Vitellogenin (Vtg) has proven to be a sensitive and simple biomarker in determining sex, sexual maturity, and xenoestrogenic effects in fish. Thus, our investigation has been focused on identification, partial characterization, and quantification of grey mullet (Mugil cephalus) Vtg through the use of a variety of biochemical and immunological analytical techniques. Mullet is considered both a promising aquaculture candidate and an important species for improving sediment quality in polyculture systems. In the first part of this work, grey mullet Vtg was purified from plasma of 17β-estradiol (E2)-induced male fish by a one-step chromatographic protocol, and partially characterized. Specific polyclonal antibodies were then raised against the mullet Vtg, and both an indirect ELISA and an optical immunosensor were set up and validated to quantify plasma Vtg. The indirect ELISA and the optical immunosensor assay developed showed linear measuring in the range 56.8–1047.1 ng mL−1 and 70–739 ng mL−1 Vtg concentrations in standard solutions, respectively. The results obtained suggest that the indirect ELISA allows Vtg detection over a wide dynamic range, thus resulting more suitable for rapid and sensitive sample screening. Therefore, we suggest that the direct immunosensor is a promising tool which needs more investigation to improve the sensitivity.  相似文献

Light competition by trees is often regarded as a major limiting factor for crops in alley-cropping agroforestry. Northern latitude farmers are usually reluctant to adopt agroforestry as they fear that light competition will be fiercer in their conditions. We questioned the light availability for crops in alley-cropping at different latitudes from the tropic circle to the polar circle with a process-based 3D model of alley-cropping agroforestry. Two tree densities and two tree line orientations were considered. The effect of the latitude was evaluated with same-sized trees. The relative irradiance of the crops was computed for the whole year or at specific times of the year when crops need more light. The heterogeneity of crop irradiance across the alley was also computed. Surprisingly, crop relative irradiance of summer crops at high latitudes is high, at odds with farmers’ fears. Best designs were highlighted for improving the crop irradiance: North–South tree lines are recommended at high latitudes and East–West tree lines at low latitudes. At medium latitudes, North–South tree lines should be preferred to achieve an homogeneous irradiance of the crop in the alley. If we assume that trees at northern latitudes grow slower when compared to southern latitudes, then alley-cropping agroforestry is highly advisable even at high latitudes with summer crops.  相似文献

This study fully describes a severe disease outbreak occurred in 2016 in black bullhead catfish farmed in Italy. Affected fish showed nervous clinical signs as well as emaciations and haemorrhagic petechiae on the skin at the fin bases, abdomen and gills. Viral isolation in cell culture allowed the subsequent identification of a rhabdovirus, tentatively named ictalurid rhabdovirus (IcRV), through electron microscopy, immunofluorescence and whole genome sequencing (WGS). The newly isolated virus, together with 14 additional viral strains stored in our repository and detected during similar mortality episodes in the period 1993–2016, was phylogenetically analysed on the basis of the nucleoprotein and the glycoprotein nucleotide and amino acid sequences. The genetic distances among Italian IcRV strains were also estimated. Our results show that all the IcRV strains belong to the genus Sprivivirus and are closely related to the tench rhabdovirus (TenRV). Italian catfish production is constantly decreasing, mainly due to viral infections, which include the newly characterized IcRV. Data presented in this work will assist to investigate the molecular epidemiology and the diffusive dynamics of this virus and to develop adequate surveillance activities.  相似文献

A field experiment was carried out in Southern Italy from 2007 to 2010 adopting a two-year rotation of tomato and maize. In this paper, the results of maize cultivation were reported, with the aim to investigate the effects of different water qualities and fertilizers on yield and soil properties. The following treatments were compared: mineral nitrogen (N) fertilizer and irrigation with fresh water (FWF); mineral N fertilizer and irrigation with saline water (SWF); Municipal Solid Waste (MSW) compost and irrigation with fresh water (FWC); MSW compost and irrigation with saline water (SWC). These treatments were compared with an unfertilized control and irrigation with fresh-water (FW0) and an unfertilized control and irrigation with saline water (SW0). At harvest, yield, grain moisture content, dry matter, grain protein, starch, fat content and soil characteristics were determined. The treatments with compost increased the average grain yield of the 11% compared to mineral fertilizer treatments. Furthermore, the grain yield in SWC increased of the 19% respect to average of SWF and SW0, indicating that MSW compost applied as amendment mitigated the adversely effects of saline water. Compost application significantly increased the Total Organic Carbon (TOC). In particular, the FWC and SWC treatments showed an average increase of the 25% compared to the mean TOC value of FWF and SWF. In the view of an urgent necessity to improve the quantity and the quality of farmed fish species, there is a strong need to improve our basic knowledge on the effects of first feeding during the developmental stages of fish larvae. High mortality, mainly due to food deprivation or inappropriate food quality, has been observed in many larval fish species, but knowledge about the morphological, biochemical and molecular processes related to this topic is still poorly understood. The understanding of the early larval ontogeny as well as the larval nutritional requirements and the molecular and cellular mechanisms elicited by fish larvae during food deprivation and starvation are thus of primary importance. At this regard, this study investigates, in Dover sole larvae, the effects of starvation and starving/re‐feeding procedures at a morphological, histological, biochemical and molecular level. The results evidenced that starved larvae progressively decrease in growth, lipid content, affected their gastrointestinal tract and muscle development and increased cortisol and heat shock protein 70 levels. On the contrary, starved and re‐fed larvae showed, after the restoration of a favourable feeding condition, a compensatory growth. In conclusion, this is the first study analysing through a multidisciplinary approach the effects of food deprivation on the development of an important economic species, the Dover sole.  相似文献

Facultative paedomorphosis, a process in which newt larvae can opt for reproduction before or after metamorphosis, is geographically heterogeneous. Despite numerous ecological studies and recent evidence of declines in paedomorphic populations, however, no attempt to model environmental variables that explain the presence of paedomorphs has been made at a multi-scale level. Our aim was to fill this gap in studying three newt species (Lissotriton vulgaris, Mesotriton alpestris, and Triturus macedonicus) of the Montenegrin karst as model species. To this end, we used multivariate analysis on three scales of habitat: the breeding pond, the land use and the climatologic features. Results show that the study area is both an important hotspot for paedomorphosis and where intraspecific diversity is quickly disappearing (20-47% extirpation) because of fish introductions. Other habitat variables (water permanency, pH or the habitat origin) were shown to act on paedomorphosis but not consistently across species, confirming complexity of the evolutionary and ecological processes. This study appeals for more long-term and detailed landscape studies of polyphenisms, a neglected but promising topic, to better understand and protect alternative modes of development. Particularly, measures should be taken to identify hotspots of intraspecific diversity at a global scale and stop fish introductions before we reach a point of no-return.  相似文献
