Experimental studies with foot-and-mouth disease virus type Asia-1, responsible for the 2005 epidemic in China

This study was carried out to investigate the biological characteristics of the foot-and-mouth disease (FMD) virus strain Asia-1 China/2005, which is responsible for the 2005 epidemic in China. The result showed that this strain is not host restricted, and could not only cause FMD in cattle and sheep but also in pigs by either inoculation or direct contact.     

Phase II Clinical Trial of Carboplatin in Canine Transitional Cell Carcinoma of the Urinary Bladder

Fourteen dogs with histologically-confirmed transitional cell carcinoma (TCC) of the urinary bladder were treated with 300 mg/m² carboplatin every 3 weeks. Response to therapy was assessed with abdominal radiography, double contrast cystography, urinary bladder ultrasonography and thoracic radiography before therapy and at 6–week intervals during therapy. Dogs were monitored for hematologic toxicity with a CBC and platelet count performed immediately before and 10 to 14 days after carboplatin treatment. Tumor responses included progressive disease in 11 dogs and stable disease in 1 dog. Two dogs were euthanized due to carboplatin toxicity before assessment of tumor response. Toxicity included thrombocytopenia with or without neutropenia in 7 dogs and gastrointestinal toxicity in 6 dogs. Carboplatin therapy was not beneficial in the treatment of TCC in the 14 dogs in this study.     

Effects of dietary lipid levels on lipid deposition and activities of lipid metabolic enzymes in hybrid tilapia (Oreochromis niloticus × O. aureus)

This study was designed to investigate the effects of dietary lipid levels on growth performance, lipid deposition and activities of lipid metabolic enzymes in hybrid tilapia (Oreochromis niloticus × O. aureus). Four isonitrogenous (300 g/kg crude protein) experimental diets containing graded levels of lipid (25, 55, 85 and 115 g/kg) were randomly assigned to triplicate groups of 180 juvenile fish. Fish were fed twice daily for 8 weeks. At the end of the experiment, the growth performance and proximate composition of fish were determined. The activities and gene expression of lipoprotein lipase (LPL) and hormone-sensitive lipase (HSL) were assessed as well. Fish fed the diets with 55 and 85 g/kg lipid had a significantly (p < 0.05) higher body weight gain than those fed the diets with 25 and 115 g/kg lipid. The whole-body and liver lipid contents were significantly (p < 0.05) elevated with increasing dietary lipid levels. Moreover, the activities and mRNA abundances of LPL and HSL in the liver, dorsal muscle and fat tissues were markedly altered by dietary lipid levels. Our data demonstrate a profound influence of dietary lipid levels on the growth and lipid deposition in hybrid tilapia, which is likely associated with the regulation of lipid metabolic enzymes including LPL and HSL.     

牦牛瘤胃内饲料蛋白质降解率的研究

用瘤胃尼龙袋技术测定了１１种常见饲料在牦牛瘤胃内的降解率。根据饲养水平,瘤胃食糜的外流速度估计为０．０６７／ｈ。测定结果表明,玉米、豌豆秸粉、菜籽饼、豌豆、鱼粉、曲拉、肉骨粉、青稞、甲醛鱼粉、甲醛菜籽饼和麸皮的粗蛋白（ＣＰ）降解率分别为０．６８、０．５６、０．４４、０．８５、０．４９、０．５５、０．８１、０,８３、０．３８、０,４８和０．７９,干物质（ＤＭ）降解率依次为０．６８、０．３０、０．４９、０．７３、０．５０、０,５４、０,５９、０．８２、０．３２、０．４９和０．７４。     

硫辛酸对海马CA1区锥体细胞凋亡的抑制作用

采用股动脉放血并双侧颈总动脉夹闭制作全脑缺血再灌注模型 ,于术后 1 d处死动物 ,取脑 ,制作石蜡切片 ,通过 HE染色、Tunel检测 ,以观察海马 CA1区锥体细胞的损伤形式 ,并探讨α-硫辛酸 (α-lipoic acid,L A)对实验性脑缺血再灌注的神经保护作用。结果表明 :在短暂性全脑缺血中 ,海马锥体细胞存在着凋亡和坏死两种死亡形式 ;与对照组相比 ,缺血再灌注后1 d,海马 CA1区损伤较重 ,出现了较多的凋亡细胞 ,而 L A可明显抑制锥体细胞的凋亡。实验证明 :在脑缺血再灌注损伤中 ,细胞凋亡是神经元死亡的一种重要形式 ;L A对脑缺血再灌注有明显神经保护作用     

