Due to its remarkable sediment pollution caused by past industrial activities, the Mediterranean coastal area facing the town of Cogoleto (near Genoa) has been declared of “national interest”. Seven sediment cores were analysed to provide information on history and trend of heavy metal inputs, with a particular focus on Cr. Grain-size compositions and 210Pb chronologies account for a strong influence of terrigenous inputs near the coast and the mouth of the Lerone Creek (draining the industrial settlement), with sand contents and accumulation rates varying from 40.3% and 0.7 cm year?1 near shore to 2.5% and 0.1 cm year?1 offshore. Heavy metal concentrations are high, notably Cr and Ni, and to a lesser extent, Ag, Hg, Pb, Cu and Zn. Cr, that shows past values up to 3,642 µg g?1, presents only minor recent decreases. In the area, it has both natural and anthropogenic sources and their relative contributions were distinguished on the basis of Cr/Ni ratios. These are 1–1.5 in soils and rocks of the Lerone Creek catchment area but reach very high values (up to 10) in marine sediments due to the anthropogenic contribution. The anthropogenic influence is higher near the creek mouth, gradually decreases offshore and is negligible at the most distal site (ca. 2.9 km from the coast). 相似文献

The effects of chemical (acid-heating treatment) and enzymatic (microbial transglutaminase, TGase) modification (deamidation) of gluten proteins on their physicochemical and celiac disease-related properties were studied. Ammonia release, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and sample solubility analyses were employed to check the extent of gluten modification. Among different treatments achieved, the acid-heating treatment performed at 90 degrees C for 3 h induced gluten deamidation, paralleling an increase of gluten solubility without relevant proteolysis. Changes in the immunoreactivity of celiac IgA anti-gliadin antibodies (AGAs) to modified gluten proteins were detected by using a competitive indirect enzyme-linked immunosorbent assay method. Chemical deamidation by acid-heating treatment of gluten lowered IgA-AGA immunoreactivity. IgA-AGA immunoreactivity to gliadins was increased when they were submitted to TGase-catalyzed deamidation. The acid-heating treatment of gluten reduced its cytotoxic activity on human colon adenocarcinoma LoVo cell line. These results showed that chemical deamidation of gluten may be envisaged as a way to lower the potential risk for celiac people due to widespread use of gluten as a food additive. 相似文献

Sixty-five samples of propolis were collected from eleven regions of Cuba; methanolic extracts of propolis were prepared from all samples, and a classification method was developed using a combination of NMR, HPLC-PDA, and HPLC-ESI/MS techniques. The analysis of (1)H and (13)C NMR spectra and chromatographic profiles of all propolis extracts allowed the definition of three main types of Cuban propolis directly related to their secondary metabolite classes: brown Cuban propolis (BCP), rich in polyisoprenylated benzophenones, red Cuban propolis (RCP), containing isoflavonoids as the main constituents, and yellow Cuban propolis (YCP), probably with aliphatic compounds. Subsequently, the principal compounds of the brown and red types were characterized by HPLC-ESI/MS analysis. Instrumental techniques used are complementary; NMR was shown to be a quick and informative tool for the rapid analysis of crude propolis polar extracts and allowed the identification of the main class of secondary metabolites, while LC-PDA and LC-MS techniques were useful tools for qualitative and quantitative analysis of marker compounds of Cuban propolis. 相似文献

A one-step triplex-polymerase chain reaction (PCR)-based assay was developed to discriminate between three tuna species, Thunnus albacares, Thunnus obesus, and Katsuwonus pelamis, even in highly processed food samples such as canned or cooked tuna. Diagnostic nucleotides were identified by direct sequencing and alignment of part of the mitochondrial cytochrome b gene of 30 authenticated exemplars, which allowed us to evaluate intraspecific variation and the genetic distance between three tuna species. The assay relies on a one-step triplex-PCR reaction in which in a single tube species-specific amplification products are generated only in the presence of the correct template nucleic acid and the species of origin of the DNA is indicated by the distinctive size of the PCR product. The identification of tuna species can be performed with a good accuracy, low cost, and with potential automation for large-scale high-throughput screenings in small in-house laboratories. 相似文献

Seventeen Sustainable Development Goals (SDGs) were adopted by 193 Governments at the General Assembly of the United Nations in 2015 for achievement by 2030. These SDGs present a roadmap to a sustainable future and a challenge to the science community. To guide activities and check progress, targets and indicators have been and are still being defined. The soil science community has published documents that describe the primary importance of soil for SDGs addressing hunger, water quality, climate mitigation and biodiversity preservation, and secondary relevance of soil for addressing several other SDGs. Soil scientists only marginally participated in the SDG discussions and are currently only peripherally engaged in discussions on targets or indicators. Agreement on several soil‐related indicators has still not been achieved. Involvement of soil scientists in SDG‐based studies is desirable for both developing solutions and increasing the visibility of the soil profession. Inputs into policy decisions should be improved as SDG committee members are appointed by Governments. Possible contributions of soil science in defining indicators for the SDGs are explored in this paper. We advocate the pragmatic use of soil–water–atmosphere–plant simulation models and available soil surveys and soil databases where “representative” soil profiles for mapping units (genetically defined genoforms) are functionally expressed in terms of several phenoforms reflecting effects of different types of soil use and management that strongly affect functionality. 相似文献

