Background: Brucella abortus, the major causative agent of abortion in cattle and a zoonotic pathogen, needs to be diagnosed at an early stage. Loop-mediated isothermal amplification (LAMP) test is easy to perform and also promising to be adapted at field level.

Objective: To develop a LAMP assay for specific and rapid detection of B. abortus from clinical samples of cattle.

Methods: LAMP primers were designed targeting BruAb2_0168 region using specific software tool and LAMP was optimized. The developed LAMP was tested for its specificity with 3 Brucella spp. and 11 other non-Brucella spp. Sensitivity of the developed LAMP was also carried out with known quantity of DNA. Cattle whole blood samples and aborted fetal stomach contents were collected and used for testing with developed LAMP assay and results were compared with polymerase chain reaction (PCR).

Results: The developed LAMP assay works at 61 °C for 60 min and the detection limit was observed to be 100-fold more than the conventional PCR that is commonly used for diagnosis of B. abortus. Clinical sensitivity and specificity of the developed LAMP assay was 100% when compared with Rose Bengal plate test and standard tube agglutination test. SYB® green dye I was used to visualize the result with naked eye.

Conclusion: The novelty of the developed LAMP assay for specifically detecting B. abortus infection in cattle along with its inherent rapidness and high sensitivity can be employed for detecting this economically important pathogen of cattle at field level as well be exploited for screening of human infections.  相似文献

This review converses the Zika virus which has attained global concern due to its rapid pandemic potential and impact on humans. Though Zika virus was first isolated in 1947, till the recent large-scale outbreak which occurred in Micronesia, in 2007, the virus was placed into the innocuous pathogen category. The World Health Organization on 1 February 2016 declared it as a ‘Public Health Emergency of International Concern.’ Of the note, American as well as Pacific Island strains/isolates is relatively closer to Asian lineage strains. The African and American strains share more than 87.5% and 95% homologies with Asian strains/isolates, respectively. Asian strains form independent clusters, except those isolated from China, suggesting relatively more diversity than African strains. Prevention and control are mainly aimed at the vector population (mosquitoes) with Aedes aegypti being the main species. Surveys in Africa and Asia indicated seropositivity in various animal species. However, so far its natural reservoir is unknown. There is an urgent need to understand why Zika virus has shifted from being a virus that caused mild illness to unforeseen birth defects as well as autoimmune-neurological problems. Unfortunately, an effective vaccine is not available yet. Availability of cryo-electron microscopy based on 3.8 Å resolution revealing mature Zika virus structure and the probable virus attachment site to host cell would provide critical insights into the development of antiviral treatments and vaccines.  相似文献

Purpose

The science of sediment fingerprinting has been evolving rapidly over the past decade and is well poised to improve our understanding, not only of sediment sources, but also the routing of sediment through watersheds. Here, we discuss channel–floodplain processes that may convolute or modify the sediment fingerprinting signature of alluvial bank/floodplain sources and explore the use of nonconservative tracers for differentiating sediment derived from surface soil erosion from that of near-channel fluvial erosion.

Materials and methods

We use a mathematical model to demonstrate the theoretical effects of channel–floodplain exchange on conservative and nonconservative tracers. Then, we present flow, sediment gauging data, and geochemical measurements of long- (meteoric beryllium-10, 10Be) and short-lived (excess lead-210 and cesium-137, 210Pbex and 137Cs, respectively) radionuclide tracers from two study locations: one above, and the other below, a rapidly incising knick zone within the Maple River watershed, southern Minnesota.

Results and discussion

We demonstrate that measurements of 10Be, 210Pbex, and 137Cs associated with suspended sediment can be used to distinguish between the three primary sediment sources (agricultural uplands, bluffs, and banks) and estimate channel–floodplain exchange. We observe how the sediment sources systematically vary by location and change over the course of a single storm hydrograph. While sediment dynamics for any given event are not necessarily indicative of longer-term trends, the results are consistent with our geomorphic understanding of the system and longer-term observations of sediment dynamics. We advocate for future sediment fingerprinting studies to develop a geomorphic rationale to explain the distribution of the fingerprinting properties for any given study area, with the intent of developing a more generalizable, process-based fingerprinting approach.

Conclusions

We show that measurements of conservative and nonconservative tracers (e.g., long- and short-lived radionuclides) can provide spatially integrated, yet temporally discrete, insights to constrain sediment sources and channel–floodplain exchange at the river network-scale. Fingerprinting that utilizes nonconservative tracers requires that the nonconservative behavior is predictable and verifiable.  相似文献

A 60‐day study was conducted to determine the response of juvenile bluegill Lepomis macrochirus to seven experimental diets, formulated using a blend of alternative protein sources as a replacement for fish meal. Adequate levels (digestible basis) of energy, protein and amino acids were maintained in diets 1–6, whereas slightly lower protein and energy levels were provided in diet 7. Feed cost per tonne ranged from $ 798.9 (diet 1, 550 g Kg?1 fish meal) to $ 515.8 (diet 6, 0 g kg?1 fish meal), or to $ 507.2 (diet 7, 0 g Kg?1 fish meal). Three commercial diets were included in the study as reference diets: a high‐energy and a low‐energy trout diet, as well as a catfish diet. Quintuplicate bluegill groups (~22 g, n = 10 fish group?1) were fed the experimental diets twice daily to apparent satiation. No major differences in feed consumption, feed efficiency and growth rates were detected among the bluegill groups fed the experimental diets. Trout diets generally produced higher fish fat deposition, whereas the catfish diet produced a poorer fish growth rate relative to the experimental diets. Under the reported conditions, results indicate diet 6, comprising predominantly soybean meal and porcine meat and bone meal, to be the most economical diet for juvenile bluegill.  相似文献

