Immunomodulatory properties have been described for Fasciola hepatica excretory-secretory products (FhESP), with their interaction with the innate immune cells being crucial during the early stages of infection. Previously, we demonstrated that FhESP induce eosinophil apoptosis. In this work, the ability of FhESP to induce apoptosis of peritoneal macrophages was evaluated. These parasite products were observed to induce apoptosis in peritoneal macrophages stimulated in vitro with FhESP, as well as in cells recovered from infected mice. The ability of FhESP to modify the viability of macrophages by apoptosis induction may constitute a crucial event for extending its survival in the host.  相似文献

Calcium (Ca) content of fruit is related to fruit quality. However, total content alone is insufficient to judge fruit quality; calcium partitioning and chemical form are also important. There are multiple validated methods for determining total calcium content. However, methodologies to measure calcium fractions are not widely known and tend to be complex, affecting their accuracy. Here, based on previous work, we aim to adapt and validate a method for the extraction of the various forms of calcium in fruits. The main modification was freezing samples to minimize the loss of sample material in the procedure, reducing the centrifugation time and speed, and the addition of nitric acid (HNO3) to improve digestion and eliminate interference from the matrix. An experiment with table grape berries cv. Thompson seedless in harvest and Postharvest (35 days) was carried out. The results of the total calcium were compared with the conventional methodology of calcination and inductively coupled plasma (ICP) measurement. We detected no and no significant differences between the two tested methods. Electron Microscope - Energy-dispersive X-ray spectroscopy analysis (SEM-EDS) and the presence of calcium oxalates were verified.  相似文献

Acer opalus subsp. granatense is an endemic endangered tree with a wide but fragmented distribution in the Mediterranean mountains. The persistence of its small populations often depends on just a few adults, and consequently is highly vulnerable to factors limiting recruitment. In this paper, we examined the regeneration status of this maple in 16 populations throughout its whole geographical distribution in the Iberian Peninsula. Age and size structures were analysed as indicators of the viability of the species. Additionally, we studied the effects of herbivory by ungulates and the role of shrubs as nurse plants in maple regeneration dynamic. Our results show that A. opalus subsp. granatense has active recruitment throughout its range in the Iberian Peninsula. Shrubs served as the main microsites for recruitment, not only for early establishment but also for long-term survival. However, ungulates heavily damaged maple saplings in all locations and microhabitats. A direct consequence of herbivory is the uncoupling of age and size structures, saplings becoming older but not higher, possibly affecting population turnover in the long term. We suggest that the conservation of the small populations of Acer opalus subsp. granatense heavily depends on the control of herbivory pressure throughout the maple distribution area.  相似文献

Atmospheric polycyclic aromatic hydrocarbons (PAHs) were determined in particulate matter (PM10) collected in a suburban area with industrial and vehicular emissions in the Metropolitan Area of Rio de Janeiro City (Brazil). A total of 22 samples were collected between March and August 2005 by means of a high volume PM10 sampler. The particulate matter contained in the filters was extracted ultrasonically with dichloromethane. The extracts were later analyzed by gas chromatography coupled to a mass spectrometer (GC/MS). The individual concentrations of PAHs ranged between the detection limit and 0.386 ng m?3. The PAHs concentrations observed in this study were towards the lowest end of the range of values reported for other European locations and also lower than values obtained for South America. PAHs concentrations and molecular ratios showed that light cars seem to be the main contributors to PM10 emissions, but diesel vehicles are clearly minor emission sources and industrial contributions should not be disregarded until more data are obtained.  相似文献

Little is known on the hydrological behavior of the volcanic ash soils, which are characterized by extremely high porosities and hydraulic conductivities. In this study the occurrence and hydrological effects of water repellency were investigated at a plot scale for different types of land use and volcanic soils in Mexican volcanic highlands from Michoacan, Mexico: [1] fir, pine and oak mixed forest soils developed from lavas, [2] soils developed from volcanic ashes and pyroclastic sediments under sparse fir, pine and oak forest and shrubland, [3] pine and oak forested soils developed from lavas and pyroclastic sediments, and [4] bare soils on recent ash sediments in plain surfaces. Soil water repellency was assessed using the water drop penetration time test and rainfall simulations were performed on circular plots (50 cm in diameter) during 30 min and at an intensity of 90 mm h− 1 in order to study the hydrological response of each area. The return period for storms with a similar intensity in the area is 10 years. The shape and depth of the wetting front after simulated rainfall was also analyzed. Soil water repellency showed a high variability among the different studied zones. Organic matter content, soil texture and acidity were the most important factors for developing hydrophobicity. A wide range of soil water repellency classes (hydrophilic to severely water-repellent soils) has been found in soils under dense fir, pine and oak mixed forests or shrubland, while inexistent or slight water repellency has been observed in soils under sparse forest or at bare ash-covered areas. At a plot scale, marked differences in the hydrological behavior of the studied land use and soil zones were observed after the rainfall simulations. Soil water repellency contributes to fast ponding and runoff generation during the first stages of rainstorms. Runoff was enhanced in water-repellent forested soils (average runoff coefficients between 15.7 and 19.9%), in contrast to hydrophilic or slightly water-repellent soils, where runoff rates were lower (between 1.0 and 11.7%). Shallow and irregular wetting fronts were observed at water-repellent zones, reducing the soil water storage capacity. The implications of soil water repellency in soil hydrology and erosion risk in the area shed light on the soil hydrology of the studied ecosystems, and can contribute to develop better management policies.  相似文献

