Background – Meticillin‐resistant Staphylococcus aureus (MRSA) and meticillin‐resistant Staphylococcus pseudintermedius (MRSP) infections are increasingly reported in dogs, and these bacteria may be isolated from ear infections. Hypothesis/Objectives – The main aim of the present study was to investigate the in vitro antimicrobial activity of miconazole, polymyxin B and a combination of both against 24 canine MRSA and 50 canine MRSP isolates. The minimal inhibitory concentration (MIC) values of 12 other antimicrobial agents were also determined. Methods – All MIC values were determined according to a broth microdilution assay. Results – Acquired resistance was found to all tested agents, except for linezolid, miconazole and polymyxin B. The MIC values for miconazole and polymyxin B against MRSA were in the range of 4–8 and 8–64 μg/mL, respectively, while the MIC values for miconazole and polymyxin B against MRSP were in the range of 1–2 and 0.25–4 μg/mL, respectively. Using a combination of miconazole and polymyxin B, there was no evidence for enhanced in vitro activity of the combination (i.e. synergy) of both products. Nevertheless, MIC90 values of the combination of these antimicrobial agents and of a commercial product containing both agents were at least 1000 times lower than the concentration present in the commercial product. Conclusions and clinical importance – These results indicate that the topical use of a combination of miconazole and polymyxin B in a 43.5:1 ratio may have potential for the treatment of MRSA‐mediated and MRSP‐associated otitis externa in dogs. 相似文献

This paper discusses factors affecting dystocia and birth weight in Grati cattle in three villages in the Pujon district, East Java. Records of birth weight, dystocia score, village, season, parity, sex, artificial versus natural service, and age and weight of dam, were obtained from 268 parturitions. Village and weight of the dam had a significant effect on birth weight but not on dystocia. Parity had a significant effect on dystocia score. More abnormal births occurred with AI using exotic bulls. Calves from an abnormal birth were on average more than three kg heavier. First parity births combined with Al with exotic bulls had a particularly high incidence of dystocia. It is concluded that farmers in Pujon can reduce the incidence of dystocia in heifers by using local bulls only. 相似文献

Flexor enthesopathy is a recently recognized elbow disorder in dogs and considered to be an important differential diagnosis for elbow lameness. Primary and concomitant forms of the disease have been previously described and treatments differ for the two forms. The goal of this prospective study was to compare magnetic resonance imaging (MRI) findings for dogs with primary flexor enthesopathy (n = 17), concomitant flexor enthesopathy (n = 23), elbow dysplasia alone (n = 13), and normal elbows (n = 7). Each elbow joint underwent MRI using the same low‐field scanner. Sequences included transverse and sagittal T1‐weighted (before and after IV contrast), transverse and sagittal T2‐weighted, and dorsal STIR. For each elbow, MRI lesions were recorded based on a consensus of two observers unaware of group status. Magnetic resonance imaging lesions involving flexor tendons were found in 100% of clinically affected joints with primary flexor enthesopathy and 96% of clinically affected joints with concomitant flexor enthesopathy. Thickened flexor muscles were the most common lesions, followed by hyperintense tendon signal and contrast enhancement. Irregular, thickened medial humeral epicondyle, edema, and calcified body lesions were less frequently observed. Magnetic resonance imaging characteristics of flexor enthesopathy were not found in normal joints or those affected by elbow dysplasia alone. No significant differences in frequencies and details of individual MRI characteristics were found between primary and concomitant flexor enthesopathy groups. Findings indicated that MRI is a sensitive technique for detection of flexor enthesopathy lesions in dogs, however, MRI characteristics do not allow differentiation of primary versus concomitant forms of the disease. 相似文献

Flexor enthesopathy is an important differential diagnosis for elbow lameness in dogs. The disorder can be a primary cause of elbow lameness or concomitant with other elbow pathology. Since treatment differs for primary and concomitant forms of flexor enthesopathy, a noninvasive method for distinguishing between them is needed. In the current prospective study, computed tomographic (CT) examination was performed before and after IV injection of contrast in 17 dogs with primary flexor enthesopathy, 24 dogs with concomitant flexor enthesopathy, 13 dogs with elbow dysplasia, and seven normal dogs. Dogs were assigned to groups based on results of clinical examination and at least three other imaging modalities. Computed tomographic lesions consistent with flexor enthesopathy were found in all clinically affected joints with primary flexor enthesopathy and in 29 of the 30 clinically affected joints with concomitant flexor enthesopathy. Those lesions were not found in sound elbows or joints affected by elbow dysplasia. Flexor lesions detected in dogs with primary flexor enthesopathy were not significantly different from those detected in dogs with the concomitant form. Findings indicated that CT can be applied to detect flexor enthesopathy, but a distinction between the primary and concomitant forms was not always possible. Land degradation and deforestation of natural forests are serious issues worldwide, potentially leading to altered land use and carbon storage capacity.Aims
Our objectives were to investigate if short-rotation plantations can restore carbon pools of degraded soils, without altering soil fertility.Methods
Carbon and nutrient pools in above- and below-ground biomass and soils were assessed using stand inventories, harvested biomass values, allometric relationships and selective sampling for chemical analyses.Results
Carbon pools in the total ecosystem were low in the degraded land and in croplands (6–13 Mg ha?1) and high in forests (66–94 in eucalyptus plantations; 86–126 in native forests). The soil nutrient status of eucalyptus stands was intermediate between degraded land and native forests and increased over time after eucalyptus introduction. All harvest scenarios for eucalyptus are likely to impoverish the soil but, for the moment, the soil nutrient status has not been affected.Conclusion
Afforestation of degraded land with eucalyptus can be a useful restoration tool relative to carbon storage and soil fertility, provided that non-intensive forestry is applied.86.

