In this investigation, three different methods for the harvesting of spruce under otherwise identical conditions were analysed with respect to their greenhouse gas emissions per unit of output: a partially mechanised method using motor saws and draft horses, a more highly mechanised method using motor saws and a forestry tractor and a fully mechanised method with a harvester and forwarder. All the sub-steps from the harvesting of the fallen trees to the transportation to the road were included in the investigated greenhouse gas audit, which followed the rules of a streamlined life cycle assessment. The lowest greenhouse gas emissions were produced by the partially mechanised method (305.7?kg?CO2e?ha?1), followed by the more highly mechanised method (510.5?kg?CO2e?ha?1) and by the fully mechanised method (554.3?kg?CO2e?ha?1). The greatest proportion of the greenhouse gas audit within each method was taken up by the fuel, lubricant and hydraulic oil utilisation. In the horse audit, transportation to and from the site of operation caused the greatest effect (60%). With these results, it could be confirmed that horses when used for logging could be assessed as being more positive with respect to their climate friendliness than large-scale machines despite their lower harvesting capacity per hectare of spruce. However, as this study did not take all environmental impacts into consideration, this relatively better environmental audit for draft horses could be changed when other environmental impacts (e.g. land use) are also included. Among other factors (eg, bedding material, roughage), feed has an important effect on the airborne particle concentration in the stable air, in particular directly in the breathing zone of the horse during feeding. Therefore, this study was designed to analyze and compare the generation of different airborne particle fractions of diverse feed types and feed processing techniques under standardized laboratory conditions. Oats and barley as well as two commercially formulated ready-prepared pellets and two different commercially formulated ready-prepared mixes were analyzed. Oats were subjected to four different forms of processing (whole uncleaned, whole cleaned, rolled uncleaned, rolled cleaned), barley was subjected to steam flaking in addition to the same forms of processing as oats. In total, 13 feeds were analyzed. Airborne particle concentrations were detected online with a gravimetrically measuring analyzer that was equipped successively with different inlets to measure the particle fractions PM 1, PM 2.5, PM 10, and PM 20. The measurements were carried out in a closed chamber (1 × 1 × 1.5 m) including a bowl (diameter = 0.5 m), which simulated the trough. The sample inlet was situated at a height of 1.0 m above the bowl. Rolling oats and barley led to a significantly increased generation of the PM 20 fraction and a significantly decreased generation of the PM 2.5 fraction. The mechanical cleaning of oat and barley brings a clear decrease in dust generation from straights whether they are left whole or are rolled after cleaning. Objective To determine if juvenile pearl oysters ( Pinctada maxima ) infected with Haplosporidium hinei are also infected with another haplosporidian parasite, Minchinia occulta .
Design Archived samples of pearl oysters infected with H. hinei were examined using polymerase chain reaction (PCR) assays and in situ hybridisation (ISH) to analyse and identify haplosporidians. A 144-bp and 220-bp region of Minchinia DNA were targeted by PCR and amplified DNA from formalin-fixed H. hinei -infected pearl oyster samples was sequenced. A 25-bp oligonucleotide probe targeting a variable section of the parasite's small subunit rRNA gene was used in ISH.
Results The results of DNA-based diagnostic assays supported each other. The sequences obtained by PCR were found to be almost identical to M. occulta from rock oysters and the ISH assay demonstrated infection with M. occulta in affected pearl oysters. ISH indicated a prevalence of infection of 26.7% in one of the previous outbreaks.
