Technical hexachlorocyclohexane (HCH) and lindane are obsolete pesticides whose former production and use led to widespread contaminations posing serious and lasting health and environmental risks. Out of nine possible stereoisomers, alpha-, beta-, gamma-, and delta-HCH are usually present at contaminated sites, and research for a better understanding of their biodegradation has become essential for the development of appropriate remediation technologies. Because haloalkane dehalogenase LinB was recently found responsible for the hydroxylation of beta-HCH, delta-HCH, and delta-pentachlorocyclohexene (delta-PCCH), we decided to examine whether beta- and gamma-PCCH, which can be formed by LinA from alpha- and gamma-HCH, respectively, were also converted by LinB. Incubation of such substrates with Escherichia coli BL21 expressing functional LinB originating from Sphingobium indicum B90A showed that both beta-PCCH and gamma-PCCH were direct substrates of LinB. Furthermore, we identified the main metabolites as 3,4,5,6-tetrachloro-2-cyclohexene-1-ols and 2,5,6-trichloro-2-cyclohexene-1,4-diols by nuclear magnetic resonance spectroscopy and gas chromatography-mass spectrometry. In contrast to alpha-HCH, gamma-HCH was not a substrate for LinB. On the basis of our data, we propose a modified gamma-HCH degradation pathway in which gamma-PCCH is converted to 2,5-cyclohexadiene-1,4-diol via 3,4,5,6-tetrachloro-2-cyclohexene-1-ol and 2,5,6-trichloro-2-cyclohexene-1,4-diol.  相似文献

Fusarium head blight is one of the most noxious cereal diseases. Worldwide, F. graminearum (FG) and the mycotoxin deoxynivalenol (DON) is the most dominant species/mycotoxin in barley and wheat. Barley is often produced as on farm feed and thus routine mycotoxin analyses similar to those of cereals for human consumption are not performed. Hence, an early prediction of mycotoxin levels is important for farmers to minimise the risk of contaminated feed but also of contaminated cereals entering the cereal supply chain. Therefore, climate chamber experiments with artificial FG infection of barley investigating the influence of different temperatures (10 °C, 15 °C, 20 °C) and durations (4 h, 8 h, 12 h) at 99% relative humidity were conducted to accumulate data to develop a forecasting system. An up to three times higher DON contamination in the 15 °C treatments for the feed barley variety Ascona was detected compared with the 10 °C and 20 °C treatments. For the malting barley variety Concerto, the prolonged humidity durations had a stronger effect under all tested temperatures and resulted in up to two times higher DON contaminations. In addition, field experiments where spore deposition during anthesis as well as disease incidence, fungal amount and mycotoxins were observed, showed that the overwintered straw treatment resulted, depending on the year, in a three times higher FG incidence and DON content compared with the control and freshly inoculated straw treatment.  相似文献

The Fusarium species complex of maize kernels and stem pieces as well as mycotoxin contamination of commercial grain maize hybrids for animal feed were evaluated in Switzerland. Throughout 2 years, natural Fusarium infection varied significantly between the years and the locations and it ranged from 0.4% to 49.7% for kernels and from 24.2% to 83.8% for stem pieces. Using the agar plate method, 16 different Fusarium species were isolated from kernels and 15 from stem pieces. The Fusarium species composition, prevalence and impact differed between the north and the south and between kernel and stem piece samples. The dominant species on kernels in the north were F. verticillioides (32.9%), F. graminearum (31.3%), F. proliferatum (7.3%) and F. crookwellense (7.1%), in the south F. verticillioides (57.1%), F. subglutinans (24.6%), F. proliferatum (14.8%) and F. graminearum (1.5%) and on stem pieces F. equiseti (36.0%), F. verticillioides (20.1%), F. graminearum (9.5%), F. crookwellense (6.2%) and F. subglutinans (6.2%). In the south, fumonisin concentration of most hybrids exceeded guidance values for animal feed. Other Fusarium species isolated were F. avenaceum, F. culmorum, F. oxysporum, F. poae, F. sambucinum, F. semitectum, F. sporotrichioides, F. solani, F. tricinctum and F. venenatum. Maize hybrids varied in their susceptibility to Fusarium infection. Because of the high diversity of Fusarium species encountered in Switzerland representing a high toxigenic potential, we propose to screen maize hybrids for resistance against various Fusarium species and examine maize produce for several mycotoxins in order to ensure feed safety.  相似文献

Polycrystalline MgGeO3 post-perovskite was plastically deformed in the diamond anvil cell between 104 and 130 gigapascals confining pressure and ambient temperature. In contrast with phenomenological considerations suggesting (010) as a slip plane, lattice planes near (100) became aligned perpendicular to the compression direction, suggesting that slip on (100) or (110) dominated plastic deformation. With the assumption that silicate post-perovskite behaves similarly at lower mantle conditions, a numerical model of seismic anisotropy in the D' region implies a maximum contribution of post-perovskite to shear wave splitting of 3.7% with an oblique polarization.  相似文献

