Secretory and membrane proteins carry amino-terminal signal sequences that, in cotranslational targeting, are recognized by the signal recognition particle protein SRP54 without sequence specificity. The most abundant membrane proteins on Earth are the light-harvesting chlorophyll a/b binding proteins (LHCPs). They are synthesized in the cytoplasm, imported into the chloroplast, and posttranslationally targeted to the thylakoid membrane by cpSRP, a heterodimer formed by cpSRP54 and cpSRP43. We present the 1.5 angstrom crystal structure of cpSRP43 characterized by a unique arrangement of chromodomains and ankyrin repeats. The overall shape and charge distribution of cpSRP43 resembles the SRP RNA, which is absent in chloroplasts. The complex with the internal signal sequence of LHCPs reveals that cpSRP43 specifically recognizes a DPLG peptide motif. We describe how cpSPR43 adapts the universally conserved SRP system to posttranslational targeting and insertion of the LHCP family of membrane proteins.  相似文献

Models for chemical mechanisms of hydrocarbon oxidation rely on spectrometric identification of molecular structures in flames. Carbonyl (keto) compounds are well-established combustion intermediates. However, their less-stable enol tautomers, bearing OH groups adjacent to carbon-carbon double bonds, are not included in standard models. We observed substantial quantities of two-, three-, and four-carbon enols by photoionization mass spectrometry of flames burning representative compounds from modern fuel blends. Concentration profiles demonstrate that enol flame chemistry cannot be accounted for purely by keto-enol tautomerization. Currently accepted hydrocarbon oxidation mechanisms will likely require revision to explain the formation and reactivity of these unexpected compounds.  相似文献

Spring-sown crops are expected to have a higher risk of drought during summer in the next decades in Central Europe due to expected climate change. Therefore, a two-year experiment was conducted under Pannonian growing conditions in Eastern Austria to investigate the effect of autumn- and spring-sowing of facultative wheat. Autumn-sowing of facultative wheat enhanced crop development, soil coverage, crop stand height, crop growth rate, and nitrogen (N) utilization efficiency during the vegetation period compared to spring-sowing; duration of growth stages was prolonged and crops were earlier ripe. In contrast, spring-sowing resulted in higher relative growth rates, higher N concentrations of aboveground dry matter, higher relative N uptake rates, and more mineral N in the soil. At harvest, grain yield and yield components ears m?2 and thousand kernel weight (TKW) were higher in autumn-sown than in spring-sown wheat, resulting thereby in an increased seed yield. Spring-sown wheat had higher N concentrations in grain and in straw. Anyhow, N yield was slightly higher with autumn-sowing due to the higher grain and straw yields. Grain and straw yield, plant stand height, ears m?2, and TKW were impaired in the second experimental year by a severe drought for both sowing dates as well as N concentrations and N yields of grain and straw, partial factor N use efficiency and N utilization efficiency. But the yield components harvest index, grains m?2, and grains ear?1 were strongly impaired with spring-sowing under drought conditions. Thus, autumn-sowing of wheat resulted in higher yield stability across both years, based on these yield components highlighting possible benefits of autumn-sowing with expected summer drought under climate change.  相似文献

Earthworms (Annelida: Oligochaeta) deposit several tons per hectare of casts enriched in nutrients and/or arbuscular mycorrhizal fungi (AMF) and create a spatial and temporal soil heterogeneity that can play a role in structuring plant communities. However, while we begin to understand the role of surface casts, it is still unclear to what extent plants utilize subsurface casts. We conducted a greenhouse experiment using large mesocosms (volume 45 l) to test whether (1) soil microsites consisting of earthworm casts with or without AMF (four Glomus taxa) affect the biomass production of 11 grassland plant species comprising the three functional groups grasses, forbs, and legumes, (2) different ecological groups of earthworms (soil dwellers—Aporrectodea caliginosa vs. vertical burrowers—Lumbricus terrestris) alter potential influences of soil microsites (i.e., four earthworms × two subsurface microsites × two AMF treatments). Soil microsites were artificially inserted in a 25-cm depth, and afterwards, plant species were sown in a regular pattern; the experiment ran for 6 months. Our results show that minute amounts of subsurface casts (0.89 g kg?1 soil) decreased the shoot and root production of forbs and legumes, but not that of grasses. The presence of earthworms reduced root biomass of grasses only. Our data also suggest that subsurface casts provide microsites from which root AMF colonization can start. Ecological groups of earthworms did not differ in their effects on plant production or AMF distribution. Taken together, these findings suggest that subsurface earthworm casts might play a role in structuring plant communities by specifically affecting the growth of certain functional groups of plants.  相似文献

