Purpose

Bioremediation using microorganisms is a promising strategy to remediate soil with petroleum hydrocarbons. Streptomyces sp. Hlh1, an endophytic strain, has previously demonstrated the ability to degrade crude petroleum in liquid culture. To apply this strain at field scale, it is necessary to test its ability to colonize the soil, compete with native microbiota, and remove the petroleum hydrocarbons under unfavorable conditions. Herein, a study was conducted to evaluate the performance of Streptomyces sp. Hlh1 to remove crude petroleum from contaminated sterilized and non-sterilized soils.

Materials and methods

Soils samples, contaminated with 2%, 5%, and 10% of petroleum, were inoculated with Streptomyces sp. Hlh1, and incubated at 30 °C for 4 weeks. At the end of bioremediation assays, the pollutant concentrations were determined by Gas chromatography flame ionization detector and the degradation rates were also calculated. The survival of the strain in the soil was estimated and the toxicity of metabolites was evaluated on Lactuca sativa.

Results and discussion

Streptomyces sp. Hlh1 was able to grow and remove total petroleum hydrocarbons (TPH), n-alkanes, and aromatic hydrocarbons found in soil samples. In sterilized soil samples, Streptomyces sp. Hlh1 removed up to 40% of TPH at an initial concentration of 10%. Whereas, the maximum TPH removal reached was 55% in non-sterilized soil at an initial concentration of 2%. In addition, it was observed that the degradation of aromatic hydrocarbons was more active than n-alkanes. The strain grew well and produced high biomass in contaminated soil. Lettuce seedling was found to be the adequate bioindicator to assess the toxicity of petroleum end products. Streptomyces sp. Hlh1 performed a successful bioremediation, which was confirmed through the phytotoxicity test.

Conclusions

The study shows the first insight of the contribution of free endophytic actinobacterial strain in the bioremediation of petroleum-contaminated soil; therefore, it suggests that Streptomyces sp. Hl1 can be usefully exploited at field scale.

The incidence of moniliformin (MON) producing Fusarium spp. in selected corn (Zea mays L.) samples from Mexico and the United States and the effects of alkaline cooking and the tortilla manufacturing processes on the reduction of MON were determined. The percentage of infected kernels with Fusarium spp. ranged from 0 to 22% in eight foodgrade corn samples, including six from Mexico and two from the United States. Complete (100%) reduction of MON was observed when a naturally contaminated corn sample containing 1.4 μg of MON/g of corn was used in a pilot‐scale alkaline cooking and tortilla manufacturing process. In a companion laboratory‐scale study, using a cultured corn sample containing 17.6 μg of MON/g of corn, a 71% reduction of the toxin was observed during the process. Alkaline cooking appeared to be an effective method for reduction of MON in corn.  相似文献

In 2000 Guarnera et al. proposed using ELISA in canine faeces collected from the ground to detect dogs infected with Echinococcus granulosus, thus determining sheep farms with active transmission. The objective was to evaluate the prevalence of E. granulosus infection in sheep farms of the Patagonia. Sheep farms were randomly selected in the Provinces of Río Negro, Chubut, Neuquén, Santa Cruz and Tierra del Fuego (areas with control programs) and La Pampa (comparison area). From one to three samples of fecal matter were obtained for each sheep farm, which were processed by means of copro-ELISA test with confirmation of positive samples by copro-Western blot. A total of 1042 samples were obtained from 352 sheep farms, 26 (7.3%) proving positive. Of these 5 (6.3%) were from La Pampa, 9 (13.8%) from Neuquén, 4 (4.7%) from Río Negro, 2 (2.9%) from Chubut, 1 (5.9%) from Santa Cruz and 5 (13.9%) from Tierra del Fuego. The identification of parasitized dogs is an essential activity upon which rests the strategy of control and surveillance. Arecoline tests or coproantigen test with fecal matter obtained directly from the dog contribute information on individual prevalence, while the use of coproantigens detected in ground-collected samples transfers the dog unit of observation to units of greater epidemiological value. In the present experience, the technique employed seems promising for its application in systems of epidemiological surveillance of cystic echinococcosis and in drawing a baseline on which to measure the progress of control programs in the Argentine Patagonia in subsequent years.  相似文献

In Venezuela, two non-tsetse transmitted trypanosomes, Trypanosoma evansi and Trypanosoma vivax, are the major etiological agents of animal trypanosomosis. Rodents can be experimentally infected with T. evansi in order to obtain enough parasites to prepare antigens for serological tests. On the contrary, the production of T. vivax antigens is a limiting factor in most laboratories. Since T. evansi and T. vivax have exhibited a very high immunological cross-reactivity, we have focused on the identification of antigens from T. evansi responsible for this phenomenon. The predominant 64 kDa glycosylated cross-reacting antigen was recently purified from the TEVA1 T. evansi Venezuelan isolate [Parasitology 124 (2002) 287]. Here, we purified two additional cross-reacting antigens with molecular masses of approximately 51 and 68 kDa from the cytosolic fraction of the same T. evansi isolate, by sequential chromatography on DEAE-sepharose and sephacryl S-300. Sera obtained from animals infected with T. evansi or T. vivax recognized both purified proteins, suggesting their potential use as diagnostic reagents.  相似文献

