Stand establishment and subsequent autumn development and growth are important determinants of winter wheat (Triticum aestivum L.) yield. Soil management practices change soil properties and conditions, which alter seedling emergence, crop development and growth. Pre-plant soil management practices were studied for 6 years in a wheat–fallow rotation in eastern Colorado, USA, to isolate the impacts of pre-plant tillage (PT) and residue level on winter wheat seedling emergence and autumn development and growth. A split plot design was used with PT, using a moldboard plow that incorporated surface residue, and with no-tillage (NT). The tillage systems represented the main plots and three residue levels within each tillage treatment as subplots: no residue (0R), normal residue (1R) and twice-normal residue (2R). Residue amount had little effect on emergence or autumn growth and development. PT resulted in soil water loss from the plow zone. NT plots had more favorable soil water levels in the seeding zone which resulted in faster, more uniform and greater seedling emergence in 4 out of the 6 years. This is especially critical for stand establishment in years with low rainfall after planting. Soil or air temperature did not account for differences among treatments. Earlier and greater seedling emergence in NT treatments resulted in greater autumn development and growth. Shoot biomass, tiller density and leaf numbers were greater in NT, and again residue amount had little effect. At spring green-up, NT treatments had greater soil water in the profile. Grain yield was always equal or greater in NT than in PT, and positively correlated with earlier/greater seedling emergence and autumn growth. Bycatch in fisheries has been recognized as a threat to many endangered populations of sea turtles, sea birds and marine mammals. Interactions between pelagic longline fisheries and critically endangered populations of leatherback sea turtles (Dermochelys coriacea) have led to temporary closures of the Hawaiian pelagic longline swordfish fishery and severe bycatch quotas. The negative impact of these events on both the populations of certain endangered species and the economic livelihood of the fishermen has resulted in a strong push from all sides to better understand bycatch events. Typically, analyses of longline catch and bycatch have examined fishing effort summarized over large areas (≥1°). Although aggregation of effort to this level may be necessary to account for uncertainty, confidentiality concerns, or to make comparisons across regions, it specifically limits the researcher's ability to characterize the local oceanographic factors that may drive individual bycatch events. Higher resolution analyses must be undertaken to identify such features. However, for these higher resolution analyses, the methods currently used to spatially represent pelagic longline fishing effort may significantly affect researcher's results. Here, we look at different methods to represent this fishing effort (i.e., points, centroids, polylines and polygons) at various resolutions (2 km to 5°) to better understand which method and spatial resolution are most appropriate. Our results validate the use of point features to represent fishing effort in previous low resolution studies of the Hawaiian pelagic longline fishery by showing that the set point method is suitable for studies with resolutions lower than 15 km. However, at higher resolutions (≤15 km) and in areas with more sparsely distributed fishing, aggregated effort values differed significantly between spatial representation methods. Three variations to the structure of the nicotinamide adenine dinucleotide (NAD)-dependent L-lactate dehydrogenase from Bacillus stearothermophilus were made to try to change the substrate specificity from lactate to malate: Asp197----Asn, Thr246----Gly, and Gln102----Arg). Each modification shifts the specificity from lactate to malate, although only the last (Gln102----Arg) provides an effective and highly specific catalyst for the new substrate. This synthetic enzyme has a ratio of catalytic rate (kcat) to Michaelis constant (Km) for oxaloacetate of 4.2 x 10(6)M-1 s-1, equal to that of native lactate dehydrogenase for its natural substrate, pyruvate, and a maximum velocity (250 s-1), which is double that reported for a natural malate dehydrogenase from B. stearothermophilus. 相似文献

Two new isocoumarin derivatives, including a new 5-hydroxy-8-methyl-2H, 6H-pyrano[3,4-g]chromen-2,6-dione (1) and 6,8-dihydroxy-3,7-dimethylisocoumarin (2b), a new chevalone derivative, named chevalone E (3), and a new natural product pyripyropene S (6) were isolated together with 6, 8-dihydroxy-3-methylisocoumarin (2a), reticulol (2c), p-hydroxybenzaldehyde, chevalone B, chevalone C, S14-95 (4), and pyripyropene E (5) from the ethyl acetate extract of the undescribed marine sponge-associated fungus Aspergillus similanensis KUFA 0013. The structures of the new compounds were established based on 1D and 2D NMR spectral analysis, and in the case of compound 3, X-ray analysis was used to confirm its structure and the absolute configuration of its stereogenic carbons. Compounds 1, 2a–c and 3–6 were evaluated for their antimicrobial activity against Gram-positive and Gram-negative bacteria, Candida albicans ATCC 10231, and multidrug-resistant isolates from the environment. One-day-old White Leghorn chicks were fed graded levels (0, 10, 50, 100 and 1,000 ppm) of pentachloronitrobenzene (PCNB; quintozene) up to 8 weeks. Each treatment group consisted of 90 randomly selected birds (2 replications of 45 birds each). Body weight gains were significantly lower at the 1,000-ppm treatment level. Histopathologic examination of brain, liver, pancreas, small intestine, gizzard, spleen, kidney, lung, and heart failed to reveal lesions in either control or treated groups. Bioaccumulation of PCNB or its metabolites (pentachloroaniline and pentachlorophenylmethylsulfide) in tissues only occurred in trace or very low concentrations. However, trace contaminating of PCNB, eg, hexachlorobenzene (HCB) and pentachlorobenzene (PCB) accumulated in tissues to a significant degree. Tetrachloronitrobenzene, another contaminant of PCNB, was found in only the adipose and cardiac tissues following the feeding of 1,000 ppm PCNB. Storage of PCNB, HCB, and PCB in adipose and other tissues was linearly related to the amount of PCNB in the ration. 相似文献
