The mechanism of Marek's disease (MD) vaccination to prevent the lymphoproliferative disease in chickens is not well understood. It is generally recognized that vaccination prevents disease, including the induction of T-cell tumors, but it does not prevent the pathogenic virus from infecting and replicating in the vaccinated host, nor does it prevent bird to bird spread of the oncogenic virus. The stage at which the vaccinated immune system intervenes in the process from infection to the induction of tumors remains obscure. Using a transplantable tumor induced by the Md5 strain of MD virus (MDV), we show that CVI988 vaccination does not prevent the induction of transplantable tumors in the 15I(5) x 7(1) chicken line. A monoclonal tumor with a V beta 1 T-cell receptor spectratype of 207 base pairs was used to follow the transplantable tumor in serial passages in vivo. This transplantable tumor could be passed in vaccinated birds. The length of time between vaccination and challenge (5 to 12 days) had little or no influence on the ability to transfer the tumor. There was variability in the manifestation of the disease produced by the transplanted tumor. Some chickens presented as normal but were still capable of transmitting the transplanted tumor to newly vaccinated recipients via their blood. This indicates that some chickens can control, but not eliminate, the tumor. The variables inducing health or disease in the challenged chickens remain obscure, but environmental or other factors likely depress the immune system allowing the tumor to overwhelm the immune system. 相似文献

The purposes of this retrospective study were to assess the prevalence of gallbladder sludge (GBS) in a population of cats presented for abdominal ultrasound in a teaching hospital and to determine its association with increased serum alanine aminotransferase (ALT), alkaline phosphatase (ALP), and total bilirubin (TB). Gallbladder sludge was detected in 152 (14%) of the cats undergoing abdominal ultrasound between 2004 and 2008. This population was compared to a control group of 32 cats without GBS. Alanine aminotransferase, ALP, and TB mean values were significantly higher in cats with GBS than in controls (P ≤ 0.0005) and odds for increased values in cats with GBS were 4.2 [95% confidence interval (CI): 1.6 to 11.0], 9.5 (95% CI: 2.2 to 41.7), and 4.1 (95% CI: 1.5 to 11.5), respectively (P ≤ 0.007). In conclusion, GBS is an uncommon ultrasonographic finding in cats that is predictive of increased liver enzymes and TB. More studies are needed to establish potential links between GBS and hepatobiliary disease in cats. 相似文献

Marek's disease (MD) is a disease of chickens that occurs worldwide and has serious economic consequences. MD can present as one of several forms, with the most commonly occurring forms being the lymphoproliferative diseases. Under experimental conditions, an early mortality syndrome has been recognized following infection by some but not all strains of MD virus (MDV). This is the first report of a confirmed case of mortality due to naturally occurring MDV infection in 1-week-old, nonvaccinated, chickens. Necrotizing lesions were observed in the bursa of Fabricius, lung, duodenum, jejunum, and proventriculus, and large intranuclear inclusion bodies were a striking feature in tissues with lesions in all birds. Immunohistochemical staining for the pp38 protein of MDV revealed abundant pp38 antigen in the affected tissues, confirming the presence of MDV within the lesions. PCR yielded an amplicon with 97% homology to the meq gene of MDV. No evidence of co-infection by either of the immunosuppressive agents chicken anemia virus and infectious bursal disease virus was detected. 相似文献

Prescribed burning, the planned application of fire to reduce the hazards of wildland fuels, is coming into wider use in southern California chaparral. Soil was heated to various temperatures in factorial combination with various soil moistures to determine the direct effects of fire and soil moisture on the survival of microorganisms in chaparral soil. For fungi, mild heating increased germination of dormant forms yielding significantly higher counts than those in unheated soil. With increasing temperatures, microbe populations showed an exponential decrease. For heterotrophic soil bacteria, this decrease was summarized as a function of soil moisture and temperature. Microbial groups differed significantly in sensitivity to temperature: fungi > nitrite oxidizers > heterotrophic bacteria. Physiologically-active populations in moist soil were significantly more sensitive than were dormant populations in dry soil. The mathematical model presented shows qualitatively that more of the microbial biomass will be killed when the soil is moderately moist—as during prescribed burning—than when it is dry. Mineralization of killed microbial biomass in soil and release of plant nutrients may partially explain the increased plant growth and reduced response to fertilizer at burned sites. 相似文献

