An isolate of the poty virus turnip mosaic virus (TuMV-Ab) showing severe mosaic symptoms inAbutilon theophrasti from Piedmont (northwestern Italy) in 1993, has been found to be of an unusual pathotype and serotype. The isolate was easily transmitted byAphyis gossiypii and Myzus persicae and was not seed-transmitted inA. Theophrasti. The host range of TuMV-Ab was different from that of another Piedmont isolate of TuMV fromAlliaria officinalis and from a TuMV isolate fromBrassica napus. TuMV-Ab was characterized using the reactions on the fourB. Napus lines S4, R4, 165 and S1 as the rare pathotype 7, found only once previously in Europe. Tests with polyclonal antisera indicated that TuMV-Ab was only distantly related to the two other TuMV isolates. Serological characterization with a panel of 30 monoclonal antibodies showed that TuMV-Ab belonged to one of the less common serotypes (JPN).  相似文献

Pathotype-specific and broad-spectrum resistance to turnip mosaic virus (TuMV) have been identified in the diploid A genome brassica species Brassica rapa. The pathotype-specific resistance is effective against pathotype 1 isolates of TuMV, which are the most common in Europe. It is almost identical in its specificity to that of a mapped resistance gene (TuRB01) present in the A genome of the amphidiploid species Brassica napus. A mutant of a pathotype 1 isolate of TuMV (UK 1M) that is able to overcome TuRB01 also overcame the B. rapa resistance. This, combined with the fact that a single-nucleotide mutation in the cylindrical inclusion gene of TuMV that has been shown to induce a change from avirulence to virulence against TuRB01, had an identical effect on the B. rapa resistance, suggest that the two resistances are conditioned by the same gene. A second source of resistance in B. rapa prevented systemic spread of all TuMV isolates tested. A third source of resistance that appears to provide immunity to, or severely restrict replication of most isolates of TuMV has been characterised. This resistance source also prevented systemic spread of all TuMV isolates tested. Prior to this study, no resistance to pathotype 4 or pathotype 12 isolates of TuMV had ever been identified. For each of these three resistance sources, plant lines that are not segregating for some of the resistance phenotypes and that are presumably homozygous for the genes controlling these phenotypes have been generated. Strategies for further characterising and deploying these resistances in different Brassica species are described.  相似文献

A panel of 30 monoclonal antibodies (MAbs) was produced against four isolates of turnip mosaic virus (TuMV). The panel was tested in plate-trapped antigen ELISA tests against 41 TuMV isolates (with different host and geographical origins and of differing pathotypes). The antibodies were also tested against four other potyviruses (bean common mosaic virus, bean common mosaic necrosis virus, lettuce mosaic virus and zucchini yellow mosaic virus). The reactions were assessed quantitatively (using multivariate analysis) and qualitatively (using the standard deviation obtained against healthy leaf material). The MAbs recognized 16–17 TuMV epitopes that were not present in the other potyviruses and a further two potyvirus epitopes. The isolates were grouped into three serotypes. Only one isolate did not fit this grouping. The classification of seven isolates in coat protein amino acid sequence homology groups correlated with serotypes. There was no correlation between serotype and pathotype, or between reactions to individual MAbs and single lines. There was therefore no evidence that the epitopes recognized by the MAbs are elicitors for the resistance genes present in the Brassica napus lines. However, the sensitivity and specificity of the MAbs will be useful for both routine detection of TuMV and fundamental studies on plant–virus interactions.  相似文献

Objective: To determine the physiologic response of the equine navicular bone to core decompression surgery in healthy horses. Study Design: Experimental in vivo study. Materials: Healthy adult horses (n=6). Methods: Core decompression was completed by creating three 2.5‐mm‐diameter drill channels into the navicular bone under arthroscopic control. The venous (PV), arterial (PA), articular (PDIPJ), and intraosseous pressures (IOP) were recorded before and after decompression drilling. Each IOP measurement consisted of a baseline (IOPB) and a stress test (intramedullary injection of saline solution, IOPS) recording. Lameness was assessed subjectively and using force plate gait analysis. Fluorochrome bone labeling was performed. Horses were euthanatized at 12 weeks. Navicular bone mineral density (BMD) was measured, and bone histology evaluated. Results: Peak IOP (IOPmax) after stress testing was significantly (P<.05) reduced immediately after core decompression; however, the magnitude of these effects was decreased at 3 and 6 weeks after decompression. A significant (P<.05) correlation existed between IOPmax and BMD. No lameness was observed beyond the first week after surgery. Substantial remodeling and neovascularization was evident adjacent the surgery sites. Conclusion: Navicular bone core decompression surgery reduced IOPmax, and, with the exception of a mild short‐lived lameness, caused no other adverse effects in healthy horses during the 12‐week study period.  相似文献

