Alpha-amylase, a major human salivary protein, and IB8c, a representative of the proline-rich proteins, were obtained by isolation from saliva and by solid-phase synthesis, respectively. The interactions between these proteins and condensed tannins isolated from grape seeds were studied at different protein and tannin concentrations by measuring their aggregation. Pectic polysaccharides were isolated from wine, and their effect on protein tannin aggregation was assessed. The results presented in this study showed that the most acidic fractions of arabinogalactan proteins have the ability to inhibit the formation of aggregates between the grape seed tannins and the two different salivary proteins. Rhamnogalacturonan II has the same ability toward alpha-amylase but not IB8c under the conditions of the present study. Polysaccharides show effects at concentrations at which they are present in wine, which could mean an influence in wine astringency. The interaction between condensed tannins and alpha-amylase is differently affected by ionic strength when compared with IB8c. 相似文献

为了研究风电机组尾流对下游风电机组载荷的影响,假设了几个重要尾流参数:上下游风电机组间距、上游风电机组推力系数、湍流强度等。采用Bladed软件和Matlab幅频程序,分别对1.5与3.0 MW双馈式风电机组进行各参数与载荷响应的关系计算。结果表明:风轮处风速会随着推力系数的增大非线性减小;风电机组处于尾流影响范围内时,在风速和推力系数相同的条件下,通常湍流强度受尾流影响后减小使载荷增大,但当推力系数对载荷的影响起主导作用时,虽然湍流强度受尾流影响后减小但载荷会增大;当风速大于额定风速时,应采用变桨控制减小推力系数,以减小风电机组的疲劳损伤。 相似文献

Chemical investigation of a red-type Cuban propolis sample has led to the isolation of 11 isoflavonoids (2 isoflavones, 3 isoflavans, and 6 pterocarpans), together with gallic acid, isoliquiritigenin, and (-)-liquiritigenin. Structural determination, including the absolute stereochemistry, was accomplished by spectroscopic analysis, particularly CD and 2D NMR techniques. The fragmentation behavior of pterocarpans was studied by electrospray ionization (ESI) tandem mass spectrometry (MS/MS) using an ion-trap analyzer, and a generalized fragmentation pathway, useful in the identification and structural characterization of pterocarpans, is proposed. The feasibility of Raman spectroscopy in combination with partial least-squares (PLS) regression for the determination of individual or grouped trans-monounsaturated fatty acids (trans-MUFA) and conjugated linoleic acids (CLA) in milk fat is demonstrated using spectra obtained at two temperature conditions: room temperature and after freezing at -80 °C. The PLS results displayed capability for direct semiroutine quantification of several individual CLA (cis-9,trans-11 and trans-10,cis-12 C18:2) and trans-MUFA (trans-4-15 C18:1) in minor concentrations (below 1.0 g/100 g of milk fat). Calibration models were based on reference data cross-correlation or determined by specific scattering signals in the Raman spectra. Distinct bands for trans-MUFA (1674 cm(-1)) and CLA (1653 cm(-1)) from the trans isolated and cis,trans conjugated C ═ C bonds were identified, as well as original evidence for the temperature effect (new bands, peak shifts, and higher intensities) on the Raman spectra of fatty acid methyl ester and triacylglyceride standards, are supplied. 相似文献

