本研究以抗虫水稻恢复系9311(Bt)及其杂交种F1(Bt)为研究材料,以非转基因的9311为阴性对照,利用ELISA方法研究9311(Bt)及F1(Bt)各生育时期可溶性总蛋白和Bt蛋白的时空变化规律,为转Bt基因抗虫水稻的安全监管提供科学依据。结果表明:外源基因的导入没有引起水稻组织中可溶性总蛋白含量的明显变化;9311(Bt)的Bt蛋白表达量在整个生长周期的各个部位均高于相应的F1(Bt)植株;同一植株不同组织器官中Bt蛋白表达量为:叶片跃胚乳跃颖壳及茎秆跃根;同一植株不同发育期叶片Bt蛋白的测定结果整体表现为:营养生长阶段跃生殖生长阶段跃成熟衰老阶段。研究结果为转Bt基因抗虫水稻适宜检测时期的选择提供了一定的参考。 相似文献

Urethral obstruction is a life‐threatening form of feline lower urinary tract disease. Ultrasonographic risk factors for reobstruction have not been previously reported. Purposes of this retrospective cross‐sectional study were to describe urinary tract ultrasound findings in cats following acute urethral obstruction and determine whether ultrasound findings were associated with reobstruction. Inclusion criteria were a physical examination and history consistent with urethral obstruction, an abdominal ultrasound including a full evaluation of the urinary system within 24 h of hospitalization, and no cystocentesis prior to ultrasound examination. Medical records for included cats were reviewed and presence of azotemia, hyperkalemia, positive urine culture, and duration of hospitalization were recorded. For medically treated cats with available outcome data, presence of reobstruction was also recorded. Ultrasound images were reviewed and urinary tract characteristics were recorded. A total of 87 cats met inclusion criteria. Common ultrasound findings for the bladder included echogenic urine sediment, bladder wall thickening, pericystic effusion, hyperechoic pericystic fat, and increased urinary echoes; and for the kidneys/ureters included pyelectasia, renomegaly, perirenal effusion, hyperechoic perirenal fat, and ureteral dilation. Six‐month postdischarge outcomes were available for 61 medically treated cats and 21 of these cats had reobstruction. No findings were associated with an increased risk of reobstruction. Ultrasonographic perirenal effusion was associated with severe hyperkalemia (P = 0.009, relative risk 5.75, 95% confidence interval [1.54–21.51]). Findings supported the use of ultrasound as an adjunct for treatment planning in cats presented with urethral obstruction but not as a method for predicting risk of reobstruction. 相似文献

A 30‐day feeding trial was conducted to investigate the effects of dietary lipid levels on growth performance, activities of digestive enzymes, fatty acid composition and some lipogenesis‐related gene expression of half‐smooth tongue sole (Cynoglossus semilaevis) larvae. Five isoproteic diets were formulated with graded lipid levels (6.68%, 9.84%, 13.47%, 17.89% and 21.88% dry weight) using fish oil as the main lipid source. Each diet was randomly allocated to triplicate groups of 150 larval tongue sole (35 DAH, 54 ± 1 mg). Fish were fed five times daily to apparent satiation during the feeding experiment. Results showed that, the survival rate (SR) of larvae increased significantly firstly, and thereafter decreased significantly. The specific growth rates (SGR) of larvae fed the diet with 13.47% lipid were significantly higher than other treatments. Larvae fed 9.84% or 13.47% dietary lipid showed higher trypsin, lipase, leucine aminopeptidase and alkaline phosphatase (AP) activities than other treatments, whereas amylase activity nearly showed reverse trend with them. The fatty acid composition of the tongue sole larvae was well correlated with dietary fatty acid profile. Expression of acetyl‐CoA carboxylase alpha (ACC1) was found to be slightly negatively correlated with dietary lipid level, and high dietary lipid level depressed the expressions of acetyl‐CoA carboxylase beta (ACC2) and fatty acid synthase (FAS) mRNA expression significantly, implying that larvae may cope with high dietary lipid mainly through down‐regulating lipogenesis‐related gene expression of FAS and ACC2. Besides, on the basis of SGR, the optimal dietary lipid level for larval tongue sole was estimated to be 13.56% using second‐order polynomial curve. 相似文献

The effect of partial substitution of dietary fish meal with a high‐value soybean meal (HVS) and a commercial soybean meal (CSM) on the growth performance in juvenile Japanese seabass (8.3±0.2 g body weight) was determined. Nine isonitrogenous (crude protein 44%) and isoenergetic (20 kJ g?1) practical diets replacing 0 (the control), 15%, 30%, 45% and 60% fish meal protein by soybean meal protein (HVS or CSM) were formulated. Each diet was randomly assigned to four replicate cages (1.5 × 1.5 × 2.0 m). Fish were fed twice daily for 10 weeks. Growth was significantly reduced with increased soybean inclusion in fish fed both the soybean sources. Independent of the soybean source used, increased inclusion of soybean meal decreased the growth performance. When the substitution level was 45% or more in CSM and 60% in HVS, the specific growth rate (SGR) and the feed efficiency ratio (FER) were significantly lower compared with the control. These results indicated that CSM protein could substitute for 30% fish meal protein, while the substitution level of HVS protein could be 45%, which did not influence the growth of juvenile Japanese seabass. 该研究对基于注意力机制的长短期记忆(Attention-Based Long Short Term Memory,AT-LSTM)模型对蒸散量(Evapotranspiration,ET)模拟的可行性和有效性进行验证,以提高环境数据缺失情况下的蒸散量模拟精度。基于盐池县2012-2017年的每30 min环境数据,利用不同环境因子组合构建基于注意力机制的LSTM模型,并将其与极限学习机(Extreme Learning Machine,ELM)模型、支持向量机(Support Vector Machine,SVM)模型、长短期记忆(Long Short Term Memory,LSTM)模型在日尺度、月尺度和季节尺度上进行对比分析。结果表明:与其他3种模型相比,当输入环境因子变化时,AT-LSTM模型模拟精度变化很小,模拟效果均较好。当输入空气温度、净辐射、相对湿度、土壤温度、土壤含水率所有环境因子时,基于AT-LSTM模型的模拟效果最好,均方根误差(Root Mean Square Error,RMSE)为0.013 mm/30 min,平均绝对误差(Mean Absolute Error,MAE)为0.006 mm/30 min,相关系数(Correlation Coefficient,R)值为0.905。且无论是从小时尺度、日尺度和月尺度来看,AT-LSTM模型的模拟效果均优于其他3种模型。在环境因子缺失的情况下,净辐射对盐池县ET的模拟贡献程度最大,仅输入净辐射时,AT-LSTM模型模拟得到的RMSE和MAE分别为0.014、0.007 mm/30 min,R为0.892。AT-LSTM模型模拟精度高,模型稳定性强,对蒸散量模拟预测具有一定的适用性,仅输入净辐射的AT-LSTM模型可以作为环境数据缺失条件下的蒸散量预测模型。 相似文献