Objective: To determine the effect of core decompression surgery and bone mineral density (BMD) on the mechanical properties of equine navicular bones. Study Design: Experimental, in vitro study. Sample Population: Fore limb navicular bones (n=36 pairs) from sound 2–5‐year‐old horses with no radiographic abnormalities of the distal aspect of the forelimbs. Methods: Navicular BMD was measured using dual energy X‐ray absorptiometry. One randomly assigned navicular bone from each pair served as control. The contralateral test specimen was allocated to 1 of 6 treatment groups defined by drill bit size (3.2 versus 2.5 mm diameter) and by the number of drill channels (1, 2, or 3) created in the proximal border of the bone. Bones were then tested until failure in 3‐point bending. Data were statistically analyzed using ANOVA and regression analysis. Results: There were significant (P<.001) positive correlations between BMD and biomechanical data. A significant (P<.001) reduction in breaking strength was noted between intact and drilled bone pairs; however, the diameter and number of decompression channels did not significantly (P>.05) influence the extent of the reduction in mechanical strength. Conclusion: In vitro core decompression significantly decreases the breaking strength of the equine navicular bone.  相似文献

Type I diabetes may be an autoimmune disorder, although the evidence is largely circumstantial. The natural history of the disease after diagnosis includes partial remission in most patients, but only about 3 percent achieve transient insulin independence. beta Cell function, as indicated by the plasma concentration of C-peptide, is lost over 6 to 30 months and islet cell antibodies disappeared over 1 to 2 years. This article describes a pilot study in which 41 patients were treated with the immunosuppressive agent cyclosporine for 2 to 12 months. Of 30 patients treated within 6 weeks of diagnosis, 16 became insulin independent with concentrations of plasma C-peptide in the normal range and decreasing titers of islet cell antibodies. Of 11 patients who entered the study 8 to 44 weeks after diagnosis, two achieved this state. These results indicate that a controlled trial of the effects of cyclosporine in type I diabetes should be conducted.  相似文献

Flufenoxuron (“Cascade”) is a novel acylurea with acaricidal and insecticidal properties. It acts in a similar manner to diflubenzuron (DFB) by impairing chitin incorporation into insect cuticle. Chitin can be localised cytochemically using the lectin, wheat germ agglutinin (WGA), which binds specifically to N-acetylglucosamine-containing polymers. WGA adsorbed to monodisperse colloidal gold (WGA-gold) was used as an electron-dense marker for localising chitin in the cuticle of sixth-instar Spodoptera littoralis. In the cuticle of control insects, the deposition zone and endocuticle were heavily labelled. In flufenoxuron- and DFB-treated insects, only the endocuticle (formed before treatment) was labelled, the deposition zone being devoid of label. These results show that flufenoxuron and DFB act in similar manner in reducing chitin incorporation in the cuticle of S. littoralis.  相似文献

Granule-bound starch synthase, also known as the waxy protein catalyses the synthesis of amylose in wheat endosperm starch. In durum wheats, the genes encoding GBSS are present at the two Wx loci on chromosome 7A and 4A (a segment of 7B that has been translocated). Several null Wx-B1 (missing GBSS protein from chromosome 4A) durum lines were produced from crosses with null-4A bread wheats backcrossed to durum wheats. Semolina milled from 4 normal and 7 null-4A durum wheat lines grown over two seasons (1999 and 2000) in South Australia were analysed for amylose content, starch pasting properties as measured by the Rapid Viscoanalyzer (RVA), swelling power and starch damage, protein content and electrophoretic protein analysis. Spaghetti was prepared with a micro-scale extruder and the cooked pasta evaluated for cooking loss, firmness, stickiness and water absorption. The null-4A lines had significantly lower (ca. 5%) amylose content, higher starch peak viscosities and semolina swelling power. The pasta derived from the null-4A lines had lower cooking loss and in 1999 was more adhesive than the non-waxy lines. Cooking loss was correlated with amylose content, peak starch viscosity, swelling power of semolina and cooked pasta adhesiveness. Semolina swelling power was highly correlated with RVA peak viscosity. Waxy durum wheats appear to have an advantage over the normal types in terms of lower cooking loss, widely used as an indicator of pasta cooking quality.  相似文献
