This is a review of current post-harvest entomology research conducted by the Agricultural Research Service, the research branch of the US Department of Agriculture. The review covers both durable and perishable commodities. Research on biochemistry, genetics, physiology, monitoring and control of insects infesting stored grain, dried fruits and nuts, and processed commodities is reviewed. Research on development of quarantine treatments, particularly for fruit flies, is also reviewed, including research on thermal and irradiation treatments and a discussion of risk management for quarantine pests. Two areas of research are covered more extensively: a project to map the genome of the red flour beetle, Tribolium castaneum, and the use of near-infrared spectroscopy for detection of hidden infestations in grain, quantification of insect fragments in food, determination of quality in dried fruits, identification of insect species and age-grading insects. Future research directions are identified.  相似文献

In a previous study, turkey coronavirus (TCV) and enteropathogenic Escherichia coli (EPEC) were shown to synergistically interact in young turkeys coinfected with these agents. In that study, inapparent or mild disease was observed in turkeys inoculated with only TCV or EPEC, whereas severe growth depression and high mortality were observed in dually inoculated turkeys. The purpose of the present study was to further evaluate the pathogenesis of combined TCV/EPEC infection in young turkeys and determine the role of these agents in the observed synergistic interaction. Experiments were conducted to determine 1) effect of EPEC dose, with and without concurrent TCV infection, and 2) effect of TCV exposure, before and after EPEC exposure, on development of clinical disease. Additionally, the effect of combined infection on TCV and EPEC shedding was determined. No clinical sign of disease and no attaching and effacing (AE) lesions characteristic of EPEC were observed in turkeys inoculated with only EPEC isolate R98/5, even when turkeys were inoculated with 10(10) colony forming units (CFU) EPEC (high dose exposure). Only mild growth depression was observed in turkeys inoculated with only TCV; however, turkeys inoculated with both TCV and 10(4) CFU EPEC (low dose exposure) developed severe disease characterized by high mortality, marked growth depression, and AE lesions. Inoculation of turkeys with TCV 7 days prior to EPEC inoculation produced more severe disease (numerically greater mortality, significantly lower survival probability [P < 0.05], increased frequency of AE lesions) than that observed in turkeys inoculated with EPEC prior to TCV or simultaneously inoculated with these agents. Coinfection of turkeys with TCV and EPEC resulted in significantly increased (P < 0.05) shedding of EPEC, but not TCV, in intestinal contents of turkeys. These findings indicate that TCV infection predisposes young turkeys to secondary EPEC infection and potentiates the expression of EPEC pathogenicity in young turkeys.  相似文献

Domestic houseflies (Musca domestica Linnaeaus) were examined for their ability to harbor and transmit turkey coronavirus (TCV). Laboratory-reared flies were experimentally exposed to TCV by allowing flies to imbibe an inoculum comprised of turkey embryo-propagated virus (NC95 strain). TCV was detected in dissected crops from exposed flies for up to 9 hr postexposure; no virus was detected in crops of sham-exposed flies. TCV was not detected in dissected intestinal tissues collected from exposed or sham-exposed flies at any time postexposure. The potential of the housefly to directly transmit TCV to live turkey poults was examined by placing 7-day-old turkey poults in contact with TCV-exposed houseflies 3 hr after flies consumed TCV inoculum. TCV infection was detected in turkeys placed in contact with TCV-exposed flies at densities as low as one fly/bird (TCV antigens detected at 3 days post fly contact in tissues of 3/12 turkeys); however, increased rates of infection were observed with higher fly densities (TCV antigens detected in 9/12 turkeys after contact with 10 flies/bird). This study demonstrates the potential of the housefly to serve as a mechanical vector of TCV.  相似文献

