Antiproliferation effects of 188Re labeled monoclonal antibody on prostate cancer cells in vitro

AIM: To investigate the effect of 188Re labeled monoclonal antibody on prostatic specific membrane antigen 7E11C5.3,radioimmunotherapy for the treatment of human prostate cancer cell line LNCaP in vitro.METHODS: 188Re-7E11C5.3 was prepared by direct 2-mercaptoethanol reduction method.Labeling efficiency and radiochemical purity was measured by paper chromatography.Immunoreactive fraction was determined by linear extrapolation.Cytotoxicity to LNCaP cells was determined by MTT assay.RESULTS: The labeling yield of 188Re-7E11C5.3 was (93.16±2.18)%,the radiochemical purity was (95.62±0.48)%,and the immunoreactive fraction was (74.86±1.86)%.The inhibitory effect of 188Re-7E11C5.3 on cell proliferation of LNCaP cells was significantly higher than that of 188Re-mIgG or 188ReO-4.The 50% inhibitory doses (IC50) of 188Re-7E11C5.3,188Re-mIgG,and 188ReO-4 were (23.38±3.73)×107 Bq/L,(59.21±8.02)×107 Bq/L and (68.89±10.91)×107 Bq/L,respectively.CONCLUSION: 188Re-7E11C5.3 can effectively inhibit the growth of in vitro cultured prostate cancer cells and shows much potential for prostate cancer radioimmunotherapy.     

Thrombolytic properties of a conjugate of liposomal urokinase with a monoclonal antibody specific for cross-linked fibrin in the model of rabbit artery thrombosis

AIM: To observe the targeting thrombolytic effect of a monoclonal antibody specific for cross-linked fibrin connected with liposomal urokinase（UK） in the model of rabbit artery thrombosis.METHODS: Preparation of thrombolytic solutions: with the method of controllable dialysis eradicator in liposomat,empty liposomes,liposomally entrapped urokinase and liposomally entrapped urokinase linked with D-dimer antibody were made.Experiment in vivo: a rabbit thrombosis model of abdominal aorta was induced by ferric chloride.When the blood pressure fall to the lowest,5 different solutions were separately imported (PBS,maximal-level UK,Ab/Lip/UK,Ab/UK-Lip,Ab-UK-Lip) and observed for 40 min continuously.RESULTS: The varieties of 5 groups blood pressures were analyzed with q-test.Significant differences were observed among Ab-UK-Lip group,maximal-level UK group and others (P<0.01).There were marked differences between Ab-UK-Lip group and others in wet weight of thrombuses (P<0.05).However,the quantity of UK in Ab-UK-Lip group was 1/3 of that in maximal-level of UK group.CONCLUSION: The thrombolytic effect on Ab-UK-Lip group is similar to that on maximal-level of UK group,but the dosage of UK is less,and the side-effect is light.This shows,as a homing-device of liposomal urokinase,the D-dimer antibody has targeting effect and the agent of Ab-UK-Lip will be attached importance by-and-by.     

Significance of expression of extracellular matrix metalloproteinase inducer and endothelin converting enzymes in lymphoid metastasis and prognosis in non-small-cell lung carcinoma

AIM: To investigate the expression of extracellular matrix metalloproteinase inducer (EMMPRIN) and endothelin converting enzymes (ECE) in non-small-cell lung carcinoma (NSCLC), and their relationship with lymphoid metastasis and prognosis.METHODS: Expression of EMMPRIN and ECE in 77 cases of patients with NSCLC was detected immunohistochemically.The relationship of the expression of EMMPRIN and ECE with tumor size, histological type, differentiation, lymphoid metastasis, clinical stage, and prognosis were analyzed.RESULTS: The expressive rate of EMMPRIN and ECE was 66% and 45%, respectively.The expression of EMMPRIN and ECE was associated positively with lymphoid metastasis (both P<0.01, r=0.371 and 0.467, respectively), and inversely with survival time (P<0.01 and P<0.01).No relationship was found in the expression of EMMPRIN and ECE with tumor size, histological type, and differentiation (P>0.05).The expression of EMMPRIN was associated with the expression of HGF in NSCLC (P<0.05, r=0.243).CONCLUSION: The expression of EMMPRIN and ECE is associated with lymphoid metastasis and prognosis in NSCLC.Overexpression of EMMPRIN and ECE implies infavourable prognosis in NSCLC.

Meteorus rubens—Steinernema carpocapsae interaction, field studies

The entomogenous nematode,Steinernema carpocapsae was applied in three forms in two fields cultivated with cotton and tomato, under each cotton seedling or tomato plant: a bait like form, a suspension and in irrigation water. High concentrations of 2000 and 1000 IJS/seedling induced 100% mortality (after 5 days), while lower concentrations showed the same result but after 15 days or later. Under all doses of nematode, bait form application was more effective than suspension or irrigated water.
