The effects of drip irrigation on the yield and crop water productivity responses of four tea (Camellia sinensis (L.) O. Kuntze) clones were studied four consecutive years (2003/2004-2006/2007), in a large (9 ha) field experiment comprising of six drip irrigation treatments (labelled: I1-I6) and four clones (TRFCA PC81, AHP S15/10, BBK35 and BBT207) planted at a spacing of 1.20 m × 0.60 m at Kibena Tea Limited (KTL), Njombe in the Southern Tanzania in a situation of limited water availability. Each clone × drip irrigation treatment combination was replicated six times in a completely randomized design with 144 net plots each with an area of 72 m2. Clone TRFCA PC81 gave the highest yields (range: 5920-6850 kg dried tea ha−1) followed by clones BBT207 (5010-5940 kg dried tea ha−1), AHP S15/10 (4230-5450 kg dried tea ha−1) and BBK35 (3410-4390 kg dried tea ha−1) and drip irrigation treatment I2 gave the highest yields, ranging from 4954 to 6072 kg dried tea ha−1) compared with those from other treatments (4113-5868 kg dried tea ha−1). Most of these yields exceeded those (4200 kg dried tea ha−1) obtained from overhead sprinkler irrigation system in Mufindi also Southern Tanzania, and Kibena Estate itself. Results showed that drip irrigation of tea not only increased yields but also gave water saving benefits of up to 50% from application of 50% less water to remove the cumulative soil water deficit (treatment I2), and with labour saving of 85% for irrigation. The yield of dried tea per mm depth of water applied, i.e., “the crop water productivity” for drip irrigation of clones TRFCA PC81, BBT207 and BBK35, in 2003/2004 for instance, were 9.3, 8.5 and 7.1 kg dried tea [ha mm]−1, respectively. The corresponding values in 2004/2005 were 2.7, 4.5 and 2.0 kg dried tea [ha mm]−1 while the yield responses from clone AHP S15/10 were linear decreasing by 1 and 1.6 kg dried tea [ha mm]−1 in 2003/2004 and 2004/2005, respectively. In 2005/2006 the crop water productivity from clones TRFCA PC81, AHP S15/10, BBK35 and BBT207 were 4.5, 0.4, 5.2 and 6.9 kg dried tea [ha mm]−1, respectively with quadratic yield response functions to drip irrigation depth of water application. The results are presented and recommendations and implications made for technology-transfer scaling-up for increased use by large and smallholder tea growers.  相似文献

The study assessed the effect of hybridization on reproductive performance of Oreochromis karongae, Oreochromis shiranus and Oreochromis mossambicus for a period of 120 days. Number of eggs per batch, number of females spawned, relative fecundity, egg size, hatching period, hatchability and hatchling survival were assessed. The results revealed higher number of eggs per batch and relative fecundity in the crosses where O. shiranus and O. mossambicus were females than female O. karongae. The results also revealed that egg size, hatching period, hatchability and hatchling survival were species specific. Significant differences were noted for number of eggs and larvae produced by the three species (p < .05). Interspecific and pure crosses for the same female species did not differ significantly for the number of eggs per batch, relative fecundity, egg size, hatching period, hatchability and hatchling survival. However, number of female spawned was found to be higher in the interspecific crosses of female O. karongae than in pure cross of O. karongae. Findings from this study suggest that hybridization can be used to improve reproductive performance of O. karongae as the females spawned increased, and it is a value addition in aquaculture management of indigenous species.  相似文献

Antimicrobial therapy is the most commonly used treatment of bacterial infections in dairy cows. Polymorphonuclear neutrophil leukocytes (PMN) play an important role in the first line defence against invading bacteria and it is important that the function of PMN is not compromised by antibiotics. We investigated the in vitro effect of cephapirin, a first generation cephalosporin, and mecillinam, an amidinopenicillin with activity against mainly Gram-negative bacteria, on phagocytosis and respiratory burst activity of PMN isolated from bovine blood. After in vitro incubation of PMN with different concentrations of the antibiotics, phagocytosis was evaluated by flow cytometry and respiratory burst activity was evaluated by registration of chemiluminescence (CL) with a luminometer. None of the investigated concentrations of cephapirin and mecillinam had an effect in vitro on phagocytosis of Escherichia coli by PMN. At high concentrations (100 and 1000 μg/mL), cephapirin and mecillinam reduced the respiratory burst activity of PMN. Part of these suppressive effects could be ascribed to oxidant scavenging. Inhibitory effects of cephapirin were stronger than mecillinam. In conclusion, cephapirin and mecillinam did not seem to affect antibacterial activity of PMN isolated from bovine blood in vitro at therapeutic concentrations.  相似文献

