Clostridium difficile, associated with a wide spectrum of diseases in humans, as well as in several animal species, is an important cause of colitis in adult horses and foals. The aim of this study was to investigate by toxin gene profile and PCR-ribotyping the molecular characteristics of 14 C. difficile strains isolated from 42 faeces of healthy horses. Both toxin genes, tcdA and tcdB, were present in only 1 isolate (7.1%). Six isolates (42.9%) demonstrated tcdA/tcdB+ genotype, and seven isolates (50.0%) were tcdA−/tcdB−. All strains were binary toxin genes negative (cdtA−/cdtB−). The PCR-positive strains, except for the tcdA+/tcdB+ isolate, tested negative for, in vitro, A and/or B toxins production by EIA. Eleven distinct ribotypes were observed.In conclusion, C. difficile can be present in the normal intestinal flora of healthy adult horses, in addition to foals. These animals could therefore play an important role as potential reservoirs of toxigenic strains.  相似文献

Objectives— To compare in vitro monotonic biomechanical properties of an axial 3‐hole, 4.5 mm narrow dynamic compression plate (DCP) using 5.5 mm cortical screws in conjunction with 2 abaxial transarticular 5.5 mm cortical screws inserted in lag fashion (DCP‐TLS) with 3 parallel transarticular 5.5 mm cortical screws inserted in lag fashion (3‐TLS) for the equine proximal interphalangeal (PIP) joint arthrodesis. Study Design— Paired in vitro biomechanical testing of 2 methods of stabilizing cadaveric adult equine forelimb PIP joints. Sample Population— Cadaveric adult equine forelimbs (n=15 pairs). Methods— For each forelimb pair, 1 PIP joint was stabilized with an axial 3‐hole narrow DCP (4.5 mm) using 5.5 mm cortical screws in conjunction with 2 abaxial transarticular 5.5 mm cortical screws inserted in lag fashion and 1 with 3 parallel transarticular 5.5 mm cortical screws inserted in lag fashion. Five matching pairs of constructs were tested in single cycle to failure under axial compression, 5 construct pairs were tested for cyclic fatigue under axial compression, and 5 construct pairs were tested in single cycle to failure under torsional loading. Mean values for each fixation method were compared using a paired t‐test within each group with statistical significance set at P<.05. Results— Mean yield load, yield stiffness, and failure load under axial compression and torsion, single cycle to failure, of the DCP‐TLS fixation were significantly greater than those of the 3‐TLS fixation. Mean cycles to failure in axial compression of the DCP‐TLS fixation was significantly greater than that of the 3‐TLS fixation. Conclusion— The DCP‐TLS was superior to the 3‐TLS in resisting the static overload forces and in resisting cyclic fatigue. Clinical Relevance— The results of this in vitro study may provide information to aid in the selection of a treatment modality for arthrodesis of the equine PIP joint.  相似文献

The effects of grain texture on pastamaking and breadmaking quality were studied in three F8 soft-textured durum wheat lines (SDLs) containing wild-type alleles Pina-D1a and Pinb-D1a as compared with their hard durum sister lines (HDLs). SDLs homozygous for a small 5DS segment, less than 14.4 cM in size, accumulated puroindolines A (Pin-A) and B (Pin-B) and showed SKCS values (19.9-23.6) significantly lower than those (72.6-76.8) of their hard-textured counterparts lacking Pin-A and Pin-B. In addition, SDLs exhibited approximately 24% higher flour extraction rates compared with HDLs. Reducing the kernel hardness decreased farinograph water absorption, dough tenacity (P) and, accordingly, alveograph P/L ratio, but increased farinograph stability, mixing tolerance and dough extensibility (L). Spaghetti cooking quality, as determined by the sensory judgment of firmness, stickiness and bulkiness, was unaffected by the kernel hardness, whereas the loaf volume exhibited a 10% increase associated with kernel softening. Flour and semolina, but not spaghetti, from SDLs showed a substantial reduction in yellowness (b*) and brownness (100 − L*) likely due to their finer particle size compared with HDLs. Alleles Pina-D1a and Pinb-D1a may offer new perspectives for breeding dual purpose (pasta and bread) durum wheat varieties.  相似文献

The contribution of biofilm to water quality and as a food source for the culture of the freshwater shrimp Neocaridina heteropoda heteropoda was assessed in indoor aquaria using a zero water exchange system. Two successive phases were conducted to evaluate biofilm development on different substrates (polyethylene net: PN, plastic bottles: PET, agrovelo: AV) and the effect of biofilm to shrimps culture. The biofilm grown on all substrates helped to keep a good water quality by the uptake of nitrogen compounds and the production of high levels of dissolved oxygen associated to the proliferation of autotrophic microorganisms. High survival, reproduction and hatching of shrimps were achieved in all groups mainly associated with good water quality. Final biomass of the shrimps was significant higher for PN and AV groups; while specific growth rate and the levels of lipids of the shrimps yielded similar values for all treatments indicating that the three substrates allowed the growth of a biofilm that resulted in a healthy food source with similar nutritional value for shrimps. The results show that the production of N. heteropoda heteropoda could be successfully conducted by a biofilm‐based culture system with no water exchange, and thus contributing to a better water use. All materials tested were suitable substrates for biofilm growth, though AV and PET could reduce significantly production costs when compared to the PN. Moreover, by the recycling and reuse of waste materials (such as plastic bottles) could contribute to the development of a responsible, sustainable and environmentally friendly culture method.  相似文献

