Oxindole alkaloids in the paraherquamide/marcfortine family exhibit broad-spectrum anthelmintic activity that includes drug-resistant strains of nematodes. Paraherquamide (PHQ), 2-deoxoparaherquamide (2DPHQ), and close structural analogs of these compounds rapidly induce flaccid paralysis in parasitic nematodes in vitro, without affecting adenosine triphosphate (ATP) levels. The mechanism of action of this anthelmintic class was investigated using muscle tension and microelectrode recording techniques in isolated body wall segments of Ascaris suum. None of the compounds altered A. suum muscle tension or membrane potential. However, PHQ blocked (when applied before) or reversed (when applied after) depolarizing contractions induced by acetylcholine (ACh) and the nicotinic agonists levamisole and morantel. These effects were mimicked by the nicotinic ganglionic blocker mecamylamine, suggesting that the anthelmintic activity of PHQ and marcfortines is due to blockade of cholinergic neuromuscular transmission. The effects of these compounds were also examined on subtypes of human nicotinic ACh receptors expressed in mammalian cells with a Ca2+ flux assay. 2DPHQ blocked nicotinic stimulation of cells expressing alpha3 ganglionic (IC50 approximately 9 microm) and muscle-type (IC50 approximately 3 microm) nicotinic cholinergic receptors, but was inactive at 100 microm vs. the alpha7 CNS subtype. PHQ anthelmintics are nicotinic cholinergic antagonists in both nematodes and mammals, and this mechanism appears to underlie both their efficacy and toxicity.  相似文献

Context

Human driven land-use and land-cover change (LULC) is considered to be among the greatest ecological pressures in mountain regions. Over the past century, across the European Alps, extensive LULC changes have been observed, affecting ecosystem goods and services (ESs).

Objectives

For eight case study sites across the Alpine arc we aimed to provide a spatiotemporal explicit assessment of the impacts of LULC dynamics on ES provision and interactions, including cultivated crops, plant material, climate regulation, soil erosion control and aesthetics.

Methods

We quantified ES provision in biophysical terms at four time periods (1850, 1955, 1985, 2005) using spatially explicit LULC based assessment models. ES interactions were identified by statistically analyzing the spatiotemporal pattern among ES capacities.

Results

Over the past century forested areas have increased mainly at the cost of grasslands, while on easily accessible sites and fertile valley floors agricultural intensification occurred. ES provision shifted between 1850 and 2005, from a predominance of production ESs in 1850 to a landscape characterized by regulating ESs in 2005. Spatiotemporal analyses of ES interactions revealed trade-offs between regulating and cultural ESs and within the provisioning ES bundle and allowed to derive three different ES trajectories: regions developing from single to multifunctional sites in terms of service provision, sites reducing their service capacities and sites with rather stationary patterns over broad time periods.

Conclusions

We demonstrated that ES capacities in complex agro-ecological mountain regions are highly sensitive to long-term landscape dynamics. We conclude that assessing ES capacities and interactions in an explicitly spatiotemporal manner can help to guide evidence-based environmental measures.

There is a great need for animal models of osteoporosis and sheep are a suitable large animal that meets most requirements. Since it is known that bone mass in humans responds to seasonal changes, this study investigated natural bone metabolism in sheep in order to better define the sheep as a model for osteoporosis. Bone mineral density (BMD), trabecular structure, biochemical markers of bone formation and resorption and estrogen were analysed over a period of 18 months. The lowest BMDs, measured by peripheral quantitative computed tomography (pQCT), were observed during winter. Thereafter, a 5.1% increase in BMD was observed during spring and summer (P<0.05). Bone resorption markers showed a variable pattern, with higher values in spring compared to autumn (P<0.001). The physiological estrus phase during autumn was detected by serum estrogen levels. The findings show that it is necessary to take seasonal variations into account if sheep are used to establish an animal model for osteoporosis.  相似文献

