Previous our studies have shown that CD44, the principal receptor for hyaluronan, is present on cumulus cells during oocyte maturation. Although hyaluronan-CD44 interaction has been implicated in cumulus expansion and/or oocyte maturation, the full significance of CD44 remains unknown. The objective of the present study was to further investigate the role of CD44 in cumulus expansion and oocyte maturation in pigs. We demonstrate here in that CD44 has a key role in oocyte maturation but not in cumulus expansion. Previous studies have reported the physiological significance of cumulus expansion in oocyte maturation. However, our results suggest that cumulus expansion is a necessary condition for oocyte maturation, but that it is not sufficient on its own. Furthermore, western blot analysis demonstrated that the CD44 of the in vitro-matured cumulus-oocyte complexes (COCs) had a larger molecular weight and more terminal sialic acid, which has been proven to inhibit the hyaluronan-binding ability of the receptor, than the CD44 of the in vivo-matured COCs, indicating that the hyaluronan-CD44 interactions during in vitro maturation might be insufficient compared with those in vivo. The insufficient interactions of hyaluronan-CD44 during in vitro maturation may cause the inferior capacity of fertilization and development of oocytes matured in vitro.  相似文献

One hundred dogs that were positive for canine distemper virus antigen and inclusion bodies in the tonsils were examined for the distribution of inclusion bodies in various tissues. Inclusion bodies were found in the lungs (70 dogs), brains (20 dogs), urinary bladders (73 dogs), stomachs (78 dogs), spleens (77 dogs), and lymph nodes (81 dogs) of the dogs. Based on these results, the tonsils may be the most suitable tissue for detection of inclusion bodies in canine distemper.  相似文献

ABSTRACT:   Neuronal activities were investigated in the cerebellum of immobilized and swimming goldfish Carassius auratus . Extracellularly recorded neural activities of the cerebellum in immobilized goldfish were characterized and classified into five types. Based on the waveforms and recording depths, these five neural activity types were estimated to originate from three identified classes of cerebellar neurons: Purkinje cells, eurydendroid cells, and granule cells. Chronic recording of cerebellar neuron activities in unrestrained goldfish was performed for more than 100 h. During the chronic recordings, a submersible amplifier attached to the goldfish head, and a multielectrode array developed for the present study were used. Neuronal activities in the cerebellum of free-swimming fish could also be classified into five types as in the immobilized condition. Firing patterns of two neurons identified as Purkinje cells and eurydendroid cells were analyzed during turning movements of the goldfish. The firing patterns of these neurons changed in relation to turning movements. Although some improvements are required, the chronic recording method developed in the present study can be applied to further investigations concerning the direct relationship between brain neural activities and certain behavior.  相似文献

ABSTRACT:   Sporophyll formation in two alariaceous plants, Undaria pinnatifida and Alaria crassifolia , was studied in relation to the nutrient requirements. The sporophylls of U. pinnatifida formed zoosporangia when they had N and P contents greater than 1.4 kgN/m3 and 0.74 kgP/m3. This indicates that these values are the critical nutrient levels for zoospore formation in U. pinnatifida . In the sporophytes of A. crassifolia , many sporophylls with zoosporangia showed nutrient contents higher than 6.25 kgN/m3 and 1.70 kgP/m3. These results suggest that the U. pinnatifida can form zoospores at lower levels of N and P contents than A. crassifolia . Both species often formed zoosporangial sori on the blades in the late period of each reproductive season. The fertile parts of the blade showed N and P contents higher than the critical levels. This phenomenon indicates that the blades have the capability to form zoosporangial sori if there is a sufficient accumulation of nutrients for zoospore formation. The zoospore formation on the blade seems to be accomplished by an overflow of excess nutrients from sporophylls into the blade, or by accumulating sufficient nutrients to form sori even if the sporophylls are not formed.  相似文献

Sequence analysis based on the anterior part of the mitochondrial DNA control region was carried out to reveal the genetic diversity, stock structure, and historical demography of the red tilefish. Nucleotide sequences of 388 bp in length were determined for 280 individuals taken from eight localities. The molecular diversity, haplotype diversity, and nucleotide diversity were relatively high (average h = 0.929 ± 0.011; average π = 0.008 ± 0.005). The mismatch distribution was not significantly different from the expected distribution for a rapidly expanding population (P = 0.453). The minimum spanning network connecting with full-sequence haplotypes contained star-like topologies derived from multiple ancient lineages, supporting the mismatch distribution analysis. No significant genetic differentiation was observed among eight localities (maximum pairwise F ST = 0.023, reduced-sequence data set). Our results suggest a large panmictic population of the red tilefish along the coast of Honshu to the East China Sea.  相似文献

