A 14-yr-old female eastern black rhinoceros (Diceros bicornis michaeli) presented with progressive suppurative osteomyelitis in her left hind lateral toe. beta-Hemolytic Streptococcus sp. was isolated. The animal was treated with multiple systemic antibiotics, and topical wound cleansing. Repeated debridements and nail trimmings were performed for 5 mo prior to electing amputation. The toe was surgically amputated under general anesthesia between the first and second phalanges. Analgesia was diffused into the wound topically via a catheter and elastomeric pump. The open amputation site was covered with adherent drapes and a negative-pressure wound therapy device provided vacuum-assisted closure (V.A.C.) for 72 hr. Three months later this animal developed a deep dermal ulcer on the lateral aspect of the right hind limb, at the level of the stifle. Methicillin-resistant Staphylococcus aureus was isolated. The wound was managed by initial daily lavage, followed by 1 mo of V.A.C. therapy, with 72 hr between dressing changes. Clinically, this therapy expedited the formation of healthy granulation tissue and overall healing was accelerated. The animal tolerated the machine and bandage changes well via operant conditioning. The use of negative-pressure wound therapy appeared to shorten time to resolution of slow-healing wounds in black rhinoceros. 相似文献

The aim of the study is to determine the efficacy of indigenous soil fungi in removing (PAHs) from creosote-contaminated soil with a view to developing a bioremediation strategy for creosote-contaminated soil. Five fungal isolates, Cladosporium, Fusarium, Penicillium, Aspergillus and Pleurotus, were separatelyinoculated onto sterile barley grains and incubated in the dark. Thecolonized barley was inoculated onto creosote-contaminated (250 000 mg kg?1) soil in 18 duplicate treatments and incubated at 25 °C forseventy days. The soil was amended with nutrient supplements to give a C:N:Pratio of 25:5:1 and tilled weekly. Creosote removal was higher (between 78and 94%) in nutrient supplemented treatments than in the un-supplementedones (between 65 and 88%). A mixed population of fungi was more effective(94.1% in the nutrient amended treatment) in creosote removal than singlepopulations wit a maximum of 88%. Barley supported better fungal growthand PAH removal. Pleurotus sp. removed the creosote more than the other isolates. Two andthree-ring PAHs were more susceptible to removal than the 4- and 5-ringPAHs, which continued to remain in small amounts to the end of thetreatment. Goats housed in microbiologically secure facilities were experimentally endobronchially infected with Mycoplasma capricolum subsp. capripneumoniae (Mccp), causal agent of contagious caprine pleuropneumonia (CCPP). The animals were monitored over an 8-week period post-infection (p.i.). Elevated temperatures were observed 2-7 days p.i., reaching a maximum of 41.5 degrees C in one animal (1884). By 8 weeks p.i. the infection was successfully cleared, with no Mccp being recovered from the lungs, serum or nasal passages. Mccp was not isolated from serum throughout the experiment, either directly by culture or indirectly via polymerase chain reaction (PCR). Humoral immune responses against Mccp capsular polysaccharide (CPS) were generally poor when measured by ELISA. CPS antigen was present in the serum of all infected animals early in the infection (day 14 p.i.), although in one animal (1855) CPS antigen persisted throughout. This was the only animal to exhibit a serious cough (day 5-19 p.i.). Successful diagnosis of CCPP was achieved using two different types of latex agglutination test (CPS antibody and CPS antigen detection test), immunoblotting and a blocking ELISA, although the latter lacked sensitivity until later in the infection (35-40 days p.i.). Only a single animal (1855) was detected positive using the current complement fixation test (CFT). Strong immune responses to protein antigens were detected by IgG and IgM immunoblotting from the first time point at day 14 p.i. IgM immunodominant bands of 220, 85, 62 and 40kDa were observed in the 3 infected animals and from CFT-positive CCPP field sera. Band intensity gradually diminished throughout the experiment. IgG immunodominant bands of 108, 70, 62, 44, 40 and 23kDa were shared between experimentally-infected and field sera, with band intensity either remaining unchanged or increasing from day 14 p.i. These bands were not present using pre-infection sera. Of the diagnostic tests used, only the CPS antibody detection latex agglutination test and IgG immunoblotting gave positive diagnoses throughout the entire period post-infection (days 14-53 p.i.). 相似文献

