In this paper, we analyzed a very large field data set on intramammary infections (IMI) and the associated somatic cell count (SCC) in dairy cows. The objective of the study was to analyze the impact of coagulase-negative staphylococci (CNS) IMI on cow SCC, both mean and variability, and on the potential of these infections to have a major impact on the bulk milk SCC (BMSCC). Data and milk samples for bacterial culture were collected by Quality Milk Production Services (QMPS) between 1992 and March of 2007. The QMPS program services dairy farms in New York State and other states in the Northeastern USA and operates in conjunction with Cornell University. Only records from cows where SCC and milk production data were available, and where only one organism was isolated from bacterial cultures of milk samples (or where culture was negative) were used for this analysis. A total of 352,614 records from 4200 whole herd mastitis screening sampling qualified for this study. Within herds an average of 15% (S.D. 12%) of cows sampled were infected with CNS, ranging between 0 and 100%. Average within herd prevalence of cows with a CNS IMI and an SCC over 200,000 cells/ml was 2% (S.D. 4%) with a minimum of 0% and a maximum of 50%. Results of linear mixed models showed three distinct populations of IMI statuses: negative cultures with the lowest SCC; CNS and Corynebacterium bovis with a moderate increase in SCC, and Streptococcus agalactiae, Streptococcus spp. and Staphylococcus aureus showing an important increase in SCC. Surprisingly, milk production was slightly but significantly higher in CNS infected cows compared to culture-negative cows, whereas it was strongly reduced in cows with a major pathogen IMI. The percentage contribution of CNS infections to the BMSCC was 17.9% in herds with a BMSCC less than 200,000 cells/ml. This value decreased to 11.9 and 7.9% in herds with bulk milk SCC between 200,000 and 400,000 and over 400,000 cells/ml, respectively. We concluded that very few herds with milk quality problems would have an important increase in BMSCC that could be mostly attributed to CNS infections. On the other hand, in herds with low BMSCC, CNS infections may be an important contributor to the total number of somatic cells in the bulk milk.  相似文献

The structure of Escherichia coli succinate dehydrogenase (SQR), analogous to the mitochondrial respiratory complex II, has been determined, revealing the electron transport pathway from the electron donor, succinate, to the terminal electron acceptor, ubiquinone. It was found that the SQR redox centers are arranged in a manner that aids the prevention of reactive oxygen species (ROS) formation at the flavin adenine dinucleotide. This is likely to be the main reason SQR is expressed during aerobic respiration rather than the related enzyme fumarate reductase, which produces high levels of ROS. Furthermore, symptoms of genetic disorders associated with mitochondrial SQR mutations may be a result of ROS formation resulting from impaired electron transport in the enzyme.  相似文献

BACKGROUND: Cercospora leaf spot (CLS), caused by the fungus Cercospora beticola, is the most serious foliar disease of sugar beet (Beta vulgaris L.) worldwide. Disease control is mainly achieved by timely fungicide applications. In 2011, CLS control failures were reported in spite of application of quinone outside inhibitor (QoI) fungicide in several counties in Michigan, United States. The purpose of this study was to confirm the resistant phenotype and identify the molecular basis for QoI resistance of Michigan C. beticola isolates. RESULTS: Isolates collected in Michigan in 1998 and 1999 that had no previous exposure to the QoI fungicides trifloxystrobin or pyraclostrobin exhibited QoI EC50 values of ?0.006 µg mL?1. In contrast, all isolates obtained in 2011 exhibited EC50 values of > 0.92 µg mL?1 to both fungicides and harbored a mutation in cytochrome b (cytb) that led to an amino acid exchange from glycine to alanine at position 143 (G143A) compared with baseline QoI‐sensitive isolates. Microsatellite analysis of the isolates suggested that QoI resistance emerged independently in multiple genotypic backgrounds at multiple locations. A real‐time PCR assay utilizing dual‐labeled fluorogenic probes was developed to detect and differentiate QoI‐resistant isolates harboring the G143A mutation from sensitive isolates. CONCLUSION: The G143A mutation in cytb is associated with QoI resistance in C. beticola. Accurate monitoring of this mutation will be essential for fungicide resistance management in this pathosystem. Copyright © 2012 Society of Chemical Industry  相似文献

OBJECTIVE: To identify factors associated with increased risk of being bitten by a dog or cat in a veterinary teaching hospital. DESIGN: Unmatched case-control study. STUDY POPULATION: 207 animal caregivers. PROCEDURE: Case subjects (n = 75) were any caregiver that reported being bitten by a dog or cat. Control subjects (n = 132) were randomly selected from a list of all caregivers interacting with dogs or cats. Information on the characteristics of the caregivers, characteristics of the dogs and cats, and the nature of the interaction between the dog or cat and the caregiver was obtained by use of self-administered questionnaires. RESULTS: Caregivers were more likely to be bitten by dogs or cats that had warning signs on their cages indicating the potential to bite or that were considered difficult to handle. Caregivers interacting with cats or with older dogs and cats were more likely to be bitten. Only 37 to 55% of dogs and cats that had characteristics traditionally associated with biting or were considered likely to bite were muzzled. CONCLUSIONS AND CLINICAL RELEVANCE: Muzzling dogs and cats should be considered more frequently. Dogs and cats considered to have the propensity to bite frequently do bite, and precautions, such as muzzling, should be taken if the medical condition or conformation of the dog or cat is amenable to this type of restraint.  相似文献

