Ceratocystis platani causes canker stain of plane trees, and it represents a serious disease of Platanus spp. both in the United States and Europe. Current chemical or biological controls do not effectively manage C. platani, so new preventive methods need to be developed in order to limit this pathogen spreading. In this work, we have characterized the main volatile organic compounds (VOC) emitted in vitro from pure cultures of C. platani and other common pathogenic fungal species of hosts plants growing in the same ecosystems as plane trees. We found that C. platani emitted a similar blend of VOC compared with phylogenetically similar species C. populicola. In particular, C. platani was characterized by emission of isoamyl acetate and isobutyl acetate while C. populicola by ethyl acetate and isobutyl acetate, which were not released by any of the other out‐group fungal species grown on the same medium. Moreover, following a targeted approach based on the main VOC found in vitro, we have successfully validated in vivo that VOC uniquely emitted by C. platani (i.e. isobutyl acetate along with isoamyl alcohol) were released from the bark of plane trees following C. platani inoculation. Our results highlight the possibility to exploit VOC emitted specifically by C. platani as biomarkers to recognize Platanus x acerifolia plants infected by this pathogen.  相似文献

In this study, the impact of sourdough fermentation on the biochemical, rheological and bread-making performances of buckwheat flour was investigated. In order to assess the effects of the solely acidification, a chemically acidified batter with the same pH of the sourdough was prepared. Extensive hydrolysis of the globulin fraction and release of small polypeptides occurred upon fermentation. A major reduction in the extent of interactions was observed in the sourdough, mainly due to the modification of the major structural components of buckwheat during the fermentation. The hydrolysis of proteins and, possibly, starch was in turn responsible for the major decrease in elasticity observed in the sourdough. In the batters destined for bread-making, the presence of acids was the major cause for reduced elasticity and increased strength, which could be related to the enhanced water-holding capacity of the proteins and/or protein/starch complexes. The addition of sourdough induced dramatic inhibition of the CO2 production by the baker’s yeasts during proofing, resulting in lower volume and harder crumb of the sourdough bread. On the other hand, the solely acidification induced hardening of the starch gel upon cooking, which was responsible for lower volume and irregular crumb grain in buckwheat bread.  相似文献

The nutritional quality of wild and cultivated species ofVigna has been evaluated. Wild species showed significantly higher protein content (P<0.01), antitryptic activity (P<0.005) and tannin content (P<0.025) and significantly lower protein digestibility (P<0.005). No significant differences were found with regard to protein quality, measured as chemical score corrected by the digestibility; sulphur amino acids turned out to be limiting in both groups. In addition no correlations were found between sulphur amino acids and protein levels, or between sulphur amino acid and trypsin inhibitor levels.Selected lines showed an improvement from the nutritional point of view linked to lower antitryptic factors and tannin content, which may give rise to a decreased resistance to pests. For all the properties considered, whether physical or biochemical, wild samples presented wider variations and the types considered most primitive were those that differed most from the cultivated ones, whose properties were altogether more consistent. Little information is available on the prevalence of swine enteropathogens in Cuba where diarrheic diseases are responsible for 31% and 37% of the total mortality during the neonatal and postweaning periods. F4+ and F18+ enterotoxigenic Escherichia coli and F18+ verotoxigenic E. coli induce diarrhea and edematous disease in pigs, but their distribution has never been thoroughly studied in the Cuban swine population. Therefore, the present study estimated the prevalence of F4- and F18-specific antibodies in sera of 1,044 6-month-old gilts distributed in 34 piggeries spread over the Cuban territory. For the data analysis, which included the optical density of individual samples tested by ELISA, random-effects models and a mixture model in R (package “mixAK”; Komárek, Computational Statistics and Data Analysis 53:3932–3947, 2009) were fitted. Low, moderate, and high levels of F4-specific antibodies were found in 67.6%, 26.8%, and 5.6% of the gilts, while 66.4% and 33.6% of gilts showed low and high levels of F18-specific antibodies. Hereby, we show that F4+ and F18+ E. coli are highly prevalent as potential enteropathogens in Cuban piggeries.  相似文献

This study describes the clinical, diagnostic, and pathological characteristics of canine nasal polyps and how they responded to medical, endoscopic, and surgical treatments. The database of a multi-center veterinary endoscopy group was searched from 2010 to 2018. All dogs with a histological diagnosis of nasal polyposis that were undergoing endoscopic investigation (N = 23), were included. Clinical signs at presentation were sneezing (91%), nasal discharge (83%), stertor (74%), and frontonasal deformation (17%). Skull radiography on 13 dogs had alterations in 77% of cases, including turbinate lysis (6/13), increased radiopacity of one (4/13) or both (6/13) nasal cavities, and lysis of the nasal vomer bone (3/13). Nasal polyposis had a characteristic endoscopic appearance. There were clinical and diagnostic similarities between this cohort of dogs and dogs with nasal neoplasia, although dogs with nasal polyps were often younger and polypoid tissue was external to the nose. Steroid therapy alone was not effective in treating polyposis in dogs; however, endoscopic debulking with a laser and forceps was more effective.  相似文献

The adsorption-desorption performances of commercial resins (two polystyrene-divinylbenzene copolymers and one methacrylic) were tested in column studies for the selective recovery of anthocyanins and hydroxycinnamates from pigmented pulp wash (PW), a byproduct of blood orange juice processing. Methanol, ethanol, and their mixtures with different percentages of water as eluents were tested in order to investigate the selective desorption of these natural antioxidants with the goal of minimizing the presence of other compounds, mostly flavanones and limonoids, in the concentrated eluates. The results indicated that polystyrene-divinylbenzene resins were able to adsorb a major amount of anthocyanins and hydroxycinnamates and to desorb them in more-concentrated fractions. No selectivity was observed using pure methanol and ethanol which resulted in concentrates with no further separation of components from the starting raw PW. The ethanol/water concentrates showed higher yields than the fractions attained using methanol/water; however, a lower selectivity toward anthocyanin pigments was observed. The best performing resin, EXA-118, together with the mixture methanol/water 50:50 (v/v) as best eluent, appears to be the most suitable system to obtain highly concentrated extracts. Thus, it was chosen to perform a larger experiment, to analyze the selectivity of the removal upon an increase in elution volume.  相似文献

Fatty acid steryl esters (FASE) in whole meal of 14 genotypes of tetraploid wheats (Triticum dicocconand T. durum) and 17 genotypes of hexaploid wheats (T. spelta and T. aestivum) were analyzed using different chromatographic strategies. By both GC-FID and HPLC-ELSD, tetraploid wheats are lacking two major peaks. The amounts of FASE, calculated on the basis of the GC-FID analysis, were double in hexaploid species as compared to tetraploids (40 and 20 mg/100 g db, respectively). HPLC with ESI-MS detection enabled the identification of FASE by the characteristic fragmentations and ion-adducts of each molecule. The distribution of steryl residues was not different between the wheat species: the main class of steryl derivatives found was the beta-sitosteryl derivatives, followed by campesteryl derivatives with small amounts of stigmasteryl esters. The esterified fatty acids explain the difference between the hexaploid and tetraploid wheats. In particular, small amounts of campesteryl and beta-sitosteryl, while no trace of stigmasteryl palmitates, were found in T. durum or its hulled ancestor T. dicoccon. Steryl oleates were not detectable in T. aestivum or its hulled ancestor T. spelta, which is consistent with the filogenesis of tetraploid and hexaploid species. Both chromatographic techniques (GC and HPLC) showed that FASE are useful to discriminate between hexaploid and tetraploid wheats from both qualitative and quantitative points of view.  相似文献