BACKGROUND: The codling moth, Cydia pomonella (L.), is one of the most important pests of apple worldwide. Use of insecticides for management of this insect has been extensive and has resulted in resistance development. There are a number of different bioassay methods to monitor for codling moth resistance; however, many are not applicable to new insecticides and most are time consuming. A novel 16‐well plasticware bioassay plate containing lyophilized diet was developed for rapid resistance monitoring of codling moth. RESULTS: The contact insecticides acetamiprid and azinphosmethyl were significantly more toxic to neonates than to fourth instars. However, there was no significant difference in LC50 values between neonates and fourth instars to the ingestion insecticides chlorantraniliprole, methoxyfenozide, novaluron and spinetoram. Field colonies of codling moth were significantly more resistant to methoxyfenozide than susceptible populations. A diagnostic dose of 20 µg mL?1 (LC99) was established to monitor for codling moth resistance to methoxyfenozide. CONCLUSIONS: The results presented here demonstrate that a novel and rapid bioassay can be used to monitor for codling moth resistance to methoxyfenozide. The bioassay method is relevant to both ingestion and contact insecticides, but a single diagnostic dose, regardless of larval age, is only relevant to ingestion insecticides. ABSTRACT In preparasitic second-stage juveniles (J(2)) of potato cyst nematode Globodera rostochiensis, six proteins with molecular masses of 30, 31a/b, 32, 39, and 49 kDa were recognized on Western blots by a monoclonal antibody (MGR48) specific for the subventral esophageal glands. All of these subventral gland proteins (svp's) focused in the basic range (pI 6.8 to 8.6) of an immobilized pH gradient. Western blotting showed that the svp's were present in preparasitic and parasitic J(2) and not in later juvenile stages and adult females. Minor svp quantities also were observed in adult males. Immunogold labeling of preparasitic J(2) showed that the svp's were localized in the rough endoplasmic reticulum and secretory granules of the subventral esophageal glands. Potato root diffusate triggered the secretion of svp's through the stylet, and 5-methoxy-N,N-dimethyltryptamine-hydrogen-oxalate had only a quantitative, additional effect. The forward flow of svp's through the metacorporal pump chamber was confirmed by the presence of svp's in the circular lumen of the esophagus (procorpus), as established by immunoelectron microscopy. Our data provide conclusive evidence that secretory proteins of the subventral glands of G. rostochiensis can be secreted through the stylet and support the hypothesis that the subventral esophageal glands play an important role in the early events of this nematode-plant interaction. 相似文献

