The common bean (Phaseolus vulgaris L.) is widely cultivated in Brazil and is known as a very important crop for families in this country. Fusarium wilt severely harms common beans and has become a big issue for this crop. In order to assist the breeding programs that target resistance to this disease, the evaluation of genetic diversity of the pathogen and its molecular characterization are crucial. Thus, the present goal was to identify Fusarium isolates obtained from several places in Brazil using molecular tools; select molecular markers for these isolates; and analyze their diversity. All of isolates were molecularly identified as Fusarium oxysporum f. sp. phaseoli (Fop). By using seven selected SSR markers, the results of diversity obtained by the dendrogram and the Bayesian analysis formed four groups where a large diversity of this fungus was found within each state. However, the groups were more homogenous according to the collection source and the pathogenicity test. More specifically, group 2 was composed of the most virulent strains and originated from Minas Gerais State – UFV, and group 3 was mostly composed by isolates from Goias state. Group I was also more diverse in terms of location and virulence. The overall results indicated a positive correlation between Fusarium diversity and its virulence to common bean. Furthermore, the use of these markers was effective in molecular identification and in detecting polymorphism within F. oxysporum f. sp. phaseoli.  相似文献

BACKGROUND: Key climatic factors influencing the transport of pesticides to drains and to depth were identified. Climatic characteristics such as the timing of rainfall in relation to pesticide application may be more critical than average annual temperature and rainfall. The fate of three pesticides was simulated in nine contrasting soil types for two seasons, five application dates and six synthetic weather data series using the MACRO model, and predicted cumulative pesticide loads were analysed using statistical methods. RESULTS: Classification trees and Pearson correlations indicated that simulated losses in excess of 75th percentile values (0.046 mg m(-2) for leaching, 0.042 mg m(-2) for drainage) generally occurred with large rainfall events following autumn application on clay soils, for both leaching and drainage scenarios. The amount and timing of winter rainfall were important factors, whatever the application period, and these interacted strongly with soil texture and pesticide mobility and persistence. Winter rainfall primarily influenced losses of less mobile and more persistent compounds, while short-term rainfall and temperature controlled leaching of the more mobile pesticides. CONCLUSIONS: Numerous climatic characteristics influenced pesticide loss, including the amount of precipitation as well as the timing of rainfall and extreme events in relation to application date. Information regarding the relative influence of the climatic characteristics evaluated here can support the development of a climatic zonation for European-scale risk assessment for pesticide fate.  相似文献

