The occurrence of Bovine Spongiform Encephalopathy (BSE, so called mad cow diseases) that was first identified in England in 1986 was considered as being limited to only European countries, including England. However, the outbreak in Asia as well as North America since 2001 has amplified the fear that there isn't any nation in the world that is a safe area. In order to assess the risk of BSE outbreak in each country, the Office International des Epizooties (OIE) and EU have respectively established criteria, where OIE has set 5 levels and EU has set 4 levels. The Scientific Steering Committee (SSC) of the European Commission conducted a Geographical BSE Risk(GBR) assessment for 64 nations, such as the United States, etc., as of April 29, 2003. However, as of July 1, 2005, the duty of GBR assessment is expected to be transferred to a newly established body called EFSA (European Food Safety Authority, located in Parma, Italy). As Korea has not undergone a GBR assessment up to now, this study analyzed the risk of BSE outbreak in Korea by reviewing BSE prevention measures, etc., that have been put in place. Determining the immune responses to the development of endometritis during the peripartum period may assist in the development of more efficient reproductive management regimens for dairy herds. In this study, we compared the peripartum immune responses of dairy cows that develop endometritis by 4 weeks postpartum (n = 11) to cows that did not develop this disease (n = 19). Blood samples were collected 1 week before calving, just after or during calving, and then at weeks 1, 2, 3, and 4 postpartum. Cows that developed endometriris had significantly higher total leukocyte, neutrophil, lymphocyte, and monocyte counts than the control cows (P < 0.05) at all time points. The leukocytes from cows that developed endometritis were significantly less phagocytic than those from control cows at all sampling time points (P < 0.01). The serum TNFalpha concentrations of the control cows decreased linearly from the prepartum time point (P = 0.0029), but the endometritis cows showed a different profile (P > 0.05). As a result, the serum TNFalpha concentrations were greater in the endometritis group (P < 0.01) than in the control group during the third and fourth weeks postpartum. The greater total leukocyte numbers and neutrophil, lymphocyte and monocyte counts, and the maintenance of elevated serum TNFalpha levels in the cows with endometritis may be due to infection in the postpartum period. Furthermore, the decreased phagocytic capacity of leukocytes during the peripartum period, including at the prepartum time point, makes cows more susceptible to postpartum endometritis. 相似文献

This study was performed to evaluate the efficacy of acupuncture on induced chronic arthritis of the dog by thermography. Complete Freund's adjuvant was injected into the left knee joint of 8 dogs to induce arthritis. Acupuncture was applied to BL-40, GB-33, GB-34, and LIV-8 once a week for 4 consecutive weeks, from 3 weeks after induction of chronic arthritis, in treatment group. At 3 weeks of acupuncture treatment, skin temperature difference (DeltaT) of treatment group returned to normal range (< 0.3 degrees C), while DeltaT remained high in non-treatment group. Infrared thermography (IRT) is useful to evaluate the treatment of acupuncture for induced canine chronic arthritis. Therefore, it is considered that clinical application of IRT in arthritis treatment would be also valuable. 相似文献

ObservationsA 9 year-old, 40 kg, female spayed Bouvier des Flandres was anesthetized for surgical removal of an intra-cardiac mass. Pre-anesthetic work-up included thoracic radiographs, which revealed moderate pleural effusion, and cardiac ultrasound, which identified a mass attached to the wall of the right ventricular outflow tract (RVOT). The mass caused dynamic obstruction of the RVOT during systole. The dog was pre-medicated with intravenous (IV) hydromorphone (0.05 mg kg?1). Following pre-oxygenation, anesthesia was induced with ketamine (3.75 mg kg?1, IV) and diazepam (0.18 mg kg?1, IV). Anesthesia was maintained with isoflurane in oxygen, an intravenous constant rate infusion (CRI) of fentanyl (10–30 μg kg?1 hour?1) and a CRI of lidocaine (50–200 μg kg?1 minute?1). A right lateral thoracotomy was performed. The heart was stopped transiently with a cold cardioplegic solution for 7.83 minutes to allow the removal of the mass through an open-heart procedure. No cardiopulmonary bypass was used. The heart was successfully restarted after cardiopulmonary resuscitation with internal cardiac massage and internal defibrillation. The dog recovered uneventfully from anesthesia without any apparent neurological sequelae. Post-operative analgesia consisted of intercostal nerve blocks with bupivacaine, CRIs of fentanyl (2–5 μg kg?1 hour?1) and lidocaine (40 μg kg?1 minute?1) and with oral meloxicam (0.1 mg kg?1). Five days following surgery, the dog was discharged from the hospital. Histopathology and immunohistochemistry of the mass identified an ectopic thyroid carcinoma.ConclusionsThis case showed the feasibility of whole body hypothermia and using a cold cardioplegic solution to induce cardiac arrest for a short open-heart procedure. 相似文献

The deleterious effects of heat stress on animal health are being increasingly recognized. This study aimed to determine hair cortisol (HC) and serotonin levels in lactating Holstein cows under heat stress conditions with different coat and hair‐cut color. Forty‐five multiparous lactating Holstein cows (days in milk = 130 ± 47, body weight = 753 ± 85 kg) were divided to two main groups of over 80% black coat color (BC) and over 85% white coat color (WC) visually observed based on registry certificates and subdividing to black hair sample (BH) and white hair samples (WH) in 2 × 2 factorial arrangements. Hair samples were taken from the forehead of the individuals. Higher HC levels were observed in BC than WC cows (P < 0.05). No differences were found in HC levels between BH and WH groups (P > 0.05). Serotonin levels showed no difference between BC and WC (P > 0.05). Interaction between coat color and hair color was not significant (P > 0.05). The cortisol levels in hair are not affected by pigmentation. However, pigmentation within the coat alters cortisol levels. In conclusion, white coat color retains less cortisol than the black coat. Therefore, white coats are preferable for dairy cows under heat stress conditions. 相似文献

