Evidence for disease-related amphibian decline in Colorado   总被引:8,自引:0,他引:8  
The recent discovery of a pathogenic fungus (Batrachochytrium dendrobatidis) associated with declines of frogs in the American and Australian tropics, suggests that at least the proximate cause, may be known for many previously unexplained amphibian declines. We have monitored boreal toads in Colorado since 1991 at four sites using capture–recapture of adults and counts of egg masses to examine the dynamics of this metapopulation. Numbers of male toads declined in 1996 and 1999 with annual survival rate averaging 78% from 1991 to 1994, 45% in 1995 and 3% between 1998 and 1999. Numbers of egg masses also declined. An etiological diagnosis of chytridiomycosis consistent with infections by the genus Batrachochytrium was made in six wild adult toads. Characteristic histomorphological features (i.e. intracellular location, shape of thalli, presence of discharge tubes and rhizoids) of chytrid organisms, and host tissue response (acanthosis and hyperkeratosis) were observed in individual toads. These characteristics were indistinguishable from previously reported mortality events associated with chytrid fungus. We also observed epizootiological features consistent with mortality events associated with chytrid fungus: an increase in the ratio of female:male toads captured, an apparent spread of mortalities within the metapopulation and mortalities restricted to post metamorphic animals. Eleven years of population data suggest that this metapopulation of toads is in danger of extinction, pathological and epizootiological evidence indicates that B. dendrobatidis has played a proximate role in this process  相似文献

The storage, soluble, and structural carbohydrates of two onion cultivars, the hard, pungent Pukekohe Longkeeper (PLK) and the softer, milder Houston Grano, were analyzed to determine differences that might be related to their response to sulfur nutrition received during growth as well as their postharvest attributes and end-use suitability. PLK tissue contained 1.37 times more dry matter than Grano and was composed of more fructan and sucrose and less glucose and fructose than Grano [corrected] There were also differences in neutral sugar content, especially galactose, and the amount, size, and content of pectin fractions soluble in chelator and weak alkali. These two onion cultivars differed in their capacity to take up sulfur, but there was no statistical association between sulfur supply and any measured dry matter component.  相似文献

Market and policy incentives that encourage agricultural intensification, such as incentives for bioenergy, may contribute to biodiversity decline when they encourage a large-scale conversion of native and semi-natural ecosystems to production fields. In order to appreciate the impact of these incentives on biodiversity, it is imperative to better understand how native and semi-natural ecosystems contribute to plant diversity and composition. We studied the five most common types of managed grasslands in Northeastern Kansas, a region undergoing agricultural intensification. We analyzed plant community data recorded at three spatial scales in 98 managed grassland sites, and examined patterns of diversity and composition of plant species and functional groups; and spatial turnover of species and functional groups measured at different spatial scales. We found reduced soil quality and plant diversity as well as a lack of scale-dependent community patterns on sites that were historically cultivated. Forage management practices (haying or grazing) altered plant diversity and composition in native grassland remnants but not in non-native grasslands. We identified several opportunities where changes to existing management practices could benefit both conservation and bioenergy production objectives. Recommendations to conserve biodiversity include increasing the use of hay management or other biomass collection on native grassland remnants and improving the Conservation Reserve Program by increasing enrollment, adding more native species to seed mixes, and incorporating a periodic biomass collection. We also found that using measures of spatial turnover in community composition added important insights in understanding the effects of management decisions on biodiversity.  相似文献

Root growth is important to the competitive ability of plants, and understanding how herbage defoliation affects root growth has implications for development of management strategies. Objectives were to determine the effects of defoliation intensity and frequency on root characteristics and herbage production of slender wheatgrass (Elymus trachycaulus [Link.] Shinners), Nebraska sedge (Carex nebrascensis C. Dewey), and “Steadfast” birdsfoot trefoil (Lotus corniculatus L.). Plants of each species were transplanted into containers that had been placed in the ground at wet meadow field sites the prior year. There were eight replications of a control and five defoliation treatments, which were combinations of different frequencies (two or five times) and intensities (light or heavy) and haying. Treatments were applied for a single growing season, and aboveground biomass was collected. Containers were extracted in October, and plant crowns, rhizomes, and roots were separated from the soil. Defoliation treatment did not affect total root weight, length, and surface area of Nebraska sedge or birdsfoot trefoil (P>0.10). Slender wheatgrass total root weight was less when defoliated five times (4.46 g·container?1) than when defoliated twice (6.62 g·container?1) during the growing season. More frequent defoliation of slender wheatgrass also reduced length (20%) and surface area (21%) compared to less frequent defoliation. However, defoliation frequency did not affect aboveground biomass. Defoliation intensity did not affect aboveground production or root characteristics of the three species. Abundant soil moisture in meadows likely buffers negative effects of defoliation. For all species, two defoliation events (e.g., haying followed by grazing) does not appear to negatively affect root growth and herbage production.  相似文献