Tulathromycin is a new injectable macrolide antibiotic used for the treatment of pulmonary diseases of swine and cattle. In this study, 37 foals with sonographic evidence of lung abscesses were treated with tulathromycin (2.5mg/kg intramuscularly [IM] once weekly, group 1) and 33 foals (group 2) with a combination of azithromycin (10mg/kg per os [PO] once daily for the first seven days of therapy, thereafter every other day) and rifampin (10mg/kg PO twice daily). The bacterial aetiological agent was not determined. The foals were only mildly sick and the median number of pulmonary abscesses was 1.4 (group 1) and 1.6 (group 2). Thirty foals in each group were treated without modifying therapy protocols until all clinical signs of disease had subsided. Tulathromycin was administered for a mean of 53 days, and azithromycin/rifampin for 42 days. The following side effects were associated with tulathromycin (279 IM injections): self-limiting diarrhoea in 11 foals; elevated temperature in six foals, and swellings at the injection site in 12 foals. This study provides some evidence that tulathromycin is well tolerated and appears promising for the treatment of pulmonary abscesses in foals.  相似文献

To study inheritance of Malus sieboldii-derived apple proliferation resistance, 14 cross combinations were performed with the tetraploid apomictic M. sieboldii and first and second generation parental lines as donor of resistance and Malus x domestica scion cultivars and apple rootstocks as donor of pomological traits. In the progeny examined mainly three classes were present consisting of mother-like plants with the allele composition of the maternal apomict (ML), hybrids based on fertilization of an unreduced egg cell (hybrid I), and fully recombinant plants (hybrid II). Two-year screening of inoculated plants in the nursery revealed that progeny classes ML and H I responded similarly to infection and that about half of the progeny showed satisfactory resistance. No appropriate resistance was identified in progeny class H II. This might be due to the fact that in fully recombinant offspring M. sieboldii haplotypes have been reduced from 4n to 1-2n or were entirely lost. Following nursery-growing, promising trees were evaluated for six more years in the orchard. Nearly all of them showed satisfactory resistance but were mostly less productive and more vigorous than trees on clonal standard rootstock M9. However, mainly among the offspring of progeny 4608 × M9, resistant genotypes were identified showing pomological properties similar to M9.  相似文献

A molecular epidemiological survey of the protozoal parasites that cause equine piroplasmosis was conducted using samples collected from horses and zebra from different geographical locations in South Africa. A total of 488 samples were tested for the presence of Theileria equi and/or Babesia caballi using the reverse line blot hybridization assay. Ten percent of the samples hybridized to the Theileria/Babesia genus-specific probe and not to the B. caballi or T. equi species-specific probes, suggesting the presence of a novel species or genotype. The small subunit of rRNA gene (18S; ∼1600 bp) was amplified and sequenced from 33 of these 488 samples. Sequences were compared with published sequences from the public sequence databases. Twelve distinct T. equi and six B. caballi 18S rRNA sequences were identified. Alignments demonstrated extensive sequence variation in the V4 hypervariable region of the 18S rRNA gene within T. equi. Sequence variation was also found in B. caballi 18S rRNA genes, although there was less variation than observed for T. equi. Phylogenetic analysis based on 18S rRNA gene sequences revealed three T. equi clades and two B. caballi clades in South Africa. The extent of sequence heterogeneity detected within T. equi and B. caballi 18S rRNA genes was unexpected since concerted evolution is thought to maintain homogeneity within repeated gene families, including rRNA genes, in eukaryotes. The findings reported here show that careful examination of variants of the 18S rRNA gene of T. equi and B. caballi is required prior to the development of molecular diagnostic tests to detect these parasites in horses. Species-specific probes must be in designed in regions of the gene that are both conserved within and unique to each species.  相似文献

Forest thinning and prescribed fires are practices used by managers to address concerns over ecosystem degradation and severe wildland fire potential in dry forests. There is some debate, however, about treatment effectiveness in meeting management objectives as well as their ecological consequences. The purpose of this study was to assess changes to forest stand structure following thinning and prescribed fire treatments, alone and combined, in the eastern Cascade Mountains of Washington State. Treatments were applied to 12 management units, with each treatment combination replicated three times (including untreated controls). Thinning modified forest structure by reducing overall tree density by >60% and canopy bulk density by 50%, and increased canopy base height by ∼4 m, thereby reducing susceptibility to crown fire. The prescribed fire treatment, conversely, did not appreciably reduce tree density or canopy fuel loading, but was effective at increasing the density of standing dead trees, particularly when combined with thinning (37 snags/ha increase). Prescribed fire effects were more pronounced when used in combination with thinning. Thinning was more reliable for altering stand structure, but spring burning was lower in intensity and coverage than desired and may have led to results that downplay the efficacy of fire to meet forest restoration goals.  相似文献