Sorption of dissolved organic matter (DOM) by poorly crystalline minerals during their formation may protect large amounts of carbon in soils from mineralization. We investigated the bioavailability of carbohydrates in DOM and after co-precipitation with short-range ordered aluminosilicates. Carbohydrates originated from soil solutions collected in situ at two depths of a Dystric Cambisol, and from litter extracts. Quantification of substrate-specific degradability was achieved by the addition of β-glucosidase at an optimal concentration and subsequent determination of glucose release. Depending on DOM composition, 0.6–41.4 mg g−1 C−1 of glucose was enzymatically released from dissolved carbohydrates. Co-precipitated carbohydrates were partially accessible, resulting in a glucose release of 0.7–5.2 mg g−1 C−1. Restricted enzymatic depolymerization due to co-precipitation may contribute to accumulation of easily degradable substrates in soils.  相似文献

Background

Glyphosate (GLP) is a widely used herbicide with possible adverse effects on human health and the environment. In soils, GLP strongly adsorbs on clay-sized minerals, depending on pH, the amount of organic carbon, as well as the contents and properties of Al and Fe oxyhydroxides and clay minerals. Many clay-sized minerals have already been investigated regarding GLP adsorption behavior, but information on minerals commonly found in volcanic soils is still lacking.

Aim

The aim of this study was to investigate for the first time the pH-dependent adsorption of GLP on allophane and halloysite, typical minerals found in volcanic soils.

Methods

GLP adsorption was studied in batch experiments at three pH values (5, 6, and 7). Synthetic allophanes with two different initial Al:Si ratios (1.4 and 1.8) and a halloysite were used as adsorbents.

Results

The adsorption capacity (AC) increased with rising Al:Si ratio and decreasing pH. The AC of allophane was significantly higher than that of halloysite. GLP adsorption on allophane was larger than that reported for other clay minerals and Al and Fe oxyhydroxides, especially at low pH. The AC of halloysite was higher than reported for most other clay minerals.

Conclusion

Different mineral formation pathways in volcanic soils, notably the formation of halloysite versus allophanes, strongly affect the soils’ retention capacity for GLP. The high AC of allophanes may induce the low mobility of GLP in allophane-containing soils. Long-term use of GLP may accumulate the herbicide in these soils with potential effects on biodiversity and ecosystem services.  相似文献

Non-timber forest products (NTFPs) strongly contribute to livelihood security in the semi-arid tropics. There is increasing concern about the population status of NTFP-providing trees and therefore a need for their sustainable use. Thus, this study examines the impact of land-use type on the multipurpose baobab tree (Adansonia digitata L.) in Burkina Faso, combined with rates and patterns of bark- and leaf-harvesting, and their impact on fruit production. We compared stands in a protected area (W National Park of Burkina Faso) with those of surrounding communal area (fallows, croplands and villages) to obtain an indication on the status of the baobab population, to assess its harvesting tolerance and to estimate to what extent their actual use is sustainable. Our results reveal that land-use type has an impact on the population structure of the baobab. The size class distribution curve of park stands was inverse J-shape which indicates good rejuvenation, while the curve of fallows, croplands and villages stands was bell-shaped, indicating a lack of recruitment. However, a high number of seedlings were recorded in villages. Nearly all baobabs were pruned and debarked in villages, croplands and fallows while half of the individuals were harvested in the park. Most of the trees were pruned and debarked moderately. Debarking and pruning were slightly size specific. Pruning in interaction with tree-size had a significant impact on fruit production. In contrast, debarking had no effect on fruit production. We conclude that despite the land-use impact and the intense harvesting, baobabs are still well preserved in the communal area due to their longevity, extremely low adult mortality rates and traditional management practices. However, land-use intensifications may lead to increasing pressure on baobab populations in the future. Therefore, adapted management strategies are needed to guarantee the persistence of this important species and to avoid a shortage of baobab products.  相似文献

ABSTRACT: Staphylococcus aureus is one of the most important causal agents of bovine mastitis. Population studies on bovine Staphylococcus aureus isolates have identified considerable genetic heterogeneity among strains causing mastitis. The aim of this study was to investigate the contribution of different clonal complexes and the occurrence of virulence factors and resistance determinants within bovine Staphylococcus aureus isolates.A total of 189 Staphylococcus aureus isolates obtained from milk samples of 34 dairy herds in the German Federal State of Thuringia were characterised by microarray technology.The isolates were assigned to eleven different clonal complexes with CC151, CC479 and CC133 being dominant (together 80.5%). The methicillin resistance gene mecA was found in four isolates (2.1%), which belonged to CC398. Enterotoxin genes could be detected in 79.3% of analysed Staphylococcus aureus and 19 isolates (10.1%) harboured a distinct allele of the toxic shock syndrome toxin gene, tst-RF122. The most striking finding of the present study was that almost all except one isolate (151/152) belonging to CC151, CC479 and CC133 harboured the leukocidin genes lukF-P83 and lukM, whereas no further isolates from other lineages possessed these genes.The consistent occurrence of lukF-P83/lukM in the dominating clonal complexes suggests an essential role of this leukocidin in the etiology of bovine mastitis.  相似文献