Inhibitor of apoptosis (IAP) proteins are negative regulators of cell death. IAP family members contain RING domains that impart E3 ubiquitin ligase activity. Binding of endogenous or small-molecule antagonists to select baculovirus IAP repeat (BIR) domains within cellular IAP (cIAP) proteins promotes autoubiquitination and proteasomal degradation and so releases inhibition of apoptosis mediated by cIAP. Although the molecular details of antagonist-BIR domain interactions are well understood, it is not clear how this binding event influences the activity of the RING domain. Here biochemical and structural studies reveal that the unliganded, multidomain cIAP1 sequesters the RING domain within a compact, monomeric structure that prevents RING dimerization. Antagonist binding induces conformational rearrangements that enable RING dimerization and formation of the active E3 ligase.  相似文献

CASE DESCRIPTION-13 equids (10 horses, 2 donkeys, and 1 pony) were examined for signs of colic (n = 7), weight loss (6), anorexia (3), and diarrhea (2). Ten equids were evaluated in the fall (September to November). Seven equids had a history of persimmon ingestion. CLINICAL FINDINGS-A diagnosis of phytobezoar caused by persimmon ingestion was made for all equids. Eight equids had gastric persimmon phytobezoars; 5 had enteric persimmon phytobezoars. Gastroscopy or gastroduodenoscopy revealed evidence of persimmon ingestion in 8 of 10 equids in which these procedures were performed. TREATMENT AND OUTCOME-2 of 13 equids were euthanatized prior to treatment. Supportive care was instituted in 11 of 13 equids, including IV administration of fluids (n = 8) and treatment with antimicrobials (5), NSAIDs (5), and gastric acid suppressants (4). Persimmon phytobezoar-specific treatments included dietary modification to a pelleted feed (n = 8); oral or nasogastric administration of cola or diet cola (4), cellulase (2), or mineral oil (2); surgery (4); and intrapersimmon phytobezoar injections with acetylcysteine (1). Medical treatment in 5 of 7 equids resulted in resolution of gastric persimmon phytobezoars. Seven of 8 equids with gastric persimmon phytobezoars and 1 of 5 equids with enteric persimmon phytobezoars survived > 1 year after hospital discharge. CLINICAL RELEVANCE-Historical knowledge of persimmon ingestion in equids with gastrointestinal disease warrants gastroduodenoscopy for evaluation of the presence of persimmon phytobezoars. In equids with gastric persimmon phytobezoars, medical management (including administration of cola or diet cola and dietary modification to a pelleted feed) may allow for persimmon phytobezoar dissolution.  相似文献

Objective: To evaluate stifle joints of dogs for synovitis, before development of joint instability and cranial cruciate ligament rupture (CrCLR). Study Design: Cross‐sectional study. Animals: Dogs (n=16) with CrCLR and stable contralateral stifles; 10 control dogs with intact CrCL. Methods: Arthritis and tibial translation were graded radiographically. Synovitis severity and cruciate pathology were assessed arthroscopically. Presence of inflammatory cells in synovial membrane biopsies was scored histologically. CrCLR stifle pairs and control stifles were compared. Results: Radiographic evidence of arthritis, cranial tibial translation, and arthroscopic synovitis were increased in unstable stifles, when compared with stable contralateral stifles in CrCLR dogs (P<.05). Arthroscopic synovitis in both joints of CrCLR dogs was increased compared with controls, was correlated with radiographic arthritis (SR=0.71, P<.05), and was present in all stable contralateral stifles. Arthroscopically, 75% of stable stifle joints had CrCL fiber disruption, which correlated with severity of synovitis (SR=0.56, P<.05). Histologic evidence of synovitis was identified in all CrCLR dogs, but was only significantly correlated with arthroscopic observations in stable stifles (r2=0.57, P<.005). Conclusion: Synovitis is an early feature of the CrCLR arthropathy in dogs before development of joint instability clinically. Severity of synovitis is correlated with radiographic arthritis in joints with minimal to no clinically detectable CrCL damage.  相似文献

Objective: To evaluate lymphocyte populations in stifle synovium and synovial fluid of dogs with degenerative cranial cruciate ligament rupture (CCLR). Study Design: Prospective clinical study. Animals: Dogs (n=25) with stifle arthritis and CCLR, 7 dogs with arthritis associated with cartilage degeneration (osteoarthritis [OA]), and 12 healthy Beagle dogs with intact CCL. Methods: Arthritis was graded radiographically in CCLR dogs. After collection of joint tissues, mononuclear cells were isolated and subsequently analyzed using flow cytometry for expression of CD3, CD4, CD8, and CD21. Results: The proportions of CD4+ T helper lymphocytes, CD8+ cytotoxic T lymphocytes, and CD3+CD4?CD8? T lymphocytes were increased in synovium from dogs with CCLR compared with synovium from healthy Beagle dogs (P<.05). The proportion of CD3+CD4?CD8? T lymphocytes in synovial fluid was increased in dogs with CCLR compared with dogs with OA (P<.05). In dogs with CCLR, the proportion of CD3+CD4?CD8? T lymphocytes in synovial fluid was inversely correlated with radiographic arthritis (SR=?0.68, P<.005). Conclusion: Lymphocytic inflammation of stifle synovium and synovial fluid is an important feature of the CCLR arthropathy. Lymphocyte populations include T lymphocytes expressing CD4 and CD8, and CD3+CD4?CD8? T lymphocytes. Presence of CD3+CD4?CD8? T lymphocytes was associated with development of stifle synovitis. Further work is needed to fully identify the phenotype of these cells.  相似文献