Despite the wide availability of liquid herbal extracts using mixtures of alcohol, glycerin, and water, or glycerin and water as solvents, no data on the chemical composition of such extracts is readily available. In this study, the amount and the stability of the major saponins in Panax quinquefolius root extracts, made either with 50% (v/v) aqueous ethanol, a mixture (v/v/v) of 20% ethanol, 40% glycerin, and 40% water, or with 65% (v/v) aqueous glycerin, were evaluated by HPLC-UV analysis. The amount of total saponins was highest in the 50% aqueous ethanol extract (61.7 +/- 0.1 mg/g dry root), although similar to the ethanol-glycerin-water extract (59.4 +/- 0.5 mg/g dry root). Saponins were significantly lower in the 65% aqueous glycerin extract (51.5 +/- 0.2 mg/g dry root). Interestingly, the amounts of individual saponins were quite variable depending on the solvent. This is in part due to enzymatic cleavage of ginsenosides in the glycerin containing extracts during the maceration process. Storage of the extracts at 25 degrees C over the period of a year led to a 13-15% loss of saponins with all three types of extractions.  相似文献

The aim of this work was to investigate the influence of the organic matter on copper and lead adsorption in soils with different physiochemical and mineralogical attributes. Suspensions (pH 6.0) of a Latosol, a Neosol and a Vertisol containing increasing amounts of copper or lead were used to obtain sorption isotherms while identical experiments were carried out with the soils previously treated with H2O2 to remove organic matter (OM). For the undisturbed soils, L-type and H-type isotherms were predominant for copper and lead respectively, showing that lead interacts more strongly with adsorption sites. For both metals, the non-linear Freundlich adsorption model revealed higher concentration of adsorption sites for Vertisol due to 2:1 clays. For the OM-removed soils, C-type isotherms were observed for copper with the permanence of less stable and more homogeneous sites. For this metal, a high correlation (R2 = 0.997) was observed between the decrease of adsorbent sites and the loss of organic carbon, evidencing the central role of organic matter on copper complexation, while lead may be able to interact efficiently with both organic matter and soil minerals.  相似文献

Mushroom tyrosinase exhibits catalase activity with hydrogen peroxide (H(2)O(2)) as substrate. In the absence of a one-electron donor substrate, H(2)O(2) is able to act as both oxidizing and reducing substrate. The kinetic parameters V(max) and K(m) that characterize the reaction were determined from the initial rates of oxygen gas production (V(0)(O)()2) under anaerobic conditions. The reaction can start from either of the two enzyme species present under anaerobic conditions: met-tyrosinase (E(m)) and deoxy-tyrosinase (E(d)). Thus, a molecule of H(2)O(2) can reduce E(m) to E(d) via the formation of oxy-tyrosinase (E(ox)) (E(m) + H(2) <==> O(2) right harpoon over left harpoon E(ox)), E(ox) releases oxygen into the medium and is transformed into E(d), which upon binding another molecule of H(2)O(2) is oxidized to E(m). The effect of pH and the action of inhibitors have also been studied. Catalase activity is favored by increased pH, with an optimum at pH = 6.4. Inhibitors that are analogues of o-diphenol, binding to the active site coppers diaxially, do not inhibit catalase activity but do reduce diphenolase activity. However, chloride, which binds in the equatorial orientation to the protonated enzyme (E(m)H), inhibits both catalase and diphenolase activities. Suicide inactivation of the enzyme by H(2)O(2) has been demonstrated. A kinetic mechanism that is supported by the experimental results is presented and discussed.  相似文献

Real-time polymerase chain reaction is currently being used for the identification and quantification of plant and animal species as well as microorganisms in food or feed samples based on the amplification of specific sequences of low copy genes. We report here the development of a new real-time PCR method for the detection and quantification of the pea (Pisum sativum) based on the amplification of a specific region of the legS gene. The specificity was evaluated in a wide range of plant species (51 varieties of Pisum sp., and 32 other plant species and varieties taxonomically related or nonrelated). The method allows the detection and quantification of as low as 21.6 pg of DNA, which corresponds to 5 haploid genome copies. The system has been shown to be sensitive, reproducible and 100% specific for the rapid detection and quantification of pea DNA in processed food and feed samples, being therefore suitable for high-throughput analysis.  相似文献

Currently, the use of by-products generated from fish processing is a relevant issue for the sustainable development of the fisheries. Fins are the main commercialized product of cartilaginous species. However, the livers contain a high content of polyunsaturated fatty acids (PUFAs), mainly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). These fatty acids have been exhaustively studied due to their remarkable benefits to human health. In the present work, different methodologies were investigated to extract oil from the livers of Zearaja flavirostris and Atlantoraja castelnaui. Enzyme-assisted processes, cold extractions, and high temperatures were studied. The extraction with enzymes was the most efficient in both species. The oils obtained presented physicochemical and quality parameters suitable for its refinement and commercialization. Palmitic (C16:0) and oleic acid (C18:1) were the most abundant saturated fatty acids (SFAs) and monounsaturated fatty acids (MUFAs), respectively. Both species showed similar content of DHA; however, the oil of Z. flavirostris had twice as much EPA than A. castelnaui. The results suggest that the livers of the species studied can be used as a source of valuable oils, rich in PUFAs, as a viable alternative for the integral use of these resources.  相似文献