短时高温处理对意大利蝗卵子发生期HSP70蛋白表达的影响

本研究采用免疫组织化学分析方法,测定了HSP70蛋白在意大利蝗卵子发生期的表达定位及33~42℃高温处理对其相对表达量的影响。结果表明,1)在卵黄发生前期,卵母细胞和滤泡细胞均表达HSP70蛋白,而在卵黄发生期和卵黄发生后期,HSP70蛋白阳性表达位于滤泡细胞;2)在33~42℃温度范围内,随着温度升高,意大利蝗卵子发生期HSP70蛋白相对表达量呈现先上升后下降的变化趋势;其中36℃处理组HSP70蛋白相对表达量最高,显著高于27℃对照组(P0.05);42℃处理组HSP70蛋白相对表达量最低,但与27℃对照组差异不显著(P0.05)。短时高温处理对意大利蝗卵子发生期HSP70蛋白相对表达量有显著影响,推测HSP70蛋白的表达在意大利蝗抵抗短时高温胁迫过程中具有重要作用。     

利用质量控制图监控不同实验条件对布鲁氏菌试管凝集实验结果的影响

运用建立的质量控制图,监控不同实验条件对布鲁氏菌试管凝集实验结果可能造成的影响。在每次日常检测时对标准阳性血清进行试管凝集实验并使用浊度仪测量其上清液的光密度(OD值),在收集足够有效数据后,绘制当前条件下的质量控制图。通过模拟日常实验条件下孵育温度、时间等容易产生偏离的条件,记录不同实验条件下标准阳性血清试管凝集实验后上清液的OD值,绘制控制用控制图。运用绘制的质量控制图对不同实验条件进行监控,发现孵育温度和孵育时间均对实验结果产生较大影响,从而导致失控。采用质量控制图对布鲁氏菌试管凝集实验进行监控,可提高质量监控的有效性、科学性。严格控制实验条件是保证实验结果的重要前提。     

安卡纯系肉鸡六个微卫星位点遗传多样性研究

安卡肉鸡是生长速度最快的有色羽肉鸡之一,具有适应性强、耐应激、饲料报酬高等特点。为了了解其群体遗传结构,实验利用6个微卫星标记对3个品系共150只个体进行遗传多样性检测。结果表明3个品系都具有较高的多态性,反映了群体基本的遗传特征,为安卡肉鸡的育种和利用提供了基本的研究数据。     

Expression and Hormonal Regulation of Hoxa10 in Canine Uterus During the Peri-implantation Period

Hoxa10, a homeobox gene, is necessary for endometrial receptivity to blastocyst implantation. The aim of this study was to investigate the differential expression of Hoxa10 in canine uterus during early pregnancy and its regulation under different conditions by in situ hybridization. Hoxa10 mRNA was mainly localized in glandular epithelium and myometrium in canine uterus. There was a low level of Hoxa10 expression in the glandular epithelium on days 6, 12 and 17 of pregnancy. On day 20 of pregnancy when embryo implanted, Hoxa10 mRNA was highly expressed in the glandular epithelium surrounding the embryo, but not in the luminal epithelium. The expression of Hoxa10 mRNA gradually declined from day 23 and reached a low level on day 28. In the myometrium, a low level of Hoxa10 mRNA signal was seen on days 6, 12 and 17 of pregnancy and reached a high level on day 20 of pregnancy. During the estrous cycle, a high level of Hoxa10 mRNA expression was seen in the estrous uterus. Either estrogen or progesterone significantly induced the expression of Hoxa10 mRNA in the ovariectomized canine uterus. These results suggest that Hoxa10 expression is closely related to canine embryo implantation and upregulated by estrogen and progesterone.     

不同养蜂合作组织形式对蜂蜜质量控制效果探讨

本文对2006年至2008年不同养蜂合作组织形式提供的蜂蜜原料进行了质量跟踪,对蜂蜜原料中的果糖和葡萄糖、蔗糖、羟甲基糠醛、淀粉酶活性、氯霉素、四环素族抗生素、碳-4植物糖含量等指标进行了检测分析,研究结果表明：不同的养蜂合作组织形式,由于对组织内养蜂户管理力度不同,其所生产的蜂蜜质量也有所不同。以亲情纽合的联合方式、产地公司＋当地固定养蜂户的联合方式和供应商＋转地固定养蜂户的联合方式生产的原料蜂蜜质量可靠性较强;公司＋无固定养蜂户的联合方式效果较差。
