In November 1997, Cryptosporidium andersoni, for the first time, was isolated from a Danish heifer. The isolate was characterised morphologically, molecularly, and furthermore inoculated into mice and one calf. Data on the distribution of cryptosporidia in the herd of origin were obtained at two separate visits in December 1997 and April 1998. C. andersoni was detected in 27 (19.0%) of 142 cattle examined at the first visit, whereas C. parvum was found in six (4.2%). At the following visit 42 (28.0%) of 150 cattle excreted C. andersoni, while 25 (16.7%) were positive for C. parvum. Oocysts of the Danish C. andersoni isolate were ovoid, 7.3(6.5-8.0) x 5.7(5.0-7.0) microm(2) (n=25), with smooth, colourless, single layer oocyst wall and distinct oocyst residuum. The length to width ratio was 1.27 (1.14-1.40, n=25). The identification was verified by sequencing of a 246bp fragment of the rDNA, which was identical to Cryptosporidium muris, the calf genotype (AF093496). The Danish C. andersoni isolate was not transmissible to mice, whereas oocysts were detected in the faeces of one experimentally infected calf from 25 days post-infection (DPI) and shed intermittently at low numbers until 165 DPI, the day of euthanasia. No macroscopic or microscopic changes that could be attributed to infection with C. andersoni were seen in the gastro-intestinal tract of the experimentally infected calf following necropsy and histological examination. This is to our knowledge the first report of C. andersoni in Scandinavia.  相似文献

Mycoplasma hyospnoviae strains from Denmark, Germany, Japan, Sweden, the Netherlands and the UK were examined for variations in the genomic DNA and within the 16S ribosomal RNA (rRNA) gene. Variations in the chromosomal DNA among 57 isolates recovered from the respiratory tract and joints of pigs, were investigated by analysis of amplified fragment length polymorphisms of the Bg/II and MfeI restriction sites and by pulsed-field gel electrophoresis of a BssHII digest of chromosomal DNA. Both methods allowed unambiguous differentiation of the analysed strains and showed similar discriminatory potential for the differentiation of M. hyosynoviae isolates. Concordant results obtained with the two whole-genome fingerprinting techniques evidence the considerable intraspecies genetic heterogeneity of M. hyosynoviae. Sixteen field strains of M. hyosynoviae and the type strain S16(T) were further examined for variation within the 16S rRNA gene. Ten field strains possessed the 16S rDNA sequences identical to the type strain, while the remaining six strains had sequences that differed by one to two nucleotides from that obtained from the type strain.  相似文献

We address the problem of optimal size‐selective exploitation in an age‐structured fish population model by systematically examining how density and size dependency in growth, mortality and fecundity affect optimal harvesting patterns when judged against a set of fisheries objectives. The study offers five key insights. First, while minimum‐length limits often maximize the biomass yield, exploitation using harvest slots (i.e. regulations that protect both immature and very large individuals) can generate within 95% of maximum yield; harvest slots also generally maximize the number of fish that are harvested. Second, density dependence in growth and size‐dependent mortality predict more liberal optimal size limits than those derived under assumptions of no density and size dependence. Third, strong density dependence in growth maximizes the catch of trophy fish only when modest harvest is introduced; the same holds for numbers harvested, when the stock–recruitment function follows the Ricker type. Fourth, the inclusion of size‐dependent maternal effects on fecundity or egg viability has only limited effects on optimal size limits, unless the increase in fecundity with mass (“hyperallometry”) is very large. However, large hyperallometry in fecundity shifts the optimal size limit for biomass yield from the traditional minimum‐length limit to a harvest slot. Fifth, harvest slots generally provide the best compromises among multiple objectives. We conclude that harvest slots, or more generally dome‐shaped selectivity to harvest, can outperform the standard minimum‐length selectivity. The exact configuration of optimal size limits crucially depends on objectives, local fishing pressure, the stock–recruitment function, and the density and size dependency of growth, mortality and fecundity.  相似文献

This study was conducted to investigate the effects of soil structure on bromide (Br) transport through three soils with granular, prismatic, and single-grain structures. The breakthrough curve (BTC) of the single-grain structure was sigmoidal, symmetrical and similar to a piston flow, showing the dominance of mass flow. In contrast, the BTCs of the granular and prismatic structures were initially steep, becoming more gradual at high pore volumes (PVs). The stable structure and preferential pathways caused the early breakthrough of Br in the leachate of these columns. The convection–dispersion equation (CDE), mobile–immobile water (MIM), and dual-permeability (DP) models were fitted to observed data using the program HYDRUS-1D. The equilibrium transport model (CDE) was not as successful as non-equilibrium (MIM and DP) models in describing the Br transport in prismatic and granular soil columns, although it was able to describe the Br transport in single-grain column well. Overall, the results demonstrated the importance of soil structure in pollutant transport through soils.  相似文献