Conclusion Pearl oyster spat are susceptible to infection by a Minchinia parasite, most likely M. occulta , which was recently identified in rock oysters within the pearl-producing zones of Western Australia and is associated with mortalities of up to 80% in this species. The occurrence of haplosporidian co-infections in pearl oysters suggests the immunocompetence of juvenile oysters may be an important factor in preventing infection and therefore preventing mortalities such as those occurring in the recent outbreaks of pearl oyster oedema disease. 相似文献

The herd sensitivity (HSe) and herd specificity (Hsp) of clinical diagnosis of an infection with classical swine fever (CSF) virus during veterinary inspection of breeding sows in a herd was evaluated. Data gathered from visits to herds during the CSF outbreak in 1997-1998 in The Netherlands were used for the analysis. Herds were visited one or more times by the same or by different veterinarians. On the basis of the veterinarians' reports, each visit was coded as 0 (negative clinical diagnosis) or 1 (positive clinical diagnosis). The HSe for clinical diagnosis of CSF was modelled as a function of days elapsed since introduction of the virus. The moment of introduction of the CSF virus in the CSF-positive herds was unknown, so for each herd, a probability distribution for the unknown number of days since introduction was derived from serum samples collected at depopulation. The information from the reports of the veterinarians and from the test results of the serum samples at depopulation was combined in a Bayesian analysis. Data from CSF-negative herds were analysed to estimate HSp of clinical diagnosis of CSF. The HSe of clinical diagnosis was 0.5 at 37 days after virus introduction (95% CI: 31, 45) and reached 0.9 at 47 days after virus introduction (95% CI: 41, 54). The estimated herd specificity was 0.72 (95% CI: 0.64, 0.79). Dependence of HSe and HSp on characteristics of the veterinarians and the herds also was studied. P-selectin (CD62P), an adhesion molecule expressed on activated endothelial cells and platelets, mediates the initial attachment of leukocytes to the stimulated endothelium upon inflammation and the interaction between leukocytes and platelets. A soluble form of P-selectin is present in the serum of healthy individuals as a circulating protein and high levels have been described in various pathological situations. The aim of this study was to characterize P-selectin on porcine platelets and investigate the soluble form of this protein, which are uncharacterized in several animal species including pigs. A new monoclonal antibody (mAb) (SwPsel.1.9) against porcine P-selectin was produced using a mouse cell line transfected with pig P-selectin cDNA. This mAb together with a previously described mAb (P-sel.KO.2.5), produced in our laboratory, was used to develop an ELISA to quantify porcine P-selectin. No significant levels of soluble-porcine P-selectin were observed in healthy animals. However, the total amount of P-selectin measured in porcine platelets was similar to that found in humans. Increased levels of this circulating protein were detected in the plasma from pigs after allograft implantation. In vitro, P-selectin expression on platelet membrane was rapidly induced by PMA and thrombin, as assessed by flow cytometry. However, these activators did not stimulate the release of soluble P-selectin. Analysis of the proteolytic cleavage of this protein from COS-transfected cells revealed that PMA treatment failed to cause the shedding of membrane-bound P-selectin. These data suggest that porcine P-selectin is a suitable marker for inflammation and that the mechanism involved in the generation of circulating P-selectin is not proteolytic release. 相似文献

The objective of this study was to validate a new blocking enzyme-linked immunosorbent assay (ELISA) (designated M108 for milk and S108 for serum samples) for detecting bovine leukemia virus (BLV) infection in dairy cattle. Milk, serum, and ethylenediaminetetraacetic acid-blood samples were collected from 524 adult Holstein cows originating from 6 dairy herds in Central Argentina. The M108 and S108 were compared with agar gel immunodiffusion (AGID), polymerase chain reaction and a commercial ELISA. Because there is currently no reference test capable of serving as a gold standard, the test sensitivity (SE) and specificity (SP) were evaluated by the use of a latent class model. Statistical inference was performed by classical maximum likelihood and by Bayesian techniques. The maximum-likelihood analysis was performed assuming conditional independence of tests, whereas the Bayesian approach allowed for conditional dependence. No clear conclusion could be drawn about conditional dependence of tests. Results