OBJECTIVE: To evaluate L-selectin (CD62L) and Mac-1 (CD11b) expression at the surface of blood and milk neutrophils during the early inflammatory response to Escherichia coli-induced mastitis in cows. ANIMALS: 6 healthy Holstein heifers in early lactation. PROCEDURE: Blood and milk samples were collected before and after intramammary administration of 10(4) CFU's of E coli in the left mammary gland quarters. Bacterial counts and electrolyte concentrations in milk, rectal temperature, differential blood leukocyte counts, milk somatic cell counts, neutrophil viability, and the expression of CD62L and CD11b on blood and milk neutrophils were determined longitudinally. RESULTS: Bacteria grew during the first 6 hours after inoculation with a pronounced leukocytic influx. Coincident with neutrophil influx was an increase in CD62L+ and CD11b+ milk neutrophils, as well as an improved viability of milk neutrophils. The peak of the inflammatory reaction was reached approximately 12 hours after E coli inoculation. From that time forward, changes in CD62L and CD11b expression were opposed to each other, with a decrease in CD62L expression and an increase in CD11b expression on blood and milk neutrophils; the magnitude of the differences in CD62L and CD11b expression between blood and milk neutrophils decreased. Percentages of CD62L+ and CD11b+ milk neutrophils increased to percentages that were similar to blood neutrophils (ie, approx 92%). CONCLUSIONS AND CLINICAL RELEVANCE: The presence of adhesion molecules on a large percentage of milk neutrophils during the acute inflammatory response, together with the changes in receptor density, suggest a major role for CD62L and CD11b in neutrophil function during coliform mastitis.  相似文献

OBJECTIVE: To determine whether proinflammatory mediators and glucocorticoids affect CD62L(L-selectin) expression on peripheral blood neutrophils from cows in various stages of lactation. ANIMALS: 100 healthy dairy cows during early (13.1 +/- 0.79 days after parturition; n = 31), peak (58.7 +/- 1.64 days after parturition; 31), and mid (137.2 +/- 2.59 days after parturition; 38) lactation. PROCEDURE: In vitro effects of relevant proinflammatory mediators that are released in response to mastitis caused by gram-negative bacteria such as lipopolysaccharide (endotoxin), tumor necrosis factor-alpha, and platelet-activating factor (PAF) on CD62L expression on bovine neutrophils were assessed by flow cytometry. Influences of cortisol and dexamethasone on CD62L expression on bovine neutrophils were also investigated. RESULTS: Basal CD62L expression on neutrophils from cows during early, peak, and mid lactation were similar. Lipopolysaccharide and tumor necrosis factor-alpha had no effect on CD62L expression on neutrophils from cows at any stage of lactation. Conversely, PAF elicited a time- and dose-dependent, down regulatory effect on CD62L expression. However, no differential shedding of CD62L from neutrophils of cows at any stage of lactation were detected. In addition, no effects on CD62L expression on bovine neutrophils after whole blood incubation with cortisol or dexamethasone were observed. Incubation with glucocorticoids did not prevent the down regulatory effect of PAF on CD62L expression. CONCLUSIONS AND CLINICAL RELEVANCE: Comparable basal CD62L expression on bovine neutrophils and equal amounts of CD62L shedding from bovine neutrophils during all stages of lactation suggest that variations in CD62L density are not a likely cause of susceptibility of cows to coliform-induced mastitis during early lactation.  相似文献