History, origin and extent of soil erosion on Easter Island (Rapa Nui)   总被引:2,自引:1,他引:2  
The isolated Easter Island (Rapa Nui) is an outstanding example of land degradation caused by land use in a sensitive ecosystem. The focus of the investigation was placed on Poike peninsula, the most eastern part of Rapa Nui. While Poike peninsula was once supplied with fertile soils, in large areas desertification takes place today. Detailed analysis of soil profiles allowed the reconstruction of the history and of causes and effects of soil erosion and gullying in the context of land use history and cultural evolution. The results of the stratigraphic analysis prove that from the beginning of human settlement around AD 300/600 until AD 1280 the agriculture on Poike peninsula was characterised by sustainable land use and a traditional type of agro-forestry. Soil erosion was not significant. At around AD 1280 the woodland on Poike, dominated by the endemic palm Jubaea sp., was cleared by slashing and burning. Intensive farming on the upper slopes of the volcanic peninsula resulted in sheet erosion lasting until the 20th century. Settlements and ceremonial places which were built around AD 1300 on downslope areas were buried soon by sediments. Agriculture ceased around AD 1400 on downslope areas as the fertile soils were completely eroded. From AD 1400 until the late 19th century sheet erosion and the accumulation of fine-layered sediments migrated upslope. On average 8.6 Mg of soil per hectare and per year were reworked by erosion (eroded and accumulated within the catchment). Gullying began on Rapa Nui with the sudden increase in the number of sheep during the early 20th century. Gullies are still developing on the island and their ongoing enlargement created extended badlands on Poike which pose a significant problem for ecological and archaeological conservation strategies. The application of enantioselective catalytic methods for the technical preparation of chiral agrochemicals is illustrated for three active ingredients of the acylanilide type. The key step for the technical synthesis of the herbicide (S)-metolachlor is the enantioselective hydrogenation of an imine intermediate using a novel iridium ferrocenyldiphosphine catalyst with an unprecedented high activity and 80% ee. (R)-metalaxyl and (αS,3R)-clozylacon were synthesized via the enantioselective hydrogenation of corre-sponding enamide precursors with Rh and Ru/binap catalysts with >95% and 99% enantiomeric purity, respectively. © 1998 Society of Chemical Industry  相似文献

Deoxynivalenol and zearalenone are among the most prevalent toxins produced by Fusarium spp. They have been investigated in food and feed products for decades but rarely in the environment. We therefore established solid-phase extraction and liquid chromatography-mass spectrometry (LC-MS) methods to quantify these mycotoxins at trace concentrations in aqueous natural samples. In a model emission study, we inoculated a winter wheat field with Fusarium graminearum and subsequently monitored deoxynivalenol and zearalenone in its drainage water. Before during and after harvest in June and July 2007, these toxins were emitted in concentrations from 23 ng/L to 4.9 microg/L for deoxynivalenol and from not detected to 35 ng/L for zearalenone. Simultaneously, in July and August 2007, deoxynivalenol was also detected in a number of Swiss rivers in concentrations up to 22 ng/L and zearalenone was present in several river samples below the method quantification limit. Other mycotoxins might be emitted from Fusarium-infected fields as well, because some of them are produced in similar amounts as deoxynivalenol and zearalenone and exhibit similar or even higher water solubility than deoxynivalenol. The ecotoxicological consequences of the presence of mycotoxins in surface waters remain to be elucidated.  相似文献

 将4.0%五倍子水悬浮液喷施田间马铃薯品种Bintje的叶片,于不同处理时间后,剪下叶片带回室内人工接种致病疫霉孢子囊悬浮液,保湿培养后调查各处理的发病情况。结果表明,喷施五倍子水悬浮液后48h内未经历降雨的处理,病情指数仅为7.41,与清水对照相比,防效为92.59%;喷施后96h接种的处理,由于在田间经历了降雨,完全丧失了防效,病情指数达100。室内毒力测定结果显示,0.4%五倍子水悬浮液喷施96h后,对致病疫霉游动孢子释放的抑制率仍在80%以上,在室外避雨不避光条件下五倍子水悬浮液96h内抑菌效果无显著降低,表明日光的影响作用较小。在室内盆栽条件下,通过喷淋水滴模拟田间降雨对药液的冲刷作用。结果表明,冲刷作用可导致五倍子水悬浮液对马铃薯晚疫病的防效显著降低。进一步说明,降雨是导致五倍子水悬浮液对马铃薯晚疫病田间防治效果降低的主要气象因素。  相似文献

It has widely been acknowledged that the diversity of arbuscular mycorrhizal fungi (AMF) is greatly affected by climate, land use intensity, and soil parameters. The objective of this study was to investigate AMF diversity in multiple agricultural soils (154 sites; 92 grasslands and 62 croplands) distributed over all agricultural regions in Switzerland and differing in a number of soil parameters (e.g., land use type and intensity, and altitude). We highlighted the main factors responsible for major AMF community shifts and documented specific distribution patterns for each AMF species. AMF spores were morphologically identified and counted for each species. In total, 17,924 spores were classified and 106 AMF species were identified. In general, AMF species richness (SR) was higher in grasslands than in croplands. In croplands, SR increased with altitude but this trend was not observed in grasslands. Some species occurred at virtually all sites, while others were rarely detected, and for others, species-specific distribution patterns were revealed. Some species were affected by land use type or intensity, or related factors like soil organic matter, soil microbial biomass and respiration or nutrient availability. Other species were more affected by soil pH and related parameters like base saturation and carbonate contents, by soil texture, or by altitude, or by a combination of two to several of all these parameters. We conclude that a high number of AMF species may serve as indicator species for specific habitats and land use. These species might deliver certain ecosystem services at their habitats and deserve further investigation about their functional diversity.  相似文献
