This study aimed to develop a system of in vitro assays based on zona pellucida binding and in vitro fertilization for predicting male fertility in buffalo bulls. Frozen–thawed semen from nine bulls was tested for motility, viability index, acrosomal integrity, zona pellucida binding and in vitro fertilizing ability. Differences in post-thaw sperm motility between bulls were not significant. Differences in viability indices and percentage of spermatozoa with detached acrosome between bulls was highly significant (P < 0.001). Sperm attached per ovum, fertilization rates and polyspermy percentages varied significantly (P < 0.01) among buffalo bulls. A significant (P < 0.01) positive correlation coefficient of 0.69 was evident between normal acrosome and sperm attached per ovum, while between normal acrosome and fertilization efficiency it was 0.72. Sperm from different buffalo bulls differs in their ability to bind and fertilize oocytes. This study provides a basis to predict and maximize the in vitro fertilization performance of individual bulls. 相似文献

We investigated the effects of grazing on natural regeneration, quantity, and diversity of woody species and dominant herb species in Kheyroud forest in northern Iran. We sampled vegetation in 5m2 plots in custom units, which are demarcated resource areas traditionally used by local livestock producers. The authors quantified number of species, height of seedlings, and diameter of seedlings. Height classes were 0-30 cm, 30-130 cm, and >130 cm, and diameter classes were 0-2.5 cm, 2.5-5 cm and 5-7.5 cm. The density of seedlings declined with distance from corral until reaching the custom unit boundary. Most seedlings had diameters of 0-2.5 cm and heights of 0-30 cm. Predominant species, Carpinus betulus and Acer capadocicum, were in plots near the centers of custom units, Fagus orientalis, Acer velutinum, Quercus castanifolia species were dominant in plots near the custom unit boundary. Plant species such as Oplismenus undulatifolius, Euphorbia amygdaloides, Rubus fruticos and Pteridium aquilinum were dominant in plots nearer to forest corral. Healthy seedlings were more numerous in plots nearest the corral, while defective and deformed seedlings were more abundant away from the corral. We conclude that grazing had negative effects on the quantity and quality of vegetative regeneration. Continuation of overgrazing will not only endanger the sustainability of forest ecosystems, but also will increase the challenge of sustainable forest management. 相似文献

The interactions between soil P availability and mycorrhizal fungi could potentially impact the activity of soil microorganisms and enzymes involved in nutrient turnover and cycling, and subsequent plant growth. However, much remains to be known of the possible interactions among phosphorus availability and mycorrhizal fungi in the rhizosphere of berseem clover (Trifolium alexandrinum L.) grown in calcareous soils deficient in available P. The primary purpose of this study was to look at the interaction between P availability and an arbuscular mycorrhizal (AM) fungus (Glomus intraradices) on the growth of berseem clover and on soil microbial activity associated with plant growth. Berseem clover was grown in P unfertilized soil (−P) and P fertilized soil (+P), inoculated (+M) and non-inoculated (−M) with the mycorrhizal fungus for 70 days under greenhouse conditions. We found an increased biomass production of shoot and root for AM fungus-inoculated berseem relative to uninoculated berseem grown at low P levels. AM fungus inoculation led to an improvement of P and N uptake. Soil respiration (SR) responded positively to P addition, but negatively to AM fungus inoculation, suggesting that P limitation may be responsible for stimulating effects on microbial activity by P fertilization. Results showed decreases in microbial respiration and biomass C in mycorrhizal treatments, implying that reduced availability of C may account for the suppressive effects of AM fungus inoculation on microbial activity. However, both AM fungus inoculation and P fertilization affected neither substrate-induced respiration (SIR) nor microbial metabolic quotients (qCO2). So, both P and C availability may concurrently limit the microbial activity in these calcareous P-fixing soils. On the contrary, the activities of alkaline phosphatase (ALP) and acid phosphatase (ACP) enzymes responded negatively to P addition, but positively to AM fungus inoculation, indicating that AM fungus may only contribute to plant P nutrition without a significant contribution from the total microbial activity in the rhizosphere. Therefore, the contrasting effects of P and AM fungus on the soil microbial activity and biomass C and enzymes may have a positive or negative feedback to C dynamics and decomposition, and subsequently to nutrient cycling in these calcareous soils. Cobalt ferrite nanocrystals were prepared from an aqueous solution containing metal nitrates and various of concentrations of poly(vinyl pyrrolidone), followed by calcinations temperature. X-ray diffraction (XRD) analysis was performed to determine the degree of crystallinity of the ferrite nanoparticles. By transmission electron microscopy (TEM), the morphology and average particle size of the cobalt ferrite nanoparticles were evaluated which had good agreement with XRD results. Fourier transform infrared spectroscopy (FT-IR) suggested the presence of metal oxide bands in all samples as well as the effective elimination of organic constituents after calcinations. High-intensity exercise and competition are associated with depressed immune function. Young horses, which participate in high-intensity exercise and competitions, are at increased risk for the development of infectious disease due to depression of immune function. The effects of branched-chain amino acid (BCAA) supplementation on the immune status of young racing horses were evaluated, determining whether BCAA might help to avoid or reduce immune suppression during exercise and competitions. Twenty horses (10 male and 10 female) were treated with BCAA supplementation; another twenty untreated horses (10 male and 10 female) constituted control group. Peripheral blood was collected from each animal and evaluated for lymphocyte subsets, phagocytosis analysis of monocytes and granulocytes, lymphocyte proliferative response, and expression of cytokine-encoding messenger ribonucleic acids (mRNAs). The numbers of CD4+, CD8+, and major histocompatibility complex (MHC) class II+ cells in females of the treated group were significantly higher than those in females of the control group. The lymphocyte proliferative response in female of the treated group also was significantly higher than that in females of the control group. In addition, expression of mRNAs encoding interleukin-1β (IL-1β) and interferon-γ (IFN-γ) in females of the treated group was significantly higher than that in females of the control group. There were no significant differences between males of the treated and control groups. The results of this study indicated the positive effects of BCAA supplementation in counteracting immunosuppression in young female racing horses during and following high-intensity exercise. 相似文献

