Background

Measurement of salivary cortisol has been used extensively as a non-invasive alternative to blood sampling to assess adrenal activity in ruminants. However, there is evidence suggesting a considerable delay in the transfer of cortisol from plasma into saliva. Previous studies in cattle have used long sampling intervals making it difficult to characterise the relationship between plasma and salivary cortisol (PLCort and SACort, respectively) concentrations at different time points and determine whether or not such a time lag exist in large ruminants. Therefore, the objective of this study was to characterise the relationship between plasma and salivary cortisol and determine if there is a significant time lag between reaching peak cortisol concentrations in plasma and saliva across a 4.25 h time-period, using short sampling intervals of 10–15 min, following social separation in dairy cattle.Five cows were separated from their calves at 4 days after calving, and six calves were separated from a group of four peers at 8 weeks of age. Following separation, the animals were moved to an unfamiliar surrounding where they could not see their calves or pen mates. The animals were catheterised with indwelling jugular catheters 1 day before sampling. Blood and saliva samples were obtained simultaneously before and after separation.

Results

In response to the stressors, PLCort and SACort increased reaching peak concentrations 10 and 20 min after separation, respectively. This suggested a 10 min time lag between peak cortisol concentrations in plasma and saliva, which was further confirmed with a time-series analysis. Considering the 10 min time lag, SACort was strongly correlated with PLCort (P < 0.0001).

Conclusions

Salivary cortisol correlates well with plasma cortisol and is a good indicator of the time-dependent variations in cortisol concentrations in plasma following acute stress. However, there is a time lag to reach peak cortisol concentrations in saliva compared to those in plasma, which should be considered when saliva samples are used as the only measure of hypothalamic-pituitary-adrenal axis response to stress in cattle.  相似文献

Key message

Foliar phosphorus (P) resorption in Quercus variabilis Blume was significantly lower at a P-rich than at a P-deficient site. Moreover, P resorption strongly decreased, and nitrogen:phosphorus and carbon:phosphorus resorption ratios increased with soil P content. This demonstrates a strong link between foliar P resorption and P content in soils, and emphasizes the importance of P resorption in leaves of trees growing in soils with contrasted P content.

Context

Subtropical ecosystems are generally characterized by P-deficient soils. However, P-rich soils develop in phosphate rock areas.

Aims

We compared the patterns of nutrient resorption, in terms of ecological stoichiometry, for two sites naturally varying in soil P content.

Methods

The resorption efficiency (percentage of a nutrient recovered from senescing leaves) and proficiency (level to which nutrient concentration is reduced in senesced leaves) of 12 elements were determined in two oak (Q. variabilis) populations growing at a P-rich or a P-deficient site in subtropical China.

Results

P resorption efficiency dominated the intraspecific variation in nutrient resorption between the two sites. Q. variabilis exhibited a low P resorption at the P-rich site and a high P resorption at the P-deficient site. Both P resorption efficiency and proficiency strongly decreased with soil P content only and were positively related to the N:P and C:P ratios in green and senesced leaves. Moreover, resorption efficiency ratios of both N:P and C:P were positively associated with soil P.

Conclusion

These results revealed a strong link between P resorption and P stoichiometry in response to a P deficiency in the soil, and a single- and limiting-element control pattern of P resorption. Hence, these results provide new insights into the role of P resorption in plant adaptations to geologic variations of P in the subtropics.

Tomato (Solanum lycopersicum) is susceptible to gray mold (Botrytis cinerea). Quantitative resistance to B. cinerea was previously identified in a wild relative, S. neorickii G1.1601. The 122 F3 families derived from a cross between the susceptible S. lycopersicum cv. Moneymaker and the partially resistant S. neorickii G1.1601 were tested for susceptibility to B. cinerea using a stem bioassay. Three putative quantitative trait loci (pQTL) were detected: pQTL3 and pQTL9 reducing lesion growth (LG) and pQTL4 reducing disease incidence (DI). For each pQTL, a putative homologous locus was identified recently in another wild tomato relative, S. habrochaites LYC4. pQTL3 was confirmed by assessing disease resistance in BC3S1 and BC3S2 progenies of S. neorickii G1.1601. pQTL4 was not statistically confirmed but the presence of the S. neorickii resistance allele reduced DI in all three tested populations. The reduction in LG of pQTL9 was not confirmed but rather, this locus conferred a reduced DI, similar to observations in the QTL study using S. habrochaites. The results are discussed in relation to other disease resistance loci identified in studies with other wild tomato relatives.  相似文献

Medroxyprogesterone acetate (MPA)-contaminated feed arrested the onset of farrowing, and induced post-lactational anoestrus in sows. Sixty percent of the sows developed cystic ovaries after weaning following exposure to pharmaceutical waste of MPA in glucose syrup. This waste ended up in acidified feed of by-products of a sow farm, and proved to be the cause of the disorders. Analysis by thin layer chromatography and Gas Chromatography/Mass Spectrometry of renal fat from 10 slaughter sows demonstrated residues of 2.5-8 ppb of MPA. Within the European Union use of MPA is illegal as growth promoter in production animals, and therefore MPA-exposed farms were placed under official control by the general inspection service. Clinical signs and diagnostic procedures of the initial case are presented and the role of the veterinary practitioner in detecting potential food safety hazards is discussed.  相似文献

