Upland cotton, Gossypium hirsutum L. is the most widely planted cultivated cotton in the United States and the world. The other cultivated tetraploid species G. barbadense L. is planted on considerable less area; however, it produces extra long, strong, and fine fibers which spins into superior yarn. The wild cotton tetraploid species G. tomentosum Nuttall ex Seemann, native to the Hawaiian Archipelago also exhibits traits, such as drought tolerance, that would also be desirable to transfer to Upland cotton. Long-term breeding efforts using whole genome crosses between Upland and these species have not been successful in transferring very many desirable alleles into Upland cotton. Our chromosome substitution lines (CSL) have one chromosome or chromosome arm from an alien species backcrossed into the Upland cotton line,TM-1, via aneuploid technology. Five Upland cultivars were crossed with CS-B01, CS-T01, CS-B04, CS-T04, CS-B18 and CS-T18 and TM-1 the recurrent parent of the CSLs. This provided an opportunity to determine the effects of chromosomes 01, 04, and 18 from the three species in crosses with the five cultivars. Predicted genotypic mean effects of the parents, F2, and F3 generations for eight agronomic and fiber traits of importance were compared. The predicted hybrid mean effects for the three chromosomes from each species were different for several of the traits across cultivars. There was no single chromosome or species that was superior for all traits in crosses. Parental and hybrid lines often differed in the effect of a particular chromosome among the three species. The predicted genotypic mean effects for F2 and F3, with a few exceptions, generally agree with our previous results for additive and dominance genetic effects of these CSL.  相似文献

Summary F2 hybrid cultivars continue to occupy a small portion of the cotton (Gossypium hirsutum L.) production are in the United States, but occupy a larger proportion of the production area in some other countries. Sixty-four F2 hybrids resulting from crosses of four commercial cultivars and 16 pest-resistant germplasm lines were evaluated for five fiber and four yield traits in four environments at Mississippi State, MS. An additive-dominance genetic model was employed for these traits. The minimum norm quadratic unbiased estimation (MINQUE) method was used with a mixed model approach for estimating genetic variance and covariance components and for predicting genetic correlations. This study investigated genetic variances, heritabilities, and genetic and phenotypic correlations between agronomic and fiber traits among these 64 F2 hybrid populations and discussed the usefulness of these populations for use as hybrids or for selections for pure lines.Dominance variance accounted for the major proportion of the phenotypic variances for lint yield, lint percentage, and boll size indicating that hybrids should have an advantage for these traits compared to pure lines. A low proportion of additive variance for fiber traits and the significant additive x environment variance components indicated a lack of substantial useful additive genetic variability for fiber traits. This suggests that selections for pure lines within these F2 populations would have limited success in improving fiber traits. Genetic and phenotypic correlation coefficients were of comparable magnitude for most pairs of characters. Fiber strength showed a positive additive genetic correlation with boll weight. Dominance genetic correlations of fiber strength with elongation and 2.5% span length were also significant and positive; however, the additive genetic correlation of length and strength was zero.Contribution of the USDA-ARS in cooperation with the Mississippi Agric. and Forestry Exp. Genetic data collected from various plant breeding and genetic studies may not be replicated in field designs although field variation is always present. In this study, we addressed this problem using spring wheat (Triticum aestivum L.) trial data collected from two locations. There were no intralocation replications and an extended additive-dominance (AD) model was used to account for field variation. We numerically evaluated the data from simulations and estimated the variance components. For demonstration purposes we also analyzed three agronomic traits from the actual spring wheat data set. Results showed that these data could be effectively analyzed using an extended AD model, which was more comparable to a conventional AD model. Actual data analysis revealed that grain yield was significantly influenced by systematic field variation. Additive effects were significant for all traits and dominance effects were significant for plant height and time-to-flowering. Genetic effects were predicted and used to demonstrate that most spring wheat lines developed by the South Dakota State University breeding program (SD lines) exhibited good general combining ability effects for yield improvement. Thus, this study provides a general framework to appropriately analyze data in situations where field crop data are collected from non-replicated designs.  相似文献