Productivity and sustainability of cacao (Theobroma cacao L.) in tropical soils are affected by levels of iron. Information is lacking on the cacao response to various sources of iron (Fe). A greenhouse experiment was conducted to evaluate the effects of five iron sources iron sulfate heptahydrate, ferric ethylenediamine-N,N’-bis(2-hydroxyphenylacetic acid), ferric diethylenetriaminepentaacetic acid, ferric ethylenediaminetetraacetic acid, fiesta herbicide (FeSO4 · 7H2O, FeEDDHA, FeDTPA, FeEDTA,) at 10 mg Fe kg?1 soil on growth, photosynthesis, content of photosynthetic pigments and starch and macro- and micronutrient nutrition of cacao. The various iron sources had significant effects on shoot and root dry biomass accumulation, leaf chlorophyll a and b content, carotenoid levels, SPAD index and PN. These parameters were significantly correlated with concentration, uptake, influx, and transport and use efficiency of Fe. In cacao net photosynthesis, stomatal conductance, internal carbon dioxide (CO2), and transpiration in leaf level responded differently to the sources of Fe. Invariably, macro and micronutrient uptake, influx, transport, and use efficiency showed differential responses to sources of iron but significant effects were only observed for copper (Cu), Fe, manganese (Mn), and zinc (Zn). Overall, FeDTPA, FeEDTA and FeHEDTA could be the best sources of Fe in improving, growth, photosynthesis and macro and micro nutrition of cacao. 相似文献

In this work a study of critical storage temperatures on pigment degradation of green beans (Phaseolus vulgaris, cvs. Perona and Boby) was conducted. In this way, green beans kept better quality at 4 degrees C than either 8 or 12 degrees C, maintaining a bright green color and good texture. Nevertheless, temperatures of 4 degrees C induced chilling injury (CI) after eight days of storage, which became evident when the pods were transferred to 20 degrees C. Cold storage temperatures, 12, 8, and 4 degrees C, produced different changes on the green beans chlorophyll profile. Green beans of both cultivars, Perona and Boby, stored at 4 and 12 degrees C showed a continuous degradation of chlorophyll pigments during storage, while samples stored at 8 degrees C showed an increase of chlorophyll content at the first 15 days. Carotenoid pigments also suffered different changes during cold storage. Perona was the green beans cultivar which maintained the higher level of lutein, mainly when samples were stored at the most suitable temperature (8 degrees C). 相似文献

Histomoniasis or blackhead is a disease of gallinaceous birds, caused by the protozoon Histomonas meleagridis. Since traditional diagnostics for the detection of this disease are complex and far less sensitive than molecular tools, a PCR would provide a more rapid and sensitive alternative. However, intestinal material and droppings, which are preferably used in epidemiological studies of histomoniasis, often contain PCR inhibitory substances. To detect these false negative results, the use of an internal amplification control is essential. Nevertheless, the recently developed PCR tests lack this internal control. Therefore, a new PCR assay with H. meleagridis specific primers was developed which does include an internal amplification control. The diagnostic value of the PCR assay was evaluated in comparison to three other conventional H. meleagridis specific PCR tests (HIS5, HM1 and HM2). None of the organ samples originating from uninfected turkeys, showed positive PCR results in any of the tests. Among the lesion-positive, inhibition-free samples, 95.4% were positive by our PCR assay, while only 50, 66.7 and 83.3% of the lesion-positive organs tested positive by the HM1, the HIS5 and the HM2 PCR respectively. In conclusion, our PCR offers the use of the internal control to detect false negative results and an increased sensitivity, and thus should be useful for routine diagnosis of H. meleagridis in poultry. 相似文献

The objectives of this study were: (1) to predict the rumen fermentation pattern from milk fatty acids using a machine learning technique, i.e. artificial neural networks (ANN) combined with feature selection and (2) to compare the prediction accuracy of the resulting model to that of a statistical multi-linear regression model, based on odd and branched chain milk fatty acids. Data were collected from 10 experiments with rumen fistulated dairy cows, resulting in a dataset of 138 observations. Feature selection was based on correlation and principal component analysis, and background physiological knowledge. Different ANN architectures and training algorithms were assessed. The evaluation of the model performance, based on the test dataset, showed a root mean square prediction error, expressed relative to the observed mean, of 2.65%, 7.67% and 7.61% of the observed mean for acetate, propionate and butyrate, respectively. Compared to a multi-linear regression model, the ANN revealed not to perform significantly better. However, the results confirm that milk fatty acids have great potential to predict molar proportions of individual volatile fatty acids in the rumen. 相似文献
