The present study is aiming at evaluating the hepatoprotective and antioxidant effects of Astragalus polysaccharide (APS) on the carbon tetrachloride (CCl4)-induced hepatocyte and liver injury in common carp in vitro and in vivo. In vitro, APS (200, 400 and 800 μg/ml) was added to the carp primary hepatocytes before (pre-treatment), after (post-treatment) and both before and after (pre- and post-treatment) the incubation of the hepatocytes with CCl4 at 8 mM in the culture medium. APS at concentrations of 200, 400 and 800 μg/ml significantly improved cell viability and inhibited the elevation of glutamate pyruvate transaminase (GPT), glutamate oxalate transaminase (GOT), lactate dehydrogenase (LDH) and malondialdehyde (MDA) and significantly increased the reduced level of superoxide dismutase (SOD). In vivo administration of APS at the doses of 1.5 and 3 g/kg in the diet for 60 days prior to CCl4 intoxication significantly reduced the elevated activities of GPT, GOT and LDH and increased the reduced levels of total protein and albumin in the serum; meanwhile, the reduced levels of SOD, glutathione and total antioxidant capacity (T-AOC) were markedly increased and the MDA formation was significantly inhibited in liver tissue. Overall results proved the hepatoprotective action of APS, which is likely related to its antioxidant activity. The results support the use of APS as a hepatoprotective and antioxidant agent in fish.  相似文献

In recent years, policy‐makers have sought the development of appropriate tools to prevent and manage introductions of invasive species. However, these tools are not well suited for introductions of non‐target species that are unknowingly released alongside intentionally‐introduced species. The most compelling example of such invasion is arguably the topmouth gudgeon Pseudorasbora parva, a small cyprinid species originating from East Asia. A combination of sociological, economical and biological factors has fuelled their rapid invasion since the 1960s; 32 countries (from Central Asia to North Africa) have been invaded in less than 50 years. Based on a combination of monitoring surveys (2535 populations sampled) and literature reviews, this paper aims to quantify and characterise important invasion parameters, such as pathways of introduction, time between introduction and detection, lag phase and plasticity of life history traits. Every decade, five new countries have reported P. parva introduction, mainly resulting from the movement of Chinese carps for fish farming. The mean detection period after first introduction was 4 years, a duration insufficient to prevent their pan‐continental invasion. High phenotypic plasticity in fitness related traits such as growth, early maturity, fecundity, reproductive behaviour and the ability to cope with novel pathogens has predisposed P. parva to being a strong invader. The Pseudorasbora parva invasion has provided quantitative data for the development of 1) early warning systems across different spatial scales; 2) rapid eradication programmes prior to natural spread in open systems and 3) sound risk assessments with emphasis on plasticity of life history traits.  相似文献

The aim of this study was to determine the impact of feeding juvenile pikeperch diets with medicinal herb adjuvants on the growth performance, proximate body composition, fatty acids profile (whole fish, muscle tissues, viscera) and cytological and histological indicators of the liver and middle intestine. The fish (mean body weight of ca. 110 g) were fed diets with a 0.1% supplement of Astragalus radix (group A), Lonicera japonica (group L) or a mixture of these herbs ( A. radix + L. japonica ; group A/L) for 8 weeks. The herbal supplementation was not noted to have had an impact on the analysed indicators of fish growth performance, condition or feed conversion ratio ( P >0.05). Statistically significant intergroup differences were noted in the value of the hepatosomatic index, hepatocyte size, their nucleus and nucleus/cytoplasm diameter ratio ( P <0.05). Significant intergroup differences were also noted in the appearance of the hepatic parenchyma. Statistically significant intergroup differences were also noted in the protein content of the whole fish body. The analysis of the proximal composition of the fish viscera, in turn, indicated significant differences in the fat content ( P <0.05). Among the analysed group of fatty acids (saturated – SFA, monoenoic – MUFA, polyenoic – PUFA) contained in the whole fish, the fillets and the viscera, significant intergroup differences were noted with regard to SFA (viscera) and MUFA (whole fish) ( P <0.05). The total PUFA content was stable, although significant intergroup differences were noted with regard to a few of the acids that belong to this group ( P <0.05).  相似文献

