. Multipole solutions yield non-dipolar contributions of 22% to 52% of the dipole field magnitude. Magnetopause and tail currents account for part of the high-order field, and plasma pressure effects may explain the remainder, so that a pure centered dipole cannot be ruled out.  相似文献

Abstract

The objective of this research was to determine the difference on growth between a rice cultivar with Clearfield® technology (Only Rice 228) and a hybrid (Benja 1); to characterize nutrient uptake, distribution, accumulation and removal between these two commercial genotypes. Tests under shade house and field conditions were performed to estimate macro and micronutrient uptake patterns. Plants were sampled at nine growth stages (emergence, initiation of tillering, active tillering, initiation of panicle primordia, booting, flowering, milky, soft dough and mature grain) and divided into different organs for nutrient determination. The results showed that “Benja 1” plants (92 d) had a shorter cycle than “Only Rice 228” (OR 228) plants (118 d). “OR 228” exhibited a greater biomass production (16.575?kg ha?1 vs. 12.621?kg ha?1) in field. The nutrient acquisition was faster in the hybrid Benja 1 between tillering initiation and the milky grain stage in which the N, K, Ca, Mg, Mn, B, and Cu uptake was more evenly and highly distributed throughout these stages in both conditions. “Benja 1” showed a higher nutrient harvest index (HI). HI values above 50% (P (62%), N (61%), Cu (67%), S (55%), and Mg (52%)) were found in Benja 1 under field conditions. The results also highlight Si removal in both rice genotypes, in which Benja 1 stands out. These results provide information on the nutrient uptake and partitioning of modern rice genotypes, and give the knowledge to optimize fertilizer programs and timing recommendations for rice biomass and grain production in Colombia.  相似文献

The main objective of this study was to evaluate the effect of sample preparation on cell-wall density and porosity using gas pycnometry. Native and thermally modified twin samples of Norway spruce (Picea abies (L.) Karst.), sycamore maple (Acer pseudoplatanus L.), and European ash (Fraxinus excelsior L.) were analyzed. The samples differed in terms of shape, geometry, and climatic state. On the one hand, the samples were oven dry as usual and, on the other hand, conditioned at 22 °C and 95 % relative humidity. Furthermore, the samples were processed using solvent exchange drying. In addition to helium, nitrogen was used as a displacement gas. The tests show i.a. how this can lead to misinterpretation of the cell-wall density or porosity of wood determined by gas pycnometry. The results show that native spruce has a lower cell-wall density and higher porosity compared with native maple and ash. Due to thermal modification, the cell-wall densities are decreased. The investigations show that the determined cell-wall density and porosity of the wood are strongly dependent on the sample geometry and climatic state. The cell-wall densities of all investigated wood species in the conditioned state at 22 °C and 95 % relative humidity are significantly higher compared with the oven-dry cell-wall densities.  相似文献

Resonance frequencies of bars with various types of end supports were examined in terms of flexural vibration. Sugi (Japanese cedar, Cryptomeria japonica D. Don) was used for the specimens. Small rectangular bars with dimensions of 25?mm (R)?×?5?mm (T)?×?300?mm (L), small round bars of 25?mm in diameter and 250?mm in length, actual-size rectangular bars with the dimensions of 45?mm (R)?×?95?mm (T)?×?1500?mm (L) and actual-size round bars of 180?mm in diameter and 2000?mm in length were made. Varying compression stress was applied to the parts around both ends of test specimen, whereupon flexural vibration tests were performed. The measured resonance frequency rose rapidly early in the increasing compression load process and was nearly stable for the larger compression load. The significant increases in the resonance frequency for round bars are thought to be derived from the wide area between the round bar and jig. It is necessary to compare the resonance frequency of a bar tightly fixed to a post with that of the same bar loosely fixed to the post.  相似文献

The performance of female calves in creep feeding under different strategies of supplementation and milk production, intake, and digestibility of grazing Nellore and crossbred cows (Nellore?×?Holstein) during the dry-rainy transition season were assessed. Forty-four female beef suckling calves, with initial age between 90 and 150?days and average initial body weight of 117.7?±?4.3?kg, and their respective dams (24 Nellore and 20 crossbred) with average initial body weight of 417.5?±?8.3?kg, were used. The experimental treatments consisted of: control group-mineral mixture only; strategy 1-supplementation from 112?days prior to weaning (0.375?kg/animal/day); strategy 2-supplementation from 112?days prior to weaning, in increasing amounts of 0.15, 0.30, 0.45, and 0.60?kg/animal/day through the four experimental periods, respectively; and strategy 3-supplementation from 56?days prior to weaning (0.750?kg/animal/day). Calves from strategy 1 had greater (P??0.05) for nutrient digestibility among genetic types. It can be concluded that strategies of supplementation that present an equitable distribution of supplement provides greater weight gain in suckling female beef calves. Crossbred cows produce more milk and present greater DMI than Nellore cows. There are no differences in the nutrient digestibility between Nellore and Nellore × Holstein crossbred cows.  相似文献

The reference strains representing serotypes 1 to 12 of Actinobacillus pleuropneumoniae biotype 1 were examined for their ability to utilize porcine hemoglobin (Hb) or porcine hemin (Hm) as iron sources for growth. In a growth promotion assay, all of the reference strains were able to use porcine Hb, and all strains except 2 were able to use porcine Hm. Using a preliminary characterization procedure with Hm- or Hb-agarose, Hm- and Hb-binding outer membrane proteins (OMPs) of approximately 75 kDa were isolated from A. pleuropneumoniae serotype 1 strain 4074 grown under iron-restricted conditions. Matrix-assisted laser desorption ionization/time-of-flight (MALDI-TOF) analysis revealed a number of common tryptic peptides between the Hb-agarose- and Hm-agarose-purified 75 kDa OMPs, strongly suggesting that these peptides originate from the same protein. A database search of these peptide sequences revealed identities with proteins from various Gram-negative bacteria, including iron-regulated OMPs, transporter proteins, as well as TonB-dependent receptors. Taken together, our data suggest that A. pleuropneumoniae synthesizes potential Hm- and Hb-binding proteins that could be implicated in the iron uptake from porcine Hb and Hm.  相似文献