Degenerative disorders of motor neurons include a range of progressive fatal diseases such as amyotrophic lateral sclerosis (ALS), spinal-bulbar muscular atrophy (SBMA), and spinal muscular atrophy (SMA). Although the causative genetic alterations are known for some cases, the molecular basis of many SMA and SBMA-like syndromes and most ALS cases is unknown. Here we show that missense point mutations in the cytoplasmic dynein heavy chain result in progressive motor neuron degeneration in heterozygous mice, and in homozygotes this is accompanied by the formation of Lewy-like inclusion bodies, thus resembling key features of human pathology. These mutations exclusively perturb neuron-specific functions of dynein.  相似文献

Inclusion of potato-processing waste (PW) from the frozen potato products industry in high-grain beef cattle finishing diets was evaluated in two studies. In a randomized complete block design, 125 crossbred yearling heifers (365 +/- 0.3 kg initial BW; five pens per treatment; five heifers per pen) were used to evaluate PW level on feedlot performance and meat quality. Heifers were fed for 85 (two blocks) or 104 d (three blocks). In a digestion study, four ruminally, duodenally, and ileally cannulated Holstein steers (474.7 +/- 26.6 kg initial BW) were used in a 4 x 4 Latin square design to evaluate effects of PW level on ruminal fermentation, site of digestion, and microbial protein synthesis. The control diet for both studies contained 80% corn, 10% alfalfa hay, 5% concentrated separator by-product (CSB), and 5% supplement (DM basis). Potato waste replaced corn and separator by-product (DM basis) in the diet at 0, 10, 20, 30, and 40% in the feedlot study, and at 0, 13, 27, and 40% in the digestion study. In the feedlot study, DMI decreased (linear; P = 0.007) with increasing inclusion of PW. Increasing PW decreased ADG and feed efficiency from 0 to 30% and then increased at 40% (quadratic; P < 0.01). Calculated dietary NEg concentrations did not differ among treatments (P = 0.18). Hot carcass weight decreased as PW increased from 0 to 30% and then increased at 40% PW (cubic; P < 0.01). Fat thickness and longissimus muscle area decreased with increasing PW (linear; P < 0.05). Level of PW did not affect marbling or liver scores (P > 0.30). No difference (P > 0.20) was observed for Warner-Bratzler shear force at 0, 10, 20, and 30% PW levels; however, 40% PW resulted in lower (P = 0.05) shear force values. Taste panel scores for juiciness and flavor intensity did not differ with increasing PW (P > 0.30). Steaks from cattle fed 0% were scored less tender than 10 and 40% PW (cubic; P < 0.05). In the digestion study, DMI decreased (quadratic; P < 0.01) with increasing PW. Ruminal pH and total VFA concentration increased (linear; P < 0.05) and true N disappearance from the stomach complex and apparent total-tract N disappearance decreased with increasing level of PW (linear; P < 0.01). Starch intake and ruminal disappearance decreased with increasing level of PW (quadratic; P < 0.05). Inclusion of PW decreased feedlot performance, with little effect on carcass characteristics or meat quality. Optimal inclusion of PW in finishing diets may depend on the cost of transportation and other dietary ingredients.  相似文献

Two hexaploid species of Solanum sect. Solanum are present in Africa: Solanum scabrum and S. nigrum. Solanum scabrum is a widely cultivated species and is used as a leafy vegetable, as a source of medicine and as a source of ink dye. In previous studies a wide range of morphological diversity has been reported in this species and in some studies subspecies have been proposed. Subspecies are also recognized in S. nigrum. However, it has not been established whether or not the morphological differences are reflected at the genomic level. The present study applies AFLPs to study the genetic diversity in S. scabrum and its relationship to geographical provenance, morphological differences and the possible existence of subspecies within S. scabrum and S. nigrum. The data obtained were analyzed with cluster analysis (using UPGMA and NJ). The results indicate that the genetic variation within S. scabrum was higher within accessions than between accessions. Accessions did not cluster according to their geographical provenance, indicating that accessions from different geographical areas were not significantly different genetically. The clustering reflected neither morphological differences nor domestication status (cultivated or wild). The morphological differences exhibited by S. scabrum could be due to selection by farmers for different plant types. The AFLP derived clustering pattern did not segregate the subspecies recognized in S. scabrum and S. nigrum into separate subclusters.  相似文献

Using Atlantic cod, Gadus morhua, as a model for the stress response in gadoid fish, the changes in the expression of some stress and immune genes as well as the profiles of plasma cortisol were examined. Adult fish were kept at a density of ca. 100 kg m?3 by lowering the water level in the rearing tank for 1 h and this short‐term crowding stress was repeated thrice over a 12‐h interval period. Blood samples were collected before exposure and at 2, 24 and 72 h post crowding. Plasma cortisol level significantly increased at 2 h post crowding but returned to pre‐crowding levels 24 h after exposure. The relative expression of the stress response genes, glucose transporter‐3 and a putative heat shock protein 70 significantly increased at 2 and 24 h post crowding respectively. Significant up‐regulation of the pro‐inflammatory cytokines, interleukin (IL)‐1β and IL‐8, as well as anti‐bacterial genes, g‐type lysozyme and bactericidal permeability‐increasing protein/lipopolysaccharide‐binding protein (BPI/LBP) was also observed at 2 h and the levels were maintained until 72 h post exposure, except for BPI/LBP which had maximum up‐regulation at 24 h. The present observations have implications with respect to fish welfare and assessment of the health status of the farmed fish.  相似文献
