The behavior of β-lactoglobulin (β-Lg) during heat treatments depends on the environmental conditions. The influence of the presence or absence of a reducing sugar, namely, glucose, on the modification of the protein during heating has been studied using fluorescence, polyacrylamide gel electrophoresis (PAGE), size-exclusion chromatography (SEC), and transmission electron microscopy. Glycated products were formed during heating 24 h at 90 °C and pH 7. The fluorescence results revealed an accumulation of the advanced Maillard products and the formation of aggregates during heating. PAGE and SEC data suggested that the products in the control samples were essentially composed of covalently linked fibrillar aggregates and that their formation was faster than that for glycated samples. We showed that glucose affected the growing step of covalent aggregates but not the initial denaturation/aggregation step of native protein. Glucose-modified proteins formed a mixture of short fibrils and polydisperse aggregates. Our results revealed that β-Lg forms fibrils at neutral pH after heating and that glucose slows the formation of these fibrils.  相似文献

The lipid profiles of wheat flour doughs containing exogenous lipase were studied under different mixing conditions using a microscale mixer. An experimental design comparing the effects of dough water content (52–68%), the speed of mixing (50–100 rpm), and the mixer temperature (18–32°C) showed that the hydrolysis levels were positively influenced by temperature and speed of mixing and negatively influenced by water content. The positive effect of temperature was enhanced both by highspeed mixing and low water content. The lipid oxidation levels were positively influenced by the speed of mixing and negatively influenced by the water content. The positive effect of temperature on the oxidation levels was less important. A series of experiments conducted with different types of industrial and semi-industrial mixers with equal concentrations of lipase added to the dough showed large differences among the rates of lipid hydrolysis and oxidation. However, the mixing conditions proposed by bakers to obtain doughs with similar handling properties led to similar dough lipid profiles. Sodium chloride did not change the lipid profile when added to dough. Conversely, calcium chloride promoted a large increase of lipid hydrolysis and oxidation due to its activation of lipase activity. Addition of yeast increased the lipid hydrolysis and slightly decreased lipid oxidation.  相似文献

In control dough, endogenous wheat lipase was inactive, because the triacylglycerol (TAG), 1,2-diacylglycerol (DAG1,2), and 1,3-diacylglycerol (DAG1,3) fractions of nonpolar lipids were not affected by mixing. Conversely, the free fatty acid (FFA) and monoacylglycerol (MAG) fractions decreased, mainly due to the oxidation of polyunsaturated fatty acids (PUFA) catalyzed by wheat lipoxygenase. Addition of exogenous lipase to flour (15 lipase units [LU] per gram of dry matter) resulted in substantial modification of nonpolar lipids during dough mixing. Due to the 1,3 specificity of the lipase used in this experiment, the TAG and DAG1,3 fractions decreased, whereas the MAG and FFA fractions increased. The DAG1,2 fraction increased at the beginning of mixing and decreased after 40 min of mixing. Moreover, part of the PUFA released by lipase activity was oxidized by wheat lipoxygenase, resulting in major losses of PUFA. Conversely, the net content of the saturated and monounsaturated fatty acids (SMUFA) remained constant, because the free SMUFA content increased primarily at the expense of the esterified forms. For a constant mixing time of 20 min, increasing the amount of lipase added to dough (from 2.5 to 25 LU/g of dry matter) resulted in a linear decrease in the TAG fraction and a linear increase in the SMUFA content in the FFA fraction. At the same time, the PUFA content of the FFA fraction increased only for additions of lipase to flour of >5 LU/g of dry matter, due to partial oxidation by wheat lipoxygenase.  相似文献

Seedlings of two sympatric oak species, Quercus robur L. and Quercus petraea (Matt.) Liebl., were grown in common garden conditions to test for potential interspecific differences in intrinsic water-use efficiency (WUE). Intrinsic water-use efficiency was estimated based on carbon isotope composition of shoots (delta13C) and on gas exchange measurements (ratio of net CO2 assimilation rate to stomatal conductance (A/g(sw))). In addition, genotype x environment interactions were tested by subjecting the seedlings to four irradiance treatments (8, 18, 48 and 100% of incident solar irradiance) imposed by neutral shading nets, and, in the 100% irradiance treatment, two watering regimes. In all treatments, initial growth of Q. robur was faster than that of Q. petraea. In both species, there was a tight correlation between delta13C and A/g(sw). Intrinsic water-use efficiency increased with increasing irradiance (almost doubling from 8 to 100% irradiance), and this effect paralleled the increase in A with increasing irradiance. In full sun, WUE of Q. petraea seedlings was 10-15% higher than in Q. robur seedlings, with the difference attributable to a difference between the species in g(sw). The interspecific difference in WUE was maintained during drought, despite the appreciable increase in WUE and decrease in growth imposed by drought. No interspecific differences in WUE were observed at low irradiances, suggesting a strong genotype x environment interaction for WUE. These findings confirm the existence of interspecific genetic differences in WUE, but also show that there is large intraspecific variability and plasticity in WUE. The initially greater height and biomass increments in Q. robur seedlings illustrate the ability of this species to out-compete Q. petraea in the early stages of forest regeneration. For adult trees growing in closed canopies, the high WUE of Q. petraea may contribute significantly to its survival during dry years, whereas the low WUE of Q. robur may account for the frequently observed declines in adult trees of this species following drought.  相似文献

