Compact growth is an important quality criterion in horticulture. Many Campanula species and cultivars exhibit elongated growth which is suppressed by chemical retardation and cultural practice during production to accommodate to the consumer’s desire. The production of compact plants via transformation with wild type Agrobacterium rhizogenes is an approach with great potential to produce plants that are non-GMO. Efficient transformation and regeneration procedures vary widely among both plant genera and species. Here we present a transformation protocol for Campanula. Hairy roots were produced on 26–90% of the petioles that were used for transformation of C. portenschlagiana (Cp), a C. takesimana × C. punctata hybrid (Chybr) and C. glomerata (Cg). Isolated hairy roots grew autonomously and vigorously without added hormones. The Cg hairy roots produced chlorophyll and generated plantlets in response to treatments with cytokinin (42 µM 2iP) and auxin (0.67 µM NAA). In contrast, regeneration attempts of transformed Cp and Chybr roots lead neither to the production of chlorophyll nor to the regeneration of shoots. Agropine A. rhizogenes strains integrate split T-DNA in TL- and TR-DNA fragments into the plant genome. In this study, regenerated plants of Cg did not contain TR-DNA, indicating that a selective pressure against this T-DNA fragment may exist in Campanula. 相似文献

Transformation of Kalancho? blossfeldiana ‘Molly’ using the naturally occurring bacterium Agrobacterium rhizogenes is a non-GMO strategy to breed compact plants. In the present study, crosses resembling a commercial breeding strategy were made to determine if the improved ornamental quality observed in the T1 generation was inherited to the offspring. F1 lines were produced by crossing the commercial Kalancho? cultivar ‘Sarah’ with a selected T1 line. Subsequently, F2 populations were produced by self-pollination of individual selected lines. The rol-genes were inherited to the progeny and the presence of rol-genes was confirmed in all F1 and many F2 plant lines exhibiting dwarfism. Screening of F1 and F2 plants showed that the rol-genes were inherited together. Besides decreased plant height, several F1 and F2 lines containing rol-genes exhibited changes in plant diameter, number of branches, flower diameter and time to first open flower and duration of flowering compared to the WT ‘Molly’. Furthermore, increased ethylene tolerance was observed in several lines containing rol-genes compared to the WT ‘Sarah’. Screening of three selected F2 populations derived from self-pollination of F1 lines containing rol-genes indicated a shift in distribution towards a lower mean plant height within the entire plant population compared to a control population of plants without rol-genes. Moreover, one of these F2 populations also exhibited earlier flowering compared to the control population. Compact potted plants and lines without delayed flowering and with improved ethylene tolerance were obtained and are valuable in commercial breeding programmes without using recombinant DNA technology. 相似文献

For low-input crop production, well-characterised varieties increase the possibilities of managing diseases and weeds. This
analysis aims at developing a framework for analyzing grain yield using external varietal information about disease resistance,
weed competitiveness and yield potential and quantifying the impact of susceptibility grouping and straw length scores (as
a measure for weed competitiveness) for predicting spring barley grain yield under variable biotic stress levels. The study
comprised 52 spring barley varieties and 17 environments, i.e., combinations of location, growing system and year. Individual
varieties and their interactions with environments were analysed by factorial regression of grain yield on external variety
information combined with observed environmental disease loads and weed pressure. The external information was based on the
official Danish VCU testing. The most parsimonious models explained about 50% of the yield variation among varieties including
genotype-environment interactions. Disease resistance characteristics of varieties, weighted with disease loads of powdery
mildew, leaf rust and net blotch, respectively, had a highly significant influence on grain yield. The extend to which increased
susceptibility resulted in increased yield losses in environments with high disease loads of the respective diseases was predicted.
The effect of externally determined straw length scores, weighted with weed pressure, was weaker although significant for
weeds with creeping growth habit. Higher grain yield was thus predicted for taller plants under weed pressure. The results
are discussed in relation to the model framework, impact of the considered traits and use of information from conventional

