Swine influenza monitoring programs have been in place in Italy since the 1990 s and from 2009 testing for the pandemic H1N1/2009 virus (H1N1pdm) was also performed on all the swine samples positive for type A influenza. This paper reports the isolation and genomic characterization of a novel H1N2 swine influenza reassortant strain from pigs in Italy that was derived from the H1N1pdm virus. In May 2010, mild respiratory symptoms were observed in around 10% of the pigs raised on a fattening farm in Italy. Lung homogenate taken from one pig showing respiratory distress was tested for influenza type A and H1N1pdm by two real time RT-PCR assays. Virus isolation was achieved by inoculation of lung homogenate into specific pathogen free chicken embryonated eggs (SPF CEE) and applied onto Caco-2 cells and then the complete genome sequencing and phylogenetic analysis was performed from the CEE isolate. The lung homogenate proved to be positive for both influenza type A (gene M) and H1N1pdm real time RT-PCRs. Virus isolation (A/Sw/It/116114/2010) was obtained from both SPF CEE and Caco-2 cells. Phylogenetic analysis showed that all of the genes of A/Sw/It/116114/2010, with the exception of neuraminidase (NA), belonged to the H1N1pdm cluster. The NA was closely related to two H1N2 double reassortant swine influenza viruses (SIVs), previously isolated in Sweden and Italy. NA sequences for these three strains were clustering with H3N2 SIVs. The emergence of a novel reassortant H1N2 strain derived from H1N1pdm in swine in Italy raises further concerns about whether these viruses will become established in pigs. Boldenone and its precursor Boldione are illegally used for anabolic purposes in humans, horses and cattle. To develop more effective policies and programs to maximize food security, Italian Public Health Services investigate all indicators capable of assisting the recognition of treated animals, and prioritize research and the formulation of action strategies for the promotion of healthy eating. Thus, an experimental administration of boldenone and boldione at anabolic dosages in veal calves was carried out to evaluate the changes in target organs by qualitative and semi-quantitative morphological analysis. The lesions resembled the effects already observed after the administration of androgen hormones to cattle. Main findings were represented by prostate hypersecretion, increased rate of apoptotic cells and decreased rate of Ki67 positive cells in the germ cell line of treated animals, particularly in boldione group and finally some new features like hypertrophy of the prostate urothelial cells. 相似文献

Influenza is a highly contagious disease that has burdened both humans and animals since ancient times. In humans, the most dramatic consequences of influenza are associated with periodically occurring pandemics. Pandemics require the emergence of an antigenically novel virus to which the majority of the population lacks protective immunity. Historically, influenza A viruses from animals have contributed to the generation of human pandemic viruses and they may do so again in the future. It is, therefore, critical to understand the epidemiological and molecular mechanisms that allow influenza A viruses to cross species barriers. Objective— To determine thoracolumbar spinal movement in dogs and the influence of subclinical radiographic changes involving the lumbosacral junction. Study Design— Experimental study. Animals— Clinically sound Malinois dogs (n=22). Methods— Kinematic analysis of markers on the spinal processes of C7, T6, T13, L3, L7, and S3 was performed while dogs were walking on a treadmill. Range of motion (ROM) in the transverse and vertical direction and the time of occurrence (TOO) of the maximal marker position were calculated. ROM and TOO of angulations formed by the corresponding markers were calculated. Initial kinematic analysis was performed without knowledge of the radiographic changes, and then data were reanalyzed to determine whether vertebral changes influenced back motion. Based on the results of radiographic analysis of the lumbosacral junction, dogs were divided into 3 groups: 1=no radiographic changes; 2=shortened L7 vertebra; and 3=transitional vertebrae, spondylosis, subluxations, and spondylarthrosis of the lumbosacral junction. ROM and TOO were compared using ANOVA for repeated measures and a Bonferroni's post hoc test; P<.05 was considered significant. Results— The highest transverse ROM was achieved by markers T6, T13, and L3, and in the vertical direction by S3; however, there were no significant differences in ROM in horizontal angulations. In the sagittal plane, T13–L3–L7 had a lower angulation than L3–L7–S3. In Group 3, transverse ROM for C7 was significantly higher than in Group 1; the horizontal angular maximum of T13–L3–L7 occurred significantly earlier. Conclusions— Significant kinematic changes were detected between clinically sound dogs with radiographic lumbosacral changes and dogs with no radiographic abnormalities. Clinical Relevance— Kinematic data from clinically sound dogs can be used for comparison with data from dogs with gait disturbances associated with orthopedic or neurologic disease or changes associated with therapy. 相似文献

Grassland extensification is followed by a change of soil organic‐matter (SOM) contents. In order to give a better assessment of these developments on grassland sites in SW Germany, the CENTURY Soil Organic Matter Model was used on five long‐term experimental sites under three different management practices (“Mowing”, “Mulching” (mowing without removal of the phytomass), and “Natural succession”). On these sites, soil‐organic‐carbon (SOC) monitoring was continuously done for soil depths of 0–4 and 4–8 cm from 1975 to 2002. The contents of organic carbon (OC) were at steady state or showed a slight decrease for the mulched and succession plots. Carbon contents of the mowed plots were decreasing. Measured C contents were transferred into stocks and compared with the simulated OC stocks. Linear regressions between observed and simulated C stocks were calculated separately for mulched and succession plots. The regressions for OC yielded significant relationships (R2 = 0.8) for both kinds of plots. However, the model did not reproduce the short‐term dynamics of C stocks. Whereas SOC stocks on mulched and succession plots are expected to stay stable for the next decades according to the simulation, they are expected to decrease for a couple of years before stabilization on mowed plots. 相似文献

Coculture is a productive technique to trigger microbes’ biosynthetic capacity by mimicking the natural habitats’ features principally by competition for food and space and interspecies cross-talks. Mixed cultivation of two Red Sea-derived actinobacteria, Actinokineospora spheciospongiae strain EG49 and Rhodococcus sp. UR59, resulted in the induction of several non-traced metabolites in their axenic cultures, which were detected using LC–HRMS metabolomics analysis. Antimalarial guided isolation of the cocultured fermentation led to the isolation of the angucyclines actinosporins E (1), H (2), G (3), tetragulol (5) and the anthraquinone capillasterquinone B (6), which were not reported under axenic conditions. Interestingly, actinosporins were previously induced when the axenic culture of the Actinokineospora spheciospongiae strain EG49 was treated with signalling molecule N-acetyl-d-glucosamine (GluNAc); this finding confirmed the effectiveness of coculture in the discovery of microbial metabolites yet to be discovered in the axenic fermentation with the potential that could be comparable to adding chemical signalling molecules in the fermentation flask. The isolated angucycline and anthraquinone compounds exhibited in vitro antimalarial activity and good biding affinity against lysyl-tRNA synthetase (PfKRS1), highlighting their potential developability as new antimalarial structural motif. 相似文献
