Cotyledons from dissected sterile embryos of salgareño pine (Pinus nigra Arn. ssp. salzmannii (Dunal) Franco) were inoculated with different disarmed Agrobacterium tumefaciens strains harbouring the binary vector p35SGUSint. The transient expression of a β-glucuronidase gene (uidA) was studied, using a histochemical staining procedure. Nineteen days after inoculation, the activity of β-glucuronidase was detected in epidermal and subepidermal layers of cotyledonary explants. The EHA105 strain harbouring a disarmed agropine-type Ti-plasmid (pTiBO542) was the most effective for gene transfer of the uidA gene. The effects of exudates and extracts from 0-day-old embryos on induction of vir gene expression in A. tumefaciens were also examined. The results of this study showed that salgarño pine embryo exudates contain a substance(s) that induce vir gene expression, in similar way to that observed with 100 μM acetosyringone (AS).All these findings suggest that T-DNA processing and transfer might take place when Agrobacterium infects suitable tissues of salgareño pine. 相似文献

RAPD (randomly amplified polymorphic DNA) technology was applied to analyze the genetic variability of sweet potato germplasm
existing in Chile and elsewhere. Analysis of 28 cultivars from all over the world showed polymorphic bands with all 18 primers
tested. A total of 124 RAPD bands were scored with an average of 6.9 polymorphic bands per primer. These results confirm that
sweet potato exhibits high genetic variation. Two groups were distinguished: one containing Peruvian cultivars, and another
containing cultivars from the rest of the world. Analysis of 14 accessions from Central Chile and one from Northern Chile
showed polymorphic bands with 24 of 26 primers tested, but almost all of the 140 polymorphic bands merely showed the distinctness
of the Northern accession. The almost complete uniformity of the other 14 accessions shows that sweet potato germplasm collected
in Central Chile has very little genetic variability and may be derived from a single cultivar.
Based on these results and on historical records, some hypotheses are proposed to explain the origin of sweet potatoes cultivated
in Chile.

Molecular marker analysis can be an effective tool when searching for new fire blight resistance donors. It can speed up the breeding process as well, even though many of the available markers linked to fire blight resistance QTLs have not yet been tested by screening a large number of cultivars. The aim of this study was to search for alternate sources of the three major QTLs of fire blight resistance; FBF7, FB_MR5 and FB_E, as well as to test the efficiency of some markers linked to minor QTLs. Altogether, nine primer pairs were used on 77 genotypes including new Hungarian cultivars and old apple cultivars from the Carpathian basin. Several marker alleles of FB resistance QTLs have been detected in the screened genotypes, most importantly the alleles coupling with FB_MR5 in the old cultivars ‘Kéresi muskotály’, ‘Szabadkai szercsika’ and ‘Batul’. We propose these cultivars as the first available resistance donors of FB_MR5 instead of the crabapple Malus × robusta 5. The results also bring new information regarding the resistance alleles of new Hungarian cultivars and selections. 相似文献

The odor-active compounds of dry-fermented sausages with added nitrite or nitrate as curing agents were identified by gas chromatography-olfactometry (GC-O) applying the detection frequency (DF) method. The quantification of these compounds in the sausage was determined by multiple headspace solid-phase microextraction (multiple HS-SPME). There were no specific odor-active compounds related to the use of nitrite or nitrate although there were differences in the DF value of several compounds. The nitrite-added sausages presented higher DF values for ethanol, 1-hexanol, propanoic acid, 2-heptenal, and nonanal while the nitrate-added sausages had higher DF values for phenylacetaldehyde and 3-methyl-butanal. Eighteen compounds were quantified by multiple HS-SPME. Most of them were above their air detection thresholds, but only hexanal, heptanal, and 1-octen-3-ol were in a concentration higher than their oil threshold values. In this paper we report on an outbreak of reovirus, herpesvirus (Pacheco disease), and/or mycosis infection (Aspergillus spp. and Zygomyces spp.) affecting a batch of young African grey parrots (Psittacus erithacus), with 80% morbidity and 30% mortality. Study material was taken from five birds (four dead and one euthanatized) with a range of clinical symptoms (depression, diarrhea, respiratory symptoms). Diagnosis was confirmed by immunohistochemical detection of avian reovirus, electron microscopy, and virus isolation. Viral antigen of reovirus was detected mainly in large mononuclear cells in the bursa of Fabricius and the spleen, pancreas epithelial cells, and circulating cells; lymphoid organs displayed the largest number of immunopositive cells and severe lymphocyte depletion. Bacteriologic study was negative. Reovirus infection was common in all birds studied, whereas Pacheco disease and mycosis were found in only some, suggesting that reovirus could be the initial cause triggering the outbreak and facilitating infection by other agents and their swift spread through the batch. 相似文献

The extrusion process allows the production of nixtamalized corn flour rich in arabinoxylans, which help to prevent cardiovascular and intestinal diseases. During extrusion, physiochemical properties of nixtamalized corn flour are negatively modified. The use of enzymes such as xylanase in order to obtain nixtamalized corn flour using extrusion has been studied as an alternative to reduce these changes in corn flour tortilla. The aim of this research was to evaluate changes in protein solubility of extruded nixtamalized corn flour with and without different concentrations of xylanase enzyme (0.05, 0.075, and 0.1 %, w/w). Soluble proteins of each corn flour were extracted and analyzed by SE-HPLC, while insoluble proteins were determined by the combustion method. In addition, each corn flour was analyzed by scanning electron microscopy (SEM). Results showed that the extruded nixtamalized corn flour, with and without xylanase, increased the protein solubility, and this effect was lower in extruded nixtamalized corn flour with xylanase. Insoluble protein diminished in corn flours either with or without xylanase enzyme. The addition of xylanase reduces the effect that the extrusion process has on the solubility proteins of extruded nixtamalized corn flour. 相似文献

The objective of the study was to evaluate an efficacy of sows vaccination protocols in the herd with serious problems affecting efficacy of reproduction. The study was performed in a large pig herd with about 1200 sows. Before vaccination against PCV2, farrowing rate in this farm was about 65%. Sows, boar and replacement gilts were immunized using Circovac vaccine (Merial, France) according to producer's recommendations. Parameters of production were analyzed since 2007 until 2010 in selected batches of sows inseminated at the same weeks of the year (17th, 18th, 19th and 20th) to eliminate seasonal variability. In total, 940 sows were subjected to the study. No significant changes in management during these years were introduced. The applied protocol of sow herd long-term vaccination proved to be very efficient. All measured production parameters: reproduction rate, number of piglets born alive, birth weight of piglets and number of piglets weaned per a litter improved after implementation of immunization program. Moreover, further improvement was observed with vaccination in the following reproduction cycles. The most spectacular effect of vaccination regarded average farrowing rate that increased from 64.76% in control group to 86.93% after basic vaccination. Two years after implementation of vaccination program this parameter reached 93.6%. Number of piglets weaned per sow per a litter improved from 10.31 to 11.74 after one year of vaccination and remained relatively stable through the following year. Simultaneously, the percentage of newborn piglets with birth weight < 1 kg decreased significantly (p < 0.05). To summarize, vaccination against PCV2 influenced positively the insemination rate, number of piglets born alive and weaned per litter as well as birth body weight and percentage of piglets weighing < 1 kg. 相似文献