ABSTRACT:   A field experiment was conducted in the Matsumae area of Hokkaido, Japan, during June and July 2002, to investigate the effects of different entrance designs on the catch efficiency of fish traps by fishing with commercial traps (entrance inclination angle [α] = 37°; funnel length of entrance [ L f ] = 22 cm) and experimental traps. The experimental traps were of the same size and similar design as commercial traps, with different entrance inclination angles (trap E1: α = 46°; E2: α = 27°; E3: α = 0°; all L f  = 22 cm) or funnel lengths (E4: α = 37°, L f  = 8 cm). In total, 2200 fish during 200 trap hauls were captured. The catch was significantly higher using both traps E2 and the commercial trap than with trap E3 ( P  < 0.05), and the catch of trap E2 was higher than that of the commercial trap. There were no significant differences in mean fish body length or the frequency distributions of body length among trap types (E1, E2, E3 and commercial). The funnel length of the entrance also affected the catch of traps. Trap E4 had significantly higher catches than the commercial trap ( P  = 0.04) when traps were deployed for a 1-day soak time. Fish body length frequency distributions did not differ between trap E4 and the commercial trap. The results showed that catch can be greatly affected by trap entrance designs.  相似文献

The development of a closed recirculating aquaculture system that does not discharge effluents would reduce a large amount of pollutant load on aquatic bodies. In this study, eel were reared in a closed recirculating system, which consisted of a rearing tank, a foam separation unit, a nitrification unit and a denitrification unit. The foam separation unit has an inhalation-type aerator and supplies air bubbles to the rearing water. The growth of eel, which were fed a commercial diet, was satisfactory, with gross weight increases of up three times in 3 months. The survival rate under the congested experimental conditions was 91%. The foam separation unit maintained oxygen saturation in the rearing water at about 80%. Furthermore, fine colloidal substances were absorbed on the stable foam formed from eel mucus and were removed from the rearing water by foam separation. Ammonia oxidation and the removal of suspended solids were accomplished rapidly and simultaneously in the nitrification unit. The ammonia concentration and turbidity were kept at less than 1.2 mg of N per litre and 2.5 units, respectively. When the denitrification process was operated, nitrate that accumulated in the rearing water (151 mg of N per litre) was reduced to 40 mg of N per litre. The sludge was easily recovered from the nitrification and denitrification tanks, and the components were found suitable as compost. Based on these results, the intensive aquaculture of freshwater fish such as eel can be achieved using a closed recirculating system without emission.  相似文献

The present study investigated the effects of elevated carbon dioxide concentration ([CO2]) and air temperature on the germination of seed bulbils and the seedling vigour of two Chinese yam lines. Plants were grown under two [CO2] levels, ambient and elevated (ambient + 200 μmol mol?1), and two mean air temperature regimes, 22.2 °C (ambient + 1.4 °C) and 25.6 °C (ambient + 5.2 °C). Elevated [CO2] did not affect bulbil germination under both air temperature regimes. During the early growth stage, the dry weight (DW) of leaves, vines, shoots, roots, belowground parts (roots + tubers) and whole plants were higher under elevated [CO2] than ambient [CO2] for both lines under the low- and high-temperature regimes. The values of vigour indexes (index I = germination % × seedling length and index II = germination % × seedling DW) were also higher under elevated [CO2] than ambient [CO2] for both lines. These results indicated that Chinese yam seedlings respond positively to elevated [CO2] during the early growth stage. The above:belowground DW ratios were lower under elevated [CO2] than ambient [CO2] in seedlings with very small new tubers for both yam lines, indicating that elevated [CO2] strongly affected the root growth in the early growth stage. The DWs of post-treatment seed bulbils were higher in the elevated [CO2] under both air temperature regimes. The results showed that Chinese yam used a smaller amount of the reserves in seed bulbils under elevated [CO2] than under ambient [CO2].  相似文献

The populations of red alga Gelidium elegans along the coast of Miyakejima Island were severely damaged by a volcanic eruption in 2000. The effect of this volcanic eruption has been long lasting, and populations of this red alga still have not recovered. We investigated the effect of seabed sediment particles derived from volcanic ash on the substrate adhesion of G.?elegans spores. The analysis provides evidence that increasing amounts of sediment particles result in lower adhesion rates of G.?elegans spores, and that smaller sediment particles have a greater influence on adhesion. The amount of seabed sediment particles around Miyakejima Island was 9.3?C1815.4?mg/cm2. This amount has changed greatly from year to year. The adhesion rate of G.?elegans spores in water around Miyakejima Island was 0?% at all points in 2008 and 2010, but it was estimated as 6.3?C38.6?% in 2009. These results suggest that there is significant inhibition of algal spore adhesion by seabed sediment particles derived from volcanic ash around Miyakejima Island.  相似文献