Chemical composition of propolis depends on the specificity of the local flora at the site of collection and thus on the geographic and climatic characteristics of this place. This paper describes a comparative analysis of Cuban red propolis (CRP), Brazilian red propolis (BRP), and Dalbergia ecastophyllum exudates (DEE) by high-performance liquid chromatography with diode-array detection and tandem mass spectrometry. The aim of this study was to investigate the overall chemical profile and the botanical origin of red propolis and to suggest similarities and differences between samples collected in different tropical regions. Isoliquiritigenin (1), liquiritigenin and naringenin (2 and 17), isoflavones (3-4 and 16), isoflavans (5-7 and 18), and pterocarpans (8-13) were detected in CRP, BRP, and DEE, whereas polyisoprenylated benzophenones (PPBs) guttiferone E/xanthochymol (14a,b) and oblongifolin A (15) were detected only in BRP. Pigments responsible for the red color of DEE and red propolis were also identified as two C30 isoflavans, the new retusapurpurin B (19) and retusapurpurin A (20). PPBs and pigments were isolated and unambiguously characterized by 1D and 2D NMR analysis. These results show that red propolis samples from different tropical zones have a similar chemical composition. DEE is the main red propolis source, but the presence of PPBs in BRP suggests the contribution of different botanical sources for Brazilian samples. This chemical information is important for quality control of red propolis and its commercial products and for biological study. 相似文献

Dicotyledons cope with ion (Fe) shortage by releasing low-molecular-weight organic compounds into the rhizosphere to mobilize Fe through reduction and complexation mechanisms. The effects induced by these root exudates on soil mineralogy and the connections between Fe mobilization and mineral weathering processes have not been completely clarified. In a batch experiment, we tested two different kinds of organic compounds commonly exuded by Fe-deficient plants, i.e., three organic acids (citrate, malate, and oxalate) and three flavonoids (rutin, quercetin, and genistein), alone or in combination, for their ability to mobilize Fe from a calcareous soil and modify its mineralogy. The effect of root exudates on soil mineralogy was assessed in vivo by cultivating Fe-deficient and Fe-sufficient cucumber plants (Cucumis sativus L.) in a RHIZOtest device. Mineralogical analyses were performed by X-ray powder diffraction. The batch experiment showed that citrate and, particularly, rutin (alone or combined with organic acids or genistein) promoted Fe mobilization from the soil. The combinations of rutin and organic acids modified the soil mineralogy by dissolving the amorphous fractions and promoting the formation of illite. These mineralogical alterations were significantly correlated with the amount of Fe mobilized from the soil. The RHIZOtest experiment revealed a drastic dissolution of amorphous components in the rhizosphere soil of Fe-deficient plants, possibly caused by the intense release of phenolics, amino acids, and organic acids, but without any formation of illite. Both batch and RHIZOtest experiments proved that exudates released by cucumber under Fe deficiency concurred to the rapid modification (on a day-scale) of the mineralogy of a calcareous soil. 相似文献

Purpose
The peatland carbon store is threatened by climate change and is expected to provide positive feedback on air temperature. Most studies indicate that enhanced temperature and microbial activities result in a rise of dissolved organic carbon (DOC) as a consequence of higher peat decomposition. Few of them, however, have investigated the impact of in situ experimental warming on DOC response.Material and methods
We studied the response of DOC, dissolved organic nitrogen (DON), phenol oxidase, and fluorescein diacetate activities (FDA) to a 3-year in situ experimental warming using open-top chambers (OTCs) in a Sphagnum-dominated peatland.Results and discussion
No significant warming of soil was recorded, implying that the simultaneous decrease in DOC and DON and the rise in FDA at the depths of 25 and 40 cm were not caused by the direct effect of OTCs on water temperature, but might instead have been mediated by plant root exudates. The water chemistry suggests that DOC production was compensated by in situ mineralization. We hypothesize that an increased hydrolysis of organic matter (OM) was counterbalanced by the mineralization of dissolved organic matter (DOM) and that microorganisms preferentially used labile compounds originating from increased root exudates.Conclusions
This trade-off between production of DOC through hydrolysis and consumption in the process of mineralization shows (1) the limitation of using only DOC as an indicator of the sensitivity of peat decomposition to climate warming and (2) the need to improve our understanding of the indirect impact of root exudates. 相似文献180.

A general synthetic procedure leading to isotopomeric dihydro-2(3H)furanones (gamma-butyrolactones) containing two, four, or six deuterium atoms has been developed. The labeled dihydro-2(3H)furanones were synthesized in quantitative yield from the saturated diacid C4 (succinic) or unsaturated diacids C4 (fumaric, maleic, or acetylendicarboxylic) in the presence of Ru4H4(CO)8(PBu3)4 using a deuterium pressure of 180 bar at 180 degrees C. This methodology was applied to the total synthesis of a hexadeuterated matairesinol lignan: The 3,4-bis[[3-methoxy-4-(phenylmethoxy)phenyl]methyl]dihydro-2(3H)furanone-[7,7',8,8',9',9'-D6] (benzyl-protected matairesinol-D6) was fully characterized. 相似文献
