The effectiveness of applying Ovaprim [(D-Arg6, Pro9NEt)-sGnRH + domperidone] and Ovopel [(D-Ala6, Pro9NEt)-mGnRH + metoclopramide] to male barbel Barbus barbus (L.) 6, 12 and 24 h after hormonal stimulation was analyzed. The control group (Control) during each time interval was stimulated with 0.9 % NaCl. Milt was collected from seven fish only once (n = 7) for Ovopel, Ovaprim and Control group in order to determine total volume of milt, volume of milt per kg of body weight, sperm concentration, total sperm production, seminal plasma osmotic pressure, pH of milt and pH of seminal plasma. Woynarovich’s solution (68 mM NaCl + 50 mM urea) with the addition of 0.5 % BSA (pH 7.7; 181 mOsm kg?1) was used as the activating liquid. Selected parameters of sperm motility (MOT %) and progressively motile sperm (%), curvilinear velocity (VCL, μm s?1), straight-line velocity (μm s?1), movement linearity (%), wobbling index (%), amplitude of lateral head displacement (μm) and beat cross frequency (Hz) were determined using the Computer-assisted sperm analysis system. A time of 6 h proved to be too short to obtain milt from barbel following hormonal stimulation with Ovaprim and Ovopel. Extending the time to 12 h, however, resulted in 100 % spermiation in males, regardless of hormonal preparation used for stimulation. The stimulation of spermiation in barbel is best performed using Ovopel 12 h upon application. Extending the latency period to 24 h following the application of this preparation results in a significant decrease in the volume of milt obtained, sperm count and motility parameters, including MOT and VCL, which may influence sperm fertilization ability.  相似文献

Twenty-four female BALB/c mice were orally inoculated with 10(8) CFU Escherichia coli ATCC 25922 and euthanized 2.5, 7, 13 and 25 h post-inoculation. The levels of organ nitric oxide (NO) and plasma endotoxin, TNF-alpha and nitrite/nitrate (NO(x)) were compared to those found in sham-inoculated mice, to evaluate systemic host-response to a low-level oral exposure to Gram-negative bacteria. Organ bacterial culture and immunohistochemistry for iNOS were performed on lungs, liver, kidneys and brain from all mice. Organ NO and plasma TNF-alpha levels were higher in E. coli-inoculated animals, but no differences were detected in plasma endotoxin levels, NO(x) or iNOS immunostaining for any of the animal groups. Single oral gavage with live E. coli stimulates an early systemic immune response in clinically healthy mice as evidenced by increased plasma TNF-alpha and organ NO levels, but bacteremia and endotoxemia are not related to this inflammatory response.  相似文献

Leaves with typical symptoms of downy mildew were found on common agrimony in the Czech Republic in 2014 and 2015 and at several locations in Germany from 2010 to 2014. The causal agent of downy mildew of agrimony was often reported as Peronospora agrimoniae, but sometimes also as P. sparsa. Morphological characteristics of the pathogens found in both countries are in the range of previous works for P. agrimoniae, but also other downy mildews parasitic on Rosaceae, rendering their discrimination based on published observations difficult. For molecular identification sequencing of several loci (nrITS rDNA, cox1 and cox2) was performed. Phylogenetic analyses based on nrITS rDNA clearly separated P. agrimoniae from other Peronospora species infecting Rosaceae. Thus, considering P. agrimoniae as separate species seems justified. Two German specimens were identical to two Czech samples in both nrITS rDNA and cox1 mtDNA sequences, but differed in a single nucleotide substitution in cox2 region. To our knowledge, this is the first verified record of P. agrimoniae on common agrimony in the Czech Republic.  相似文献

This study discusses a novel approach for fabrication of floating photocatalyst which can be efficiently exploited for photodegradation of dyes in aqueous solutions. A fabrication of the floating photocatalyst consisted of two steps: transformation of the poly(ε-caprolactone) beads (PCLb) into poly(ε-caprolactone) foam (PCLf) with porous structure in supercritical carbon-dioxide and subsequent loading of PCLf with TiO2 nanoparticles (NPs). Morphological characterization of the PCLf before and after TiO2 NPs loading was carried out by FESEM. The presence of titanium on the surface and inside the PCLf was detected by EDX. Photocatalytical activity of the floating photocatalyst was investigated in aqueous solution of textile dyes C.I. Acid Orange 7 (AO7) and C.I. Basic Yellow 28 (BY28) which were exposed to lamp that simulates the sun light. In addition to sustainable floatability for a long period of time, developed floating photocatalyst exhibited high rate of photodegradation since the complete discoloration of AO7 and BY28 solutions and photocatalysts alone occurred after 300 and 180 min of illumination, respectively. Its photocatalytic activity was preserved after three repeated photodegradation cycles with unchanged chemical structure that was confirmed by FTIR analysis.  相似文献