Natural occurring amorphous silica is ubiquitous on the earth in soils and sediments. Both, natural derived silica and synthetically produced amorphous silica are used against stored product pests and parasites in animal houses. New formulations, active even under higher relative humidity's can be possible used against horticultural arthropod pests. Since many horticultural pests hide on the undersides of leaves and silica is a physical contact insecticide, an appropriate application method is needed. This paper reviews electrostatic application of natural derived amorphous silica to Brassica chinensis leaves. Due to the inherent high volume resistivity of silica powder, induction charging was ineffective for electrostatic powder coating. Corona charging involves a reliable current source, but typically is inefficient in charging the powder particles and creates a high density of free ions between the gun and grounded substrate as well as within the coating layer. These factors resulted in poor coating quality and inefficient particle deposition. The best leave coating was archived using tribo charging. Here we have been able to apply coatings with different thickness. While a silica coating of 25–50?μm in petri dish experiments resulted in a reduced mortality of the granary weevil, Sitophilus granarius (L.), there was no difference between mortality with coatings of 50–100?μm and 100–125?μm. Silica applications on B. chinensis leaves resulted within two hours in a 45% reduction of the CO2-absorption. Absorption level stayed reduced over a period of seven days and was between 45 and 73% compared to the control. Even after removing silica dust using pressurized air CO2-absorption was between 19 and 28% reduced. This might be the case because silica particles where so small that they could possible clog the stomata opening resulting in a reduced transpiration and photosynthesis rate.  相似文献

Straw mulch application after high severity wildfire has gained favor in recent years due to its efficacy in reducing soil erosion hazards. However, possible collateral effects of mulching on post-fire vegetation recovery have received relatively little study. We assessed mulching effects on plant cover and species richness, tree seedling establishment, and exotic species densities in the second year following the 2006 Tripod Wildfire in north-central Washington State, USA, by observing vegetation responses to spatial variability in mulch cover and depth. Mulch cover averaged about 35%, with a median depth of 0.5 cm. Vegetation recovery was generally slow, with median plant cover of only 10%. Tree seedling densities were low and spatially variable. Vegetative cover, species richness, and seedling densities all declined with increasing elevation. Mulch cover was positively associated with plant cover, plant species richness, and conifer seedling densities when second year mulch cover did not exceed 40%. Only when mulch cover exceeded 70% did mulching begin to negatively affect vegetation recovery relative to areas with no mulch. Vegetation responses to mulch depth were minimal at depths under 3 cm, but quite strong when mulch depth exceeded 5 cm. Exotic plant frequency and density were positively associated with mulch cover, but exotic plant cover was low on average (<1%). In this study, mulch added significant cover to sites with slow natural recovery of vegetation, thereby likely reducing erosion hazard. Mulching also appears to have facilitated native plant recovery and conifer seedling establishment except at very high application levels, easing management concerns about longer-term impacts of mulching treatments on post-fire vegetation recovery.  相似文献

Maintaining understory plant species diversity is an important management goal as forest restoration and fuel reduction treatments are applied extensively to dry coniferous forests of western North America. However, understory diversity is a function of both local species richness (number of species in a sample unit) and community heterogeneity (beta diversity) at multiple spatial scales, while studies of restoration treatment effects often only examine local species richness at one or two spatial scales. We studied experimental thinning and prescribed fire treatment effects on understory plant species richness and community heterogeneity at three spatial scales using additive diversity partitioning. We also evaluated treatment effects on understory plant species colonization and extirpation at two spatial scales. There was no evidence that active restoration treatments reduced species richness or increased local extirpation of species. Restoration treatments significantly increased herbaceous species richness at the treatment-unit level primarily by increasing community heterogeneity among sampling points within the units. The combination of thinning and burning produced the greatest increase in community heterogeneity, and increased colonization by species that were not sampled prior to treatment. These results suggest that restoration treatments designed primarily to reduce fire hazard and promote sustainable conditions in these fire-adapted ecosystems can also increase community heterogeneity and facilitate colonization by new understory species without significant local extirpation of extant species.  相似文献