Total extract from the fruit of Szechuan pepper (Zanthoxylum piperitum DC), the volatile components of the extract and a non‐volatile fraction containing alkylamides (NVA fraction) are feeding deterrents for rats. The present study investigated the effectiveness of these natural repellents in prairie voles (Microtus ochrogaster Wagner). Two‐choice feeding trials were conducted during which food‐deprived voles were offered choices between oat‐bran wafers. In Experiment 1, 10 voles were given three sets of feeding trials, each 2 h long. Baseline consumption was established during the first set of two trials by offering a choice between two oat‐bran wafers dipped in ethanol, the control solvent. During the second set of two trials the voles were given a choice between an oat‐bran wafer dipped in ethanol and a wafer dipped in Zanthoxylum extract. During the third set the voles were given a choice between a wafer served on top of a screened dish containing a sample of ethanol and a wafer served on top of a dish containing a sample of extract. In this manner the voles were exposed to volatile compounds emanating from the extract but could not contact it. Wafers dipped in extract were almost completely avoided. The volatile components of extract also significantly reduced food intake. In Experiment 2, habituation to the volatile constituents of extract was examined in 16 Zanthoxylum‐naïve voles. Baseline consumption was established by offering two wafers served on top of screened dishes containing ethanol. This was followed by twelve tests during which a choice between a wafer served above a sample of ethanol and a wafer served above a sample of extract was given. The voles failed to habituate to the volatile components of extract, consistently consuming less of the wafers served above extract. In Experiment 3 a dose‐response curve to Zanthoxylum extract was established, using 12 stimulus‐naïve voles. After baseline consumption was established, the animals were given two tests each, presenting a choice between a control wafer and a wafer dipped in a dilution of extract (0.001–100 g liter?1). Only concentrations of 10 and 100 g liter?1 reduced food intake. In Experiment 4 the effects of the non‐volatile fraction of extract were compared to those of whole extract. Vegetable oil was used as solvent. Eight stimulus‐naïve voles were given two tests with a choice between an oil‐dipped and an extract‐dipped wafer. A second group of eight voles received two tests with a choice between an oil‐dipped and NVA‐dipped wafer. Extract‐dipped wafers were avoided, but the NVA fraction had no effect on food consumption. With the worldwide increase of fisheries, fish wastes have had a similar increase, alternatively they can be seen as a source of novel substances for the improvement of society’s wellbeing. Elasmobranchs are a subclass fished in high amounts, with some species being mainly bycatch. They possess an endoskeleton composed mainly by cartilage, from which chondroitin sulfate is currently obtained. Their use as a viable source for extraction of type II collagen has been hypothesized with the envisaging of a biomedical application, namely in biomaterials production. In the present work, raw cartilage from shark (Prionace glauca) and ray (Zeachara chilensis and Bathyraja brachyurops) was obtained from a fish processing company and submitted to acidic and enzymatic extractions, to produce acid-soluble collagen (ASC) and pepsin-soluble collagen (PSC). From all the extractions, P. glauca PSC had the highest yield (3.5%), followed by ray ASC (0.92%), ray PSC (0.50%), and P. glauca ASC (0.15%). All the extracts showed similar properties, with the SDS-PAGE profiles being compatible with the presence of both type I and type II collagens. Moreover, the collagen extracts exhibited the competence to maintain their conformation at human basal temperature, presenting a denaturation temperature higher than 37 °C. Hydrogels were produced using P. glauca PSC combined with shark chondroitin sulfate, with the objective of mimicking the human cartilage extracellular matrix. These hydrogels were cohesive and structurally-stable at 37 °C, with rheological measurements exhibiting a conformation of an elastic solid when submitted to shear strain with a frequency up to 4 Hz. This work revealed a sustainable strategy for the valorization of fisheries’ by-products, within the concept of a circular economy, consisting of the use of P. glauca, Z. chilensis, and B. brachyurops cartilage for the extraction of collagen, which would be further employed in the development of hydrogels as a proof of concept of its biotechnological potential, ultimately envisaging its use in marine biomaterials to regenerate damaged cartilaginous tissues.  相似文献

The effects of cryopreservation on the viability, morphology and capability of spermatozoa in Atlantic cod, Gadus morhua L., were studied. The sperm was cryopreserved in straws using Hanks' balanced salt solution, hens' egg yolks and glycerol in the vapor of liquid nitrogen. Straws of cryopreserved sperm were stored in liquid nitrogen and thawed in seawater (35 C) for 8 sec before use. The motility of cryopreserved sperm was low (range 8–19%) compared to motility before freezing (range 69–76%). The fertilization rate (range 94–95%) in control groups using fresh sperm was significantly higher (P < 0.05) than in test groups (range 48–72%). In cryopreserved sperm, a relatively high percentage (range 82–93%) of the spermatozoa had changes in morphology. Many spermatozoa had no mitochondria; when mitochondria were present, the observed number varied from one and five in cryopreserved spermatozoa, and from two and seven in noncryopreserved spermatozoa. In groups where cryopreserved sperm was used, the hatching rate was lower (range 18–38%) than in control groups (range 41–63%), indicating higher mortality during embryonic development. Paternal effects on progeny performance were noted in the proportion of abnormalities but no negative effects were identified in newly hatched larvae produced using cryopreserved sperm.  相似文献

Summary It is shown in biometrical studies performed for two years and concerning the statistical diversity of fruitshape within one plant having two fruits that fruitshape is inherited as a reaction norm, the mother fruit being without influence on the progeny. The shape of the first fruit is extremely modifiable. Realization of the various fruit shapes is influenced at the end of the vegetative period by the position of the fruit on the plant, by weather, by infection and other environmental conditions. Experiments were done with someCucurbita pepo convar.patissonina Greb. inbred lines of the same origin.