A previous study indicated decreased DNA content of chromosome 4A in the wheat (Triticum aestivum L. cv. Tähti) compared to cvs. Chinese Spring and Rennan. Here we show that the lower 4A DNA content is associated with a specific haplotype in the distal part of 4AL. In 41 cultivars of bread wheat (T. aestivum L.), including cv. Tähti, a common haplotype was identified in the linkage disequilibrium region on the long arm of chromosome 4A (4AL). The haplotype (haplotype A) is characterized by 7 SSR and 5 EST marker alleles, including five zero-alleles. Haplotype A was found in 46 % of the Swedish/Finnish/Estonian spring wheat genotypes, while only one of the modern wheat accessions from Germany carried the same haplotype. Fluorescent cytometry analysis linked haplotype A to diminished DNA content of chromosome 4A. The haplotype was introduced into the Canadian and US breeding programs at the beginning of the twentieth century (cvs. Marquis, Thatcher, Ruby) from the common progenitor, the Polish landrace Fife, and it is still found in modern wheat germplasm in these countries. Zero-alleles characteristic for haplotype A were also detected in several accessions of European spelt (Triticum spelta L.), and in two accessions of tetraploid Triticum timopheevii Zhuk. The presence of haplotype A in European spelt indicates the considerable antiquity of the haplotype, as it must have been inherited from the hexaploid or tetraploid parent of spelt in at least one hybridization event.  相似文献

An overview of a research on Vitis vinifera plants treated with a chemical (calcium and magnesium ions) and biological (Trichoderma viride) agents simultaneously loaded in alginate microspheres was presented. Microspheres were applied at two growth stages: before flowering and berries pea-size. Physicochemical characteristics of leaves after the two growth stages and grapes were measured in terms of bioactive components content and antioxidant activity. After the treatments, vine leaves reached a significant increase in almost all measured parameters (polyphenols, antioxidant capacity, β-carotene, and chlorophyll) compared to the control. The highest total chlorophyll content was found after the treatment with microspheres containing Mg2+/Ca2+ cations, and T. viride. The treatments enhanced Vitis vinifera leaves in terms of bioactive potential and can be further used as a functional food. As compared to the control, somewhat elevated values of total polyphenols and antioxidant activity were found on the grape samples.  相似文献

Leaf senescence in a recent maize (Zea mays L.) hybrid is delayed relative to that in an older maize hybrid and the trait is associated with an improvement of the ratio of assimilate supply (i.e., source) and demand (i.e., sink) during grain filling. This study examined whether effects of source : sink ratio of leaf longevity in an old and more recent hybrid are associated with changes in leaf nitrogen (N) concentration and N uptake during grain filling. A 3-year field study was conducted with maize hybrids Pride 5 (old) and Pioneer 3902 (recent) grown at two soil-N levels: 150 kg−1 N ha−1 was broadcast in the high N treatment while none was added to the low N treatment. Four imposed source : sink treatments ranged from partial defoliation to no grain. Leaf N of the control treatments did not differ between the two hybrids, but the decline in leaf N from the control to the no-sink treatment was larger for Pioneer 3902 than for Pride 5. Total N uptake in above-ground portions was 10 and 18% greater in the new than in the old hybrid under low and high soil-N conditions, respectively. The difference in the total N uptake between the two hybrids could be attributed to post-silking N uptake. The proportion of N in the grain derived from post-silking N uptake was 60% for Pioneer 3902 and 40% for Pride 5 and this proportion was positively associated with the source : sink ratio. Higher rates of N uptake in Pioneer 3902 vs. Pride 5 appear to be, in part, the result of higher rates of dry matter accumulation of the newer hybrid during grain filling.  相似文献

The effects of temperatures and growth regulators on deformations in tomato fruits, a common occurrence in winter, were examined in greenhouses and in controlled-climate conditions with cultivars ‘Arava’ and ‘Orith’. The malformations could be attributed both to low-temperature conditions during flower development and to the use of growth regulators. Deformations caused by low temperatures differed from those caused by growth regulators. As a result of flower development under low-temperature conditions, fasciated fruits, fruits with a navel-like scar at the blossom end, puffy (hollow) fruits, and seedless fruits were obtained, where as with growth regulators there were more seedless fruits, sharply elongated blossom ends and green jelly. The 2 cultivars tested differed in their susceptibility to low-temperature conditions and growth regulators.  相似文献