Soy protein elastomer (SPE) exhibits elastic, extensible, and sticky properties in its native state and displays great potential as an alternative to wheat gluten. The objective of this study was to better understand the roles of soy protein subunits (polypeptides) contributing to the functional properties of SPE. Six soy protein samples with different subunit compositions were prepared by extracting the proteins at various pH values on the basis of the different solubilities of conglycinin (7S) and glycinin (11S) globulins. Soy protein containing a large amount of high molecular weight aggregates formed from α' and α subunits exhibited stronger viscoelastic solid behavior than other soy protein samples in terms of dynamic elastic and viscous modules. Electrophoresis results revealed that these aggregates are mainly stabilized through disulfide bonds, which also contributed to higher denaturation enthalpy as characterized by DSC and larger size protein aggregates observed by TEM. Besides, the most viscoelastic soy protein sample exhibited flat and smooth surfaces of the protein particles as observed by SEM, whereas other samples had rough and porous particle surfaces. It was proposed that the ability of α' and α to form aggregates and the resultant proper protein-protein interaction in soy proteins are the critical contributions to the continuous network of SPE.  相似文献

The focus of the present research work is on evaluation of the antimicrobial finished textile material from the extract of Aerva lanata plant and optimization of process parameters to get the maximum antimicrobial efficiency. The microencapsulated extracts of Aerva lanata plant (leaves and stalk) using citric acid as a cross linking agent were applied on plain-woven cotton fabric by pad-dry-cure method. The Aerva lanata extract treated fabric samples were tested for antibacterial activity against bacterial strains of Staphylococcus aureus and Escherichia coli under Agar diffusion test and AATCC 100–2004. It is observed that the treated cotton fabric shows a clear microbial resistance of about 19–25 mm zone of inhibition in the agar diffusion test against the above-mentioned bacterial strains. The treated samples showed 80 % of reduction against Staphylococcus aureus and 70 % reduction against Escherichia coli as per AATCC (100–2004). Process parameters like concentration of extract, citric acid and curing temperature were optimized using Box-Behnken design for better performance of antimicrobial treated material. The optimum values of 21.14 % extract concentration, 2.39 % crosslinking agent and 101.2 °C curing temperature were determined.  相似文献

Soil contaminated by chromium (Cr) is a major concern for sustainable agriculture. Considering this as a basis, the present study was designed to isolate Cr(VI)-reducing and plant growth-promoting bacterial strain from contaminated sampling sources. In this study, Rhizobium strain ND2 was isolated from the root nodules of Phaseolus vulgaris grown in leather industrial effluent contaminated soil. The strain ND2 exhibited strong resistance to different heavy metals and reduced 30 and 50 µg ml?1 concentrations of Cr(VI) completely after 80 and 120 h of incubation, respectively, as well as chromium adsorption and immobilization were confirmed by scanning electron microscopic equipped with energy X-ray spectroscopy. In addition, the strain produced 21.73 and 36.86 µg ml?1 of indole-3-acetic acid at 50 and 100 µg ml?1 of L-tryptophan supplimentations, respectively. Strain ND2 positively affected the exo-polysaccharide, ammonia, protease and catalase production and stimulated root length of various test crops under Cr(VI) stress. Moreover, Rhizobium strain ND2 has the potential to colonize the diverse agricultural crops. Thus, the present findings strongly suggested that the multipotential properties of ND2 could be exploited for bioremediation of contaminated sites with Cr(VI) as well as potential bio fertilizer for enhancing the agricultural productivity.  相似文献

Mastitis (intramammary inflammation) caused by infectious pathogens is still considered a devastating condition of dairy animals affecting animal welfare as well as economically incurring huge losses to the dairy industry by means of decreased production performance and increased culling rates. Bovine mastitis is the inflammation of the mammary glands/udder of bovines, caused by bacterial pathogens, in most cases. Routine diagnosis is based on clinical and subclinical forms of the disease. This underlines the significance of early and rapid identification/detection of etiological agents at the farm level, for which several diagnostic techniques have been developed. Therapeutic regimens such as antibiotics, immunotherapy, bacteriocins, bacteriophages, antimicrobial peptides, probiotics, stem cell therapy, native secretory factors, nutritional, dry cow and lactation therapy, genetic selection, herbs, and nanoparticle technology-based therapy have been evaluated for their efficacy in the treatment of mastitis. Even though several strategies have been developed over the years for the purpose of managing both clinical and subclinical forms of mastitis, all of them lacked the efficacy to eliminate the associated etiological agent when used as a monotherapy. Further, research has to be directed towards the development of new therapeutic agents/techniques that can both replace conventional techniques and also solve the problem of emerging antibiotic resistance. The objective of the present review is to describe the etiological agents, pathogenesis, and diagnosis in brief along with an extensive discussion on the advances in the treatment and management of mastitis, which would help safeguard the health of dairy animals.  相似文献