Soil Taxonomy was developed primarily for the practical purpose of supporting the National Cooperative Soil Survey Program in the United States. The previous system of classification was limited in its usefulness and could not be applied consistently by a large cadre of soil scientists with diverse levels of skill and experience. The first edition of Soil Taxonomy, published in 1975, was developed under the leadership of Dr. Guy Smith over a period of about 25 years. The magnitude and complexity of the information favored the use of a hierarchical system presented as a key for consistent application. Operational definitions were used to identify taxonomic limits and diagnostic horizons and features, so that each taxon conveyed the same meaning to every user. The application of Soil Taxonomy had several important impacts on the soil survey program in the United States. The emphasis on objective, measurable diagnostic horizons and features to define classes made all competent soil scientists, regardless of experience and rank, equally capable of accurately and consistently classifying soils. The use of quantitative class differentia improved the quantity and quality of the field data collection. The property ranges of soil series and their geographic distribution have generally been narrowed over time, thus, allowing more precise interpretations to be made. Soil Taxonomy has benefited the soil correlation process by grouping the nearly 22 000 series currently established in the United States in ways that allow us to efficiently compare and differentiate competing soil series and coordinates their use among survey areas. After publication in 1975, Soil Taxonomy was further refined through the work of nine international committees and many individuals. This work culminated in the publication of the second edition in 1999 and, most recently, the 9th edition of the Keys to Soil Taxonomy in 2003. Today, two international committees are considering revisions to the moisture and temperature regimes and improvements to the classification of anthropogenic soils. Four trials, three in the United States and one in South Africa, were conducted to evaluate the potential value of a novel self-medicating applicator in the passive control of gastrointestinal nematodes in cattle and deer, and of files and ticks on cattle using oil-based treatments. The results of the trials demonstrated that this applicator is an effective and practical device for the passive treatment of both deer and cattle for trichostrongyle infections using the endectocide, moxidectin (Cydectin. Fort Dodge Animal Health, USA), of cattle for hom fly (Haemotobia irritans) infestations using the insecticide, cyfluthrin (CyLence, Bayer AG, Germany) and of cattle for tick infestations (in particular Amblyomma hebraeum and Rhipicephalus appendiculatus) using the acaricides deltamethrin and amitraz (Delete All, Intervet, South Africa).  相似文献

The efficacy of the classical swine fever (CSF) subunit marker vaccine Porcilis Pesti based on baculovirus expressed envelope glycoprotein E2 of CSF virus (CSFV) was evaluated in pregnant sows. Ten gilts were vaccinated with one dose of marker vaccine, followed by a second dose 4 weeks later. Four gilts remained unvaccinated and received a placebo at the same times. Thirty-three days after the second vaccination all animals were artificially inseminated. Neither local or systemic reactions nor an increase of body temperature were observed after vaccinations. All gilts showed a normal course of pregnancy. Thirty-five days after first vaccination all animals developed E2 specific neutralising antibodies with titres in the range of 5.0 and 7.5 log(2). No antibodies to CSFV-E(rns) were found in ELISA.On day 65 of gestation (126 days after the first immunisation) all sows were infected intranasally using 2ml (10(6.6) TCID(50)/ml) of the low virulent CSFV strain "Glentorf". After challenge in two of the unvaccinated control sows a slight transient increase of body temperature was observed, whereas leukopenia was demonstrated in all control animals. In addition all controls became viraemic. Vaccinations with the CSFV subunit vaccine protected the animals from clinical symptoms of CSF. In two sows a moderate decrease of leukocyte counts was detected on day 5 post infection. In contrast to the unvaccinated control sows in none of the vaccinated animals virus was isolated from the nasal swabs or the blood.Approximately 40 days after challenge all sows were killed and necropsy was done. The sows and their offspring were examined for the presence of CSFV in blood, bone marrow and different organs. No virus was found in any of the sows. In contrast, in all litters of the control sows CSFV was found in the blood as well as in the organ samples. Nine out of 10 litters of the vaccinated sows were protected from CSFV infection. Blood samples, lymphatic organs and bone marrow of these animals were all virologically negative. When sera were tested for CSFV-antibodies all sows had developed E(rns)-specific antibodies but no CSFV-specific antibodies were found in any of the progeny.It was concluded that vaccination with CSF subunit marker vaccine Porcilis((R)) Pesti protected 90% of the litters from viral infection when sows were challenged mid-gestation using the CSFV-strain "Glentorf".  相似文献
