We previously induced protective immune response by oral immunization with yeast expressing the ApxIIA antigen. The ApxI antigen is also an important factor in the protection against Actinobacillus pleuropneumoniae serotype 5 infection; therefore, the protective immunity in mice following oral immunization with Saccharomyces cerevisiae expressing either ApxIA (group C) or ApxIIA (group D) alone or both (group E) was compared with that in two control groups (group A and B). The immunogenicity of the rApxIA antigen derived from the yeast was confirmed by a high survival rate and an ApxIA-specific IgG antibody response (p < 0.01). The highest systemic (IgG) and local (IgA) humoral immune responses to ApxIA and ApxIIA were detected in group E after the third immunization (p < 0.05). The levels of IL-1β and IL-6 after challenge with an A. pleuropneumoniae field isolate did not change significantly in the vaccinated groups. The level of TNF-α increased in a time-dependent manner in group E but was not significantly different after the challenge. After the challenge, the mice in group E had a significantly lower infectious burden and a higher level of protection than the mice in the other groups (p < 0.05). The survival rate in each group was closely correlated to the immune response and histopathological observations in the lung following the challenge. These results suggested that immunity to the ApxIA antigen is required for optimal protection.  相似文献

The expression of caveolin-1 and -2 in the retina was examined; Western blot analysis showed that both were present. Immunohistochemistry indicated that caveolin-1 was expressed in the majority of retinal layers, including the ganglion cell layer, inner plexiform layer, outer plexiform layer, and in the vascular endothelial cells of the retina. Caveolin-2 was primarily immunostained in the vessels, but in a few other elements as well. This is the first demonstration of caveolin differential expression in the retina of rats, and suggests that caveolin plays an important role in signal transduction in glial cells and neuronal cells.  相似文献

The gene constitution of polymorphisms of the four calpain genes (µ‐calpain, m‐calpain, p94, and µ/m‐calpain) were analyzed in South‐East Asian native chickens, White Leghorn and Broiler commercial chickens, and Red and Green jungle fowl. Polymorphisms were detected at all loci in chickens and Red jungle fowl, but only for CAPN1 (µ‐calpain gene) in Green jungle fowl. CAPN2 and CAPN1.5 are linked on chicken chromosome 3, and the genotype for these loci were treated as haplotype. Some combinations of calpain loci were tested using principal component analysis, and the best combination (CAPN1, CAPN3, and CAPN1.5) was determined. The proportion of polymorphic loci (Ppoly) and heterozygosity (H?) were 1.00 and 0.316–0.465 in domestic chickens and red jungle fowl, and 0.33 and 0.137 in Green jungle fowl, respectively. GST values suggested that the degree of subdivision among native chickens was relatively low except for Thailand, which was highest. Pair‐wise FST testing, dendrogram and principal component analysis from the results of calpain loci showed that the four South‐East Asian native and commercial chicken populations were close genetically.  相似文献

To obtain the basic information on fruit set regulation, effects of several RNases including S-RNase on pollen tube growth and RNA degradation in the tube were studied in the pear. Purified S-RNase from the Japanese pear ‘Kosui’ (S4S5) predominantly inhibited the growth of ‘Kosui’ pollen tubes (self) in vitro at 0.28 unit μL−1, but it inhibited ‘Chojuro’ (S2S3) pollen (cross) only slightly. The same unit of RNase T1 (EC 3.1.27.3) clearly inhibited the pollen tube growth, but the action was significantly weaker than that of the S-RNase against the self-pollen. Inhibitory effect of RNase T2 (EC 3.1.27.1) and RNase A (EC 3.1.27.5) was only slight. The proteins other than the S-RNase extracted from pear style did not have any inhibitory action, though they possessed RNase activity 3.8 times higher than S-RNase. Thus, RNases tested here could not substitute for the S-RNase in specific inhibition against the self-pollen tube growth. Total RNA degradation by each RNase occurred in the pollen tubes as following order; S-RNase (self) ≥T1 > T2 ≥ A > S-RNase (cross). Degradation degree of 28S and 18S rRNA was as follows; S-RNase (self) > A > T1 > T2 > S-RNase (cross). The degradation of 5.8S and 5S rRNA was; S-RNase (self) > S-RNase (cross) > A > T2 > T1. The degree of rRNA degradation was, thus, not always in parallel with the degree of pollen growth inhibition. The S-RNase may degrade not only rRNA but also mRNA essential for pollen tube growth, and may be specifically adapted to inhibit the growth of self-pollen tubes. Therefore, controlling S-RNase amount in the style will produce self-thinning cultivars efficiently, which are unnecessary not only for hand-pollination but fruit-thinning practices in the pear. Practically, cultivar with weak self-incompatibility and small amount of S-RNase, such as ‘Okusankichi’, may be an expecting candidate for breeding self-thinning cultivars.  相似文献