ABSTRACT Forest productivity is a crucial variable in forest planning, usually expressed as site index (SI). In Nordic commercial forest inventories, SI is commonly estimated by a combination of aerial image interpretation, field assessment and information obtained from previous inventories. Airborne laser scanning (ALS) and digital aerial photogrammetry (DAP) data can alternatively be used for SI estimation, however the economic utilities of the inventory methods have not been compared. We compared seven methods of SI estimation in a cost-plus-loss analysis, by which we added the expected economic losses due to sub-optimal treatment decisions to the inventory costs. The methods comprised direct and indirect estimation from combinations of ALS, DAP and stand register data, and manual interpretation from aerial imagery supported by field assessment and information from previous inventories (conventional practices). The choice of method had great impact on both the accuracy and the economic value of the produced estimates. Direct methods using bitemporal ALS and DAP data gave the best accuracy and the smallest total cost. DAP was a suitable and low-cost data source for SI estimation. Estimation from single-date ALS and DAP data and age obtained from the stand register provided practical alternatives when applied to even-aged stands. 相似文献

BACKGROUND: Clinical studies investigating platelet function in dogs have had conflicting results that may be caused by normal physiologic variation in platelet response to agonists. OBJECTIVES: The objective of this study was to investigate platelet function in clinically healthy dogs of 4 different breeds by whole-blood aggregometry and with a point-of-care platelet function analyzer (PFA-100), and to evaluate the effect of acetylsalicylic acid (ASA) administration on the results from both methods. METHODS: Forty-five clinically healthy dogs (12 Cavalier King Charles Spaniels [CKCS], 12 Cairn Terriers, 10 Boxers, and 11 Labrador Retrievers) were included in the study. Platelet function was assessed by whole-blood aggregation with ADP (1, 5, 10, and 20 microM) as agonist and by PFA-100 using collagen and epinephrine (Col + Epi) and Col + ADP as agonists. Plasma thromboxane B(2) concentration was determined by an enzyme immunoassay. To investigate the effect of ASA, 10 dogs were dosed daily (75 or 250 mg ASA orally) for 4 consecutive days. RESULTS: A higher platelet aggregation response was found in CKCS compared to the other breeds. Longer PFA-100 closure time (Col + Epi) was found in Cairn Terriers compared to Boxers. Plasma thromboxane B(2) concentration was not statistically different between groups. Administration of ASA prolonged the PFA-100 closure times, using Col + Epi (but not Col + ADP) as agonists. Furthermore, ASA resulted in a decrease in whole-blood platelet aggregation. CONCLUSIONS: Platelet function is influenced by breed, depending upon the methodology applied. However, the importance of these breed differences remains to be investigated. The PFA-100 method with Col + Epi as agonists, and ADP-induced platelet aggregation appear to be sensitive to ASA in dogs. 相似文献

In a six-year crop rotation trial organically and integrated grown vegetables were produced according to current good agricultural practices, taking quality and quantity aspects into consideration. The raw materials assessed focussed on materials used for industrial food production. Nutritional, sensory and agricultural aspects were evaluated. Carrot, cabbage, onion, pea and potato are possible to grow organically for industrial purposes. Depending on crop, the yield was lower (65-90%) for organically grown compared to integrated grown. Cultivation of organic spinach and dill turned out to be difficult due to problems with weed and discoloration. The chemical analyses included pesticide residues, nitrate, glycoalkaloid, dry matter, vitamin C and 25 different minerals and trace elements. Overall, the organically grown crops had higher dry matter content than the integrated grown. However, when examining the data for the different crops contradictory results were noted. No significant differences due to growing system were noticed for vitamin C and the other nutrients except for 4 of the trace elements. The sources and mechanisms regulating fluoride (F) in Norwegian lake waters are studied using data from regional surveys of precipitation chemistry, lake water chemistry and bedrock geology. Fluoride concentrations in Norwegian lakes range from < 5 to 560 μg L?1. Fluoride content in the bedrock is the most important factor controlling F levels in lake waters, as shown by significant differences in median value of F concentrations between lakes situated in different geological provinces. There are also weak but significant correlations between F in the lakes and components typical for weathering such as non-marine Ca, Mg, Na and K. The regional picture of F concentration in lake water shows elevated F concentrations in the acidified areas in southern and southeastern Norway compared to other regions of the country with comparable geology. There is a weak but significant correlation between F and SO4, a typical indicator of acidification in surface water. Mass balance calculations in three catchments show that F is retained in soils in pristine areas, while F output exceeds precipitation input in acidified areas. This both demonstrates the strong retention capacity for F in soils and indicates that anthropogenic F added through polluted rain is a minor source of F in surface waters.