Organic nitrogen (N) uptake, rather than solely inorganic N (DIN), is considered a significant pathway for plant nutrition, especially in arctic, alpine and boreal ecosystems. Assays of plant-available N in these ecosystems might therefore be improved with measures of dissolved organic N (DON). We examined DON and DIN abundance from an in situ 5-week incubation across plant associations that represent the widest range in site potential in southern boreal forests of British Columbia, Canada. The supply of N from forest floors and mineral soils (20 cm depth) was measured separately and then combined (kg ha−1) to facilitate comparisons of sites. DON was the predominant form of extractable N, and was increasingly supplemented, rather than replaced, by NH4+ and NO3− on productive sites. The amount of DIN produced in the soils was very low, perhaps too small to support forest needs, and the correlation of DIN to asymptotic stand height (a measure of site potential) was significant but nonlinear. The combined amount of DON+DIN was considered a more effective index of plant-available N because it was strongly significant as a linear correlation to stand height and more typical of annual forest N uptake. The relative shift in N forms, from a predominance of DON to progressively greater ratios of DIN:DON, was consistent with the current paradigm of N forms across gradients of N availability, although the actual amounts of DON increased, rather than decreased, with site potential. Based on this, we suggest organic N uptake has the potential to contribute to plant nutrition across the entire productivity gradient of soils in southern boreal forests. Although other N indices were effective in characterizing forest productivity, a combined assay of DON+DIN production could provide new insights into functional differences in plant-available N. 相似文献

Light synchronizes mammalian circadian rhythms with environmental time by modulating retinal input to the circadian pacemaker-the suprachiasmatic nucleus (SCN) of the hypothalamus. Such photic entrainment requires neither rods nor cones, the only known retinal photoreceptors. Here, we show that retinal ganglion cells innervating the SCN are intrinsically photosensitive. Unlike other ganglion cells, they depolarized in response to light even when all synaptic input from rods and cones was blocked. The sensitivity, spectral tuning, and slow kinetics of this light response matched those of the photic entrainment mechanism, suggesting that these ganglion cells may be the primary photoreceptors for this system. 相似文献

Statistical tools are discussed for the analysis of data collected from tractor guidance systems. The importance of both accuracy and precision is discussed, and statistical tools for analysis are considered which incorporate important features of the data. In particular, accuracy is modelled using a generalized least squares model incorporating autocorrelation, and variances (inverse of precision) using a gamma generalized linear model. The methods are applied to data collected during an experiment conducted with a Trimble receiver used with a Beeline tractor guidance system. Three different scenarios are considered, then compared: a tractor simulating ploughing a field; the tractor pulling a plough with the receivers on the tractor; the tractor pulling a plough with the Trimble receiver on the plough. The change in the precision and accuracy between the scenarios is discussed. Data were recorded over repeated swaths for each scenario. After discussing specific statistical techniques for analysis of this type of data, the collected data are analysed; major conclusions are: The data from the Trimble receiver showed evidence of autocorrelation in the offsets; the plough recorded a variance about three times that recorded by the tractor.This research was completed while the second author was at the University of Southern Queensland. 相似文献

Measurements from the Near Earth Asteroid Rendezvous (NEAR)-Shoemaker Laser Rangefinder (NLR) indicate that asteroid 433 Eros is a consolidated body with a complex shape dominated by collisions. The offset between the asteroid's center of mass and center of figure indicates a small deviation from a homogeneous internal structure that is most simply explained by variations in mechanical structure. Regional-scale relief and slope distributions show evidence for control of topography by a competent substrate. Impact crater morphology is influenced by both gravity and structural control. Small-scale topography reveals ridges and grooves that may be generated by impact-related fracturing. 相似文献

Disposable plasticware such as test tubes, pipette tips, and multiwell assay or culture plates are used routinely in most biological research laboratories. Manufacturing of plastics requires the inclusion of numerous chemicals to enhance stability, durability, and performance. Some lubricating (slip) agents, exemplified by oleamide, also occur endogenously in humans and are biologically active, and cationic biocides are included to prevent bacterial colonization of the plastic surface. We demonstrate that these manufacturing agents leach from laboratory plasticware into a standard aqueous buffer, dimethyl sulfoxide, and methanol and can have profound effects on proteins and thus on results from bioassays of protein function. These findings have far-reaching implications for the use of disposable plasticware in biological research. 相似文献
