Lady beetles are among the most successful predators of aphids in different environments. The functional responses of different life stages of Hippodamia variegata (Goeze) towards cotton aphids were examined in two different set-ups, a two-dimensional Petri dish set-up with detached leaves and a three-dimensional set-up with whole plants. In addition, the functional responses in two-dimensional set-ups towards cotton aphids and the pea aphids were compared. H. variegata exhibited a functional type II response to both cotton aphids and pea aphids irrespective of life stage and spatial scale of the set-up. Females and fourth instars generally had higher search rates than third instars whereas handling times were consistently lower for the fourth instar stage compared with the preceding juvenile stage and with females. The spatial dimensions did not have any significant influence on the functional responses towards cotton aphids, except for third instars which in the three-dimensional set-up reduced their search rate and increased their handling time. Fourth instars reacted in the same way to both Aphis gossypii and Acyrthosiphon pisum whereas the functional response parameters for third instars and females were significantly different on the two prey species. Our study, a part of this first-step evaluation of H. variegata as a biocontrol agent against A. gossypii and A. pisum under field condition, suggest that the voracity of all tested stages of H. variegata towards both aphid species hold good promises for a use of especially fourth instars and females in inundative biocontrol.  相似文献

An in vitro study was conducted to quantitatively investigate the metabolism of pipecolic acid (Pip), a neuromodulator, by mixed rumen bacteria (B), mixed rumen protozoa (P), a combination of B and P (BP), species‐enriched rumen protozoal suspension (Polyplastron sp., Diploplastron sp., entodinia and Entodinium caudatum) and pure cultures of several isolates of rumen bacteria (Prevetolla bryantii, Prevetolla albensis, Streptococcus bovis, Veillonella parvula, Megasphaera elsdenii and Ruminococcus albus). Only P produced Pip from L‐lysine (1.0 mmol/L L‐Lys) at a rate of 83.5 ± 1.6 µmol/L/h and even in BP, Pip was produced from L‐Lys by P and increased at a rate of 31.2 ± 3.8 µmol/L/h. Pip production by P was highest when the substrate (L‐Lys) concentration was 6 mmol/L and then the rate was 580 ± 36 µmol/L/h. Pipecolic acid production by P suspension enriched with different species of protozoa showed that Polyplastron sp. had the highest Pip production rate of 0.907 ± 0.092 µmol/L/mg protozoal protein per h, and Diploplastron sp. had the lowest rate of 0.55 ± 0.13 µmol/L/mg protozoal protein per h. The addition of D‐Lys (1.0 mmol/L) as a substrate to the P suspension revealed that P were also able to produce Pip from D‐Lys, though at a lower rate (1/3) compared with L‐Lys (1.0 mmol/L), suggesting the presence of epimerases in P. It was confirmed that B were unable to produce Pip from L‐ or D‐Lys. Only B degraded Pip (1.0 mmol/L) after a lag phase at a rate of 56.0 ± 1.5 µmol/L/h. The B suspension was able to degrade D‐Lys, though the products were not identified. Pip degradation by pure culture of some species of rumen bacteria showed that P. bryantii and R. albus had the highest rate followed by P. albensis, S. bovis and M. elsdenii with a low rate of Pip degradation. Veillonella parvula showed no ability to degrade Pip. The results suggest that a fairly large proportion of rumen‐produced Pip is likely to be absorbed by the host animal before degradation by rumen bacteria.  相似文献

Depleted soil fertility and high-yielding cultivars have been associated with low nutrient contents in vegetables. This study explored if elemental nutrient concentrations in tomato (Lycopersicon esculentum Mill.) can be increased through selection of cultivars and nutritional regimes. Cultivars with different phenotypes of heirloom and modern origins were studied in field experiments in two years. Chemical fertilizer (10-10-10), compost, and an organic fertility regime of soybean meal, bone meal, and potassium sulfate were assessed for their effects on growth and composition of fruits of the cultivars. Differences in nutrient concentrations between modern or heirloom cultivars or among fertility treatments were small or non-significant. Differences among individual cultivars for each element were large with some cultivars having nearly twice the concentrations of nutrients of others and with considerable uniformity in cultivar rankings among the elements. This work suggests that cultivars can be selected for production of nutrient-dense tomatoes.  相似文献

Using Bacillus stearothermophilus AB-1 isolated from air, the production of lipase was attempted along with its purification and characterization studies. When different carbon and nitrogen sources were supplemented in the culture medium, xylose, tryptophan, alanine, phenylalanine and potassium nitrate were found to be the best. During cultivation, the strain secreted most of its lipase content after 48 h. In particular, the lipase produced in the culture broth showed 300 U mL(-1) when cultivated at optimal temperature and pH of 35 degrees C and 7.5, respectively. The enzyme was purified using 60% ammonium sulfate precipitation and sephadex G200 column chromatography. The enzyme was stable up to 40 degrees C and in the range of pH 7-8. This research reports for the first time the characterization of mesophilic lipase from Bacillus stearothermophilus AB-1 isolated from air.  相似文献