Across the physiognomic types of the Orinoco llanos, periodic inventories and changes in land-use between 1982–1992 are estimated. Results indicate that the area under pastures and forest plantations is increased by 0.005337×106 km2, whilts reducing the area of croplands by 0.000119×106 km2. This is a net increase of 0.005218×106 km2. The gross carbon release is 174.66 Tg C per year to the atmosphere and transferring from cultivated and native vegetation to wood products (1.62 Tg C per year) and slash (1.18 Tg C per year). The processes of land preparation contribute 1.40 Tg C per year to the atmosphere. From the tree savannas, woodlands and forests 0.73 Tg C per year are estimated to have been transferred to the soil following clearance and burning over this period, and 1.05 Tg C per year from herbaceous savannas when were buried and decomposed at 0.84 Tg C per year. The estimate of carbon balance here by inventories and changes in land-use approach indicates that the Orinoco llanos is a sink of −17.53 Tg C per year. The carbon turnover time in the Orinoco system is 68 years, which provides a limited route for carbon sequestration. The calculated potential of the Orinoco llanos for storing carbon is 8300 Tg C. Ecological options to achieve this potential value are addressed. However, nutrient deficiency and seasonal water supply are serious drawbacks to take into account for increasing carbon accretion. These results are particular for the Orinoco llanos, even though described processes could be similar to world-wide savannas, where a gradient of carbon heterogeneity exists.  相似文献

Japanese red pine (Pinus densiflora) seedlings taken from a nursery were grown in liquid culture with all roots entirely submerged in water. The aims of the present study were to observe the reactions of preformed ectomycorrhizae to liquid culture and to examine new infections by ectomycorrhizal fungi on newly formed root tips in liquid culture. Inoculation levels were controlled by trimming the ectomycorrhizal roots to one of three selected root lengths. The results showed that the mantles of pre-formed ectomycorrhizae were lost during the 8 weeks of the liquid culture and that these preformed root tips became blackish and wrinkled, but Hartig nets remained in these blackish root tips. On the newly formed lateral root tips, no ectomycorrhizal mantles were formed, although Hartig nets were found. In addition, the level of inoculum did not affect the frequency of Hartig nets in newly formed root tips, suggesting that the density of inoculum was more important than the amount of inoculum for new infection. In conclusion, ectomycorrhizal fungi endured and infected new roots while submerged; however, only Hartig nets were formed and not mantles.  相似文献

This article analyzes the relationship between forest resources, refugees, and the host population. The findings of the research suggest that the host population are heavily dependent on the local forest for their daily needs such as fuelwood, timber, grazing area, fodder for domestic animals, foods, and medicine in addition to cultural and esthetic needs. The forest has also been relied upon for agricultural needs such as manufacture of agricultural tools, maintenance of irrigation water systems, erosion control, and fertilizer needs. The forest was under a sustained demand as any other Terai forest of Nepal. After the arrival of refugees in 1992, the demand for forest resources increased substantially. Initially, the construction of the refugee camps decreased the total forest area and also required some felling of trees. More significantly, the refugees themselves became active users of the forest resource, which generated extra pressure on the forest and created scarcity of forest resources. Before the arrival of the refugees, forest management and monitoring of illegal use of the forest resources were carried out by the government through its local forester office. The local residents were active users of the forest resources, but were passive in managing and maintaining the forest resource. However, competition from the refugees instilled a desire in the local population to safeguard and protect the dwindling resource against the external threat by creating the Humse Dumse Community Forest.  相似文献

A half-scale model of a light-frame shear wall was developed to evaluate the racking performance of a fullscale shear wall (prototype). The effect of nail size on the performance of the shear wall was also investigated using models constructed with three types of nail. Materials for the model were determined through experimental methods, which included nail-head push-through, stud-to-sheathing nail connection, and static bending tests. Materials with which the model was made to be in similarity to the prototype were three-layer 4.8-mm plywood, 39.72-mm long nails, and 1 × 2 lumber cut from 2 × 4 studs. In accordance with ASTM E 72 and ASTM E 564, racking resistance tests were conducted on 20 shear walls. The results showed that the maximum load capacities of the prototype walls could be evaluated by the model without significantly different failure modes.