RNA interference was used to suppress protein expression of three S-class genome segments of avian reovirus (ARV). Viral progeny titer was successfully down-regulated by RNA interference. Suppression of S1 genome segment, which has three open reading frames, not only decreased the expression level of the structural protein σC but also reduced cell fusion and the level of Ser15-phosphorylated p53 protein caused by the nonstructural proteins p10 and p17, respectively. Suppression of S2 or S4 genome segment by RNA interference could also reduce the expression level of σA or σNS. Interestingly, suppression of σNS resulted in down regulation of the expression of other viral products. In terms of variability of different genes among viral strains and of the impact after their suppression, it seems that the viral products involved in construction of viroplasm or core particles, like σNS, are considerable choices to efficiently inhibit ARV multiplication by RNA interference. Using a GFP reporter system, it was discovered that ARV could not inhibit activated RNA interference, suggesting that RNA interference may be used in the suppression of ARV infection.  相似文献

Female reproductive technologies such as multiple ovulation and embryo transfer (MOET) and juvenile in vitro fertilization and embryo transfer (JIVET) have been shown to accelerate genetic gain by increasing selection intensity and decreasing generation interval. Genomic selection (GS) increases the accuracy of selection of young candidates which can further accelerate genetic gain. Optimal contribution selection (OCS) is an effective method of keeping the rate of inbreeding at a sustainable level while increasing genetic merit. OCS could also be used to selectively and optimally allocate reproductive technologies in mate selection while accounting for their cost. This study uses stochastic simulation to simulate breeding programmes that use a combination of artificial insemination (AI) or natural mating (N), MOET and JIVET with GS. OCS was used to restrict inbreeding to 1.0% increase per generation and also to optimize use of reproductive technologies, considering their effect on genetic gain as well as their cost. Two Australian sheep breeding objectives were used as an example to illustrate the methodology—a terminal sire breeding objective (A) and a dual‐purpose self‐replacing breeding objective (B). The objective function used for optimization considered genetic merit, constrained inbreeding and cost of technologies where costs were offset by a premium paid to the seedstock breeder investing in female reproductive technologies. The premium was based on the cumulative discounted expression of genetic merit in the progeny of a commercial tier in the breeding programme multiplied by the proportion of that benefit received by the breeder. With breeding objective B, the highest premium of 64% paid to the breeder resulted in the highest allocation of reproductive technologies (4%–10% for MOET and 19%–54% for JIVET) and hence the highest annual genetic gain. Conversely, breeding objective A, which had a lower dollar value of the breeding objective and a maximum of 5% mating types for JIVET and zero for MOET were optimal, even when highest premiums were paid. This study highlights that the level of investment in breeding technologies to accelerate genetic gain depends on the investment of genetic improvement returned to the breeder per index point gain achieved. It also demonstrates that breeding programmes can be optimized including allocation of reproductive technologies at the individual animal level. Accounting for revenue to the breeder and cost of the technologies can facilitate more practical decision support for beef and sheep breeders.  相似文献

Reference populations for genomic selection usually involve selected individuals, which may result in biased prediction of estimated genomic breeding values (GEBV). In a simulation study, bias and accuracy of GEBV were explored for various genetic models with individuals selectively genotyped in a typical nucleus breeding program. We compared the performance of three existing methods, that is, Best Linear Unbiased Prediction of breeding values using pedigree‐based relationships (PBLUP), genomic relationships for genotyped animals only (GBLUP) and a Single‐Step approach (SSGBLUP) using both. For a scenario with no‐selection and random mating (RR), prediction was unbiased. However, lower accuracy and bias were observed for scenarios with selection and random mating (SR) or selection and positive assortative mating (SA). As expected, bias disappeared when all individuals were genotyped and used in GBLUP. SSGBLUP showed higher accuracy compared to GBLUP, and bias of prediction was negligible with SR. However, PBLUP and SSGBLUP still showed bias in SA due to high inbreeding. SSGBLUP and PBLUP were unbiased provided that inbreeding was accounted for in the relationship matrices. Selective genotyping based on extreme phenotypic contrasts increased the prediction accuracy, but prediction was biased when using GBLUP. SSGBLUP could correct the biasedness while gaining higher accuracy than GBLUP. In a typical animal breeding program, where it is too expensive to genotype all animals, it would be appropriate to genotype phenotypically contrasting selection candidates and use a Single‐Step approach to obtain accurate and unbiased prediction of GEBV.  相似文献