Seven captive eastern box turtles (Terrapene carolina carolina) from a large collection of North American chelonians in North Carolina became acutely ill in the fall of 2002. Five of the turtles died. Clinical signs included cutaneous abscessation, oral ulceration or abscessation (or both), respiratory distress, anorexia, and lethargy. The predominant postmortem lesion was fibrinoid vasculitis of various organs, including skin, mucous membranes, lungs, and liver. No inclusion bodies were detected by histopathology or electron microscopy of formalin-fixed tissue. An iridovirus was isolated from tissues obtained postmortem from two of the box turtles that died. The virus was characterized by electron microscopy, polymerase chain reaction, and sequence analysis of a portion of the major capsid protein as a member of the genus Ranavirus.  相似文献

In a topical bioassay with pupae and adults of the mealwormTenebrio molitor (Coleoptera: Tenebrionidae), an important pest in stored products worldwide, three insect growth inhibitors (IGRs) were tested: the two chitin synthesis inhibitors diflubenzuron and flucycloxuron, and the ecdysone agonist halofenozide (RH-0345). To address differences in toxicity, we determined with the use of a14C-labeled isotope, the pattern of absorption through the cuticle and then the distribution in the different body tissues of gut, hemolymph, integument and reproductive organs (ovaries, testis). The rate of absorption through the cuticle was highest for flucycloxuron, and this concurred with its high toxicity. In general, the patterns of absorption followed a similar trend in young and old pupae and in the adult stage. Accumulation in the reproductive system of females and males was relatively high, which may explain the strong reproductive effects of the IGRs tested. In addition, we recorded large differences in the clearance of insecticide from the insect bodyvia the excrements, with the chitin synthesis inhibitors exhibiting higher rates than halofenozide.  相似文献

Animal response to landscape heterogeneity directs dispersal and affects connectivity between populations. Topographical heterogeneity is a major source of landscape heterogeneity, which is rarely studied in the contexts of movement, dispersal, or connectivity. The current study aims at characterizing and quantifying the impacts of topography on landscape connectivity. We focus on ‘hilltopping’ behavior in butterflies, a dispersal-like behavior where males and virgin females ascend to mountain summits and mate there. Our approach integrates three elements: an individual-based model for simulating animal movements across topographically heterogeneous landscapes; a formula for the accessibility of patches in homogenous landscapes; and a graphical analysis of the plots of the simulation-based vs. the formula-based accessibility values. We characterize the functional relationship between accessibility values and landscape structure (referred to as ‘accessibility patterns’) and analyze the influence of two factors: the intensity of the individuals’ response to topography, and the level of topographical noise. We show that, despite the diversity of topographical landscapes, animal response to topography results in the formation of two, quantifiable accessibility patterns. We term them ‘effectively homogeneous’ and ‘effectively channeled’. The latter, in which individuals move toward a single summit, prevails over a wide range of behavioral and spatial parameters. Therefore, ‘channeled’ accessibilities may occur in a variety of landscapes and contexts. Our work provides novel tools for understanding and predicting accessibility patterns in heterogeneous landscapes. These tools are essential for linking movement behavior, movement patterns and connectivity. We also present new insights into the practical value of ecologically scaled landscape indices.  相似文献

Study of microbial genomes has provided new insight into the functions that pathogens require for survival in the animal host. Small genome bacterial pathogens, defined as those < or = 1/3 the size of Escherichia coli, include chlamydiae, rickettsiae and ehrlichiae, mycoplasmas, and spirochetes. The small genome size is believed to result from reductive evolution, a process of initial mutation with loss of function followed by progressive accumulation of mutations and eventual gene deletion. This is most notable in the 1.1 Mb genome of Rickettsia prowazekki in which 24% of the genome is non-coding, as compared to approximately 10% in the 4.4 Mb E. coli. Consequently, these pathogens are thus presumed to retain only the most important functions for survival and propagation. There is consistent evidence from small genomes that the genetic deletion is primarily related to the loss of metabolic function and especially reduction of multiple overlapping pathways and duplicated genes. Thus, these pathogens undergo progressive reduction in their genomes yet maintain the ability to infect, survive within, and cause disease in animals. In the face of this reductive process, what genes and associated functions are maintained? Strikingly, these pathogens devote a high percentage of their genomes to paralogous families of polymorphic surface molecules. This retention suggests that evasion of the immune response is the highest priority of obligate microbial pathogens and provides a strategy for identifying protective antigens for vaccine development to control disease.  相似文献

