Plant health regulations to prevent the introduction and spread of Phytophthora ramorum and P. kernoviae require rapid, cost effective diagnostic methods for screening large numbers of plant samples at the time of inspection. Current on-site techniques require expensive equipment, considerable expertise and are not suited for plant health inspectors. Therefore, an extensive evaluation of a commercially available lateral flow device (LFD) for Phytophthora species was performed involving four separate trials and 634 samples. The assay proved simple to use, provided results in a few minutes and on every occasion a control line reacted positively confirming the validity of the test. LFD results were compared with those from testing a parallel sample, using laboratory methods (isolation and real-time PCR). The diagnostic sensitivity of the LFD (87·6%) compared favourably with the standard laboratory methods although the diagnostic specificity was not as stringent (82·9%). There were a small number ( n = 28) of false negatives, but for statutory purposes where all positive samples must be identified to species level by laboratory testing, overall efficiency was 95·6% as compared with visual assessment of symptoms of between 20-30% for P. ramorum and P. kernoviae . This work demonstrates the value of the LFD for diagnosing Phytophthora species at the time of inspection and as a useful primary screen for selecting samples for laboratory testing to determine the species identification. 相似文献

An asymptomatic adult female rabbit had intestinal cryptosporidiosis. The ileum had blunted villi, a decrease in villus-crypt ratio and a mild edema in the lamina propria. Transmission electron microscopy showed the parasite to be a Cryptosporidium similar to those reported in mouse, guinea pig, lamb, calf, horse, monkey and man. This organism is referred to as Cryptosporidium cuniculus. Scanning electron microscopy on ileal mucosa showed altered intestinal microvilli in the attachment of the cryptosporidia. It is postulated that the organism was enveloped by the microvilli of the ileal epithelial cells which then fused and formed a continuous double membrane around the parasite. 相似文献

OBJECTIVE: To determine whether vaccination with a monovalent Leptospira borgpetersenii serovar Hardjo strain hardjobovis vaccine coupled with parenteral long-acting oxytetracycline treatment would improve the reproductive performance of beef cows. DESIGN: Randomized controlled field trial. ANIMALS: 1,446 Angus-cross beef cows in 8 locations. PROCEDURES: Cows were given 2 doses, 4 weeks apart, of vaccine (treatment; n=741) or placebo (control; 705). Cows in the treatment group were also given a single dose of a long-acting oxytetracycline formulation (20 mg/kg [9.1 mg/lb], SC) at the time of booster vaccination. Estrus synchronization was performed, followed by fixed-time artificial insemination; bulls were introduced 14 days after insemination. Serum and urine samples were obtained on a random subset of cows (n=194) for detection of serologic titers and leptospiruria, and locations were categorized as positive or negative for leptospirosis on the basis of detection of leptospiruric cows. RESULTS: No differences in pregnancy and calving rates between treatment and control groups were detected. Pregnancy rate following artificial insemination was lower among cows in locations positive for leptospirosis than among cows in locations negative for leptospirosis; however, overall pregnancy rate, calving rate following artificial insemination, and overall calving rate did not differ between groups when cows were grouped on the basis of location leptospirosis status. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that under the conditions of the present study, administration of a monovalent Leptospira vaccine in conjunction with systemic administration of oxytetracycline did not significantly improve reproductive performance of beef cows. 相似文献
