Random amplified polymorphic DNA fingerprinting was performed on single females of root-knot nematodes, Meloidogyne spp., using a new procedure for DNA isolation. One-fourth of the total DNA amount isolated from a single female proved to be sufficient as a template in a polymerase chain reaction. Electrophoretic patterns of the amplified fragments were reproducible between replicates from a single female or sister females from the same progeny, and identical to those obtained with genomic DNA purified from a large number of nematodes. Moreover, a comparative analysis over three successive generations showed stability of the amplification patterns, thus demonstrating the utility of this procedure for epidemiological and ecological studies on root-knot nematodes.  相似文献

Pseudomonas syringae pv.pisi, pea bacterial blight agent, is seed-transmitted. Some aspects of its life cycle and its biology were investigated. The colonization of pea plants obtained from naturally infected seeds was studied in natural conditions while high populations of bacteria developed on plants showing no symptoms. Two streptomycin-resistant mutants were used to study the epiphytic life of the pathogen. Populations were monitored in different host-parasite compatibilities. When race 2 or race 6 of the pathogen was surface-inoculated on susceptible cultivars, a decrease of population size was observed during the following one to three days but was followed by an increase to levels 1000 times greater than the initial number detected, without symptoms for most of the plants. When race 2 was surface-inoculated on resistant genotypes or race 6 on non-host plants, bacteria did not multiply but population levels slightly decreased.Pseudomonas syringae pv.pisi shows a resident phase and its development is race-specific. Weeds collected in naturally contaminated pea fields, diseased or not, often harboured the pathogen but with levels smaller than those observed on peas. Pea crop debris and volunteers kept high levels of bacteria for at least eight months after the harvest of a diseased crop. As long as two pea crops are not grown one after the other in the same field, it is unlikely that debris and volunteers will act as an important inoculum source. The development of this pathogen during the growing season is considered as an important parameter to take into account for controlling the disease through seed health testing.  相似文献

Inoculation of leaves of resistantPlatanus occidentalis and susceptiblePlatanus acerifolia leaves withCeratocystis fimbriata f. sp.platani, the canker stain disease agent, induced foliar necrosis and biosynthesis of two phytoalexins, scopoletin and umbelliferone. Foliar symptoms keep localized and accumulation of coumarin phytoalexins was rapid for incompatible interactions. Necrosis spread widely and accumulation of these phenolic compounds was much later and lower for compatible interactions. The differential response could be used in a genetic improvement program for resistance against canker stain.  相似文献

Potato seed tubers are imported annually to Israel from northern Europe. Although the seed is registered as certified, a survey carried out over a 9-year period indicated that most lots were affected by latent or active bacterial and fungal infections. Latent infection byErwinia carotovora subsp.atroseptica, the causal agent of blackleg, at a level of 103 cells/g peel, was present in 30% of the lots in most years. Black scurf caused byRhizoctonia solani was present in 20–70% of the imported lots, with a moderate to high level of infection in all years except 1985. In contrast, although many lots were affected by powdery scab, common scab, and Fusarium dry rot in most years, disease incidence within lots was generally low. The gangrene pathogen (Phoma exigua) was rarely detected. The survey findings are of marked importance, due to the extensive use of soil fumigation in Israeli agriculture.  相似文献

During a five-year survey including studies on the parasite fauna of bream (Abramis brama), four gill-parasitic Myxobolus species (M. bramae, M. hungaricus, M. impressus and M. macrocapsularis) were recorded in a total of 313 breams from Lake Balaton. The commonest species, M. bramae showed a prevalence of 33%, while the other species occurred sporadically. Myxobolus bramae and M. macrocapsularis infected the tips of the gill filaments and caused both intralamellar and interlamellar infection. Intralamellar plasmodia of small size developed in the capillary network of the gill lamellae whereas the much larger interlamellar plasmodia were formed in the arteria afferens. The intralamellar plasmodia of M. hungaricus always infected the basal or central part of the gill filaments. In contrast to the above species developing in the blood vessels, M. impressus proved to be an epithelial parasite, as its plasmodia always developed in the adjacent gill filaments of two opposite haemibranchia, in the stratified epithelium between the respiratory plates, causing changes of the haemibranchium which were well visible even by the naked eye.  相似文献

During a recent investigation of parasites infecting fishes from the Okavango River and Delta, Botswana (southern Africa) fourteen sharptooth catfish, Clarias gariepinus (Burchell, 1822) (Siluriformes: Clariidae) were examined for the presence of myxozoan infections. Results revealed the presence of two species of the genus Henneguya Thélohan, 1895 and one species of the genus Myxobolus Bütschli, 1882 infecting this fish host. Two of the sampled fish exhibited large plasmodia of Henneguya suprabranchiae Landsberg, 1987 in the cartilage of the accessory breathing organ, another two individuals were infected with H. samochimensis sp. n. plasmodia in the gills and another three individuals revealed an infection with Myxobolus gariepinus sp. n. plasmodia in the ovaries.  相似文献

ABSTRACT Pepper huasteco virus (PHV) and Pepper golden mosaic virus (PepGMV) are found in mixtures in many horticultural crops in Mexico. This combination constitutes an interesting, naturally occurring model system to study several aspects of virus-virus interactions. Possible interactions between PHV and PepGMV were studied at four levels: symptom expression, gene expression, replication, and movement. In terms of symptom expression, the interaction was shown to be host-dependent because antagonism was observed in pepper, whereas synergism was detected in tobacco and Nicotiana benthamiana. PHV and PepGMV did not generate viable pseudorecombinant viruses; however, their replication is increased during mixed infections. An asymmetric complementation in movement was observed because PHV was able to support the systemic movement of PepGMV A whereas PepGMV did not support the systemic distribution of PHV A. Heterologous transactivation of both coat protein promoters also was detected. Several conclusions can be drawn from these experiments. First, viruses coinfecting the same plant can interact at several levels (replication, movement) and in different manners (synergism, antagonism); some interactions might be host dependent; and natural mixed infections could be a potential source of geminivirus variability by generating viable tripartite combinations that could facilitate recombination events.  相似文献