From March 1996 to February 1997, slaughtered-sheeps' heads were examined for the metacestode of Taenia multiceps. Out of 451 sheep heads, only 12 (3%) were infested. All cysts but one were in the age group 0.5-2.5 years old, with prevalence of 10% (11 of 108). Eleven of 12 infested heads were found during the spring and autumn seasons. Out of 95 flocks involved in questionnaire survey unrelated to the abattoir survey, all the farmers reported the existence of the clinical signs known for coenurosis and described it in locally known names and diseases. Twenty-five of them had recent cases that were diagnosed by the veterinarian as coenurosis cases. Each flock reported one or two cases at the time of survey. A variety of clinical signs were reported from these cases (n = 42). These included circling (86%), head pressing (52%), blindness (29%) and paresis (40%). This study was designed to evaluate effects of different combinations of cryoprotectants on the ability of vitrified immature buffalo oocytes to undergo in vitro maturation. Straw and open‐pulled straw (OPS) methods for vitrification of oocytes at the germinal vesicle stage also were compared. The immature oocytes were harvested from ovaries of slaughtered animals and were divided into three groups: (i) untreated (control); (ii) exposed to cryoprotectant agents (CPAs); or (iii) cryopreserved by straw and OPS vitrification methods. The vitrification solution (VS) consisted of 6 m ethylene glycol (EG) as the standard, control vitrification treatment, and this was compared with 3 m EG + 3 m dimethyl sulfoxide (DMSO), 3 m EG + 3 m glycerol, and 3 m DMSO + 3 m glycerol. Cryoprotectants were added in two steps, with the first step concentration half that of the second (and final) step concentration. After warming, oocyte samples were matured by standard methods and then fixed and stained for nuclear evaluation. Rates of MII oocytes exposed to CPAs without vitrification were lower (54.3 ± 1.9% in EG, 47.5 ± 3.4% in EG + DMSO, 36.8 ± 1.2% in EG + glycerol and 29.9 ± 1.0% in DMSO + glycerol; p < 0.05) than for the control group (79.8 ± 1.3%). For all treatments in each vitrification experiment, results were nearly identical for straws and OPS, so all results presented are the average of these two containers. The percentages of oocytes reaching telophase‐I or metaphase‐II stages were lower in oocytes cryopreserved using all treatments when compared with control. However, among the vitrified oocytes, the highest maturation rate was seen in oocytes vitrified in EG + DMSO (41.5 ± 0.6%). Oocytes cryopreserved in all groups with glycerol had an overall low maturation rate 19.0 ± 0.6% for EG + glycerol and 17.0 ± 1.1% for DMSO + glycerol. We conclude that the function of oocytes was severely affected by both vitrification and exposure to cryoprotectants without vitrification; the best combination of cryoprotectants was EG + DMSO for vitrification of immature buffalo oocytes using either straw or OPS methods. 相似文献

An outbreak of cerebrospinal microfilariosis in 23 kids, representing a whole kid crop, occurred on a farm in Qassim, Central Saudi Arabia. The morbidity and mortality rates were 100%. Clinical signs, in kids from the fourth to sixth week of age, manifested as weakness in the hind legs followed by incoordination of movement and feet dragging. Seven to ten days later, the hind legs were completely paralyzed, and within a week the front legs were also paralyzed. The animals continued to feed, drink and suckle normally, and were in good health. Paralyzed kids were helped to suckle by the farm attendants. With time, loss of control of motor functions occurred and the ability to hold the head in upright position was lost, ability to eat or drink was lost. The infected kids health deteriorated and they eventually died. Two dead kids were postmortemed. With the exception of a meningeal haemorrhage seen in one kid, no obvious macroscopic lesions were seen in any of the autopsied animals. Histopathological examination of the spinal cord and brain showed different microfilaria of different sizes between the meninges and the nervous tissue. This indicated that development of the parasite occurred in the nervous tissue. The larger microfilaria contained black deposits, most likely to be digested blood, in their guts. It is likely that the parasites are haematophagus. The main lesions caused by the microfilaria were migratory traumatic haemorrhagic tracts in the spinal cord and the brain. These sometimes contained parts of the microfilariae. In the brain malacic lesions and sporadic aggregates of lymphocytes were also noted. 相似文献

The aim of the present study was to investigate the effect of kisspeptin-10 (Kp10) injection on semen characteristics, testosterone (T) production and sperm rheotaxis using microfluidic devices in immature ram. Computer-assisted sperm analysis (CASA) with controlled flow velocity was used to explore the kinetic parameters of sperm and positive rheotaxis (PR %). PR % was defined as the number of PR sperms over the number of motile sperms. Healthy Ossimi rams were randomly divided into two groups; a saline-treated control group and Kp10-treated one (5 µg/kg body weight). Treatments were given by intramuscular injection once a week for 1 month. After 1 month, the semen was collected and evaluated weekly for 6 weeks, while the blood samples were collected every 2 weeks for the next 8 weeks. Semen properties were significantly affected by Kp10 injection (p < .01). The Kp10 increased the volume, sperm concentration and percentages of live sperm compared with those of control. Additionally, sperm trajectories and rheotaxis get improved by the injection of Kp10 with time. Furthermore, kisspeptin improved the secretion of testosterone levels throughout the period of study. In conclusion, injections of the Kp10 had a positive impact on semen characteristics as well as improved sperm rheotaxis of Ossimi rams in subtropics. 相似文献