Aquaporins are transmembrane channels found in cell membranes of all life forms. We examine their apparently paradoxical property, facilitation of efficient permeation of water while excluding protons, which is of critical importance to preserving the electrochemical potential across the cell membrane. We have determined the structure of the Escherichia coli aquaglyceroporin GlpF with bound water, in native (2.7 angstroms) and in W48F/F200T mutant (2.1 angstroms) forms, and carried out 12-nanosecond molecular dynamics simulations that define the spatial and temporal probability distribution and orientation of a single file of seven to nine water molecules inside the channel. Two conserved asparagines force a central water molecule to serve strictly as a hydrogen bond donor to its neighboring water molecules. Assisted by the electrostatic potential generated by two half-membrane spanning loops, this dictates opposite orientations of water molecules in the two halves of the channel, and thus prevents the formation of a "proton wire," while permitting rapid water diffusion. Both simulations and observations revealed a more regular distribution of channel water and an increased water permeability for the W48F/F200T mutant.  相似文献

Biodegradable polymers, such as poly(lactic acid) (PLA) have attracted a lot of attention in the scientific community recently due to a rapid growth of intensive interest in the global environment for alternatives to petroleum-based polymeric materials. Fatty nitrogen compounds (FNCs), fatty amides (FA), fatty hydroxamic acids (FHA), and carbonyl difatty amides (CDFA), which were synthesized from vegetable oils, were used as one of organic compounds to modify natural clay (sodium montmorillonite). The clay modification was carried out by stirring the clay particles in an aqueous solution of FA, FHA, and CDFA, by which the clay layer thickness increased from 1.23 to 2.61, 2.84 and 3.19 nm, respectively. The modified clay was then used in the preparation of the PLA/epoxidized soybean oil (ESO) blend nanocomposites. They were prepared by incorporating 2% of CDFA-MMT and 3% of both FA-MMT and FHA-MMT. The interaction of the modifier in the clay layer was characterized by X-ray diffraction (XRD), and Fourier transform infrared (FTIR). Elemental analysis was used to estimate the presence of FNCs in the clay. The nanocomposites were synthesized by solution casting of the modified clay and a PLA/ESO blend at the weight ratio of 80/20, which has the highest elongation at break. The XRD and transmission electron microscopy (TEM) results confirmed the production of nanocomposites. PLA/ESO modified clay nanocomposites show higher thermal stability and significant improvement of mechanical properties in comparison with those of the PLA/ESO blend. The novelty of this study is use of FNCs which reduces the dependence on petroleum-based surfactants.  相似文献

To improve nutrition intake of humans, it is recommended that fruits and vegetables constitute a major portion of diets. An improvement in the mineral nutrient contents of fruits and vegetables is needed to offset apparent declines in the elements in these foods. This research addressed the enrichment of cabbage (Brassica oleracea var. capitata L.) through the selection of nutrient-rich cultivars and soil fertility practices. Conventional fertilizers, organic fertilizers, and compost were evaluated with modern F1 hybrids in field experiments. Crop production was greater with the chemical or organic fertilizers than with the compost. Mineral nutrient composition varied among cultivars but not with fertility practices, suggesting that cultivar selection could lead to production of nutrient-rich cabbage. Neither the mass of heads or days to maturation of crops affected nutrient composition.  相似文献

Surface irrigation is the oldest and the most widely used method of irrigation. One disadvantage of surface irrigation is soil erosion. New technology employing water-soluble polymers may provide a technique that is effective and affordable to control soil erosion. Water-soluble anionic organic compound known as polyacrylamide polymer (PAM) is the most successful polymer in controlling soil erosion. This study investigated the effect of spraying PAM on the soil surface to control soil erosion and to increase soil infiltration on a Jordanian clay loam soil. Different PAM concentrations, namely 5, 10, and 20 mg/l in addition to the control (0 mg/l) were used in this study. The highest effect of PAM on the measured properties was attained at 20 mg/l. We noticed that PAM's efficiency was decreased with subsequent irrigations. The reduction in soil erosion was 72 and 47.6%, the reduction in runoff water turbidity was 83 and 35%, the increase in water advance time was 6 and 0.9% and the increase in soil infiltration was 36 and 20.8% for the first and fourth irrigation, respectively. PAM's efficiency in flocculating soil particles was studied in the lab where we noticed that its efficiency in sedimentation was increased as its concentration increased.  相似文献