Genetic mapping is an essential tool for cotton (Gossypium hirsutum L.) molecular breeding and application of DNA markers for cotton improvement. In this present study, we evaluated an RI population including 188 RI lines developed from 94 F2-derived families and their two parental lines, ‘HS 46’ and ‘MARCABUCAG8US-1-88’, at Mississippi State, MS, for two years. Fourteen agronomic and fiber traits were measured. One hundred forty one (141) polymorphic SSR markers were screened for this population and 125 markers were used to construct a linkage map. Twenty six linkage groups were constructed, covering 125 SSR loci and 965 cM of overall map distance. Twenty four linkage groups (115 SSR loci) were assigned to specific chromosomes. Quantitative genetic analysis showed that the genotypic effects accounted for more than 20% of the phenotypic variation for all traits except fiber perimeter (18%). Fifty six QTLs (LOD > 3.0) associated with 14 agronomic and fiber traits were located on 17 chromosomes. One QTL associated with fiber elongation was located on linkage group LGU01. Nine chromosomes in sub-A genome harbored 27 QTLs with 10 associated with agronomic traits and 17 with fiber traits. Eight chromosomes in D sub-genome harbored 29 QTLs with 13 associated with agronomic traits and 16 with fiber traits. Chromosomes 3, 5, 12, 13, 14, 16, 20, and 26 harbor important QTLs for both yield and fiber quality compared to other chromosomes. Since this RI population was developed from an intraspecific cross within upland cotton, these QTLs should be useful for marker assisted selection for improving breeding efficiency in cotton line development. The addition of small or trace amounts of carbon to soils can result in the release of 2-5 times more C as CO2 than was added in the original solution. The identity of the microorganisms responsible for these so-called trigger effects remains largely unknown. This paper reports on the response of individual bacterial taxa to the addition of a range of 14C-glucose concentrations (150, 50 and 15 and 0 μg C g−1 soil) similar to the low levels of labile C found in soil. Taxon-specific responses were identified using a modification of the stable isotope probing (SIP) protocol and the recovery of [14C] labelled ribosomal RNA using equilibrium density gradient centrifugation. This provided good resolution of the ‘heavy’ fractions ([14C] labelled RNA) from the ‘light’ fractions ([12C] unlabelled RNA). The extent of the separation was verified using autoradiography. The addition of [14C] glucose at all concentrations was characterised by changes in the relative intensity of particular bands. Canonical correspondence analysis (CCA) showed that the rRNA response in both the ‘heavy’ and ‘light’ fractions differed according to the concentration of glucose added but was most pronounced in soils amended with 150 μg C g−1 soil. In the ‘heavy RNA’ fractions there was a clear separation between soils amended with 150 μg C g−1 soil and those receiving 50 and 15 μg C g−1 soil indicating that at low C inputs the microbial community response is quite distinct from that seen at higher concentrations. To investigate these differences further, bands that changed in relative intensity following amendment were excised from the DGGE gels, reamplified and sequenced. Sequence analysis identified 8 taxa that responded to glucose amendment (Bacillus, Pseudomonas, Burkholderia, Bradyrhizobium, Actinobacteria, Nitrosomonas, Acidobacteria and an uncultured β-proteobacteria). These results show that radioisotope probing (RNA-RIP) can be used successfully to study the fate of labile C substrates, such as glucose, in soil.  相似文献

BACKGROUND: The redlegged earth mite, Halotydeus destructor Tucker, is an important pest of broad‐acre farming systems in Australia, New Zealand and South Africa. In 2006, high levels of resistance to synthetic pyrethroids were discovered in this species in Western Australia. RESULTS: Between 2007 and 2010, the authors monitored resistance in field populations and found it had spread considerably within the state of Western Australia. Twenty‐six paddocks from 15 individual properties were identified with resistance, and these paddocks ranged over 480 km. To date, resistance has not been detected in any other Australian state. Resistance in H. destructor appears to be found across the entire pyrethroid group, but not to other chemical classes such as organophosphates and carbamates, or other chemistries with novel modes of action. CONCLUSION: The high levels of resistance occurring in Western Australia have caused considerable economic losses due to ineffective chemical applications and mortality of crop plants at seedling establishment. These findings highlight the need for a comprehensive resistance surveillance programme to be developed for H. destructor within Australia. Growers need to consider non‐chemical approaches for pest control and should be encouraged to implement pesticide resistance management programmes for H. destructor. The effect of dietary energy restriction on serum insulin, insulin-like growth factor I (IGF-I), growth hormone, (GH), cortisol, plasma urea nitrogen (PUN) and nonesterified fatty acid (NEFA) concentrations was examined. Angus bulls and steers (10 mo) were allotted to two groups of 12 animals and assigned a treatment order. In a switchback design, animals in order 1 were fed a high grain diet, then fasted, while order 2 animals were fasted, and then fed. Animals were allowed 60 hr to acclimate between treatments. Serum and plasma were obtained at 20 min intervals and 60 min, respectively, for 6 hr after feeding and for the last 6 hr of a 30 hr fast. Serum was assayed for insulin, IGF-I, GH, and cortisol (total and free). Plasma was assayed for PUN and NEFA. Mean insulin (ng/ml) differed between fed (.95 +/- .08) and fasted (.26 +/- .08) animals (P less than 01). Both mean total and free cortisol (ng/ml) were lower in fed (11.48 +/- .99) (1.06 +/- .12) than in fasted (17.10 +/- .93) (1.62 +/- .12) animals, respectively (P less than .01). Animals in order 1 differed in mean IGF-I (ng/ml) between fed (199.0 +/- 8.0) and fasted (116.5 +/- 7.2) treatments (P less than .01). Mean IGF-I for animals in order 2 was 146.7 +/- 7.2 in fed and 213.9 +/- 7.2 in fasted animals (P less than .01). Mean GH did not differ between treatments. Mean PUN and NEFA were higher in fasted than in fed animals (P less than .01). Abstract.— Juvenile white bass Morone chrysops (age 158 d, 110.4 mm total length) were reared at salinity levels of 0, 4, 8, 12, 16, and 20 parts per thousand (ppt). Fish (10/tank) were stocked in 64-L tanks with four replicates/treatment ( N = 40heatment) and fed a pelleted feed daily. Fish were sampled semimonthly for 70 d. Fish reared at salinities of 12 ppt and below were significantly ( P ≤ 0.05) larger (mean = 151.5 mm total length; range 146.8–155.7 mm) than fish reared at salinities of 16 and 20 ppt (mean 133.7 mm total length; range 131.5–135.8 mm) after 70 d. Survival was also higher ( P ≤ 0.05) among fish reared at salinities of 12 ppt or lower (mean 85.6%; range 77.5%–92.5%) compared to those reared at the two highest salinities (42.5% survival at 16 ppt; 5.0% survival at 20 ppt). Thus, although typically found in freshwater habitats, white bass can survive and grow in brackish water.  相似文献