Aerosols serve as cloud condensation nuclei (CCN) and thus have a substantial effect on cloud properties and the initiation of precipitation. Large concentrations of human-made aerosols have been reported to both decrease and increase rainfall as a result of their radiative and CCN activities. At one extreme, pristine tropical clouds with low CCN concentrations rain out too quickly to mature into long-lived clouds. On the other hand, heavily polluted clouds evaporate much of their water before precipitation can occur, if they can form at all given the reduced surface heating resulting from the aerosol haze layer. We propose a conceptual model that explains this apparent dichotomy.  相似文献

The carbonization process causes different physical and chemical transformations to wood, affecting the final properties of the charcoal produced. In Brazil, the cutting and use of Ocotea porosa (Nees & Mart. Ex Nees) L. Barroso is legally prohibited. But the difficulty of identifying the species used to produce charcoal hinders enforcement efforts to protect this and other endangered tree species. The aim of this work was to characterize the anatomy of charcoal made from O. porosa at different temperatures to provide information to facilitate differentiation of the species by the authorities in charge of suppressing illegal commerce. Samples were carbonized by direct heating and final temperatures of 350, 450 and 650 °C, besides a group under a ramp heating regime also with a final temperature of 450 °C. The qualitative characteristics of the O. porosa wood were preserved at the carbonization temperatures studied permitting the use of its anatomical structures for identification of carbonized wood to support control of the illegal trade in charcoal of this species.  相似文献

Atmospheric polycyclic aromatic hydrocarbons (PAHs) were determined in particulate matter (PM10) collected in a suburban area with industrial and vehicular emissions in the Metropolitan Area of Rio de Janeiro City (Brazil). A total of 22 samples were collected between March and August 2005 by means of a high volume PM10 sampler. The particulate matter contained in the filters was extracted ultrasonically with dichloromethane. The extracts were later analyzed by gas chromatography coupled to a mass spectrometer (GC/MS). The individual concentrations of PAHs ranged between the detection limit and 0.386 ng m?3. The PAHs concentrations observed in this study were towards the lowest end of the range of values reported for other European locations and also lower than values obtained for South America. PAHs concentrations and molecular ratios showed that light cars seem to be the main contributors to PM10 emissions, but diesel vehicles are clearly minor emission sources and industrial contributions should not be disregarded until more data are obtained.  相似文献

The effects of the commonly used processing techniques of soaking (at different pH values) and cooking on the digestive and nutritive utilization of calcium, phosphorus, and magnesium from common beans (Phaseolus vulgaris L.) were studied. Before the cooking step, the beans were soaked in solutions of acid (2.6 and 5.3) or basic (8.4) pH. Chemical and biological methods were used to determine nutritional parameters in growing rats, and the fiber content of the beans was established. As the pH of the soaking solution increased, so did mineral absorption and the apparent digestibility coefficient, which reached suitable values for growing rats, due to the reduced losses of soluble minerals and the increased food intake. Metabolic utilization also improved with increased pH of the soaking solution, although the values were, in general, low as a result of urinary losses under the experimental conditions. For the experimental period of 10 days, the femur and the muscle seem to be good metabolic indicators for calcium, but not for phosphorus or magnesium. The increased amount of cellulose in the soaked seed did not have a negative effect on the digestive utilization of minerals.  相似文献

Plant tissues may show chemical changes following herbivory. In aromatic plants such changes could affect the specific compounds on which commercial exploitation is based. This possibility was analyzed for Mintosthachys mollis, a member of the Lamiaceae native to Central Argentina with medicinal and aromatic uses in the region, and two types of insect herbivores: a leaf miner and a gall insect. Analysis of the essential oils of mined/undamaged leaves, as well as leaves from stems with and without galls, revealed changes in concentrations of the two main monoterpenes. A decrease in pulegone concentration was associated with both types of insect damage, whereas menthone increased significantly only in mined leaves. Inducible chemical changes in aromatic and medicinal plants may be common and widespread; their economic implications deserve investigation.  相似文献

The African tulip tree, Spathodea campanulata, the most common tree in Puerto Rico, forms novel forest types with mixtures of native and other introduced tree species. Novel forests increase in area in response to human activity and there is no information about their biomass accumulation and nutrient cycling. We established allometric relationships and chemically analyzed plant parts of African tulip trees to determine the concentration and standing stock of chemical elements (C, N, P, K, Ca, Mg, S, Mn, Al, Fe, Na), and ash. Trees ranged in diameter at breast height from 8 to 85 cm and in height from 8.8 to 28 m. The concentrations of N, P, K, and Ca in leaves of the African tulip tree were similar to those of the native pioneer Cecropia schreberiana and higher than those of mature forest tree species in Puerto Rico. The over bark wood volume of African tulip trees in nine forest stands where it was dominant ranged from 163 to 849 m3/ha. Aboveground biomass ranged from 60 to 296 Mg/ha, and N and P stocks ranged from 190 to 988 and 32 to 137 kg/ha, respectively. Novel forests on abandoned agricultural lands can store more biomass and elements than native and plantation forest stands of similar age.  相似文献