OBJECTIVE: To evaluate the effect of feeding aspirin and supplemental vitamin E on growth performance, lung lesions, plasma concentrations of 3-methylindole (3MI), and 3-methyleneindolenine (3MEIN)-adduct concentrations in blood and pulmonary tissues of feedlot cattle. ANIMALS: 256 crossbred steers; 64 cattle were used in experiment 1 and 192 cattle were used in experiment 2. PROCEDURES: A 2 X 2 factorial design was used for each experiment. Treatment factors were aspirin (0 or 3 g daily) and vitamin E (200 or 1,500 IU daily). Steers were housed in pens (8 steers/pen). Steers were slaughtered on days 59 and 138 for experiments 1 and 2, respectively. Lungs were grossly evaluated. 3MEIN-adduct concentrations were determined, and blood and pulmonary tissues. RESULTS: Treatment was not associated with improvement or adverse effects on weight gain, dry-matter intake, or feed efficiency in experiment 2. In experiment 1, 36 of 63 (57.1%) steers had lung lesions. Lesions were not associated with treatment or concentrations of 3MI and 3MEIN-adduct. Plasma 3MI concentration and concentrations of 3MEIN-adduct in blood and pulmonary tissues were 3.11 microg/mL, 0.51 U/microg of protein, and 0.49 U/microg of protein, respectively. Aspirin was associated with increased blood concentrations of 3MEIN-adduct for diets that did not contain supplemental vitamin E. CONCLUSIONS AND CLINICAL RELEVANCE: Differences n performance of feedlot steers were not associated with treatment diet. It is possible that concurrent exposure of feedlot cattle to other factors typically associated with development of respiratory tract disease would affect these findings.  相似文献

OBJECTIVE: To evaluate signalment, surgical treatment, postoperative complications, and future breeding success or semen production in a group of bulls with naturally occurring disease of the scrotum or testis. STUDY DESIGN: Retrospective study. ANIMALS: 21 bulls that underwent unilateral castration after evaluation for scrotal swelling. PROCEDURE: A computer-assisted search of medical records at 2 veterinary teaching hospitals was performed. Historical, diagnostic, surgical, and follow-up data were collected and analyzed for those bulls with scrotal swelling that underwent unilateral castration. RESULTS: Four of 5 pasture breeding bulls and 9 of 10 semen collection-center bulls successfully bred cows or produced viable semen within 6 months of surgery. Fourteen of 21 surgical procedures were performed after induction of general anesthesia. Sixty-six percent of procedures were performed as open castrations. Seventy-one percent of bulls developed postoperative complications, most of which were mild swellings. Unilateral castration returned 13 of 15 bulls with unilateral disease of the scrotum or testis to productive service by 6 months after surgery. CONCLUSION AND CLINICAL RELEVANCE: Unilateral castration is an effective treatment for unilateral disease of the,scrotum or testis in bulls, allowing return to reproductive function.  相似文献

Three pregnant cows were each orally challenged at 10 weeks of gestation with 600 sporulated oocysts of Neospora caninum. The number of oocysts was limited by those available. In concurrent bioassays, one oocyst per os infected each of two gerbils. Challenged cattle developed Neospora-specific antibody, cell proliferation and gamma-interferon responses. N. caninum specific PCR demonstrated persisting infection in the brains of cows 4 months after calving. Abortion was not induced and there was no evidence of transplacental infection in the healthy calves born at full-term. This experiment suggests that the dose threshold for induction of abortion exceeds 600 oocysts.  相似文献