The objective of the study was to investigate the hypothesis that perioperative lymphocytopenia was due to apoptosis of these cells induced by either halothane or epidural anaesthesia in dogs. The relationship between apoptosis induction and plasma concentrations of the stress hormone cortisol and the cytokines TNF-alpha and IL-10 was examined as well. The study was performed on 22 healthy mongrel dogs, equal numbers from both genders, weighing 18.3 ± 2.9 kg, and aged between 3–5 years. Dogs were divided in three groups. Eight of the animals were anaesthetized with halothane, another eight received epidural anaesthesia using lidocaine, and six served as controls. Venous blood samples were obtained immediately before (0 minute) anaesthesia, during deep anaesthesia (120 minute), and on the next day (24 hour) in order to determine the following parameters: the total lymphocyte counts, the percentage of apoptotic peripheral blood mononuclear cells (PBMC) by flow cytometry, plasma concentrations of the cytokines tumor necrosis factor – alpha (TNF-alpha) and interleukin – 10 (IL-10) by enzyme-linked immunosorbent assay (ЕLISA), and plasma cortisol levels by radioimmune assay. Both halothane and epidural anaesthesia in dogs induces apoptosis of PBMC with slight decrease in total lymphocyte counts. These immunomodulatory effects were transient and faded till the 24th hour. Concerning the mechanism of inducing lymphocyte apoptosis by general or epidural anaesthesia, it seemed that neither cortisol, nor the tested cytokines TNF-alpha and IL-10 were implicated in this process. With the objective of evaluating the effect of methionine supplementation prior to a breeding program, thirty one heifers (Bos taurus x Bos indicus) were used averaging 386  ±  29 days of age and a mean body weight of 402.6  ±  28 kg. Fifteen of the animals received a supplement (SG) during 45 days with molasses-urea mixture (2 kg molasses + 407 g urea/head/day), plus 10 g of ruminally protected methionine. The other sixteen heifers did not receive supplement (CG). Fecal and pastures samples were collected to assess dry herbage intake and digestibility. Serial ultrasound measurements from the ovary were performed in both groups to evaluate follicular dynamics. The heifers were categorized according to their follicular size and presence of a CL. Forage intake and dry matter digestibility were reduced (P  <  0.05) and body condition tended to improve (P  =  0.07) in the supplemented heifers, however, total intake, final weight, daily gain and dorsal back fat were not affected. After the supplementation period, the percentage of females in the categories < 3 mm and 3 to < 6 mm, was greater (P  <  0.05) in CG (25% and 43.7%) that in SG (0% and 26.6%) but in the follicle category of ≥  9 mm, the percentage of animals was 60% in SG and 18.8% in CG (P  <  0.05). The percentage of ovulation for the SG and CG was 86.7% and 62.5%, respectively (P  <  0.05). The combination of supplementation with methionine-urea and molasses at the end of the dry season and the anticipated onset of the rainy season favored the establishment of ovarian activity and follicular dynamics.  相似文献

Objective – To assess the relationship between total plasma protein (TPP) as measured by refractometry and serum hypoalbuminemia and hypoproteinemia in hospitalized dogs. Design – Retrospective, observational study conducted over 6‐month period between March and August 2008. Setting – University teaching hospital. Animals – Four hundred and three hospitalized dogs in an ICU. Interventions – None. Measurements and Main Results – TPP, serum albumin, total protein, glucose, urea, cholesterol was measured from dogs enrolled in study. TPP was evaluated as a predictor for hypoalbuminemia defined both as albumin <25 g/L (<2.5 g/dL) and albumin <20 g/L (<2.0 g/dL), and serum hypoproteinemia, defined as serum total protein <40 g/L (<4.0 g/dL), using logistic regression. Impact of glucose, urea, cholesterol, and total bilirubin on refractometric readings were also assessed. TPP predicted hypoalbuminemia at albumin concentrations of <25 g/L (<2.5 g/dL) and <20 g/L (<2.0 g/dL) (P<0.001). A TPP<60 g/L (<6.0 g/dL) predicted albumin <25 g/L (<2.5 g/dL) with 73% sensitivity and 86% specificity. A TPP<58 g/L (<5.8 g/dL) predicted a serum albumin <20 g/L (<2.0 g/dL) with 70% sensitivity and 80% specificity. For dogs with known risk factors where specificity optimization may be appropriate, refractometer TPP<50 g/L (<5.0 g/dL) and <48 g/L (<4.8 g/dL) predicted hypoalbuminemia at each level with >95% specificity, although sensitivity was poor. Refractometer TPP<58 g/L (<5.8 g/dL) predicted serum total protein of <40 g/L (<40 g/dL) with sensitivity of 82% and specificity of 84%. Hypercholesterolemia and hyperglycemia significantly affected TPP readings; an increase in serum glucose by 10 mmol/L (180 mg/dL) was associated with an average independent increase in refractometer TPP of 2.27 g/L (0.23 g/dL) (P<0.001, 95% confidence interval=1.08–3.47) and an increase in serum cholesterol of 1 mmol/L (38.6 mg/dL) was associated with an average independent increase in refractometer TPP of 1.36 g/L (0.14 g/dL) (P<0.001, 95% confidence interval=1.12–1.59). Conclusion – Suboptimal sensitivity limits the use of refractometric TPP for prediction of hypoalbuminemia in the context of patient screening; a high proportion of false negatives may result. However, identification of a refractometric TPP<58 g/L is strongly indicative of both serum hypoalbuminemia and hypoproteinemia, with high specificity, and warrants further investigation. Refractometric readings may be falsely increased in patients with hyperglycemia or hypercholesterolemia.  相似文献