Because carbon dioxide (CO2) concentration is rising, increases in plant biomass and productivity of terrestrial ecosystems are expected. However, phosphorus (P) unavailability may disable any potential enhanced growth of plants in forest ecosystems. In response to P scarcity under elevated CO2, trees may mine deeper the soil to take up more nutrients. In this scope, the ability of deep horizons of forest soils to supply available P to the trees has to be evaluated. The main objective of the present study was to quantify the relative contribution of topsoil horizons and deep horizons to P availability through processes governed by the activity of soil micro-organisms. Since soil properties vary with soil depth, one can therefore assume that the role of microbial processes governing P availability differs between soil layers. More specifically, our initial hypothesis was that deeper soil horizons could substantially contribute to total plant available P in forested ecosystems and that such contribution of deep horizons differs among sites (due to contrasting soil properties). To test this hypothesis, we quantified microbial P and mineralization of P in ‘dead’ soil organic matter to a depth of 120 cm in forest soils contrasting in soil organic matter, soil moisture and aluminum (Al) and iron (Fe) oxides. We also quantified microbiological activity and acid phosphomonoesterase activity. Results showed that the role of microbial processes generally decreases with increasing soil depth. However, the relative contribution of surface (litter and 0–30 cm) and deep (30–120 cm) soil layers to the stocks of available P through microbial processes (51–62 kg P ha?1) are affected by several soil properties, and the contribution of deep soil layers to these stocks vary between sites (from 29 to 59%). This shows that subsoils should be taken into account when studying the microbial processes governing P availability in forest ecosystems. For the studied soils, microbial P and mineralization of P in ‘dead’ soil organic matter particularly depended on soil organic matter content, soil moisture and, to a minor extent, Al oxides. High Al oxide contents in some sites or in deep soil layers probably result in the stabilization of soil organic compounds thus reducing microbiological activity and mineralization rates. The mineralization process in the litter also appeared to be P-limited and depended on the C:P ratio of soil organic matter. Loss on ignition (LOI) is one of the most widely used methods for measuring organic matter content in soils but does not have a universal standard protocol. A large number of factors may influence its accuracy, such as furnace type, sample mass, duration and temperature of ignition and clay content of samples. We conducted a series of experiments to quantify these effects, which enabled us to derive (i) guidelines for ignition conditions (sample mass, duration and temperature), (ii) temperature‐specific soil organic matter (SOM) to soil organic carbon (SOC) conversion factors and (iii) clay content‐dependent correction factors for structural water loss (SWL). Bulk samples of a sandy soil (4% clay) and a silt loam soil (25% clay) were used to evaluate the effects of ignition conditions. Samples with a range of clay contents (0–50%) were used to quantify conversion and correction factors. Two furnaces, one without and one with pre‐heated air, did not show significant differences in terms of within‐batch LOI variability. In both furnaces less combustion occurred close to the door, which necessitated tray turning at half‐time as this reduced the standard deviation per batch significantly. Variation in mass loss declined exponentially with sample mass (range, 0.15–20 g). The LOI increased with duration at lower temperatures (≤ 550°C) for the sandy soil. At greater temperatures (600 and 650°C), no effect of duration was found. For the silt loam soil, LOI values increased with duration for each temperature, which was attributed to SWL. The SOM to SOC conversion factor decreased strongly with temperature at an ignition duration of 3 hours from 0.70 (350°C) to 0.57 (500°C) and stabilized around 0.55 between 550 and 650°C, indicating that at temperatures ≥ 550°C all SOM had been removed. The clay correction factor for SWL increased from 0.01 to 0.09 as the temperature of ignition increased from 350 to 650°C. To minimize within‐batch LOI variation we recommend a standard ignition duration of 3 hours, tray turning at half‐time, a sample mass ≥ 20 g and temperatures equal to or greater than 550 °C. To avoid over‐estimates of SOM through structural water loss, the presented SWL correction procedure should always be applied. 相似文献

Organic farming may contribute substantially to future agricultural production worldwide by improving soil quality and pest control, thereby reducing environmental impacts of conventional farming. We investigated in a comprehensive way soil chemical, as well as below and aboveground biological parameters of two organic and two conventional wheat farming systems that primarily differed in fertilization and weed management strategies. Contrast analyses identified management related differences between “herbicide-free” bioorganic (BIOORG) and biodynamic (BIODYN) systems and conventional systems with (CONFYM) or without manure (CONMIN) and herbicide application within a long-term agricultural experiment (DOK trial, Switzerland). Soil carbon content was significantly higher in systems receiving farmyard manure and concomitantly microbial biomass (fungi and bacteria) was increased. Microbial activity parameters, such as microbial basal respiration and nitrogen mineralization, showed an opposite pattern, suggesting that soil carbon in the conventional system (CONFYM) was more easily accessible to microorganisms than in organic systems. Bacterivorous nematodes and earthworms were most abundant in systems that received farmyard manure, which is in line with the responses of their potential food sources (microbes and organic matter). Mineral fertilizer application detrimentally affected enchytraeids and Diptera larvae, whereas aphids benefited. Spider abundance was favoured by organic management, most likely a response to increased prey availability from the belowground subsystem or increased weed coverage. In contrast to most soil-based, bottom-up controlled interactions, the twofold higher abundance of this generalist predator group in organic systems likely contributed to the significantly lower abundance of aboveground herbivore pests (aphids) in these systems. Long-term organic farming and the application of farmyard manure promoted soil quality, microbial biomass and fostered natural enemies and ecosystem engineers, suggesting enhanced nutrient cycling and pest control. Mineral fertilizers and herbicide application, in contrast, affected the potential for top-down control of aboveground pests negatively and reduced the organic carbon levels. Our study indicates that the use of synthetic fertilizers and herbicide application changes interactions within and between below and aboveground components, ultimately promoting negative environmental impacts of agriculture by reducing internal biological cycles and pest control. On the contrary, organic farming fosters microbial and faunal decomposers and this propagates into the aboveground system via generalist predators thereby increasing conservation biological control. However, grain and straw yields were 23% higher in systems receiving mineral fertilizers and herbicides reflecting the trade-off between productivity and environmental responsibility. 相似文献