Previous comparative studies on patterns of reproduction in small-mammal species focus primarily on latitudinal differences in average litter size. Few studies compare reproductive patterns among northern and southern populations at the landscape scale. Our study compares differences in seasonal patterns of reproduction in northern and southern populations of the golden mouse, Ochrotomys nuttalli, and the white-footed mouse, Peromyscus leucopus. These are remarkably similar species with regard to bioenergetics, body mass, feeding behavior, home-range size, natural history, nest-site preference, and periods of activity. Both species also exhibit very similar intraspecific seasonal patterns of reproduction across their respective geographic ranges. We found that O. nuttalli and P. leucopus switch from a summer breeding season, extending from late spring through early autumn in the north to a winter breeding season extending from late autumn through early spring in the south, near the isotherm where mean annual temperature is 15.6°C (60°F), or approximately 35° N latitude. This latitudinal isotherm provides a geographic benchmark to address future changes in patterns of reproduction attributed to climate change. Findings also suggest that length of the breeding season and patterns of reproduction between species partially explain why P. leucopus is typically more abundant than O. nuttalli in similar habitat types.  相似文献

Background

Hereditary ataxias with similar phenotypes were reported in the Smooth-Haired Fox Terrier, the Jack Russell Terrier and the Parson Russell Terrier. However, segregation analyses showed differing inheritance modes in these breeds. Recently, molecular genetic studies on the Russell group of terriers found independent mutations in KCNJ10 and CAPN1, each associated with a specific clinical subtype of inherited ataxia. The aim of this study was to clarify whether or not Smooth-Haired Fox Terriers with hereditary ataxia and dogs of other related breeds harbor either of the same mutations. A sub goal was to update the results of KCNJ10 genotyping in Russell group terriers.

Findings

Three Smooth-Haired Fox Terriers with hereditary ataxia and two Toy Fox Terriers with a similar phenotype were all homozygous for the KCNJ10 mutation. The same mutation was also found in a heterozygous state in clinically unaffected Tenterfield Terriers (n = 5) and, in agreement with previous studies, in Jack Russell Terriers, Parson Russell Terriers, and Russell Terriers.

Conclusions

A KCNJ10 mutation, previously associated with an autosomal recessive spinocerebellar ataxia in Jack Russell Terriers, Parson Russell Terriers, and Russell Terriers segregates in at least three more breeds descended from British hunting terriers. Ataxic members of two of these breeds, the Smooth-Haired Fox Terrier and the Toy Fox Terrier, were homozygous for the mutation, strengthening the likelihood that this genetic defect is indeed the causative mutation for the disease known as “hereditary ataxia” in Fox Terriers and “spinocerebellar ataxia with myokymia, seizures or both” in the Russell group of terriers.  相似文献

Sonographic evaluation of the pancreas was performed in ten healthy research dogs following intraperitoneal fluid administration. Physiological saline solution was aseptically introduced into the peritoneal cavity of each dog to create an acoustic window that would facilitate sonographic delineation of normal pancreatic tissues. Abdominal ultrasound examinations were performed in all dogs before and after the production of the diagnostic hydroperitoneum. Sonograms of the pancreas and other abdominal organs were scored on the basis of image clarity by experienced and novice ultrasonographers. Statistical analysis was then used to compare the effectiveness of each preparation method. A significant difference (P < 0.05) was established between the scores from the diagnostic hydroperitoneum sonograms and the baseline scans performed without intraperitoneal fluid placement. Sonographic visualization scores for the pancreas and other abdominal organs were increased following production of the diagnostic hydroperitoneum. These results indicate that the technique of diagnostic hydroperitoneum will significantly improve ultrasonographic visualization of the pancreas, and may prove useful in the diagnosis of canine pancreatic disease.  相似文献

Anthocyanins (ACNs) are water-soluble plant pigments that have important functions in plant physiology as well as possible health effects. Over 100 common foods were screened for ACNs, and 24 of them were found to contain ACNs. Concentrations of total ACNs varied considerably from 0.7 to 1480 mg/100 g of fresh weight in gooseberry ('Careless' variety) and chokeberry, respectively. Not only does the concentration vary, but the specific anthocyanins present in foods are also quite different. Only six common aglycones, delphinidin, cyanidin, petunidin, pelargonidin, peonidin, and malvidin, were found in all of these foods. However, their sugar moieties and acylation patterns varied from food to food. Results from this study will add to the available data for the USDA Nutrient Database of flavonoids. On the basis of the concentration data and updated food intake data from NHANES 2001-2002, the daily intake of ACNs is estimated to be 12.5 mg/day/person in the United States. Of the different aglycones, cyanidin, delphinidin, and malvidin were estimated to contribute 45, 21, and 15%, respectively, of the total ACN intake. Nonacylated contributed 77% compared to 23% from acylated ACNs.  相似文献
