Iron-dependent outer membrane proteins (IROMPs) play an important role in bacterial pathogenesis and present several attributes of potential vaccine candidates. TBLASTN analysis of the Pasteurella multocida Pm70 genome using the same molecules of other bacterial pathogens as a query identified eight putative haemin and haemoglobin receptors for this organism. Quantitative binding assays have demonstrated that the proteins PM0040, PM0236, PM0741, PM1081, PM1428, PM0592 and HgbA bind both haemin and haemoglobin, whereas PM0576 and PM1282 ORFs only bind either haemoglobin or haemin, respectively. Furthermore, Western blot analysis showed that P. multocida-infected mice generate specific antibodies against PM0040, PM0236, PM0741, PM1081, PM1428, PM0592 and HgbA proteins. Nevertheless, inoculation of mice with any single one of these receptors alone did not protect against P. multocida infection.  相似文献

The fractal index by Katz and George (1985) for thecharacterization of planar curves is applied to wolf search pathsrecorded by radio-telemetry. All the sets of paths studied showspatial patterns whose complexity is between a straight line anda true random walk. Females fractal dimensions show significantchanges throughout the year, depending on the state of their lifecycle (normal, breeding and wandering). There are alsodifferences between males and females, but not between adults andnon-adults. The results are discussed with regard to wolffood-search strategies.  相似文献

Determination of F1 hybrid seed purity in pepper using PCR-based markers   总被引:5,自引:0,他引:5  
We present the potential use of two kinds of PCR-based markers, RAPDs (Random Amplified Polymorphic DNA) and SPARs (Single Primer Amplification Reactions), as tools for hybrid seed purity determination. Five F1 pepper hybrids (Capsicum annuum L.) and their parents were analyzed with 100 10-mer primers and 10 nucleotide repeat primers. We found at least one useful marker for testing the purity of all hybrids studied. Despite their dominant inheritance these markers could be an efficient implement in the process of quality testing of hybrid seeds. Winter conditions in the NW Mediterranean cause instability of the water column and non-geostrophic dynamics, such as vertical mixing and convection are significant. These events involve nutrient supply to the photic zone that can sustain high productivity. In this study, we aim to investigate the role of winter hydrodynamics on the spawning strategies of Sardina pilchardus and Micromesistius poutassou. Data were obtained on two oceanographic cruises (February 2017 and 2018) off the Catalan coast. The occurrence of S. pilchardus eggs very close to the coast indicated a clear preference of the species for spawning in coastal areas. Preflexion and postflexion larvae exhibited a slightly wider distribution showing a clear association with the cold, less saline and more productive coastal waters. Preflexion larvae of M. poutassou were found on the upper slope and over the shelf, being offshore limited by the shelf/slope front present all along the slope. The front would act as a barrier preventing their dispersion towards the open sea. M. poutassou larvae in advanced developmental stages were located close to the coast in the productive shelf waters, with instabilities of the front contributing to larval transport from offshore waters to the coast. The vertical distribution of both species showed high variability, not only related with the daily cycle or developmental stage, but also with the vertical structure of the water column. Overall, the results provide some clues on how the spawning strategies of both species may evolve under future scenarios of higher winter-stratification, because of the global warming.  相似文献

Evidence from DNA‐analysis is commonplace in human criminal investigations, and while it is increasingly being used in wildlife crime, to date, its application to control and enforcement activities in fisheries and aquaculture has only been sporadic. Contemporary DNA‐analysis tools are capable of addressing a broad range of compliance issues, species identification, mislabelling of fish products, determining the origin of catches and the farm of origin of aquaculture escapees. Such applications have the potential to ensure traceability along the fish product supply chain and to combat consumer fraud and Illegal, Unreported and Unregulated fishing. Nevertheless, DNA‐analysis is not yet used routinely in investigations into compliance with fisheries and aquaculture legislation. One potential reason for this is that DNA‐analysis techniques may have been regarded as too expensive. However, costs have plummeted over the past decade prompting us to objectively assess whether the costs associated with routine use of DNA‐analysis techniques for fisheries and aquaculture control and enforcement activities do constitute an impediment. Based on a number of recent fisheries and aquaculture compliance investigations that incorporated DNA‐analysis, our results indicate that the use of genetic analysis was justified and worthwhile in all cases examined. We therefore conclude that the costs associated with DNA‐analysis do not represent a barrier to the routine adoption of DNA‐analysis techniques in fisheries and aquaculture compliance investigations. Thus, control and enforcement agencies should be encouraged to use such techniques routinely.  相似文献

The presence of herbicide‐resistant Lolium rigidum in Mediterranean (Spanish) citrus orchards was reported in 2005 and it poses a serious threat to crop management. The main objective of this research was to investigate which components could be responsible for the persistence of annual ryegrass populations in Mediterranean mandarin and orange orchards. This is the first study regarding L. rigidum populations in Mediterranean citrus orchards. Surveys were conducted in 55 commercial citrus orchards in eastern Spain in 2013 by interviewing technicians who were working in cooperatives about crop management. The level of infestation by L. rigidum and the presence of harvester ants (Messor barbarus) then were estimated in the same orchards. The variables were subjected to a two‐dimensional analysis and both univariate and multivariate logistic regression models were fitted for each of the three L. rigidum density levels that had been established. The multivariate models showed the significant factors that were associated with various L. rigidum densities: (i) at a low density, the herbicides that were applied, the number of applications in 2013 and the type of irrigation (flood or drip); (ii) at a medium density, the presence of harvester ants; and (iii) at a high density, the herbicides that were applied in 2013. The results indicated that drip irrigation and one application of glyphosate mixed with other herbicides (or herbicides other than glyphosate) were associated with a lower L. rigidum density. The alternative management options that are presented here should help farmers to reduce weed problems in Mediterranean citrus orchards. Future research is required to better understand the presence of herbicide‐resistant populations, as well as the possible beneficial presence of granivorous ant species.  相似文献