1. Germ-free and conventional Japanese quail reared on a diet in which the nitrogen was supplied solely in the form of free amino acids grew well, but growth was significantly better in both environments when some of the amino acids were replaced by an equivalent quantity of isolated soya protein. 2. Although the small intestine was shorter in the birds given the soya protein supplement, its weight per unit of length was greater; histological examination suggested that its circumference and wall thickness were both increased. 3. Results of a small experiment with conventional quail given the diets marked with 14C-polyethylene glycol indicated that the free amino acid diet passed through the gut more rapidly than the supplemented diet. 4. It was concluded that the growth-promoting effect of the isolated soya protein was unrelated to the activities of the gut microflora but might be due to changes in the digestive and absorptive capacities of the small intestine induced by the different physical nature of the supplemented diet. 相似文献

Recently weaned pigs were infected aerogenically with Haemophilus (Actinobacillus) pleuropneumoniae, serotype 5. At three, six, 12, and 18 hours and one, two, four and seven days after exposure to haemophili a pair of animals were killed and necropsied. Pulmonary oedema with multifocal petechial haemorrhages and a diffuse neutrophilic bronchiolitis and alveolitis were observed at three and six hours after infection. Focal areas of coagulative necrosis developed in areas of intense suppuration at 12 and 18 hours after infection. At one and two days after infection, necrotic areas were surrounded by dense bands of degenerating leucocytes and contained unidentifiable round and elongated cells characteristic of this disease. In subacute lesions a thick layer of granulation tissue formed around the outer margin of developing abscesses. Most of the round and elongated cells in alveolar exudates could not be identified by enzyme histochemistry or electron microscopic examination. Neutrophils in various stages of degeneration and deterioration provided strong evidence that some of the cells were of this type. These findings suggest that neutrophils may play an early and significant role in development of lesions. 相似文献

Ovarian and gonadotropin responses to insulin and energy restriction were investigated in a 2 X 2 factorial experiment using 2-yr-old Brangus heifers. Thirty heifers were paired by weight and body condition, then assigned to treatment groups receiving 75 (LE) or 180% (HE) of NRC recommendations for dietary energy for maintenance. Diets were adjusted weekly to maintain daily .25 to .5 kg weight loss or 0 to .25 kg weight gain, respectively. On d 10 of the first estrous cycle subsequent to the initial 45 d of feeding, heifers within each dietary group were allocated to receive twice daily infusions of either 40 U insulin (I) or saline (C). Infusions began at 5 and 10 h postprandial and were given in six boluses, 20 min apart. Infusions continued daily until d 20 or estrus, whichever occurred first. On d 11, blood samples were collected at 15-min intervals for 12 h to determine luteinizing hormone (LH) and insulin concentrations. On d 16 to 20, twice daily im injections of 1 mg follicle stimulating hormone (FSH) were administered. Heifers were ovariectomized on d 11 after estrus. Number of corpora lutea (CL) in LE-I heifers was greater (P less than .05) in LE-C, HE-C or HE-I. Total CL weight (g) per heifer was greater (P less than .05) in HE-C and LE-I heifers than in LE-C. Cyclic AMP is known to play a major role in intracellular signalling during mammalian sperm capacitation. However, despite much research, many of the molecular details of cyclic AMP's involvement remain obscure. In this review, I discuss the following aspects, presenting some original data as illustration where relevant. With respect to cyclic AMP synthesis, uncertainties exist as to the number of forms of adenylyl cyclase that are present in the spermatozoon, whether they are cytosolic or bound to subcellular structures, and which physiological effectors they respond to (e.g. bicarbonate, Ca2+, or receptor‐coupled G‐proteins). While net intracellular levels of cyclic AMP in spermatozoa depend upon the relative activities of adenylyl cyclase and phosphodiesterase, there are wide between‐sample variations within species, both in basal levels and in levels attained after activation of the cyclase (e.g. after sperm treatment with bicarbonate). Moreover, minor changes in bulk cyclic AMP levels can result in large changes in cyclic AMP‐dependent functions. Finally, while cyclic AMP levels respond very rapidly to sperm treatment by effectors such as bicarbonate and Ca2+ (key components of capacitating media), there are big discrepancies between the rates of functional response. For example, enhancement of motility and collapse of phospholipid asymmetry take place within a few minutes, whereas more than 1 h of exposure to capacitating conditions is needed for cyclic AMP‐dependent protein tyrosine phosphorylation to become detectable or for the sperm population to attain a capacitated state. 相似文献
