Heat shocks, hydrostatic pressure shocks, and ultraviolet radiation were evaluated for their efficacy as methods of manipulating ploidy in yellow perch (Perca flavescens). The most effective methods of inducing triploidy were heat shocks of 28–30°C applied at a time of initiation (TI) of 5 min postfertilization for durations of 10 or 25 min, and hydrostatic pressure shocks of 9000 or 11 000 psi applied at a TI of 5 min for a duration of 12 min. These treatments resulted in triploidy induction rates that ranged from 54–100%, and embryonic survival rates of 16–80%. Cold shocks of 0°C had no effect on the ploidy or survival of embryos. For perch, hydrostatic pressure shock offered several advantages over heat shock as a method of manipulating ploidy. The most effective methods of inducing tetraploidy were hydrostatic pressure shocks of 9000 psi applied at a TI of 192 min for durations of 16 or 24 min. Ultraviolet radiation of perch sperm with doses of 3240–6480 ergs/mm2 resulted in 100% inactivation of paternal chromosomes, and perch eggs fertilized with inactivated sperm had survival rates of > 50%, thereby establishing methods for producing gynogenetic perch. Airborne in situ observations of molecules with a wide range of lifetimes (methane, nitrous oxide, reactive nitrogen, ozone, chlorinated halocarbons, and halon-1211), used in a tropical tracer model, show that mid-latitude air is entrained into the tropical lower stratosphere within about 13.5 months; transport is faster in the reverse direction. Because exchange with the tropics is slower than global photochemical models generally assume, ozone at mid-latitudes appears to be more sensitive to elevated levels of industrial chlorine than is currently predicted. Nevertheless, about 45 percent of air in the tropical ascent region at 21 kilometers is of mid-latitude origin, implying that emissions from supersonic aircraft could reach the middle stratosphere. 相似文献

How anthropogenic climate change will affect hydroclimate in the arid regions of southwestern North America has implications for the allocation of water resources and the course of regional development. Here we show that there is a broad consensus among climate models that this region will dry in the 21st century and that the transition to a more arid climate should already be under way. If these models are correct, the levels of aridity of the recent multiyear drought or the Dust Bowl and the 1950s droughts will become the new climatology of the American Southwest within a time frame of years to decades. 相似文献

Linear arrays in lead isotope space for mid-ocean ridge basalts (MORBs) converge on a single end-member component that has intermediate lead, strontium, and neodymium isotope ratios compared with the total database for oceanic island basalts (OIBs) and MORBs. The MORB data are consistent with the presence of a common mantle source region for OIBs that is sampled by mantle plumes. 3He/4He ratios for MORBs show both positive and negative correlation with the 206Pb/204Pb ratios, depending on the MORB suite. These data suggest that the common mantle source is located in the transition zone region. Microorganisms play a dominant role in Antarctic ecosystems, yet little is known about how fungal diversity differs at sites with considerable human activity as compared to those that are remote and relatively pristine. Ross Island, Antarctica is the site of three historic expedition huts left by early explorers to the South Pole, Robert F. Scott and Ernest Shackleton. The fungal diversity of these wooden structures and surrounding soils was investigated with traditional culturing methods as well as with molecular methodology including denaturing gradient gel electrophoresis (DGGE) using the internal transcribed spacer (ITS) regions of ribosomal DNA for identification. From historic wood and artifact samples and soils adjacent to the huts as well as soil samples obtained from the Lake Fryxell Basin, a remote Dry Valley location, and remote sites at Mt. Fleming and the Allan Hills, 71 fungal taxa were identified. The historic huts and associated artifacts have been colonized and degraded by fungi to various extents. The most frequently isolated fungal genera from the historic woods sampled include Cadophora, Cladosporium and Geomyces. Similar genera were found in soil samples collected near the huts. Sampling of soils from locations in the Transantarctic Mountains and Lake Fryxell Basin at considerable distances from the huts and with different soil conditions revealed Cryptococcus spp., Epicoccum nigrum and Cladosporium cladosporioides as the most common fungi present and Cadophora species less commonly isolated. DGGE revealed 28 taxa not detected by culturing including four taxa which possibly have not been previously described since they have less than 50% ITS sequence identity to any GenBank accessions. Fungi capable of causing degradation in the wood and artifacts associated with the expedition huts appear to be similar to those present in Antarctic soils, both near and at more remote locations. These species of fungi are likely indigenous to Antarctica and were apparently greatly influenced by the introduction of organic matter brought by early explorers. Aspermia was diagnosed in a 12-year-old Thoroughbred stallion with generalized lymphosarcoma. Invasion of the epididymus by neoplastic cells caused thickening and enlargement of both epididymes. Malignant melanoma in a 21-year-old, gray, Arabian gelding was manifested by rapidly deteriorating lameness of the right pelvic limb. A melanotic, cutaneous mass of small dimensions was identified in the left jugular furrow. Exploratory laparoscopy revealed widespread infiltration of melanotic masses into the structures of the abdominal cavity. Necropsy indicated the lameness to have resulted from infiltration of neoplastic cells into the sacral nerves, dorsal root ganglia, proximal ischiatic nerve, and gluteal muscle fibers.