In addition to strong natural stresses forest ecosystems in the Kola Subarctic, Russia, receive high loads of sulphur and heavy metals from the nickel smelter. To estimate soil response to acid deposition we compared the soil field data along a pollution gradient and simulated time effects. Multivariate technique was applied to investigate spatial distribution of soil field data. Time response of soils to acid deposition was evaluated with the SMART model. According to field observations there is no evidence for strong soil acidification effects close to the smelter. Concentrations of exchangeable Ca and base saturation increase, while acidity decrease in lower soil mineral horizons towards the pollution source. However, some features seem to reflect the early stages of the started acidification. Most soil profiles have low pH values. Despite increasing of exchangeable Ca and Mg towards the smelter in lower mineral horizons due to geological inheritance, they do not reveal the same trends in the upper ones. Concentration of exchangeable K in organic horizons decreases towards the smelter, thus confirming the starting acidification. As result, exchangeable base cations are depleted in the considerable part of shallow soil profiles. According to model simulation the present acid load does not effect considerably on forest soils in background areas, however, dramatic shift in soil chemistry near the smelter is expected within several decades. Due to low pool of exchangeable base cations and low weathering rate continued acid deposition can lead to increased soil acidification and nutrient imbalance.  相似文献

以环磷酰胺(CTX)为诱导物,建立罗非鱼体内外急性肝损伤模型。体外,0、5、10、15、20和25 mmol·L-1 CTX 分别作用于离体培养的罗非鱼精密肝切片(PCLS)6 h 后,测定切片培养上清中谷丙转氨酶(GPT)、谷草转氨酶(GOT)和乳酸脱氢酶(LDH)水平,同时收集肝切片,用噻唑蓝(MTT)法检测肝切片增殖活性,测定切片内总抗氧化能力(T-AOC)、丙二醛(MDA)、过氧化物歧化酶(SOD)和谷胱甘肽(GSH)水平。体内,采用胸腔注射法造模,每千克鱼体重分别注射0、10、25、75和100 mg环磷酰胺(下文以mg·kg-1表示),每隔3 d给药1次,连续3次。末次给药后第4天取血,测定罗非鱼血清中GPT、 GOT 和LDH及肝组织匀浆中T-AOC 、MDA、SOD和GSH水平。结果显示: 20~25 mmol·L-1 CTX作用体外培养精密肝切片6 h及75~100 mg·kg-1 CTX胸腔注射罗非鱼后,均可以导致GPT、GOT和 LDH 活力显著升高;T-AOC、SOD活力及GSH含量显著下降,MDA含量显著提高。同时,20 和25 mmol·L-1 CTX能导致肝切片增殖活性显著下降,分别为空白对照组的67.29%和55.41%,且有剂量依赖性。结果表明:CTX可引起罗非鱼肝损伤;分别采用20 mmol·L-1 CTX 作用体外培养PCLS 6 h或者采用75~100 mg·kg-1 CTX胸腔注射罗非鱼,每隔3 d给药1次,连续3次,均可以构建体内外急性肝损伤模型。  相似文献