Alternative sources of mesenchymal stem cells (MSCs) for replacing bone marrow (BM) have been extensively investigated in the field of bone tissue engineering. The purpose of this study was to compare the osteogenic potential of canine MSCs derived from adipose tissue (AT), BM, umbilical cord blood (UCB), and Wharton''s jelly (WJ) using in vitro culture techniques and in vivo orthotopic implantation assays. After canine MSCs were isolated from various tissues, the proliferation and osteogenic potential along with vascular endothelial growth factor (VEGF) production were measured and compared in vitro. For the in vivo assay, MSCs derived from each type of tissue were mixed with β-tricalcium phosphate and implanted into segmental bone defects in dogs. Among the different types of MSCs, AT-MSCs had a higher proliferation potential and BM-MSCs produced the most VEGF. AT-MSCs and UCB-MSCs showed greater in vitro osteogenic potential compared to the other cells. Radiographic and histological analyses showed that all tested MSCs had similar osteogenic capacities, and the level of new bone formation was much higher with implants containing MSCs than cell-free implants. These results indicate that AT-MSCs, UCB-MSCs, and WJ-MSCs can potentially be used in place of BM-MSCs for clinical bone engineering procedures. 相似文献

Salmonella Kentucky is among the most frequently isolated S. enterica serovars from food animals in the United States. Recent research on isolates recovered from these animals suggests there may be geographic and host specificity signatures associated with S. Kentucky strains. However, the sources and genomic features of human clinical S. Kentucky isolated in the United States remain poorly described. To investigate the characteristics of clinical S. Kentucky and the possible sources of these infections, the genomes of all S. Kentucky isolates recovered from human clinical cases in the State of Maryland between 2011 and 2015 (n = 12) were sequenced and compared to a database of 525 previously sequenced S. Kentucky genomes representing 12 sequence types (ST) collected from multiple sources on several continents. Of the 12 human clinical S. Kentucky isolates from Maryland, nine were ST198, two were ST152, and one was ST314. Forty‐one per cent of isolates were recovered from patients reporting recent international travel and 58% of isolates encoded genomic characteristics similar to those originating outside of the United States. Of the five isolates not associated with international travel, three encoded antibiotic resistance genes conferring resistance to tetracycline or aminoglycosides, while two others only encoded the cryptic aac(6′)‐Iaa gene. Five isolates recovered from individuals with international travel histories (ST198) and two for which travel was not recorded (ST198) encoded genes conferring resistance to between 4 and 7 classes of antibiotics. Seven ST198 genomes encoded the Salmonella Genomic Island 1 and substitutions in the gyrA and parC genes known to confer resistance to ciprofloxacin. Case report data on food consumption and travel were, for the most part, consistent with the inferred S. Kentucky phylogeny. Results of this study indicate that the majority of S. Kentucky infections in Maryland are caused by ST198 which may originate outside of North America. 相似文献

Kaempferol (KAE) is a natural flavonoid present in different plant species and exhibits anti‐inflammatory, antioxidant, and anticancer therapeutic properties. In the present study, we investigated the influence and underlying mechanisms of KAE supplementation on porcine oocytes during in vitro aging. The results show that KAE treatment can alleviate the aging‐related reduction of developmental competence. We observed that the blastocyst production rate in aged oocytes treated with 0.1 μM KAE was significantly higher than in untreated aging oocytes (36.78 ± 0.86% vs. 27.55 ± 2.60%, respectively, p < .05). The KAE‐treated aging oocytes had significantly reduced levels of reactive oxygen species (p < .05). Furthermore, the mRNA levels of the embryonic pluripotency‐related genes Oct4, NANOG, and ITGA5 were significantly increased in blastocysts derived from KAE‐treated oocytes (p < .05). During excessive oocyte culture, KAE treatment maintained the mitochondrial membrane potential and reduced apoptosis; however, this was not observed in untreated aging oocytes. In conclusion, our results suggest that KAE treatment can alleviate the aging of porcine oocytes by reducing oxidative stress and improving mitochondrial function. 相似文献

The relationship of the urease operon in the highly virulent O149 porcine enterotoxigenic Escherichia coli (ETEC) strain Ro8 to a genomic island (GI) homologous to O island (OI) 48 of O157 enterohemorrhagic E. coli (EHEC) strain EDL933 was investigated. Eighty-four of 84 O149:H10 strains were urease positive whereas 44 of 44 O149:H43 porcine ETEC strains were urease-negative. Seventeen of 17 O149:H10 strains that were tested possessed the OI-48 homolog whereas 24 of 24 O149:H43 strains lacked this OI. Transposon insertions in lipB or guaA genes in strain Ro8 eliminated urease activity while insertions in the caiF gene increased urease activity. When the O149 ure operon was cloned on a high copy number plasmid, urease expression was increased approximately 11-fold in Ro8 and 83-fold in O157 strain EDL933 compared with that in the wild type Ro8. The O149 urease activity was expressed despite the presence of the same premature stop codon in ureD that is present in ure+ O157:H7 strains that are urease-negative. The ure operon in Ro8 consists of 4 893 nucleotides with 99% identity with the ure operons in EHEC O157:H7 strains EDL933 and Sakai, and is part of a GI similar to GI-48 of strain EDL933. This OI, designated OI-48149 , is inserted in the serX tRNA gene in strain Ro8 and contains genes for urease, tellurite resistance, iha and an AIDA-I-like adhesin. The presence of a homolog of the O157:H7 OI-48 in highly virulent O149 porcine ETEC suggests that this OI may contribute to establishment of the bacteria in the intestine. 相似文献