The effects of nutraceuticals in improving growth and immune response of Nile tilapia in cool water were investigated. Fish were reared in two different environments: warm water (28°C) and cool water (16°C). Fish reared in warm water (control) were provided with a basal commercial diet, while fish reared in cool water were provided with either the basal commercial diet (cool water control) or diets supplemented with nutraceuticals (either phosphatidylcholine or β-carotene). Experiments were conducted over an 8-week period. Fish held in warm water had significantly higher growth (p < 0.05), but condition factor, blood hematocrit, plasma glucose, and phagocytic capacity of macrophage cells were similar to fish held in cool water. Within the cool water groups (basal vs. supplemented diet), fish did not show any significant difference in condition factor, blood hematocrit, and phagocytic capacity of macrophage cells. Furthermore, there were no significant differences in glucose levels until the eighth week. Collectively these data indicate that a nutraceutical supplementation to the basal diet was not significantly beneficial. Despite slower growth, fish reared in cool water remained generally healthy. The observation that phagocytic capacity of macrophage cells was not significantly different suggests that disease resistance of tilapia grown in cool water may be comparable to those reared in warm water. Thus, our experiments suggest the feasibility of stocking or rearing tilapia in water temperatures as low as 16°C during cool weather, without need for supplementation to basal commercial feed.  相似文献

BACKGROUND: Glyphosate tolerance is a dominant trait in modern biotech crops. RESULTS: A gene encoding a glyphosate-tolerant EPSP synthase (aroA(1398)) from bacterial strain ATX1398 was cloned and characterized. The protein is initiated at a GTG translational start codon to produce a protein that provides robust glyphosate resistance in Escherichia coli (Mig) Cast & Chalm. The aroA(1398) protein was expressed and purified from E. coli, and key kinetic values were determined (K(i) = 161 microM; K(m)(PEP) = 11.3 microM; k(cat) = 28.3 s(-1)). The full-length enzyme is 800-fold more resistant to glyphosate than the maize EPSP synthase while retaining high affinity for the substrate phosphoenol pyruvate. To evaluate further the potential of aroA(1398), transgenic maize events expressing the aroA(1398) protein were generated. T(0) plants were screened for tolerance to glyphosate sprays at 1.3x commercial spray rates, and T(1) plants were selected that completely resisted glyphosate sprays at 1x, 2x and 4x recommended spray rates in field trials. CONCLUSION: These data suggest that aroA(1398) is a suitable candidate for conferring glyphosate tolerance in transgenic crop plants.  相似文献

The Northern Adriatic Sea is the area of the Mediterranean Sea where eutrophication and episodes related to harmful algae have occurred most frequently since the 1970s. In this area, which is highly exploited for mollusk farming, the first occurrence of human intoxication due to shellfish consumption occurred in 1989, nearly 10 years later than other countries in Europe and worldwide that had faced similar problems. Until 1997, Adriatic mollusks had been found to be contaminated mostly by diarrhetic shellfish poisoning toxins (i.e., okadaic acid and dinophysistoxins) that, along with paralytic shellfish poisoning toxins (i.e., saxitoxins), constitute the most common marine biotoxins. Only once, in 1994, a toxic outbreak was related to the occurrence of paralytic shellfish poisoning toxins in the Adriatic coastal waters. Moreover, in the past 15 years, the Adriatic Sea has been characterized by the presence of toxic or potentially toxic algae, not highly widespread outside Europe, such as species producing yessotoxins (i.e., Protoceratium reticulatum, Gonyaulax spinifera and Lingulodinium polyedrum), recurrent blooms of the potentially ichthyotoxic species Fibrocapsa japonica and, recently, by blooms of palytoxin-like producing species of the Ostreopsis genus. This review is aimed at integrating monitoring data on toxin spectra and levels in mussels farmed along the coast of the Emilia-Romagna region with laboratory studies performed on the species involved in the production of those toxins; toxicity studies on toxic or potentially toxic species that have recently appeared in this area are also reviewed. Overall, reviewed data are related to: (i) the yessotoxins producing species P. reticulatum, G. spinifera and L. polyedrum, highlighting genetic and toxic characteristics; (ii) Adriatic strains of Alexandrium minutum, Alexandrium ostenfeldii and Prorocentrum lima whose toxic profiles are compared with those of strains of different geographic origins; (iii) F. japonica and Ostreopsis cf. ovata toxicity. Moreover, new data concerning domoic acid production by a Pseudo-nitzschia multistriata strain, toxicity investigations on a Prorocentrum cf. levis, and on presumably ichthyotoxic species, Heterosigma akashiwo and Chattonella cf. subsalsa, are also reported.  相似文献