with maximum likelihood (under conditional independence) and posterior Bayes (under conditional dependence) were practically the same. Conservative estimates of SE and SP (with 95% confidence intervals) for M108 were 98.6 (96.7; 99.6) and 96.7 (92.9; 98.8) and for S108 99.5 (98.2; 99.9) and 95.4 (90.9; 98.1), respectively. The ELISA 108 using either milk or serum to detect BLV-infected animals had comparable SE and SP with the official AGID and a commercial ELISA test, which are currently the most widely accepted tests for the serological diagnosis of BLV infection. Ph3SnCl, Ph3SnAc (Brestan), Cy3SnOH (Plictran), and (Bu3Sn)2O (TBTO) markedly reduce the weight of the spleen and the gain in body weight of young mice feeding for 7 days on diets containing 260 μequiv organotin/kg diet. With mature mice feeding for 4 days on diets containing 780 μequiv organotin/kg, these compounds result in a loss in spleen weight and body weight and an alteration in the blood composition (reduction in lymphocytes and total leucocytes and an increase in erythrocytes, hemoglobin level, and hematocrit value). Four other organotins, [(PhCMe2CH2)3Sn]2O (Vendex), PhSnCl2, Ph2SnCl2, and Ph4Sn, are much less toxic based on these parameters. Poisoning by these dietary triorganotins was not related to cerebral edema or alterations in adrenal epinephrine level, liver nonprotein -SH groups, or the activity of digestive enzymes (intestine and stomach proteases and intestine amylase and invertase). Our aim was to provide additional estimates of main parameters for the transmission of foot-and-mouth disease virus (FMDV) strain O Taiwan (3/97). We used the data of previous experiments in non-vaccinated and vaccinated pigs and combined the data of experiments with the same treatment(s). First, we quantified the reproduction ratio R for the various groups using a final-size method. Our final-size results predicted that vaccination with a four-fold vaccine dose (but not with a single dose) at 1 week before inoculation (-7 dpi) would reduce R compared to the non-vaccinated group. Secondly, we used the daily results of virus excretion to quantify the transmission rate beta (by using generalized linear modelling), and the infectious period T (by using survival analysis). We used the estimates of beta and T to estimate R more precisely as compared to the final-size method and also for the groups for which a finite estimate could not be obtained using a final-size method. Our modelling results predicted that beta for non-vaccinated, for single-dose and four-fold-dose groups would be 6.1 (3.7, 10)day(-1), 2.0 (1.0, 4.0)day(-1) and 0.4 (0.1, 1.4)day(-1), T at 6.5 (5.7, 7.3), 5.3 (4.7, 6.0) and 2.3 (0.9, 5.7) days and R at 40 (21, 74), 11 (4.9, 24) and 1.0 (0.1, 7.8), respectively. These results predicted that both vaccination with a four-fold vaccine dose and with a single dose at -7 dpi would reduce beta, T and R significantly as compared to the non-vaccinated pigs, thereby showing that vaccination will reduce transmission of FMDV significantly already 1 week post vaccination. 相似文献

Knowledge of the sensitivity of diagnostic tests for infectious diseases under field conditions can be used to design a surveillance program that increases the effectiveness of the control policy. In this study, the sensitivity of tests for the detection of classical swine fever (CSF) virus (CSFV) under field conditions was estimated without knowledge of the true disease status of the animals tested. During the CSF epidemic of 1997-1998 in The Netherlands, tonsil samples from pigs of CSF suspect farms were collected for laboratory diagnosis of CSE These specimens were tested in a fluorescence antibody test (FAT1) for the presence of CSFV antigen. When at least 1 specimen in a particular sample series from a farm was positive, this farm was declared CSFV infected. Specimens of that series, either FAT1 negative (98) or FAT1 positive (127), were subsequently tested again (FAT2). After that, a suspension was made of the remaining tissue, and this suspension was evaluated with a virus isolation test. In total, 225 tonsil specimens were examined. A statistical model was formulated, and the sensitivity of the 3 tests and the prevalence of positive specimens in the sample were estimated by the method of maximum likelihood. The sensitivity of the FAT1, the test that was used for confirmation of CSFV infection in a pig herd, was approximately 78% (95% confidence interval [CI] = 62-92%). The effectiveness of the selection process of animals on the farm by the veterinarian was estimated to be 77% (64-87%). The sensitivity of the combination of FAT1 and FAT2 (60%) indicates that at least 5 animals should be selected on a CSF-suspect farm to gain a detection probability for CSFV of 99%. 相似文献
