The concept that tumors are maintained by dedicated stem cells, the so-called cancer stem cell hypothesis, has attracted great interest but remains controversial. Studying mouse models, we provide direct, functional evidence for the presence of stem cell activity within primary intestinal adenomas, a precursor to intestinal cancer. By "lineage retracing" using the multicolor Cre-reporter R26R-Confetti, we demonstrate that the crypt stem cell marker Lgr5 (leucine-rich repeat-containing heterotrimeric guanine nucleotide-binding protein-coupled receptor 5) also marks a subpopulation of adenoma cells that fuel the growth of established intestinal adenomas. These Lgr5(+) cells, which represent about 5 to 10% of the cells in the adenomas, generate additional Lgr5(+) cells as well as all other adenoma cell types. The Lgr5(+) cells are intermingled with Paneth cells near the adenoma base, a pattern reminiscent of the architecture of the normal crypt niche. 相似文献

In this study an in vitro assay was optimized to detect feline proliferating lymphocytes as an assessment for the cell-mediated immune response. For this purpose, 5-bromo-2'-deoxyuridine (BrdU) labeling was chosen because of its sensitivity and the possibility of further characterization of proliferating cells. The assay was optimized by selecting the best batch and concentration of fetal bovine serum, β-mercaptoethanol concentration, cell density, BrdU incubation time and antigen presenting cell type. Cats were vaccinated with the attenuated Nobivac vaccine Tricat and the peripheral blood lymphocyte proliferation responses were quantified upon in vitro restimulation with inactivated and infectious feline panleukopenia virus (FPV), feline calicivirus (FCV) and felid herpesvirus 1 (FeHV-1). Proliferation signals were detected with inactivated FeHV-1 in the CD8(+) but not in the CD8(-) T lymphocyte population, with inactivated FCV and FPV in both CD8(-) and CD8(+) T lymphocyte populations. Restimulation with infectious FCV caused significant proliferation in the CD8(-) T lymphocyte population only while infectious FPV and FeHV-1 seemed to suppress lymphocyte proliferation in both T cell populations. Additional IFN-γ quantification in the culture supernatant revealed a large correlation between the proliferation signals and IFN-γ production, indicating that BrdU labeling is a very reliable technique to assess and characterize feline lymphoproliferative responses to viral antigens in vitro. 相似文献

ABSTRACT: The presence of foreign cells within the tissue/circulation of an individual is described as microchimerism. The main purpose of the present investigation was to study if microchimerism occurs in healthy sows/fetuses and if porcine reproductive and respiratory syndrome virus (PRRSV) infection influences this phenomenon. Six dams were inoculated intranasally with PRRSV and three non-inoculated dams served as controls. Male DNA was detected in female fetal sera of all dams via PCR. Male DNA was also detected in the maternal circulation. Sex-typing FISH showed the presence of male cells in the female fetal organs and vice versa. PRRSV infection did not influence microchimerism, but might misuse maternal and sibling microchimeric cells to enter fetuses. 相似文献

ABSTRACT: Domestic cats are commonly affected by viral pathogens that induce lengthy infections with fatal outcomes. Prevention of viral propagation is of primordial importance in shelters and catteries, where cats from different backgrounds have narrow contacts. Oligonucleotides (ODN) containing cytosine-phosphate-guanosine motifs of class A (CpG-A) are highly potent synthetic inducers of innate antiviral mechanisms. The aim of this study was to test their ability to modulate innate immune responses and prevent viral replication as stand-alone agents in the domestic cat. CpG-A stimulation of feline peripheral blood mononuclear cells (PBMCs) enhanced their proliferation, increased the presence of co-stimulatory molecules on their surface and influenced their gene expression profiles in an antiviral orientation. Incubation of the supernatants of CpG-A stimulated PBMCs with feline cell lines of epithelial and fibroblastic origin induced expression of the antiviral myxovirus resistance (Mx) gene in these target cells, which also showed enhanced resistance to feline viruses from five distinct families, namely Coronaviridae, Herpesviridae, Caliciviridae, Parvoviridae, and Retroviridae. Most importantly, subcutaneous administration of CpG-A in domestic cats systemically increased the expression of Mx, reaching maximal levels within 24 h. Plasma from treated cats could furthermore inhibit viral replication in vitro. Altogether, our data highlight the promising potential of CpG-A to induce a preventive antiviral state in the cat and to protect feline populations against a broad range of virus infections. 相似文献

An analytical model to predict the penetration of adhesives into hardwood is proposed. Penetration into hardwood is dominated by the vessel network which prohibits porous medium approximations. The model considers two scales: (1) a one dimensional capillary fluid transport of a hardening adhesive through a single, straight vessel with diffusion of solvent through the walls of the vessel; and (2) a mesoscopic scale based on topological characteristics of the vessel network. Given an initial amount of adhesive and applied bonding pressure, the portion of the filled structure could be calculated. The model was applied to beech samples joined with three different types of adhesive (PUR, UF, PVAc) under various growth ring angles as described by Hass et al. (2011). ‘Maschanzker’ is one of the best-known old apple cultivars in Austria and in the 19th century this variety also played an important role in the fruit production of South Tyrol (Northern Italy). ‘Maschanzker’ is believed to originate from one of the oldest German apple varieties ‘Borsdorfer’, which is even considered a synonym. However, since the denomination ‘Maschanzker’ is part of the name of a number of more or less similar types of apple, this study aimed at investigating the synonymy and homonymy as well as the relationships within and between different accessions of ‘Maschanzker’, ‘Borsdorfer’ and ‘Edelborsdorfer’ by employing 14 microsatellite DNA markers. A total of 38 accessions from several collections in Europe were analysed and 14 different genotypes were identified, which were assigned to three groups. Our data suggest a series of parent-offspring connections within the here defined Maschanzker group and the Edelborsdorfer group, while no closer relationship was evident to or among the accessions of the Borsdorfer group. Molecular genetic analyses thus provide fundamental data which can serve as a basis for the pomological revision of the ‘Maschanzker’/‘Borsdorfer’ cultivar complex. 相似文献

Blackcurrants, treated with 0.1 kg of 2,4,5-T ha?1 (as esters of mixed C4–C6 alcohols; ‘Tormona 80’), contained 0.1 mg of 2,4,5-T residues kg?1 in the berries at ripeness 29 days after treatment. Total residues in the berries were not reduced during growth and ripening, although the residue concentrations declined in the same period due to growth dilution. In spinach leaves from old plants, treated with 0.1 kg ha?1, 0.05 mg of 2,4,5-T kg?1 was found 14 days after treatment. Fodder peas showed no residues (< 0.002 mg kg?1) at harvest 62 days after treatment with 2,4,5-T esters. After application of 0.1 kg ha?1 on potato plants, the disappearance of 2,4,5-T was rapid during the first month, but residues were translocated into the tubers and reached a constant level of 0.02 mg kg?1 after 1 month until harvest at 108 days after treatment. In all crops, visible effects were observed after treatment with 0.1 kg ha?1. After the application at 0.01 kg ha?1, phytotoxic effects were observed only in blackcurrants, but negligible residues were found in all the test crops. 相似文献