In ecology, tradeoff theory has been used to understand differences among plant species in their competitive abilities. In efforts to develop weed-suppressive soybean, we found evidence of a tradeoff between the ability for rapid initial growth and the ability for sustained growth later in the season: early maturing lines displayed more rapid initial growth but ceased growth sooner. Such a tradeoff would increase the difficulty of obtaining a full-season weed-suppressive variety. To determine this tradeoff's existence and severity we examined two possible mechanisms that could lead to it. We tested whether early maturing soybean lines attain higher early relative growth rate than late-maturing soybean lines and whether early maturing soybean lines produce larger seeds by an environmentally-dependent or -independent mechanism. Early maturing lines had higher relative stem elongation rates than late lines but not higher relative dry weight or leaf area increase rates. In more northern locations and in years with shorter growing seasons, early maturity lines produced larger seeds than late maturing lines, implicating an environmentally-dependent rather than -independent mechanism causing seed size differences. Relative to early lines, when late lines mature, temperature and photoperiod are in greater decline, leading to a risk of incomplete seed fill. Resulting seed size decreases could lead to lower initial growth in late maturity lines and thus to an environmental rather than genetic or physiological cause of tradeoff between initial growth and sustained growth later in the season. We evaluated the effect of heat challenge on cell viability, concanavalin A-induced proliferation and heat shock protein (HSPs) mRNA expression in peripheral mononuclear blood cells (PBMC) isolated from Creole (CR) and Large White (LW) pigs. The PBMCs were cultured for 9 h at 37°C before being subjected to heat challenge: (1) at 42°C or 45°C for 2, 4, 6 and 9 h to monitor cell viability;(2) at 45°C for 2 and 9 h followed by stimulation for 24 h at 37°C with concanavalin A to evaluate mitogen-induced proliferation; and (3) at 45°C for 3, 6 and 9 h to measure induction of HSP70.2 and HSP90 mRNA. Cell viability was affected by breed and temperature (P < 0.01), and the viability decrease caused by heat challenge was greater for LW than CR pigs. For mitogen-stimulated PBMCs, incubation at 45°C reduced lymphoblastogenesis equally in both breeds (P < 0.01). Although heat challenge for 3 and 6 h at 45°C induced expression of HSP70.2 and HSP90 mRNA, no breed difference was observed. In conclusion, differences in heat resistance between these two breeds at the whole organism level are reflected at the cellular level. Neither HSP70.2 nor HSP90 mRNA expression levels explain this effect.  相似文献

Vegetation structure parameters are key elements in the study of ecosystem functioning and global scale ecosystemic interactions. The detailed retrieval of many of these parameters by direct measurements is impractical due to the quantity of plant material in trees. Terrestrial LiDAR Scanners (TLSs) have been shown to hold great potential as an indirect means of estimating plant structure parameters with a high level of detail, while some studies identified a number of challenges inherent to this approach. In this study we investigate the use of a voxel-based approach to retrieve leaf area distribution of individual trees. The approach is based on the contact frequency method applied to co-registered TLS returns from two or more scanning positions. The contact frequency was computed for voxels being 10, 30, and 50 cm in size and subsequently corrected for the influence of occlusion effects, leaf inclination, the presence of non-photosynthetic material, and the laser beam size. The leaf area of voxels for which occlusion effects were too pronounced was estimated using modeled values based on the availability of light. We compared the TLS derived leaf area estimates against direct measurements, obtained by the harvesting of leaves, in a broad-leaved savanna of central Mali. The measured leaf area values of the sampled trees ranged from 30 to 530 m2, and crown LAI values between 0.8 and 7.2. The leaf area estimates lay on average 14% from the reference measurements (general bias). Our method provides vertical as well as radial distributions of leaf area in individual trees, and lends itself to the estimation of savanna vegetation structural parameters with a high level of detail.  相似文献

The purpose of this study was to describe the computed tomographic (CT) features of nasal aspergillosis in dogs. Initial (n = 35) and follow-up (n = 12) CT images were available from 35 dogs. The most commonly encountered CT findings were (1) moderate to severe cavitary destruction of the turbinates with presence of a variable amount of abnormal soft tissue in the nasal passages, (2) non-specific thickening of the mucosa adjacent to the inner surface of bones of the frontal sinus, maxillary recess and nasal cavity and, (3) thickened reactive bone. The findings were consistent with a disease initially affecting one nasal cavity then progressing into the ipsilateral frontal sinus, the contralateral nasal cavity and the contralateral frontal sinus. Two dogs with associated nasal foreign body had a more localized invasion of the nasal cavity. Attenuation values and contrast enhancement were not specific. With follow-up examinations, a reduction in the amount of abnormal soft tissue was observed in all dogs except one, but this reduction could not be quantified.  相似文献
